このプロトコルは、二重蛍光トランスジェニックマウスモデルにおけるマウス眼球表面の構造全体のライブイメージング用マルチフィルター顕微鏡を使用します。カスタムマルチフォトン顕微鏡プラットフォームと二重蛍光トランスジェニックマウスの組み合わせにより、眼表面の構造と構造をリアルタイムで可視化できます。マルチフォトン顕微鏡をセットアップするには、水浸漬20X、1.00 NA目的を選択し、直立顕微鏡上の励起源としてチタンサファイアレーザーを設定します。
レーザー出力波長をEGFP用880ナノメートル、tdTomatoの場合は940ナノメートルに設定します。SHG EGFPとEGFP tdTomatoの分離のための2つの二色性ミラーを含み、SHG EGFPおよびtdTomato信号をスペクトル的に分離するために434-17、510-84および585-40ナノメーターのバンドパスフィルターを使用する。眼ホルダーを設置するには、8-12週齢のマウスでペダル反射に対する応答の欠如を確認した後、加熱顕微鏡ステージにマウスを置き、温度監視プローブを挿入します。

角膜表面のZシリアルイメージングでは、水銀光源を使用してターゲットフィールドを画像化し、顕微鏡ソフトウェアを開きます。眼表面の細胞構造を視覚化するための適切な写真乗数とデジタルゲインを選択し、画像スタックの取得を可能にする最初と最後のスライドを設定します。画像解像度を 512 x 512 ピクセルに設定し、Z ステップを 1 マイクロメートルに設定します。
次に、SHG EGFP信号収集用880ナノメートル励起を用いた1つのライブ画像を取得するZシリアル画像を収集し、各領域内のEGFP tdTomato信号コレクション用の940ナノメートルの励起で1つのライブ画像を収集するために、開始をクリックします。画像全体を通して、ステッパー電動ステージホルダーを使用して眼球を回転させ、角膜表面全体をイメージングできるようにします。すべての画像が取得されたら、フィジーにZシリアル画像をロードし、中央値3Dフィルタプラグインを選択します。
バックグラウンドノイズを除去した後、鮮明なマスクフィルターパッケージフィジーを選択して画像をシャープにし、自動輝度コントラストをクリックして画像の品質を自動的に最適化します。処理後、イメージをシリアル画像シーケンスとして保存し、適切な3D再構成ソフトウェアプログラムにイメージシーケンスをエクスポートします。マルチフォトン顕微鏡画像の全てにおいて、EGFP tdTomatoとSHG信号をそれぞれ擬似緑、赤、シアンで表示してから、スナップショットで3D構造画像をキャプチャします。
多光子顕微鏡検査により、二重蛍光トランスジェニックマウスの角膜上皮における表在翼細胞および基底細胞の可視化が可能となる。基底層からの単一細胞は、単一の六角形の表面細胞と同様に表層にマッピングすることができる。tdTomatoシグナルの細胞質発現によって明らかにされるように、ゴルジ装置および小胞体を含む膜タンパク質は、翼細胞内に散乱される。
