De gedwongen zwemmen test is gevalideerd als een experimentele benadering van potentiële antidepressieve werking te evalueren bij knaagdieren. Proefdieren worden geplaatst in een tank met water en ontsnappen met betrekking tot mobiliteit gedrag wordt gekwantificeerd. De gemeenschappelijke procedures voor de muis versie van deze test zijn beschreven.
De gedwongen zwemmen test is een knaagdier gedragstest worden gebruikt voor de evaluatie van antidepressiva, antidepressieve werkzaamheid van nieuwe verbindingen, en experimentele manipulaties die gericht zijn op rendering of het voorkomen van depressieve-achtige toestanden. Muizen worden geplaatst in een onontkoombare transparante tank die gevuld is met water en hun ontsnapping aanverwante mobiliteitsgedrag wordt gemeten. De gedwongen zwemmen test is eenvoudig om betrouwbaar te voeren en het vereist een minimale gespecialiseerde apparatuur. Succesvolle implementatie van de gedwongen zwemmen test vereist de naleving van bepaalde procedurele details en minimalisering van de ongerechtvaardigde stress bij de muizen. In het protocol beschrijving en de bijbehorende video, leggen we uit hoe u de muis versie van deze test uit te voeren met de nadruk op mogelijke valkuilen die nadelig kan zijn voor de interpretatie van de resultaten en hoe ze te vermijden. Daarnaast leggen we uit hoe het gedrag tot uiting in de test worden beoordeeld.
De FST (ook wel Porsolt zwemmen test) werd voor het eerst ontwikkeld voor ratten en vervolgens aangepast voor muizen door Porsolt en collega's 12,13. In aanvulling op de hierboven beschreven protocol succesvol in ons laboratorium, hebben een aantal grotendeels subtiele wijzigingen te testen gepubliceerd (zie Hascoet en Bourin voor een volledige herziening 1).
