Bioactive and mechanically reliable metal scaffolds have been fabricated through a method which consists of two processes, dynamic freeze casting for the fabrication of porous Ti, and coating and densification of the Ti scaffolds. The densification process is simple, effective and applicable to the fabrication of functionally graded scaffolds.
Biometal systems have been widely used for biomedical applications, in particular, as load-bearing materials. However, major challenges are high stiffness and low bioactivity of metals. In this study, we have developed a new method towards fabricating a new type of bioactive and mechanically reliable porous metal scaffolds-densified porous Ti scaffolds. The method consists of two fabrication processes, 1) the fabrication of porous Ti scaffolds by dynamic freeze casting, and 2) coating and densification of the porous scaffolds. The dynamic freeze casting method to fabricate porous Ti scaffolds allowed the densification of porous scaffolds by minimizing the chemical contamination and structural defects. The densification process is distinctive for three reasons. First, the densification process is simple, because it requires a control of only one parameter (degree of densification). Second, it is effective, as it achieves mechanical enhancement and sustainable release of biomolecules from porous scaffolds. Third, it has broad applications, as it is also applicable to the fabrication of functionally graded porous scaffolds by spatially varied strain during densification.
Terwijl metalen biomaterialen zijn op grote schaal gebruikt als dragende implantaten en interne fixatie inrichtingen vanwege hun uitstekende mechanische sterkte en veerkracht, 1-3 zij betrekking twee kritische problemen: 1) mechanische mismatch omdat metalen zijn veel stijver dan biologische weefsels, wat ongewenste schade de omringende weefsels en 2) lage bioactiviteit die vaak resulteert in een slechte koppeling met biologische weefsels, vaak veroorzakend reacties van het lichaam (bijvoorbeeld een ontsteking of trombose). hebben 4-6 poreus metallisch steigers voorgesteld om botingroei te bevorderen in de structuren, verbeteren . bot-implantaat contact terwijl de spanning afscherming effecten onderdrukt vanwege hun verminderde stijfheid 7-9 Voorts dient het metaal wijzigingen zijn aangebracht om de biologische activiteiten van metalen implantaten te verbeteren; dergelijke modificaties omvatten coating het metaaloppervlak met bioactieve moleculen (bijvoorbeeld groei facren) of drugs (bijvoorbeeld vancomycine, tetracycline). 12/10 echter problemen zoals verminderde mechanische eigenschappen van poreuze metalen steigers, verminderde stijfheid en de snelle afgifte van de bioactieve coating lagen onopgelost blijven. 13-16
In het bijzonder titaan (Ti) en Ti legeringen zijn een van de meest populaire BioMetal systemen vanwege hun uitstekende mechanische eigenschappen, chemische stabiliteit en goede biocompatibiliteit. 13,17-19 hun schuim-vormig aanvragen trok ook toenemende belangstelling omdat het 3D poreuze netwerken voor botingroei naast botachtige mechanische eigenschappen. 20-22 zijn pogingen gedaan om de mechanische eigenschappen te verbeteren door de ontwikkeling van nieuwe fabricagetechnieken zoals replicatie polymere spons, sinteren van metaaldeeltjes, Rapid Prototyping (RP) methode, en space houder methode om de verschillende functies van de poriën regelen (bijvoorbeeld poriën fractievorm, grootte, distributie en connectivity) en materiaaleigenschappen (bv metallische fase en onzuiverheid). 23-25 Onlangs heeft de vries gieten van water gebaseerde metal drijfmest heeft opgedaan veel aandacht aan mechanisch versterkte Ti vormen met goed uitgelijnd poriën produceren structuren door gebruikmaking van de unidirectionele ijs dendriet groei tijdens het stollen; echter, zuurstof vervuiling door contact van metaalpoeders met water vereist speciale zorg aan de verbrossing van Ti scaffolds minimaliseren. 14,15
Daarom hebben we een nieuwe benadering fabriceren bioactieve en mechanisch afstembare poreuze Ti scaffolds ontwikkeld. 25 De steigers ligt aanvankelijk poreuze structuur met een porositeit van meer dan 50%. De gefabriceerde poreuze steigers werden bekleed met biologisch actieve moleculen en vervolgens gecomprimeerd met behulp van een mechanische pers waarin de uiteindelijke porositeit, mechanische eigenschappen en de geneesmiddelafgifte gedrag werden gecontroleerd door de toeed stam. De verdichte poreuze Ti implantaten aangetoond lage porositeit met goede sterkte ondanks de lage stijfheid vergelijkbaar met dat been (3-20 GPa). 2 vanwege de bekledingslaag, werd de biologische activiteit van de verdichte poreuze Ti aanzienlijk verbeterd. Bovendien, vanwege de unieke platte poriënstructuren geïnduceerd door de verdichtingswerkwijze, de beklede bioactieve moleculen werden gezien geleidelijk afgegeven uit het schavot behoud van hun werkzaamheid langdurig.
In deze studie hebben we onze gevestigde methode verdichte poreuze Ti draagstructuren voor potentieel gebruik in biomedische toepassingen fabriceren geïntroduceerd. Het protocol bevat dynamische bevriezing gieten met metalen slurry en verdichting van poreuze steigers. Ten eerste, poreuze draagstructuren Ti goede vervormbaarheid de dynamische freeze gietmethode ingevoerd zoals getoond in figuur 1A fabriceren. Ti poeder werd verspreid in vloeibare kamfeen; Vervolgens, door het verlagen van de temperatuur,de vloeibare fase werd gestold, waardoor de fasescheiding tussen de Ti poeder netwerk en vaste camfeen kristallen. Vervolgens werd de gestolde Ti-camfeen groenling gesinterd waarin Ti poeders werden gecondenseerd met doorlopende Ti stutten en de camfeen fase werd volledig verwijderd om een poreuze structuur te verkrijgen. De coating en verdichtingswerkwijze de verkregen poreuze draagstructuren werd toegepast, variëren van de mate van verdichting en beginporositeit. De bekledingslaag en afgiftegedrag gevisualiseerd en gekwantificeerd met het green fluorescent protein (GFP) gecoate poreuze Ti met en zonder verdichting in vergelijking met de GFP-beklede dichte Ti. Tenslotte functionele gradient Ti scaffolds die twee poreuze structuren voorgesteld en gedemonstreerd door het variëren van de mate van verdichting van de binnenste en buitenste delen van de poreuze steigers.
Terwijl BioMetal systemen zijn op grote schaal gebruikt voor biomedische toepassingen, met name als dragende materialen, hoge stijfheid en lage bioactiviteit van metalen werden beschouwd als belangrijke uitdagingen.
