This pilot randomised controlled clinical trial aimed to evaluate the feasibility and effectiveness of using a polycaprolactone (PCL) scaffold in fresh extraction sockets for ridge preservation. The hypothesis was that the insertion of a 3D bioresorbable PCL scaffold in fresh extraction sockets allowed for normal bone healing and better maintenance of ridge dimensions after 6 months as compared to extraction sockets without the scaffold.

Despite the powerful impact gene expression markers like the green fluorescent protein (GFP) or enhanced GFP (EGFP) exert on linking the expression of recombinant protein for selection of high producers in recent years, there is still a strong incentive to develop more economical and efficient methods for isolating mammalian cell clones secreting high levels of recombinant proteins. Here we present a new method based on the co-expression of EGFP that allows clonal selection in standard 96-well cell culture plates. The genes encoding the EGFP protein and the related protein are linked by an internal ribosome entry site and thus are transcribed into the same mRNA in an independent translation process. Since both proteins arise from a common mRNA, the EGFP expression level correlates with the expression level of the therapeutic protein in each clone. By expressing recombinant porcine ?-defensin 1 in Marc 145 cells, we demonstrate the robustness and performance of this technique. The method can be served as an alternative to identify high-producer clones with various cell sorting methods.

RSV prophylaxis is not routine in infant born 33 to 35 weeks gestation. Risk scoring tool can be utilized to identify infants that have significant chance for hospitalization. Premature birth is a leading cause of infant mortality and chronic pulmonary morbidity, therefore prevention of RSV hospitalization though immune prophylaxis in late preterm infants appears attractive.

Varicella vaccine was introduced to the infant immunization schedule in each province or territory between 2000 and 2007 as a result of the Canadian Immunization Strategy. The impact of vaccinating children against this disease is potentially far reaching, as immunization may also benefit those segments of the population not immunized. The objective of this paper is to examine the effects of varicella vaccine on related hospitalizations across the entire Canadian population.

Acquired resistance to 5-fluorouracil (5-FU) is one of the important reasons for failure in 5-FU-based chemotherapy. The upregulation of dihydropyrimidine dehydrogenase (DPD) in tumours was reported as an important factor for acquired 5-FU resistance. Genetic deficiency of adenosine deaminase (ADA) can cause profound lymphopenia and result in the clinical presentation of severe combined immune deficiency (SCID). However, because of the ubiquitous expression of ADA, ADA-deficient patients often present also with nonimmunologic clinical problems, affecting the skeletal, central nervous, endocrine, and gastrointestinal systems. We now report that myeloid dysplasia features and bone marrow hypocellularity are often found in patients with ADA-SCID. As a clinical correlate to this finding, we have observed vulnerability to antibiotic-induced myelotoxicity and prolonged neutropenia after nonmyeloablative chemotherapy. We have also noted that, in the absence of enzyme replacement therapy, absolute neutrophil counts of patients with ADA deficiency vary inversely with the accumulation of deoxynucleotides. These data have significant implications for the application of standard and investigational therapies to patients with ADA-SCID and support further studies to investigate the possibility that ADA deficiency is associated with a stem cell defect. Antiangiogenic drugs inhibit tumor growth by decreasing blood supply and causing transient "normalization" of the tumor vasculature, thereby improving the delivery of systemic chemotherapy. A higher dose of antiangiogenic drugs may lead to a more marked decrease in intratumoral blood flow but may concomitantly cause a decrease in delivery of chemotherapeutic agents. The purpose of this study was to define an optimal schedule for the combination of gemcitabine with a recombinant endostatin, endostar.

In this study, we used two-dimensional gel electrophoresis and MALDI-Q-TOF-MS/MS analysis to examine the global protein expression of a pair of colorectal carcinoma cell lines, SW620 and irinotecan-resistant SW620. Of the 30 spots identified as differentially expressed proteins (±over twofold, P<0.05) between the two cell lines, 26 spots (corresponding to 26 unique proteins) were positively identified by MALDI-Q-TOF-MS/MS analysis. These proteins could be grouped into main classes including metabolism (15.38%), cell SSproliferation/differentiation (11.53%), molecular chaperone (11.53%), mRNA splicing (11.53%), and so on. The proteins, which might be involved in the development of tumor drug resistance, such as ?-enolase, cofilin, and thioredoxin-dependent peroxide 1, have been validated by western blot analysis and have been discussed. The proteins identified in this study may be useful in showing the mechanisms underlying irinotecan resistance.

Cytokines play important roles in regulating immune responses. Interleukin-2 (IL-2) has usually been used as an adjuvant to enhance antitumour immune responses. However, its crucial role in activation-induced cell death, inhibition of homeostatic proliferation of CD8+ memory T cells and its notable biological side effects impair its prospect of application. IL-15 has several similar functions to IL-2 and shows potential advantages over IL-2, and is being investigated to enhance antitumour dendritic cell (DC) vaccine strategies in our ongoing studies.

Sunitinib, a novel oral multi-targeted tyrosine kinase inhibitor for patients with metastatic renal cell carcinoma (mRCC) and advanced gastrointestinal stromal tumor, has a good prospect for clinical application and is being investigated for the potential therapy of other tumors. We observed the phenomenon that drinking tea interfered with symptom control in an mRCC patient treated with sunitinib and speculated that green tea or its components might interact with sunitinib. This study was performed to investigate whether epigallocatechin-3-gallate (EGCG), the major constituent of green tea, interacted with sunitinib. The interaction between EGCG and sunitinib was examined in vitro and in vivo. (1)H nuclear magnetic resonance ((1)H-NMR) spectroscopy and mass spectrometry (MS) were used to analyze the interaction between these two molecules and whether a new compound was formed. Solutions of sunitinib and EGCG were intragastrically administered to rats to investigate whether the plasma concentrations of sunitinib were affected by EGCG. In this study, we noticed that a precipitate was formed when the solutions of sunitinib and EGCG were mixed under both neutral and acidic conditions. (1)H-NMR spectra indicated an interaction between EGCG and sunitinib, but no new compound was observed by MS. Sticky semisolid contents were found in the stomachs of sunitinib and EGCG co-administrated mice. The AUC(0-?) and C (max) of plasma sunitinib were markedly reduced by co-administration of EGCG to rats. Our study firstly showed that EGCG interacted with sunitinib and reduced the bioavailability of sunitinib. This finding has significant practical implications for tea-drinking habit during sunitinib administration.

Skeletal muscle loss represents an established key feature yet there is no molecular understanding of the disease process. In fact, the postulated molecular regulators of cancer cachexia originate largely from pre-clinical models and it is unclear how these translate to the clinical environment.

An investigation was made on the soil physical and chemical properties in different-aged Pinus massoniana plantations in Three Gorges Reservoir Area under effects of litterfall and roots. The annual litter production in mature stand was 19.4% and 65.7% higher than that in nearly mature and middle-aged stands, respectively. The litter standing amount was in the sequence of mature stand > middle-aged stand > nearly mature stand, while the litter turnover coefficient was in the order of nearly mature stand (0.51) > mature stand (0.40) > middle-aged stand (0.36). The total root biomass, live root biomass, and dead root biomass were the highest in middle-aged stand, and the lowest in nearly mature stand. In middle-aged stand, soil total porosity was the highest, and soil bulk density was the lowest. Soil organic matter and total nitrogen contents were in the order of mature stand > middle-aged stand > nearly mature stand, soil nitrate nitrogen occupied a larger proportion of soil mineral N in nearly mature stand, while ammonium nitrogen accounted more in middle-aged and mature stands. In nearly mature stand, litter production was moderate but turnover coefficient was the highest, and soil nutrient contents were the lowest. In middle-aged stand, root biomass and soil total porosity were the highest, and soil bulk density were the lowest. In mature stand, root biomass was lower while soil nutrient contents were the highest. The increase of root biomass could improve soil physical properties.

The 12 Immunization Monitoring Program, Active (IMPACT) centers that represent 90% of pediatric tertiary care beds in Canada conducted active surveillance for varicella-related hospitalizations and complications from 1999 onward, after varicella vaccine was authorized. Publicly funded routine immunization programs at 12 or 15 months of age were introduced by 5 provinces and territories (prov/terr) in 2000 to 2002 (earlier programs, EP) and by 8 prov/terr in 2004 to 2007 (later programs, LP).

Based on the measurements of soil nutrient contents and enzyme activities and the canonical correspondence analysis (CCA), this paper studied the relationships between soil nutrient contents and soil enzyme activities in different age Pinus massoniana stands in Three Gorges Reservoir Area. Among the test stands, mature stand had the highest contents of organic matter, total nitrogen, ammonium nitrogen, and available phosphorus in 0-20 cm soil layer, followed by middle-aged stand, and nearly-mature stand. With the increase of the stand age, soil invertase activity increased after an initial decrease, cellulase and polyphenoloxidase activities decreased gradually, while urease and peroxidase activities decreased after an initial increase. CCA analysis showed that the effects of the main soil parameters on the soil enzyme activities in the stands ranked in the sequence of total nitrogen > organic matter > pH > bulk density > ammonium nitrogen > available phosphorus. Soil invertase activity had significant positive correlations with soil organic matter, total nitrogen, and total phosphorus, while soil peroxidase activity significantly negatively correlated with soil organic matter, total nitrogen, total phosphorus, and bulk density. The soil was rich in main nutrients, invertase activity was relatively high, while peroxidase activity was relatively low. The activities of soil invertase, cellulase and peroxidase could be used as the good biological indicators in evaluating soil quality and fertility.

Appetite loss has a major impact on cancer patients. It is exceedingly prevalent, is a prognostic indicator and is associated with inferior quality of life. Cachexia is a multi-factorial syndrome defined by a negative protein and energy balance, driven by a variable combination of reduced food intake and abnormal metabolism. Not all cancer patients that experience weight loss have appetite loss, and the pathophysiology between cachexia and appetite loss may thus be different. Knowledge of pathophysiology of appetite loss in cancer patients is still limited. The primary object of this study was to explore the association with 93 predefined candidate single-nucleotide polymorphisms (SNPs) and appetite loss in cancer patients to possibly generate new theories of the pathophysiology of the condition.

In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
