Type 1 Collagen

Type 1 collagen is the most abundant structural protein in the human body, providing tensile strength and organization to connective tissues such as skin, bone, tendons, ligaments, and the cornea. Cells synthesize it as procollagen, which undergoes enzymatic modification and secretion before its terminal regions are removed and the molecules assemble into cross-linked fibrils within the extracellular matrix. Studying type 1 collagen helps explain tissue development, wound healing, fibrosis, and disorders that weaken connective tissue. Its stability, biocompatibility, and ability to support cell attachment also make it valuable in biomaterials, tissue engineering, drug delivery, and regenerative medicine.

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.

Here, we present a protocol for the preparation of two different forms of culture substrates utilizing type I collagen. Depending on how collagen is handled, collagen molecules either maintain two-dimensional, non-fibrous form or reassemble into three-dimensional, fibril form. Cell proliferation on type I collagen is drastically affected by fibril formation.
