Lung Biopsy

Lung biopsy is a medical procedure that removes a small sample of lung tissue for microscopic and molecular examination, helping clinicians identify the cause of abnormal imaging findings or respiratory symptoms. Depending on lesion location and patient factors, tissue may be obtained through bronchoscopy with biopsy instruments, a needle guided through the chest wall, or minimally invasive or open surgery; the specimen is then evaluated by pathology. In medicine, lung biopsy can distinguish cancer from infection, inflammation, and fibrotic disease, while also enabling tumor classification and treatment planning. Careful method selection balances diagnostic yield against risks such as bleeding or pneumothorax.

This protocol describes the feasibility to perform miRNA profiling in exosomes, released in plasma of NSCLC patients, through a commercial exosome isolation kit with Proteinase K and RNAse treatments, in order to avoid circulating miRNAs contamination and evaluate their biomarker features in NSCLC.

Lung-resident immune cells, including dendritic cells (DCs) in humans, are critical for defense against inhaled pathogens and allergens. However, due to the scarcity of human lung tissue, studies are limited. This work presents protocols to process human mucosal endobronchial biopsies for studying lung DCs using immunohistochemistry and flow cytometry.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.
