Muscle Tissue Isolation

Muscle tissue isolation is the process of separating skeletal, cardiac, or smooth muscle from surrounding tissues to obtain samples for laboratory analysis. It typically combines careful dissection with mechanical or enzymatic dissociation, which removes connective tissue and breaks tissue into intact fragments or individual cells while preserving key cellular features. In developmental biology, isolated muscle tissue enables researchers to examine myogenesis, cell differentiation, fiber formation, and tissue organization across developmental stages. These preparations support microscopy, gene and protein analysis, cell culture, and studies of congenital muscle disorders, providing controlled systems for investigating how muscle develops, maintains structure, and responds to experimental conditions.

This video demonstrates the isolation of satellite cells from rat head branchiomeric muscle tissues through tissue dissection, enzymatic digestion, and subsequent mechanical and centrifugal purification. The cells are then seeded on extracellular matrix-coated wells for differentiation into myoblasts, and eventually into myotubes and mature muscle fibers.

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.

Mitochondrial Isolation from Skeletal Muscle

This protocol describes a procedure to study the respiration of mitochondria isolated from skeletal muscles. This method was adapted from Scorrano et al. (2007). The mitochondrial isolation procedure requires about 2 hours. The mitochondrial respiration can be completed in about 1 hour.
