Den snabba utvecklingen, liten storlek och öppenhet zebrafisk är enorma fördelar för studier av medfödda immunförsvaret kontroll av infektion<sup> 1-4</sup>. Här visar vi tekniker för att infektera zebrafisk larver med hjälp av svamppatogen<em> Candida albicans</em> Genom mikroinjektion, metodik använts nyligen att blanda fagocytsystemet aktivitet NADPH oxidas kontroll över svamp dimorphism<sup> 5</sup>.
Disseminerad candidiasis som orsakas av patogen Candida albicans är en kliniskt viktigt problem i sjukhus individer och är associerad med en 30 till 40% kan hänföras mortalitet 6. Systemisk candidiasis normalt styrs av medfödda immunitet, och individer med genetiska defekter i medfödda immunförsvaret cell komponenter såsom fagocytsystemet NADPH oxidas är mer mottagliga för candidemia 7-9. Mycket lite är känt om dynamiken av C. albicans interaktion med medfödda immunceller in vivo. Omfattande studier in vitro har visat att utanför den mottagande C. albicans gror inuti makrofager och snabbt förstörs av neutrofiler 10-14. In vitro-studier, men nyttig, inte kan rekapitulera den komplexa in vivo miljö, vilket inkluderar tidsberoende dynamiken i cytokinnivåer, extracellulära bilagor matris, och intercellulära kontakter 10, 15-18 </sup>. Att söka bidrag för dessa faktorer i värd-patogen interaktion, är det viktigt att hitta en modell organism att visualisera dessa aspekter av infektionen icke-invasivt i en levande intakt värd.
Zebrafisk larven erbjuder en unik och mångsidig vertebratvärden för studiet av infektionen. Under de första 30 dagarna av utvecklingen zebrafisk larver har bara medfödda immunförsvar 2, 19-21, vilket förenklar studier av sjukdomar som sprids candidiasis som är starkt beroende av medfödd immunitet. Den lilla storleken och insyn i zebrafisk larver möjliggör avbildning av infektion dynamik på cellulär nivå för både värd och patogen. Transgen larver med fluorescerande medfödda immunförsvaret celler kan användas för att identifiera specifika celltyper inblandade i infektioner 22-24. Modifierade anti-sense-oligonukleotider (Morpholinos) kan användas för att slå ner olika immun komponenter såsom fagocytsystemet NADPH oxidas och studera de förändringar som svar på fungal infektion 5. Förutom de etiska och praktiska fördelarna med att använda en liten mindre ryggradsdjur, erbjuder zebrafisk larverna den unika möjligheten att avbilda fältslag mellan patogen och värd både intravitally och i färg.
Zebrafisk har använts för att modellera infektion för ett antal humanpatogena bakterier, och har varit avgörande i de stora framsteg i vår förståelse av mykobakterieinfektion 3, 25. Men först nyligen har mycket större patogener såsom svampar använts för att infektera larven 5, 23, 26, och hittills har det inte funnits en detaljerad visuell beskrivning av infektionen metoden. Här presenterar vi våra tekniker för bakhjäman kammare mikroinjektion av prim 25 zebrafisk, inklusive våra ändringar tidigare protokoll. Våra fynd med hjälp av larver zebrafisk modellen för svampinfektion avviker från in vitro-studier och stärka behovet av att undersöka värd-patogen InteraInsatser som den komplexa miljö hos värden snarare än den förenklade systemet av petriskålen 5.
Zebrafisk mikroinjektion metod som presenteras här skiljer sig från Gutzman et al. 34 i att vi här visar injektion genom örat vesikeln i bakhjäman ventrikeln av 36 till 48 HPF larver. Metoden beskriver vi gör för konsekvent injektion av 10-15 jäst i bakhjäman kammaren med minskad vävnadsskada.
