We beschrijven een aangepaste werkwijze voor membraanfractie isolatie uit humane cellen en monstervoorbereiding voor de detectie van SCAP N -glycosylation en totaal eiwit door Western blot. We verdere invoering van een GFP-labeling methode om SCAP mensenhandel te monitoren met behulp van confocale microscopie. Dit protocol kan in normale biologische laboratoria.
Elevated lipogenesis is a common characteristic of cancer and metabolic diseases. Sterol regulatory element-binding proteins (SREBPs), a family of membrane-bound transcription factors controlling the expression of genes important for the synthesis of cholesterol, fatty acids and phospholipids, are frequently upregulated in these diseases. In the process of SREBP nuclear translocation, SREBP-cleavage activating protein (SCAP) plays a central role in the trafficking of SREBP from the endoplasmic reticulum (ER) to the Golgi and in subsequent proteolysis activation. Recently, we uncovered that glucose-mediated N-glycosylation of SCAP is a prerequisite condition for the exit of SCAP/SREBP from the ER and movement to the Golgi. N-glycosylation stabilizes SCAP and directs SCAP/SREBP trafficking. Here, we describe a protocol for the isolation of membrane fractions in human cells and for the preparation of the samples for the detection of SCAP N-glycosylation and total protein by using western blot. We further provide a method to monitor SCAP trafficking by using confocal microscopy. This protocol is appropriate for the investigation of SCAP N-glycosylation and trafficking in mammalian cells.
Deregulering van het vetmetabolisme is een gemeenschappelijk kenmerk van kanker en metabole ziekten 1-6. Bij deze werkwijzen, sterol-regulerende element bindende eiwitten (SREBPs), een familie van transcriptiefactoren, spelen een cruciale rol in de expressie van genen betrokken in de opname en de synthese van cholesterol, vetzuren en fosfolipiden 7-10.
SREBPs, waaronder SREBP-1a, SREBP-1c en SREBP-2, worden gesynthetiseerd als inactieve precursors die binden aan het endoplasmatisch reticulum (ER) membraan vanwege twee transmembraandomeinen 7. De N-terminus van SREBPs bevat een DNA-bindingsdomein voor de transactivatie van doelgenen 11. De C-terminus van SREBP precursors bindt aan SREBP-splitsing activerend eiwit (SCAP) 12,13, polytopisch een membraaneiwit dat een centrale rol in de regulatie van SREBP stabiliteit en activering 14-16 speelt.
de uitvoelegging van transcriptionele functie vereist de translocatie van de SCAP / SREBP complex uit het ER naar het Golgi, waar twee proteases sequentieel SREBP splitsen en laat de N-terminale fragment, dat vervolgens gaat in de kern van de transactivatie van lipogene genen 7. Bij deze werkwijzen, de niveaus van sterolen in het ER membraan controle van de uitgang van de SCAP / SREBP complex uit het ER 7,17. Onder hoge sterol omstandigheden, sterol bindt aan SCAP of ER-resident insuline-geïnduceerde gen-eiwit-1 (Insig-1) of -2 (Insig-2), het verbeteren van de associatie van SCAP met Insigs dat de SCAP / SREBP complex in het behouden ER 18-20. Wanneer sterolen verlagen, SCAP distantieert met Insigs. Dit leidt tot een SCAP conformationele verandering die SCAP interactie gemeenschappelijke manteleiwitten (COP) II complex maakt. Het complex bemiddelt de opname van de SCAP / SREBP complex in de ontluikende blaasjes en richt haar vervoer van het ER naar het Golgi 21,22. Bij Translocatie het Golgi worden SREBPs achtereenvolgens gesplitst door site-1 en Site-2 proteasen, wat leidt tot de afgifte van N-terminus 7,23-29.
SCAP eiwit draagt drie N-gebonden oligosachariden op asparagine (N) positioneert N263, N590, N641 en 15. We hebben onlangs onthuld dat glucose-gemedieerde N -glycosylation van SCAP in deze sites is een voorwaarde voorwaarde voor SCAP / SREBP mensenhandel uit het ER naar het Golgi onder lage sterolen omstandigheden 30-32. Verlies van SCAP glycosylering via mutatie van alle drie de asparagine naar glutamine (NNN naar QQQ) schakelt de handel in de SCAP / SREBP complex en resulteert in de instabiliteit van SCAP eiwit en vermindering van SREBP activering 31. Onze recente gegevens tonen ook aan dat SREBP-1 is zeer actief in glioblastoma en gereguleerd door SCAP N -glycosylation 4,31,33,34. Targeting SCAP / SREBP-1-signalering is in opkomst als een nieuwe strategie om maligniteiten en metabole s behandelenyndromes 1,3,35-38. Daarom is het belangrijk om een effectieve methode om SCAP eiwit en N -glycosylation niveaus analyseren en controleren de handel in menselijke cellen en weefsels patiënten ontwikkelen.
De luminale regio SCAP eiwit (aminozuren 540-707) in het ER bevat twee N -glycosylation sites (N590 en N641), die worden beschermd tegen proteolyse wanneer intacte membranen worden behandeld met trypsine 15. Deze luminale fragment heeft een molecuulgewicht van ~ 30 kDa die klein genoeg is om de oplossing van individuele geglycosyleerde varianten van SCAP door natriumdodecylsulfaat-polyacrylamidegelelektroforese (SDS-PAGE) 15,31 toelaten. Hier geven we een methode om N -glycosylation van SCAP en de totale hoeveelheid eiwit in menselijke cellen te detecteren. Dit protocol is afgeleid van de in de publicaties van Brown en Goldstein laboratorium 15 en onze recente publicatie 31 beschreven werkwijze. Het protocol kan worden gebruikt in thij bestuderen van SCAP eiwit uit zoogdiercellen.
In deze studie beschrijven we een protocol voor de isolatie van membraanfracties in menselijke cellen en voor de bereiding van de monsters voor de detectie van SCAP N -glycosylation en totaal eiwit door Western blot.
