Organotypic Retinal Wholemount

An organotypic retinal wholemount is an ex vivo preparation that preserves the retina’s layered architecture and cellular organization while displaying the tissue as a flattened sheet for analysis. Researchers carefully isolate and flatten the retina, maintain its orientation and, when required, culture it under controlled conditions so that neural circuits, cell morphology, and spatial relationships remain accessible; labeling or immunostaining can then reveal specific cell types and structures. This preparation supports mapping retinal neurons and projections, studying development, degeneration, and regeneration, and assessing cellular responses to disease or experimental treatments. It provides a practical bridge between intact-eye studies and cellular or molecular analysis.

Retinal explants obtained from wild-type macaques were cultured in vitro. Retinal degeneration and the cGMP-PKG signaling pathway was induced using the PDE6 inhibitor zaprinast. cGMP accumulation in the explants at different zaprinast concentrations was verified using immunofluorescence.
