Bioactive and mechanically reliable metal scaffolds have been fabricated through a method which consists of two processes, dynamic freeze casting for the fabrication of porous Ti, and coating and densification of the Ti scaffolds. The densification process is simple, effective and applicable to the fabrication of functionally graded scaffolds.
Biometal systems have been widely used for biomedical applications, in particular, as load-bearing materials. However, major challenges are high stiffness and low bioactivity of metals. In this study, we have developed a new method towards fabricating a new type of bioactive and mechanically reliable porous metal scaffolds-densified porous Ti scaffolds. The method consists of two fabrication processes, 1) the fabrication of porous Ti scaffolds by dynamic freeze casting, and 2) coating and densification of the porous scaffolds. The dynamic freeze casting method to fabricate porous Ti scaffolds allowed the densification of porous scaffolds by minimizing the chemical contamination and structural defects. The densification process is distinctive for three reasons. First, the densification process is simple, because it requires a control of only one parameter (degree of densification). Second, it is effective, as it achieves mechanical enhancement and sustainable release of biomolecules from porous scaffolds. Third, it has broad applications, as it is also applicable to the fabrication of functionally graded porous scaffolds by spatially varied strain during densification.

Därför har vi utvecklat en ny strategi för att tillverka bioaktiva och mekaniskt avstämbara porösa Ti ställningar. 25 De byggnadsställningar har inledningsvis porösa strukturer med en porositet av mer än 50%. De tillverkade porösa ställningar belades med bioaktiva molekyler och sedan komprimeras med hjälp av en mekanisk press under vilken den slutliga porositet, mekaniska egenskaper och läkemedelsfrisättningsbeteende kontrollerades tillämped-stam. De förtätade porösa Ti implantat har visat låg porositet med god styrka trots den låga styvhet som är jämförbar med den hos ben (3-20 GPa). 2 På grund av beläggningsskiktet, var bioaktiviteten hos den förtätade porösa Ti förbättrats avsevärt. I denna studie, introducerade vi vår etablerad metod att tillverka förtätade porösa Ti ställningar för potentiell användning i biomedicinska tillämpningar. Protokollet innehåller dynamiska frysning gjutning med metallslam och förtätning av porösa ställningar. Först, för att tillverka porösa Ti ställningar med god duktilitet den dynamiska frysgjutningsmetoden infördes, såsom visas i Figur 1A. Ti pulver dispergerades i flytande kamfen; sedan, genom att sänka temperaturen,vätskefasen stelnade, vilket resulterar i fasseparation mellan pulvernätet Ti och fasta kamfen kristaller. Därefter fick stelnade Ti-kamfen grönkropp sintras i vilken Ti pulver kondenserades med kontinuerliga Ti strävor och kamfen fasen avlägsnades fullständigt för att erhålla en porös struktur. Beläggningen och förtätningsprocessen med de erhållna porösa stöden användes, variera graden av förtätning och initiala porositet. Beläggningsskiktet och dess frisättning beteende visualiserades och kvantifierades med användning av grönt fluorescerande protein (GFP) -belagda porös Ti med och utan förtätning jämfört med GFP belagda tät Ti. Slutligen var funktionellt graderade Ti byggnadsställningar som har två olika porösa strukturer föreslagits och demonstrerats genom att variera graden av förtätning av de inre och yttre delarna av de porösa ställningar.
