Summary

Abstract

Introduction
在胚胎发育过程中,单个细胞层被转化为三个独立的生殖层1,2,3。为了增加在体内发生的现象的研究可能性,已经开发出三维聚集物(胚胎体)作为一种方便的模型。以这种方式形成的细胞聚集体可以暴露在各种导致细胞分化的条件下,这反映了胚胎4、5的发育。P19鼠胚胎癌细胞系(P19细胞系)常用作体外神经发生研究的细胞模型6、7、8。P19细胞系具有典型的多能干细胞特征,在细胞聚集过程中,在视黄酸(RA)存在的情况下可以分化成神经元,随后在粘附条件下生长出神经质。此外,未分化的P19细胞系还能够在二甲基硫酸盐(DMSO)9、10、11、12的影响下形成肌肉和心肌细胞样细胞。

Protocol
1. 文化维护
- 培养维护介质中的P19细胞系(Dulbeco的改性Eagle培养基,含有4,500毫克/升葡萄糖,辅以10%FBS、100单位/mL青霉素和100单位/mL链霉素)。在37°C和5%CO2下孵育。
2. 亚培养细胞
- 当细胞达到约80%汇合时,从细胞培养瓶中取出废培养基(表面积25cm2)。
- 用不含钙和镁的2 mL磷酸盐缓冲盐水(PBS)清洗细胞。
- 在细胞单层上加入1 mL的0.25%胰蛋白酶-EDTA(乙二胺四乙酸)。
- 将烧瓶放入CO2培养箱(37°C和5%CO2)2-3分钟。
- 评估与烧瓶表面的细胞附件。确保所有单元格都分离并浮动在介质中。
- 加入9 mL的维护介质,以阻止胰蛋白酶的酶活性。
- 在维护介质中重新挂起单元格。
- 将细胞转移到15 mL管和离心机5分钟,在200 x g和室温(RT)。
- 丢弃上清液,在 15 mL 管中加入 10 mL 的新鲜维护介质。
- 使用细胞悬架根据制造商的说明使用细胞计数器确定细胞编号。
- 种子细胞在2 x 104细胞/cm2在新的25厘米2烧瓶。
- 将维护介质添加到 10 mL。
- 将装有细胞的烧瓶放入CO2培养箱(37°C和5%CO2)2-3天。
3. 胰蛋白酶消化
- 从电池瓶中吸气维持介质。用2 mL的钙和无镁PBS清洗细胞一次。
- 添加 1 mL 0.25% 胰蛋白酶-EDTA。
- 将装有细胞的烧瓶在37°C下放入CO2培养箱2-3分钟。
- 使用 1 mL 移液器通过移液细胞分离细胞 10 次。
- 通过在细胞中加入9 mL的分化介质(Dulbeco的改性Eagle的培养基,4500mg/L葡萄糖辅以5%FBS、100单位/mL青霉素和100单位/mL链霉素)来中和胰蛋白酶,而不向细胞中添加RA。
- 将细胞转移到15 mL管和离心机5分钟,在200 x g和RT。

4. 聚合生成
- 将 5 μL RA(1 mM 库存溶解在 99.8% 乙醇中,储存在 -20°C) 添加到 10 mL 的分化介质中,并混合良好(最终浓度为 0.5 μM RA)。
注:RA 是光敏的。EtOH的低浓度不影响细胞分化19,20,21。 - 在 100 mm 未经处理的培养皿(专用于悬浮培养)中添加 10 mL 的分化介质(带 RA)。
- 在 100 mm 盘中播种 1 x 106细胞(碟面面积 56.5 厘米2)。
- 将装有细胞的烧瓶在37°C和5%CO2的培养箱中放置2天,以促进聚集的形成。
- 2天后,交换分化介质。使用 10 mL 移液器的包含集料的吸气介质,在 RT 处转移到 15 mL 管中。
- 允许聚合在 RT 下按重力沉降 1.5 分钟。
- 丢弃上清液。
- 使用 10 mL 血清移液器使用 0.5 μM RA 添加新的 10 mL 分化介质。

5. 聚合分离
- 使用 10 mL 移液器吸气细胞聚集体。
- 将骨料转移到 15 mL 管中。允许细胞聚合按重力沉降 1.5 分钟。
- 拆下上清液。
- 单独用DMEM清洗骨料(无血清和抗生素)。
- 在RT处,允许细胞聚集物通过重力沉降沉降1.5分钟沉降。
- 吸出上清液,加入2 mL的胰蛋白酶-EDTA(0.25%)。
- 将细胞聚集体放入水浴(37°C)10分钟,用手敲击,每2分钟轻轻搅拌一次。
- 通过添加 4 mL 的维护介质,停止胰蛋白酶化过程。
- 移液器使用 1 mL 移液器向上和向下聚合 20 次。
- 离心细胞在200 x g和RT下5分钟。
- 取出上清液,在5 mL的维护介质中重新悬浮细胞颗粒。
- 使用单元格计数器确定单元格编号。
6. 电镀电池
- 将维护介质每孔 3 mL 添加到 6 孔板中。
- 6 孔培养板中的种子细胞密度为 0.5 x 106/孔。
- 在37°C孵育,CO2浓度为5%。
- 将细胞播种在6孔培养板上的盖玻璃上,用抗MAP2抗体(20%汇合)进行免疫染色。使用6孔板分离RNA,并对Map2、NeuN、Oct4、Nanog和Gapdh(20%汇合)执行RT-PCR。

Representative Results
图1给出了P19细胞系神经发生诱导方案的简化方案。为了在未分化状态和神经发生期间定义P19细胞系的特征,采用了RT-PCR(逆转录-聚合酶链反应)方法。未分化的P19细胞系表示多能基因,如有机阳离子/肉碱转运体4(Oct4)和Nanog主源盒(Nanog)。在RA存在的情况下,悬浮培养细胞聚集引起的神经发生导致Oct4和Nanog表达的迅速减少。相反,神经元标记物的表达:微管相关蛋白2(Map2),NeuN(也称为RNA结合蛋白,狐狸-1同源3(Rbfox3))在触发神经发生后增加(图2)6 ,14,15,22.表1中用的每个基因的引源与核苷酸序列和产品大小一起指示。未分化的P19细胞系的微观图像呈现了圆形形态(图3A)。神经发生诱导后,细胞的神经元结构在电镀后4天清晰可见(图3B)。此外,图4表示在分化的P19细胞系(电镀细胞后4天)中MAP2表达的荧光图像14。

Discussion
在这里,我们描述了使用P19细胞系的神经发生的简单方案。虽然在这方面已经发表了许多报告,但使用P19细胞系进行神经发生诱导的详细方法仍不清楚。此外,我们在整个实验中使用了简单的高葡萄糖(4,500mg/L)DMEM培养基,10%FBS。这使我们能够以用户友好的方式进行神经生成实验,并在未来扩展此方法的使用范围。
