Mog 35-55

MOG 35-55 is a synthetic peptide corresponding to amino acids 35–55 of myelin oligodendrocyte glycoprotein, a central nervous system myelin protein, and serves as a defined antigen in autoimmune neuroimmunology. When administered with an appropriate adjuvant, the peptide is processed and presented by major histocompatibility complex class II molecules, activating myelin-reactive CD4+ T cells that can promote inflammatory damage in the central nervous system. MOG 35-55 is widely used to induce experimental autoimmune encephalomyelitis in susceptible mice, providing a controlled model for studying neuroinflammation, immune-cell trafficking, disease mechanisms, and potential therapies relevant to demyelinating disorders.

Experimental autoimmune encephalomyelitis (EAE) is an established animal model of multiple sclerosis. C57BL/6 mice are immunized with myelin oligodendrocyte glycoprotein (MOG) peptide 35-55 (MOG35-55), resulting in an ascending flaccid paralysis caused by autoreactive immune cells in the central nervous system. Protocols for disease induction and monitoring will be discussed.

Experimental autoimmune encephalomyelitis is one of the most widely used murine models of multiple sclerosis. In the current protocol, C57BL/6J mice of both sexes are immunized with myelin oligodendrocyte glycoprotein peptide, resulting mainly in ascending paresis of the tail and limbs. Here we discuss the protocol of EAE induction and evaluation.

Kinematic gait analysis in the sagittal plane yields highly precise information about how movement is executed. We describe the application of these techniques to identify gait deficits for mice subjected to autoimmune-mediated demyelination. These methods may also be used to characterize gait deficits for other mouse models featuring impaired locomotion.

Simple and Efficient Production and Purification of Mouse Myelin Oligodendrocyte Glycoprotein for Experimental Autoimmune Encephalomyelitis Studies

We describe a simple protocol using only basic lab equipment to generate and purify large quantities of a fusion protein that contains mouse Myelin Oligodendrocyte Glycoprotein. This protein can be used to induce experimental autoimmune encephalomyelitis driven by both T and B cells.
