Een demonstratie van de isolatie van neonatale muis ruggenmerg voor elektrofysiologische studies.
Abstract
De neonatale muis ruggenmerg is een model voor het bestuderen van de ontwikkeling van neurale schakelingen en bewegingsapparaat beweging. Tonen we aan het ruggenmerg dissectie en de voorbereiding van de opname bad kunstmatige cerebrospinale vloeistof die gebruikt wordt voor de motorische studies. Eenmaal ontleed, kan het ruggenmerg ventrale zenuwwortels worden aangesloten op een registratie-elektrode naar de elektrofysiologische signalen van de centrale patroon genereren circuit record in de lumbale snoer.
Protocol
1. Bereid kunstmatige cerebrospinale vloeistof (aCSF) 1. We hebben eerst voor te bereiden een 2L van een 10X voorraad aCSF zonder magnesium of calcium. Reagentia genoemd in millimolaire. De geïsoleerde neonatale ruggenmerg biedt een handelbaar methode van de bestudering zenuwstelsel ontwikkeling1, 2. Binnen de lumbale ruggenmerg van neonatale knaagdieren, centrale patroon genereren circuits kunnen fictieve motoriek te produceren in de aanwezigheid van neurotransmitters.
