Den raske utviklingen, liten størrelse og gjennomsiktighet sebrafisk er enorme fordeler for studiet av medfødte immunsystemet kontroll av infeksjon<sup> 1-4</sup>. Her demonstrerer vi teknikker for å infisere sebrafisk larver med sopp patogen<em> Candida albicans</em> Med mikroinjeksjon, metodikk nylig brukt til å implisere phagocyte NADPH oksidase aktivitet i kontroll av sopp dimorphism<sup> 5</sup>.
Spres candidiasis forårsaket av patogenet Candida albicans er en klinisk viktig problem i sykehus individer og er forbundet med en 30 til 40% skyldes dødelighet 6. Systemisk candidiasis er normalt kontrolleres av medfødt immunitet, og personer med genetiske defekter i medfødte immunceller komponenter som phagocyte NADPH oksidase er mer utsatt for candidemia 7-9. Svært lite er kjent om dynamikken i C. albicans samspill med medfødte immunceller in vivo. Omfattende in vitro studier har fastslått at utenfor verten C. albicans spirer innsiden av makrofager, og er raskt ødelagt av nøytrofile 10-14. In vitro-studier, men nyttig, ikke kan rekapitulere komplekse in vivo miljø, som inkluderer tidsavhengige dynamikken i cytokin nivåer, ekstracellulære matrix vedlegg og intercellulær kontakter 10, 15-18 </sup>. For å undersøke bidraget av disse faktorene i host-patogen samspill, er det avgjørende å finne en modell organisme å visualisere disse sidene ved infeksjon non-invasiv i en levende intakt vert.
Sebrafisk larve tilbyr en unik og allsidig virveldyr vert for studiet av infeksjon. For de første 30 dagene av utvikling sebrafisk larver har bare medfødte immunforsvar 2, 19-21, forenkle studiet av sykdommer som spres candidiasis som er sterkt avhengig av medfødt immunitet. Den lille størrelsen og gjennomsiktigheten sebrafisk larver gjør avbildning av infeksjon dynamikk på cellenivå for både vert og patogen. Transgene larver med fluoriserende medfødte immunceller kan brukes til å identifisere spesifikke celler typer involvert i infeksjon 22-24. Modifiserte anti-sense oligonukleotider (Morpholinos) kan brukes til å slå ned ulike immunologiske komponenter som phagocyte NADPH oksidase og studere endringer i respons til fungal infeksjon fem. I tillegg til de etiske og praktiske fordelene ved å bruke en liten lavere virveldyr, tilbyr sebrafisk larvene en unik mulighet til å avbilde pitched kampen mellom patogen og vert både intravitally og i farger.
Sebrafisk har blitt brukt til å modellere infeksjon for en rekke humanpatogene bakterier, og har vært medvirkende i store fremskritt i vår forståelse av mykobakterielle infeksjon 3, 25. Men bare nylig har mye større patogener som sopp blitt brukt til å infisere larve 5, 23, 26, og hittil har det ikke vært en detaljert visuell beskrivelse av infeksjonen metodikk. Her presenterer vi våre teknikker for hindbrain ventrikkel mikroinjeksjon av prim 25 sebrafisk, inkludert våre modifikasjoner til tidligere protokoller. Våre funn ved hjelp av larvenes sebrafisk-modellen for soppinfeksjon avvike fra in vitro studier og forsterke behovet for å undersøke host-patogen interaction i det komplekse miljøet verten snarere enn forenklet system petriskål 5.
Sebrafisk mikroinjeksjon metoden presenteres her avviker fra Gutzman et al. 34 i det her vi demonstrere injeksjon gjennom otic vesicle inn hindbrain ventrikkel 36-48 HPF larver.
