Expression Platform

An expression platform is a biological system designed to produce a selected gene product, such as a protein or RNA, in a suitable host cell. It works by placing the target sequence under compatible regulatory elements, allowing the host’s transcription and translation machinery to generate the product under controlled conditions; vectors, promoters, host strains, and culture parameters can be optimized to improve yield and quality. Expression platforms support recombinant protein production, functional studies, assay development, vaccine research, and biopharmaceutical manufacturing. Comparing platforms helps researchers match product requirements with production speed, scalability, cellular processing, and post-translational modification capabilities.

High-throughput Protein Expression Generator Using a Microfluidic Platform

We present a microfluidic approach for the expression of protein arrays. The device consists of thousands of reaction chambers controlled by micro-mechanical valves. The microfluidic device is mated to a microarray-printed gene library. These genes are then transcribed and translated on-chip, resulting in a protein array ready for experimental use.

In the post-human genomics era, the availability of recombinant proteins in native conformations is crucial to structural, functional and therapeutic research and development. Here, we describe a test- and large-scale protein expression system in human embryonic kidney 293T cells that can be used to produce a variety of recombinant proteins.

We describe the use of a multiplexed high-throughput gene expression platform that quantitates gene expression by barcoding and counting molecules in biological substrates without the need for amplification. We used the platform to quantitate microRNA (miRNA) expression in whole blood in subjects with and without irritable bowel syndrome.
