Rabbits are widely used to study the pharmacokinetics of intraocular drugs. We describe a method for conducting pharmacokinetic studies of intraocular drugs using rabbit eyes.
Den intraokulære rute for medikamentadministrasjon muliggjør levering av høye konsentrasjoner av terapeutiske legemidler, mens deres systemiske absorpsjon minimaliseres. Mange medikamenter blir administrert inn i det fremre kammer eller glasslegemet, og den intraokulære injeksjons har vært effektiv i herding forskjellige intraokulære sykdommer. Kaninøyne har blitt mye brukt for oftalmisk forskning, som dyret er lett å håndtere og økonomisk i forhold til andre pattedyr, og størrelsen på et kaninøye er lik den for et menneskelig øye. Ved å bruke en 30 G nål, kan medikamenter injiseres i intracameral og intravitreal rom av kaninøyne. Øyeeplet blir deretter frosset inntil analyse, og kan deles inn i den vandige humor, glasslegemet, og netthinnen / årehinne. Glasslegemet og netthinnen / choroid prøver kan homogenisert og oppløst før analyse. Deretter kan immunoanalyser utføres for å måle konsentrasjonene av intraokulære medikamenter i hvert kammer. Passende farmakokinetiske modeller kan værebrukes til å beregne flere parametere, slik som halveringstid og maksimal konsentrasjon av medikamentet. Kaninøyne kan være en god modell for farmakokinetiske studier av intraokulære narkotika.
Før ankomsten av intraokulært levering av legemidler, den største bekymringen av medisinsk behandling for intraokulære sykdommer var effektiviteten som stoffet kan trenge inn i øyet. Blod-okulære barrieren hindrer mange substanser, blant annet medikamenter, fra å diffundere inn i øyet. Derfor konsentrasjoner av stoffer som er ovenfor terapeutiske nivåer kan ikke lett oppnås. Den intraokulære legemiddeladministrering metode, inkludert intracameral og intravitreal injeksjon, kan direkte omgå blod-barrieren okulær 1-3, slik at det terapeutiske konsentrasjoner av medikamenter kan oppnås i øyet 4,5.
Følgelig har intravitreal medikamentavgivelse blitt en populær metode for behandling av flere sykdommer intraokulær 5,6. For eksempel er intravitreal injeksjon viden utført for aldersrelatert makuladegenerasjon, diabetisk retinopati, retinal vene okklusjon, og intraokulære infeksjoner 7-10. Spesielt, sideninnføring av anti-VEGF-medisiner, har hyppigheten av intravenøse injeksjoner merkbart øket for behandling av retinale sykdommer. Derfor er det viktig å forstå de intraokulære farmakokinetikken til slike stoffer for å evaluere effektiviteten og sikkerheten til den medisinske terapi.
Selv om intraokulære administrering av legemidler anses som et stort gjennombrudd i medisinsk behandling for øyesykdommer, overvåking av legemiddelkonsentrasjonen i øyeeplet er teknisk krevende. Fordi det menneskelige øye bare inneholde små mengder av vandig humor (ca. 200 ul) og glasslegeme (ca. 4,5 ml, tabell 1), er det teknisk vanskelig å oppnå tilstrekkelige mengder av okulære fluidet for å måle medikamentkonsentrasjonen. Videre metoder som brukes for å få væsken øyet, slik som glasslegemet tapping eller fremre kammer paracentesis, kan skade øyet vev og føre til alvorlige komplikasjoner, som for eksempel grå stær, endoftalmitt ellernetthinneavløsning 11,12. Følgelig er dyremodeller brukes i farmakokinetiske studier av brukte intraokulære narkotika 13. Blant disse dyremodeller, kaniner eller aper er de mest brukte dyr.
Kanin, som er små pattedyr av ordenen Haredyr i familien Leporidae, finnes i flere deler av verden. Fordi kaniner er ikke aggressive, de er enkle å håndtere, bruke i et eksperiment, og observere. Lavere kostnader, lett tilgjengelighet av dyret, lik øye størrelse for mennesker, og en stor database med informasjon for sammenligning fordel å utføre farmakokinetiske studier ved hjelp av kaninøyne.
