Combining cell transplantation, cytoskeletal labeling and loss/gain of function approaches, this protocol describes how the migrating zebrafish prospective prechordal plate can be used to analyze the function of a candidate gene in in vivo cell migration.
Cell migration is key to many physiological and pathological conditions, including cancer metastasis. The cellular and molecular bases of cell migration have been thoroughly analyzed in vitro. However, in vivo cell migration somehow differs from in vitro migration, and has proven more difficult to analyze, being less accessible to direct observation and manipulation. This protocol uses the migration of the prospective prechordal plate in the early zebrafish embryo as a model system to study the function of candidate genes in cell migration. Prechordal plate progenitors form a group of cells which, during gastrulation, undergoes a directed migration from the embryonic organizer to the animal pole of the embryo. The proposed protocol uses cell transplantation to create mosaic embryos. This offers the combined advantages of labeling isolated cells, which is key to good imaging, and of limiting gain/loss of function effects to the observed cells, hence ensuring cell-autonomous effects. We describe here how we assessed the function of the TORC2 component Sin1 in cell migration, but the protocol can be used to analyze the function of any candidate gene in controlling cell migration in vivo.
I flercelliga organismer, är cellmigration viktigt både för utvecklingen av embryot där det garanterar att organisera celler i vävnader och organ, och för vuxenlivet, där det tar del vävnadshomeostas (sårläkning) och immunitet. Förutom dessa fysiologiska funktioner, är cellmigration också involverad i olika patologiska situationer, inklusive i synnerhet cancermetastaser.
Cellmigration har analyserats in vitro i årtionden, vilket ger en övergripande förståelse för de molekylära mekanismer som säkerställer cellrörelser på plana ytor. In vivo dock celler inför en mer komplex miljö. Det framgick tydligt under de senaste åren att migration inom en organism kan påverkas av yttre signaler såsom den extracellulära matrisen, angränsande celler eller utsöndrade kemokiner vägledande migration, och att de mekanismer som driver cellmigration kan variera från vad som har beskrivits <em> In vitro 1,2. Mekanismerna som säkerställer in vivo cell migration har fått mindre uppmärksamhet hittills, främst på grund av den ökade tekniska svårigheter, jämfört med in vitro-studier. In vivo analys av cellmigration i synnerhet kräver direkt optisk access till migrerande celler, tekniker för att märka unika celler i för att se deras dynamik och morfologi samt vinst eller förlust av funktion metoder för att testa rollen av kandidatgener. Hittills har endast ett fåtal modellsystem som härbärgerar dessa egenskaper använts för att dissekera in vivo cell migration 3.
Vi använde nyligen migreringen av den blivande prechordal plattan i början av zebrafisk embryon som en ny bekväm modell för att bedöma funktionen av kandidatgener för att kontrollera in vivo cell migration 4,5. Prospektiv prechordal platta (även känd som främre mesendoderm) är en grupp av celler som bildar vid uppkomsten av gastrulation på den dorsala sidan av embryot. Under gastrulation denna grupp migrerar kollektivt mot djuret pol av embryot 6-8, för att bilda den prechordal platta, en mesendodermal förtjockning, anterior till notokorden, och ligger till grund för neurala plattan. Den främre delen av prechordal plattan kommer att ge upphov till kläckning körtel, medan dess bakre del bidrar sannolikt att leda mesoderm 9. Tack vare den externa utvecklingen och optisk klarhet av fisken embryot, kan cellmigration direkt och lätt observeras i denna struktur.
Celltransplantation är en mycket potent teknik som gör det möjligt att snabbt och enkelt skapa mosaik embryon 10. Att uttrycka fluorescerande cytoskelettala markörer i transplanterade celler resulterar i märkning av isolerade celler, morfologi och dynamik som lätt kan observeras. Kombinera detta till förlust eller vinst av funktion närmar tillåter analys av cellautonoma funktioner i en burkkandidaten genen.
De presenterade protokoll beskriver hur vi bedömt funktion TORC2 komponent Sin1 i att kontrollera cellmigration och aktin dynamik in vivo 5. Men, som nämnts i resultaten och ytterligare diskuteras, det skulle kunna användas för att analysera de möjliga konsekvenserna av varje kandidatgen i att kontrollera cellmigration in vivo.
Detta protokoll utgör ett enkelt sätt att studera rollen av en kandidatgen i cell migration in vivo, genom att kombinera att skapa chimära embryon med hjälp av celltransplantation med levande avbildning.
