We describe key steps for biosensing by using polysilicon nanowire field-effect transistors, including the preparation of the device and the immobilization and confirmation of a DNA molecular probe on the nanowire surface, as well as conditions for DNA sensing.
Surveillance using biomarkers is critical for the early detection, rapid intervention, and reduction in the incidence of diseases. In this study, we describe the preparation of polycrystalline silicon nanowire field-effect transistors (pSNWFETs) that serve as biosensing devices for biomarker detection. A protocol for chemical and biomolecular sensing by using pSNWFETs is presented. The pSNWFET device was demonstrated to be a promising transducer for real-time, label-free, and ultra-high-sensitivity biosensing applications. The source/drain channel conductivity of a pSNWFET is sensitive to changes in the environment around its silicon nanowire (SNW) surface. Thus, by immobilizing probes on the SNW surface, the pSNWFET can be used to detect various biotargets ranging from small molecules (dopamine) to macromolecules (DNA and proteins). Immobilizing a bioprobe on the SNW surface, which is a multistep procedure, is vital for determining the specificity of the biosensor. It is essential that every step of the immobilization procedure is correctly performed. We verified surface modifications by directly observing the shift in the electric properties of the pSNWFET following each modification step. Additionally, X-ray photoelectron spectroscopy was used to examine the surface composition following each modification. Finally, we demonstrated DNA sensing on the pSNWFET. This protocol provides step-by-step procedures for verifying bioprobe immobilization and subsequent DNA biosensing application.
Silicium nanodraad veldeffecttransistoren (SNWFETs) hebben de voordelen van ultra-hoge gevoeligheid en directe elektrische responsen milieu ladingsverandering. In n-type SNWFETs bijvoorbeeld wanneer een negatief (of positief) geladen molecuul zal het silicium nanodraad (SNW), de dragers in de SNW uitgeput (of stapel). Bijgevolg is de geleidbaarheid van de SNWFET afneemt (of verhogingen) 1. Daarom kan een geladen molecuul SNW nabij het oppervlak van het apparaat SNWFET gedetecteerd. Essentiële biomoleculen zoals enzymen, eiwitten, nucleotiden en vele moleculen op het celoppervlak zijn ladingsdragers en kan worden gecontroleerd met behulp SNWFETs. Met de juiste aanpassingen, in het bijzonder het immobiliseren van een biomoleculaire sonde op het SNW, kan een SNWFET worden ontwikkeld tot een label-free biosensor.
Surveillance gebruik biomarkers is cruciaal voor de diagnose van ziekten. Zoals getoond in Tabel 1, hebben verscheidene studies gebruikt NWFET-gebaseerde biosensoren voor label-free, ultra-hoge gevoeligheid en real-time detectie van verschillende biologische doelwitten, waaronder een virus 2, adenosine trifosfaat en kinasebinding 3, neuronale signalen 4, metaalionen 5,6, bacteriële toxinen 7, dopamine 8, 9-11 DNA, RNA 12,13, enzym en kanker biomarkers 14-19, 20 hormonen en cytokinen 21,22. Deze studies hebben aangetoond dat NWFET gebaseerde biosensoren vormen een krachtig detectieplatform voor een breed scala aan biologische en chemische species in een oplossing.
In SNWFET gebaseerde biosensoren, de probe geïmmobiliseerd op het oppervlak van SNW herkent het apparaat een specifieke biotarget. Immobiliseren van een bioprobe gewoonlijk een aantal stappen, en is het essentieel dat elke stap goed wordt uitgevoerd om de goede werking van de biosensor te waarborgen. Verscheidene technieken zijn ontwikkeld voor het analyseren van de surface samenstelling, waaronder X-ray foto-elektron spectroscopie (XPS), ellipsometrie, Contacthoekmeting, atomic force microscopie (AFM), en scanning elektronenmicroscopie (SEM). Methoden zoals AFM en SEM directe bewijs van bioprobe immobilisatie van de nanodraad apparaat, terwijl methoden zoals XPS, ellipsometrie en contact hoekmeting afhankelijk zijn van parallelle experimenten uitgevoerd op andere soortgelijke materialen. In dit rapport beschrijven we de bevestiging van een modificatiestap door twee onafhankelijke methoden. XPS wordt toegepast om de concentraties van specifieke atomen aan polysilicium wafers onderzoeken, en variaties in de elektrische eigenschappen van de inrichting worden gemeten direct naar ladingsverandering op SNW oppervlak te bevestigen. We maken gebruik van DNA biosensoren met behulp van polykristallijne SNWFETs (pSNWFETs) als een voorbeeld om dit protocol te illustreren. Immobiliseren van een DNA-probe op het SNW oppervlak bestaat uit drie stappen: aminegroep modificatie op de inheemse hydroxyl oppervlak van de SNW, aldehyde groep modificatie, en 5'-aminomodified DNA-probe immobilisatie. Bij elke wijziging stap, kan het apparaat rechtstreeks te detecteren de variatie in de lading van de functionele groep geïmmobiliseerd op het SNW oppervlak, omdat het oppervlak lasten veroorzaken lokale grensvlak potentiële veranderingen over het hek diëlektricum dat het kanaal huidige en geleiding 1 te veranderen. Kosten rond de SNW oppervlak kan elektrisch moduleren van de elektrische eigenschappen van de pSNWFET apparaat; dus de oppervlakte-eigenschappen van de SNW spelen een cruciale rol bij de bepaling van de elektrische eigenschappen van pSNWFET apparaten. In de gerapporteerde procedures, kan de immobilisatie van een bioprobe op SNW oppervlak direct worden bepaald en bevestigd door middel van elektrische metingen, en de inrichting wordt voorbereid voor biosensoren toepassingen.
Commercialiseren van de top-down en bottom-up fabricage benaderingen voor sSNWFETs wordt moeilijk beschouwd vanwege de kosten 32,33, SNW positie controle 34,35, en de geringe omvang van de productie 36. Daarentegen fabriceren pSNWFETs is eenvoudig en lage kosten 37.
