Viable Myeloid Cell Purification

Viable myeloid cell purification is a cell-separation method that isolates living myeloid cells, including monocytes, macrophages, dendritic cells, and related populations, from blood or tissue samples for downstream analysis. The process uses selective surface markers and methods such as magnetic-activated cell sorting or fluorescence-activated cell sorting to enrich target cells while maintaining membrane integrity, viability, and functional properties. In cancer research, purified myeloid cells help investigators characterize the tumor immune microenvironment, assess immunosuppressive or inflammatory phenotypes, and study interactions with cancer cells. High-quality viable isolates support flow cytometry, molecular profiling, functional assays, and evaluation of therapeutic responses.

Given the limited xenograft models available to study interactions between human CART and myeloid cells, we established in vitro and in vivo models to understand the impacts of human macrophages on CART cells. Findings can potentially be generalized to evaluate macrophage roles in the tumor microenvironment and test macrophage-targeted immunotherapies.

A protocol to utilize a poly(N-iso-propylacrylamide) (PIPAAm) coated microfilter for effective capture and thermoresponsive release of viable circulating tumor cells (CTC) is presented. This method allows capture of CTC from patients' blood and subsequent release of viable CTC for downstream off-chip culture, analyses and characterization.
