Licv Dimerization

LicV dimerization is the association of two LicV protein molecules into a defined complex, a structural event that can influence protein stability, molecular recognition, and biological regulation. The dimer forms when complementary surfaces on LicV interact through hydrophobic contacts, hydrogen bonds, and electrostatic forces, with changes in concentration, solution conditions, or specific amino acid substitutions potentially affecting the equilibrium between monomeric and dimeric states. Characterizing this process through biochemical assays, mutagenesis, and structural analysis helps identify the dimer interface and its functional consequences. Such information supports models of LicV activity and can guide studies of protein regulation, interaction networks, and structure-based biological research.

Isolating Free Carbenes, their Mixed Dimers and Organic Radicals

We present protocols for the isolation of stable heterocyclic carbenes. The synthesis of a cyclic (alkyl)(amino) carbene (CAAC) and an N-heterocyclic carbene (NHC) is demonstrated using filter cannulas and Schlenk technique. We furthermore present the synthesis of the related oxygen-sensitive, electron-rich mixed “Wanzlick dimer” and the reduced stable organic radical.

Chemical Dimerization-Induced Protein Condensates on Telomeres

This protocol illustrates a chemically induced protein dimerization system to create condensates on chromatin. The formation of promyelocytic leukemia (PML) nuclear body on telomeres with chemical dimerizers is demonstrated. Droplet growth, dissolution, localization and composition are monitored with live cell imaging, immunofluorescence (IF) and fluorescence in situ hybridization (FISH).
