Ion Mobility-massespektrometri og molekylære modelleringsteknikker kan karakterisere den selektive metal chelaterende ydeevne af designede metal bindende peptider og kobber bindende peptid methanobactin. Udvikling af nye klasser af metal Chelat peptider vil hjælpe med at føre til terapeutisk for sygdomme forbundet med metal ionfejl balance.
Elektro spray ionisering (ESI) kan overføre en vandig fase peptid eller peptid kompleks til gas-fase samtidig bevare sin masse, samlede ladning, metal bindende interaktioner, og konformationel form. Kobling ESI med ion Mobility-massespektrometri (IM-MS) giver en instrumental teknik, der giver mulighed for samtidig måling af et peptid’s Mass-to-Charge (m/z) og kollision tværsnit (CCS), der relaterer til sin støkiometri, protonation tilstand, og konformationel form. Den samlede ladning af et peptidkompleks styres af protonation af 1) peptid’s sure og grundlæggende sites og 2) oxidations tilstanden af metal ion (s). Derfor er den samlede opladningstilstand for et kompleks en funktion af pH-værdien af opløsningen, som påvirker peptider metal ionbindende affinitet. For ESI-IM-MS-analyser fremstilles peptid-og metaliumiopløsninger ud fra vandige opløsninger med pH justeret med fortyndet vandig eddikesyre eller ammoniumhydroxid. Dette gør det muligt at fastgøre pH-afhængighed og metal ionselektiviteten for et specifikt peptid. Desuden kan m/z og CCS af et peptidkompleks anvendes med B3LYP/LanL2DZ Molekylær modellering til at skelne mellem bindingssteder af metal ionkoordineringen og den tertiære struktur i komplekset. Resultaterne viser, hvordan ESI-IM-MS kan karakterisere den selektive chelerende præstation af et sæt af alternative methanobactin peptider og sammenligne dem med kobber bindende peptid methanobactin.
Kobber og zink ioner er afgørende for levende organismer og afgørende for processer, herunder oxidativ beskyttelse, vævsvækst, respiration, kolesterol, glukose metabolisme og genom læsning1. For at muliggøre disse funktioner inkorporerer grupper som cys, imidazol i2,3, (sjældnere) thioether af methionin og carboxylat af Glu og ASP selektivt metaller som cofaktorer i de aktive steder i metalloenzymer. Ligheden mellem disse koordinationsgrupper rejser et spændende spørgsmål om, hvordan hans og cys ligander selektivt indarbejde enten cu (I/II) eller Zn (II) for at sikre korrekt funktion.
Selektiv binding opnås ofte ved opkøb og handel med peptider, som kontrollerer Zn (II) eller cu (I/II) ionkoncentrationerne4. Cu (I/II) er meget reaktiv og forårsager oxidativ skader eller utilsigtet binding til enzymer, så dens frie koncentration er stramt reguleret af kobber chaperoner og kobber-regulerende proteiner, der transporterer det sikkert til forskellige steder i cellen og stramt styre sin homøostase5,6. Afbrydelse af kobber metabolisme eller homøostase er direkte impliceret i Menkes og Wilsons sygdom7 samt kræft7 og neurale lidelser, såsom prionsygdomme8 og Alzheimers sygdom9.
Wilsons sygdom er forbundet med øget kobber niveauer i øjnene, leveren og dele af hjernen, hvor redox reaktioner af cu (I/II) producerer reaktive ilt arter, forårsager hepatolloulære og neurologiske degeneration. Eksisterende chelationterapier er den lille thiol aminosyre penicillamin og triethylenetetramin. Alternativt udviser methanotrophic kobber-overtagelses peptider methanobactin (MB)10,11 terapeutisk potentiale på grund af deres høje bindingsaffinitet for cu (I)12. Når methanobactin (MB-OB3b) fra Methylosinus trichosporium OB3b blev undersøgt i en dyremodel af Wilsons sygdom, blev kobber effektivt fjernet fra leveren og udskilles gennem galde13. In vitro forsøg bekræftede, at MB-OB3b kunne Chelat kobber fra kobber metallothionein indeholdt i leveren cytosol13. Laser ablation induktions koblede plasma massespektrometri Imaging teknikker har undersøgt den rumlige fordeling af kobber i wilson’s sygdom leverprøver14,15,16 og vist, at MB-OB3b fjerner kobber med korte behandlingsperioder på kun 8 dage17.
MB-OB3b vil også binde med andre metalioner, herunder AG (I), AU (III), PB (II), MN (II), Co (II), fe (II), ni (II), og Zn (II)18,19. Konkurrence om den fysiologiske cu (I) binding site er udstillet af AG (I) fordi det kan fortrænge cu (I) fra MB-OB3b kompleks, med både AG (I) og ni (II) også viser irreversibel binding til MB, som ikke kan fordrevet af cu (I)19. For nylig er en række alternative methanobactin (Amb) oligopeptider med 2his-2cys bindende motiv blevet undersøgt20,21, og deres Zn (II) og cu (I/II) bindingsegenskaber karakteriseret. Deres primære aminosyresekvenser er ens, og de indeholder alle 2His-2Cys Motif, Pro og en acetyleret N-endestation. De adskiller sig hovedsageligt fra MB-OB3b, fordi 2His-2Cys Motif erstatter de to enethiol oxazolone bindingssteder af MB-OB3b.

Den samlede opladningstilstand af komplekset er også en funktion af pH og påvirker peptid’s metal ion binding affinitet, fordi deprotonated grundlæggende eller sure steder som carboxyl, hans, cys og tyr er også de potentielle bindende steder for metal ion. Til analyserne tilberedes peptid og metal ion i vandige opløsninger med pH justeret ved fortyndet vandig eddikesyre eller ammoniumhydroxid. Dette gør det muligt at fastgøre pH-afhængigheden og metal ionselektiviteten for peptid. Desuden kan m/z og CCS bestemt af ESI-im-MS bruges sammen med B3LYP/LanL2DZ molecular modeling til at opdage typen af metal ion koordination og tertiær struktur af komplekset. Resultaterne vist i denne artikel afslører, hvordan ESI-IM-MS kan karakterisere den selektive chelerende præstation af et sæt Amb peptider og sammenligne dem med kobber bindende peptid MB-OB3b.
