Cancer Research Functional Assessment of BRCA1 variants using CRISPR-Mediated Base Editors Ji-Eun See1,2, Ha Rim Shin1,2, Gayoung Jang1,2, Jiyeon Kweon1,2, Yongsub Kim1,2 1Department of Biomedical Sciences, Asan Medical Institute of Convergence Science and Technology, Asan Medical Center, University of Ulsan College of Medicine, 2Stem Cell Immunomodulation Research Center, University of Ulsan College of Medicine People with BRCA1 mutations have a higher risk of developing cancer, which warrants accurate evaluation of the function of BRCA1 variants. Herein, we described a protocol for functional assessment of BRCA1 variants using CRISPR-mediated cytosine base editors that enable targeted C:G to T:A conversion in living cells. This article elucidates the protocols for the decellularization of pancreatic tissue, evaluation of pancreatic tissue-derived dECM bioink, and generation of 3D pancreatic tissue constructs using a bioprinting technique. Biology A Novel Nicotinamide Adenine Dinucleotide Correction Method for Intracellular Ca2+ Measurement with Fura-2-Analog in Live Cells Jeong Hoon Lee1, Jeong Mi Ha1, Quynh Mai Ho1, Chae Hun Leem1,2,3 1Department of Physiology, University of Ulsan College of Medicine/Asan Medical Center, 2Asan Medical Center, 3Asan Medical Institute of Convergence Science and Technology Due to the spectral overlapping of the excitation and emission wavelengths of NADH and fura-2 analogs, the signal interference from both chemicals in live cells is unavoidable during quantitative measurement of [Ca2+]. Thus, a novel online correction method of NADH signal interference to measure [Ca2+] was developed.
