Her viser vi hvordan agarose-baserte vev-mimicking optisk fantomer gjøres og hvordan optiske egenskaper fastsettes ved hjelp av en konvensjonell optisk system med en integrert sfære.
Denne protokollen beskriver hvordan lage agarose-baserte vev-mimicking fantomer og demonstrerer hvordan å bestemme optiske egenskapene med en konvensjonell optisk system med en integrert sfære. Måle systemer for oppkjøpet av diffus refleksjon og totale transmisjon spectra er bygget med bredbånd hvit lyskilde, en lys guide, en achromatic linse, en integrert sfære, prøve innehaver, en optisk fiber sonde, og flerkanals spectrometer. En akryl mold som består av to rektangulære akryl stykker og en U-formet akryl stykke er konstruert for å opprette en epidermal phantom og en dermal phantom med fullblod. Anvendelsen av en natrium dithionite (Na2S2O4) løsning til dermal phantom gjør forskeren til deoxygenate hemoglobin i røde blod celler distribuert i dermal phantom. Inverse Monte Carlo simulering med diffus refleksjon og totale transmisjon spectra målt ved et spektrometer med en integrert sfæren er utført for å avgjøre den absorpsjon koeffisienten spektrum µen(λ) og redusert spredning koeffisienten spektrum μs‘ (λ) i hvert lag phantom. En to-lagdelt phantom mimicking diffus refleksjon av menneskelige huden vev er også demonstrert av piling opp epidermal phantom på dermal phantom.
Optisk phantoms objekter mimicking den optiske egenskapene av biologisk vev og brukt mye innen Biomedisinsk optikk. De er utformet slik at den optiske egenskapene, for eksempel lysspredning og absorpsjon koeffisienter, samsvarer med de av levende menneske og dyr vev. Optisk fantomer brukes vanligvis til følgende formål: simulere lys transport i biologisk vev, kalibrere en nyutviklet optisk systemdesign, vurdere kvaliteten og ytelsen av eksisterende systemer, sammenligne ytelsen mellom systemer og validere evne av optiske metoder å kvantifisere den optiske egenskaper1,2,3,4,5. Derfor kreves lett å få stoffer, en enkel fabrikasjon prosessen, en høy reproduserbarhet og en optisk stabilitet for å lage optiske fantomer.
Ulike typer optisk fantomer med forskjellige base materialer som vandig suspensjon6, gelatin gel7, agarose gel8,9,10, polyakrylamid gel11, harpiks12, 13,14,15,16, og rom-temperatur-vulcanizing silikon17 har blitt rapportert i forrige litteratur. Det har blitt rapportert at gelatin og alginate-baserte gels er nyttig for optisk fantomer med heterogene strukturer18. Alginate fantomer har en egnet mekanisk og termisk stabilitet for å vurdere photothermal effekter som laser ablasjon studier og laser-basert hypertermi studier18. Agarose gels har muligheten til å dikte heterogene strukturer, og deres mekaniske og fysiske egenskaper er stabil for en lang tid18. Høy renhetsgrad agarose gels har en svært lav turbiditet og en svak optisk absorpsjon. Derfor kan optiske egenskaper av agarose-baserte fantomer lett være utformet med riktig lys spredning og absorberende agenter. Nylig styren-etylen-butylene-styren (SEBS) blokk copolymers19 og polyvinylklorid (PVC) gels20 har blitt rapportert som interessant phantom materialer for optisk og photoacoustic teknikker.
Polymer mikrosfærer7,12,21,22, Titan oksid pulver1og lipid emulsjoner23,24,25,26 som melk og lipid emulsjon brukes som lysspredning agenter, mens svart blekk27,28 og molekylære fargestoffer29,30 brukes som lys dempere. Diffus refleksjon spektra av de fleste levende organer er dominert av absorpsjon av oksygen og deoxygenated hemoglobin i røde blod celler. Derfor hemoglobin løsninger31,32 og fullblod,8,,9,,10,,33,,36 blir ofte brukt som lys dempere i den fantomer for en diffus refleksjon spektroskopi og multispectral bildebehandling.
Metoden beskrevet i denne artikkelen brukes til å opprette en optisk phantom mimicking lys transport i biologisk vev og karakterisere optiske egenskaper. Som et eksempel, en to-lagdelt optisk phantom mimicking optiske egenskaper av menneskelig hud vev er demonstrert. Fordelene med denne metoden over alternative teknikker er muligheten til å representere diffus refleksjon spektra av levende biologisk vev i den synlige for nær-infrarøde bølgelengdeområdet, samt enkelheten å gjøre det, bruke som er lett tilgjengelig materialer og konvensjonelle optiske instrumenter. Derfor vil de optiske fantomer laget av denne metoden være nyttig for utvikling av optisk metoder basert på diffus refleksjon spektroskopi og multispectral bildebehandling.
Det viktigste trinnet i denne protokollen er temperaturkontroll grunnmaterialet. Temperaturen å opprettholde grunnmaterialet varierte fra 58 til 60 ° C. Hvis temperaturen er mer enn 70 ° C, oppstår en denaturering av både den lipid emulsjonen og hele blod. Som en konsekvens, vil den optiske egenskapene av phantom svekkes.
