G1 Checkpoint Regulation

G1 checkpoint regulation is the control system that determines whether a cell proceeds from the G1 growth phase into DNA synthesis, helping coordinate proliferation with cell size, nutrients, and genome integrity. Mitogenic signals activate cyclin D–CDK4/6 and cyclin E–CDK2, which phosphorylate the retinoblastoma protein and release E2F transcription factors; DNA damage instead activates p53, which induces p21 to inhibit cyclin-dependent kinases and delay progression. This checkpoint supports accurate cell-cycle control and prevents damaged DNA from being replicated. Its disruption contributes to uncontrolled proliferation and cancer, making checkpoint proteins important targets for biological research and therapeutic development.

The Spindle Assembly Checkpoint

2023

The spindle assembly checkpoint is a molecular surveillance mechanism ensuring the fidelity of chromosome segregation during anaphase. The checkpoint monitors the completion of all the prerequisite steps before chromosome segregation to determine whether the segregation process should proceed or be delayed. Many proteins function together to control the spindle assembly checkpoint. Mutations affecting these proteins may allow cells to proceed into anaphase prematurely, resulting in the...

The kinetochore is where the SAC initiates its signal monitoring the mitotic segregation of the sister chromatids. A method is described to visualize the recruitment and turnover of one of the kinetochore proteins and its coordination with the chromosome motion in Drosophila embryos using a Leica laser scanning confocal system.

Evaluation of the Spindle Assembly Checkpoint Integrity in Mouse Oocytes

Error in chromosome segregation is a common feature in oocytes. Therefore, studying the spindle assembly checkpoint gives important clues about the mechanisms needed to produce healthy eggs. The present protocol describes three complementary assays to evaluate spindle assembly checkpoint integrity in mouse oocytes.
