Dit protocol wordt beschreven hoe u agarose gebaseerde weefsel nabootsen fantomen en laat zien hoe om te bepalen hun optische eigenschappen met behulp van een conventionele optische systeem met een bolfotometer. Meetinstallaties voor de overname van de diffuse reflectie en de totale doorlatingsfactor spectra worden geconstrueerd met een breedband witte lichtbron, een lichtgeleider, een achromatische lens, een bolfotometer, een monsterhouder, een optische vezel sonde, en een meerkanaals spectrometer. Een acryl schimmel, bestaande uit twee rechthoekige acryl stukken en een U-vormige acryl stuk is gebouwd om te maken een epidermale phantom en een dermale phantom met volbloed. De toepassing van een natrium-dithioniet (Na2S2O4)-oplossing op de dermale phantom in staat stelt de onderzoeker naar deoxygenate hemoglobine in rode bloedcellen in de dermale phantom verspreid. De inverse Monte Carlo simulatie met de diffuse reflectie en de totale doorlatingsfactor spectra gemeten door een spectrometer met een bolfotometer is uitgevoerd om te bepalen van de absorptie coëfficiënt spectrum µeen(λ) en de verminderd verstrooiing coëfficiënt spectrum µs‘ (λ) van elke laag phantom. Optische fantomen zijn objecten nabootsen van de optische eigenschappen van biologische weefsels en hebben op grote schaal gebruikt op het gebied van de biomedische optica. Ze zijn zo ontworpen dat de optische eigenschappen, zoals licht verstrooiing en absorptie coëfficiënten, met die van levende menselijke en dierlijke weefsels overeenkomen. Optische phantoms worden meestal gebruikt voor de volgende doeleinden: het lichte transport in biologische weefsels, kalibreren van een nieuw ontwikkelt optisch systeemontwerp, evaluatie van de kwaliteit en de prestaties van bestaande systemen, vergelijken van de prestaties simuleren tussen systemen, en het vermogen van de optische methodes te kwantificeren van de optische eigenschappen1,2,3,4,5te valideren. Daarom, gemakkelijk-aan-get stoffen, een eenvoudige productie-procédé, een hoge reproduceerbaarheid en een optische stabiliteit zijn vereist voor het maken van optische fantomen.
Verschillende soorten optische phantoms met verschillende basismaterialen zoals waterige suspensie6, gelatine gel7, agarose gel8,9,10, polyacrylamidegel11, hars12, 13,14,15,16, en kamer-temperatuur-vulcanizing siliconen17 zijn gemeld in de vorige literatuur. Er werd gemeld dat gelatine en alginaat-gebaseerde gels nuttig voor optische phantoms met heterogene structuren18 zijn. Alginaat phantoms hebben een geschikte mechanische en thermische stabiliteit voor de evaluatie van de effecten van de photothermal zoals laser ablatie studies en hyperthermie laser gebaseerde studies18. Agarose gel hebben de mogelijkheid om het fabriceren van de heterogene structuren en hun mechanische en fysische eigenschappen zijn stabiel voor een lange tijd18. Hoge zuiverheid agarose gel hebben een zeer lage turbiditeit en een zwakke optische absorptie. Daarom zou de optische eigenschappen van agarose gebaseerde phantoms gemakkelijk worden ontworpen met het juiste licht verstrooiing en het absorberen van agenten. Onlangs, styreen-ethyleen-butyleen-styreen (SEBS) blok copolymeren19 en PVC (polyvinylchloride) gels20 hebben gemeld als interessante phantom materialen voor optische en photoacoustic technieken.
Polymeer microsferen7,12,21,22, titanium-oxide poeder1en lipide emulsies23,24,25,26 zoals melk en lipide emulsie zijn gebruikt als gemachtigden van de verstrooiing van licht, terwijl zwarte inkt27,28 en moleculaire kleurstoffen29,30 als lichte absorptieflessen worden gebruikt. Diffuse reflectie spectra van de meeste levende organen worden gedomineerd door de absorptie van zuurstofrijk en gedeoxygeneerd hemoglobine in rode bloedcellen. Daarom hemoglobine oplossingen31,32 en volbloed8,9,10,33,36 worden vaak gebruikt als lichte absorptieflessen in de Phantoms voor een diffuse reflectie spectroscopie en multispectrale beeldbewerking.
De in dit artikel beschreven methode wordt gebruikt voor het maken van een optische phantom nabootsen het licht vervoer in biologische weefsels en te karakteriseren van de optische eigenschappen. Als voorbeeld, een twee-lagen optische phantom nabootsen optische eigenschappen van menselijke huidweefsel wordt aangetoond.
