Nuværende ex vivo modeller af glioblastom (GBM) er ikke optimeret til fysiologisk relevant undersøgelse af humant tumorinvasion. Her præsenterer vi en protokol til generering og vedligeholdelse af organotypiske skivekulturer fra frisk human GBM væv. En beskrivelse af time-lapse mikroskopi og kvantitative celle migrationsanalyse teknikker er tilvejebragt.
Glioblastom (GBM) har fortsat en ekstremt dårlig klinisk prognose på trods af kirurgisk, kemoterapeutisk og strålebehandling. Progressiv tumorinvasion i omgivende hjerneparenchyma repræsenterer en varig terapeutisk udfordring. For at udvikle anti-migrationsterapier til GBM er det nødvendigt med modelsystemer, der giver en fysiologisk relevant baggrund for kontrolleret forsøg. Her præsenterer vi en protokol til generering af skivekulturer fra humant GBM væv opnået under kirurgisk resektion. Disse kulturer tillader ex vivo- eksperimentering uden passage gennem dyre-xenografter eller enkeltcellekulturer. Desuden beskriver vi brugen af time-lapse laser scanning konfokal mikroskopi i forbindelse med cellesporing for kvantitativt at studere tumorcellernes tilknyttede adfærd og tilhørende respons på terapeutik. Skiverne produceres reproducerbart inden for 90 minutter af kirurgisk vævsopkøb. Retroviralt medieret fluorescerende celle laBeling, konfokal billeddannelse og tumorcelle migrationsanalyser udføres efterfølgende inden for to ugers kultur. Vi har med succes brugt disse skivekulturer til at afdække genetiske faktorer i forbindelse med øget migrerende adfærd i human GBM. Desuden har vi valideret modelens evne til at detektere patientspecifik variation som svar på anti-migrationsbehandling. Flytning fremad er humane GBM skivekulturer en attraktiv platform til hurtig ex vivo vurdering af tumorfølsomhed over for terapeutiske midler for at fremme personlig neuro-onkologisk terapi.
Laboratorieundersøgelsen af glioblastom (GBM) hindres af mangel på modeller, som trofast rekapitulerer de krævede patologiske karakteristika ved den menneskelige sygdom, nemlig tumorcellemigration og invasion. Sammenligningsundersøgelser af 2D- og 3D in vitro- invasionstest samt 3D-gnavere af slagtekulturer har afdækket mekanisk forskelligartede cellulære migrationsprogrammer i disse to sammenhænge, der potentielt begrænser translatabiliteten af fund fra 2D-systemer til den humane sygdom 1 , 2 , 3 . Det organotype tumorskivekultur- og billeddannelsesparadigm, der beskrives her, tillader undersøgelse af tumorcellemigration inden for skiver af humant tumorvæv ex vivo opnået fra kirurgisk resektion. Således tilvejebringer skivekulturer af kirurgisk resekteret tumorvæv i forbindelse med time-lapse-konfokalmikroskopi en platform til at studere tumorcelle-migration i den nativeMikro-miljø uden vævsopløsning eller kulturpassage.
Der er omfattende litteratur, der anvender gnavere hjerneskivekulturmodeller af GBM genereret fra humane tumor-xenotransplantater, retrovirale inducerede tumorer og cellulære overlejringer til undersøgelse af tumorinvasion 1 , 2 , 3 , 4 , 5 . For nylig har flere grupper beskrevet genereringen af organotypiske skivekulturer direkte fra humant GBM væv 6 , 7 , 8 , 9 , 10 . Der er imidlertid markant variation blandt offentliggjorte protokoller med hensyn til udskæringsteknik og kulturmedier. Endvidere har anvendelsen af organotypiske skivekulturer fokuseret på statiske eksperimentelle endepunkter, der har medtaget ændringer i celle signalerNg, proliferation og død. Protokollen beskrevet heri udvider på tidligere skivekulturparadigmer ved at inkorporere tidsopløst observation af dynamisk tumorcelleadfærd ved hjælp af time-lapse laser scanning-konfokal mikroskopi. Nylig opdagelse af inter 11 og intratumoral 12 , 13 genetisk variation i humant GBM understreger vigtigheden af at forbinde denne heterogenitet med tumorcelleadfærd og dens implikationer på tumorrespons til terapi. Her rapporterer vi en strømlinet og reproducerbar protokol til brug af direkte skivekulturer fra et humant cancervæv til at visualisere tumorcelleindvandring i nær real-time.
Organotypiske skivekulturer fra humant kræftvæv giver en attraktiv og underudnyttet platform til præklinisk translationel eksperimentering. Forståelse af befolkningsniveauadfærd hos tumorceller med hensyn til migration, proliferation og celledød i det naturlige tumormikromiljø mangler. Kritisk kan undersøgelse af tumorrespons på terapi på en dynamisk, tidsopløst måde på celleadfærdens niveau afdække nye mekanismer for behandlingsresistens.
