This study aimed to analyze the etiology of the encephalitis outbreak in Longyan, Fujian Province, China in 2010, in order to provide valuable information for this prevention and control of this disease. Pathogens were confirmed from cerebrospinal fluid samples with fluorescent RT-PCR, virus isolation (RD cells), and neutralization tests. Then, the VP1 fragments or whole genome nucleotide sequences were determined for four virus strains using PCR. Homology was assessed using the MegAlign software, and a phylogenetic evolutionary tree was drawn using Mega 4.0 software. The results confirmed that the etiology of the outbreak was the ECHO6 intestinal virus, and the nucleotide sequence of the VP1 segment indicated that the C2 subtype was responsible. The genome sequence consisted of 7407 nucleotides, and resembled the genome of other ECHO and CoxB viruses with homology levels of 78.5%-87.3%. The encephalitis outbreak in Longyan in 2010 was caused by the ECHO6 C2 subtype intestinal virus, and its complete genome sequence length is similar to the standard strain (U16283) with a sequence homology of 80.4%.

[Effect of SDS on the adsorption of Cd2+ onto amphoteric modified bentonites].

Under different modified ratios, temperatures, pH and ionic strengths, the effect of sodium dodecyl sulfonate (SDS) on the adsorption of Cd2+ onto bentonites which modified with amphoteric modifier dodecyl dimethyl betaine (BS-12) was studied by batch experiments, and the adsorption mechanism was also discussed. Results showed that the adsorption of Cd2+ on amphoteric bentonites can be enhanced significantly by SDS combined modification, Cd2+ adsorption decreases in the order: BS + 150SDS (BS-12 + 150% SDS) > BS + 100SDS (BS-12 + 100% SDS) > BS +50SDS(BS-12 + 50% SDS) > BS + 25SDS (BS-12 + 25% SDS) > BS (BS-12) > CK (unmodified soil). The adsorption isotherm can be described by the Langmuir equation. The change of temperature effect from positive on CK and amphoteric bentonites to negative on BS + 150SDS bentonites is observed with an increase of SDS modified ratio. The pH has little influence on Cd2+ adsorption on bentonites. The adsorption of Cd2+ on bentonites decreases with ionic strength rise, but the effect of ionic strength can be reduced with an increase of SDS modified ratio also. The adsorption thermodynamic parameters demonstrated that the adsorption of Cd2+ on modified bentonites was spontaneously controlled by entropy increment. When the SDS modified ratio is lower than 100% CEC, the adsorption of Cd2+ on modified bentonites is a process with characteristics of both enthalpy increment and entropy increment, while the SDS modified ratio is equal to or higher than 100% CEC, the adsorption of Cd2+ on modified bentonites becomes a process of enthalpy decrement and entropy increment.

To study the effect of urea nitrogen on the ecosystem of Lake Taihu, we conducted urea and various nitrogen analysis for the water samples collected from the lake and surrounding rivers during summer. The ecological index analysis of 82 sites in rivers and lake yielded the following results: (1) The urea nitrogen contents in Taihu ranged from 0.011 to 0.161 mg x L(-1), which was high in the northwest and low in the southeast, related to the main pollution sources distribution of its drainage basin. (2) The dissolved nitrogen was dominated by inorganic nitrogen and the ratio between ammonia nitrogen and nitrate nitrogen was 5: 1. The average percentage of urea nitrogen in total nitrogen, dissolved nitrogen, dissolved organic nitrogen and bioavailable nitrogen was respectively 2.28%, 5.91%, 15.86%, and 6.22%, which showed a significant ecological function in Taihu. (3) Urea nitrogen concentration in river was more than twice that in lake, and the lake river concentration was slightly higher than the river into the lake. (3) In Taihu, there was a transformation relationship between urea nitrogen and the nitrogen in other forms. It showed that urea nitrogen had a significant positive correlation with permanganate index and the other forms of nitrogen, and a significant negative correlation with dissolved oxygen. In addition, urea nitrogen was weakly and positively correlated with chlorophyll a, while closely related to the spatial distribution of benthos and zooplankton species. All the results above showed that urea nitrogen was the bridge of organic and inorganic nitrogen transformation, and was the sign of nitrogen cycle of Lake Taihu, which was controlled by the circulating rate. High nitrogen content (especially the organic nitrogen) and low dissolved oxygen content were the key contributors to the increased urea nitrogen content. In Taihu, the urea nitrogen content was affected by both exogenous input and endogenous release.

Aberrant Aur-A signaling is associated with tumor malignant behaviors. However, its involvement in tumor metabolic stress is not fully elucidated. In the present study, prolonged nutrient deprivation was conducted into breast cancer cells to mimic metabolic stress in tumors. In these cells, autophagy was induced, leading to caspase-independent cell death, which was blocked by either targeted knockdown of autophagic gene ATG5 or autophagy inhibitor 3-Methyladenine (3-MA). Aur-A overexpression mediated resistance to autophagic cell death and promoted breast cancer cells survival when exposed to metabolic stress. Moreover, we provided evidence that Aur-A suppressed autophagy in a kinase-dependent manner. Furthermore, we revealed that Aur-A overexpression enhanced the mammalian target of rapamycin (mTOR) activity under metabolic stress by inhibiting glycogen synthase kinase 3? (GSK3?). Inhibition of mTOR activity by rapamycin sensitized Aur-A-overexpressed breast cancer cells to metabolic stress-induced cell death. Consistently, we presented an inverse correlation between Aur-A expression (high) and autophagic levels (low) in clinical breast cancer samples. In conclusion, our data provided a novel insight into the cyto-protective role of Aur-A against metabolic stress by suppressing autophagic cell death, which might help to develop alternative cell death avenues for breast cancer therapy.

MicroRNA-195 (miR-195) is a tumor suppressor that plays an important role in tumorigenesis. There are few studies on miR-195 expression in breast cancer patients and the results have been inconsistent; therefore, this study examined miR-195 expression in the serum of BC patients. Samples from 102 normal subjects and 210 subjects with BC who had detailed clinical follow-up information available were selected. An internal reference (miR-16) and serum miR-195 were amplified and quantitatively detected by SYBR green-based real-time RT-PCR. We analyzed the differences in miR-195 levels between BC and healthy cases and the relationships between the miR-195 level and TNM stage and other clinicopathological parameters. In addition, changes in miR-195 levels were examined for 21 BC cases using paired samples before and after neoadjuvant chemotherapy. miR-195 was downregulated in BC compared with control samples (P = 0.000, Mann-Whitney U test). The sensitivity and specificity of miR-195 in the diagnosis of BC were 69.0 and 89.2 %, respectively; whereas, the sensitivities of carcinoembryonic antigen (CEA) and carbohydrate antigen 153 (CA153) were only 15.08 and 21.1 %, respectively. Remarkably, serum miR-195 had higher sensitivity, 73.97 % (108/146), as a tumor marker in the diagnosis of early stage BC [ductal carcinoma in situ, tumor-node-metastasis (TNM) I, II] compared with the conventional tumor markers CA153 and CEA (12.41 and 7.59 %). Moreover, compared with CEA and CA153, miR-195 had a higher sensitivity for detecting the response to neoadjuvant chemotherapy and significantly increased, more than twofold, after neoadjuvant chemotherapy (P = 0.025, paired t test) in 52.381 % (11/21) of BC cases. However, there were no significant relationships between miR-195 expression and other clinicopathological parameters (TNM stage/pathotype/ER/PR/lymph node status). Our data indicate serum miR-195 is a promising tumor marker for BC diagnosis and general screening, especially for early stage BC. The high sensitivity of miR-195 to neoadjuvant chemotherapy may lay the foundation for future studies on the use of miRNA-based methods for monitoring BC treatment and therapy.

Male infertility microsurgery represents the fastest growing sub-specialty in urology and clinical andrology over the past two decades. The importance of microsurgery for male infertility has risen as a part of the urologist's armamentarium in the medical and surgical management of male infertility. Despite the advances in male infertility microsurgery in China, the lack of standardized and well-organized training programs for male infertility microsurgery remains a serious problem affecting its development. In this article, Zhao and Peng have shared their experience with the learning curve of male infertility microsurgery at the Center for Male Reproductive Medicine and Microsurgery, Weill Medical College of Cornell University, which centers on how to pay attention to the details and basic principles of microsurgery. Male infertility microsurgery is physically, technically and mentally challenging, and must be first learned in the laboratory. Clinical success depends heavily upon appropriate training in a microsurgical laboratory. Good training can significantly reduce operation time and surgical errors as well as improve the quality of outcomes.

Male infertility is a common and complex disease in urology and andrology, and for many years there has been no effective surgical treatment. With the emergence of microsurgery and assisted reproductive medicine (IVF/ICSI), rapid development has been achieved in the treatment of male infertility. The Center for Male Reproductive Medicine and Microsurgery at Weill Cornell Medical College of Cornell University has been playing an important leading role in developing microsurgical techniques for the management of male infertility. The development of microsurgical treatment of male infertility in China has experienced the 3 periods of emerging, making, and boosting ever since its systematic introduction from Weill Cornell Medical College 15 years ago. At present, many Chinese hospitals have adopted microsurgery in the management of male infertility, which has contributed to the initial establishment of a microsurgical treatment system for male infertility in China. However, some deficiencies do exist concerning microsurgical treatment of male infertility, as in normalized technical training programs for competent surgeons, unified criteria for evaluation of surgical outcomes, and detailed postoperative follow-up data. This article presents an overview on the 15-year development of microsurgical management of male infertility in China, points out the existing deficiencies, and offers some propositions for the promotion of its development.

The information about the crystal structure of porcine reproductive and respiratory syndrome virus (PRRSV) leader protease nsp1? is available to analyze the roles of tRNA abundance of pigs and codon usage of the nsp1 ? gene in the formation of this protease. The effects of tRNA abundance of the pigs and the synonymous codon usage and the context-dependent codon bias (CDCB) of the nsp1 ? on shaping the specific folding units (?-helix, ?-strand, and the coil) in the nsp1? were analyzed based on the structural information about this protease from protein data bank (PDB: 3IFU) and the nsp1 ? of the 191 PRRSV strains. By mapping the overall tRNA abundance along the nsp1 ?, we found that there is no link between the fluctuation of the overall tRNA abundance and the specific folding units in the nsp1?, and the low translation speed of ribosome caused by the tRNA abundance exists in the nsp1 ?. The strong correlation between some synonymous codon usage and the specific folding units in the nsp1? was found, and the phenomenon of CDCB exists in the specific folding units of the nsp1?. These findings provide an insight into the roles of the synonymous codon usage and CDCB in the formation of PRRSV nsp1? structure.

[Sensitivity analysis of AnnAGNPS model's hydrology and water quality parameters based on the perturbation analysis method].

Sensitivity analysis of hydrology and water quality parameters has a great significance for integrated model's construction and application. Based on AnnAGNPS model's mechanism, terrain, hydrology and meteorology, field management, soil and other four major categories of 31 parameters were selected for the sensitivity analysis in Zhongtian river watershed which is a typical small watershed of hilly region in the Taihu Lake, and then used the perturbation method to evaluate the sensitivity of the parameters to the model's simulation results. The results showed that: in the 11 terrain parameters, LS was sensitive to all the model results, RMN, RS and RVC were generally sensitive and less sensitive to the output of sediment but insensitive to the remaining results. For hydrometeorological parameters, CN was more sensitive to runoff and sediment and relatively sensitive for the rest results. In field management, fertilizer and vegetation parameters, CCC, CRM and RR were less sensitive to sediment and particulate pollutants, the six fertilizer parameters (FR, FD, FID, FOD, FIP, FOP) were particularly sensitive for nitrogen and phosphorus nutrients. For soil parameters, K is quite sensitive to all the results except the runoff, the four parameters of the soil's nitrogen and phosphorus ratio (SONR, SINR, SOPR, SIPR) were less sensitive to the corresponding results. The simulation and verification results of runoff in Zhongtian watershed show a good accuracy with the deviation less than 10% during 2005- 2010. Research results have a direct reference value on AnnAGNPS model's parameter selection and calibration adjustment. The runoff simulation results of the study area also proved that the sensitivity analysis was practicable to the parameter's adjustment and showed the adaptability to the hydrology simulation in the Taihu Lake basin's hilly region and provide reference for the model's promotion in China.

[Comparative metabonomics study on urine in rat treated by Angelica sinensis volatile oil].

Metabonomics was employed to investigate the effect of Angelica sinensis volatile oil (ASVO) to the endogenous metabolites of normal rats, and to reveal the possible ways of metabolism in rats caused by ASVO. The fifty male Waster rats were randomly divided into five groups (each consists of 10 rats), such as control group, high dose group of ASVO, middle dose group of ASVO, low dose group of ASVO, and Aspirin group. They were given 0.9% saline, 0.352 mL x kg(-1) ASVO, 0.176 mL x kg(-1) ASVO, 0.088 mL x kg(-1) ASVO and ASP respectively with the equal volume of 0.2 mL. Drugs and vehicle were given for 3 successive days. The urine was collected at 12, 24, 36, 48 h after modeling with metabolic cages. Rat urine metabolic fingerprint in different stages was analyzed using GC-MS, based on which the principal component analysis (PCA)and orthogonal partial least-squares discriminant analysis (OPLS-DA) models were established for metabonomic analysis. Potential biomarkers were screened by using variable importance in the projection (VIP) and T test. It was revealed that the middle dose of ASVO at 36 h induces a substantial change in rat urine. Compared with control group, seven kinds of endogenous metabolites in ASP group and ASVO group change significantly (P < 0.05), among which aconitic acid, succinic acid, citric acid, alpha-ketone glutaric acid, glycine and malic acid content had an upward trend (P < 0.05) and prostaglandin content had a downward trend (P < 0.01). The mechanism of ASVO and ASP have the similarity. It is likely that ASVO intervenes the metabolic process by affecting the energy, amino acid and lipid metabolism. Our work also indicates that rats administrated with ASVO can increase the energy metabolism of the body, induce the production of inflammatory substances and strengthen the body's immune ability. The result has also provide a proof for futher interpret ASVO pharmacological effects.

miRNA expression in African American compared to Caucasian PCa patients has not been widely explored. Herein, we probed the miRNA expression profile of novel AA and CA derived prostate cancer cell lines. We found a unique miRNA signature associated with AA cell lines, independent of tumor status. Evaluation of the most differentially expressed miRNAs showed that miR-132, miR-367b, miR-410, and miR-152 were decreased in more aggressive cells, and this was reversed after treatment of the cells with 5-aza-2'-deoxycytidine. Sequencing of the miR-152 promoter confirmed that it was highly methylated. Ectopic expression of miR-152 resulted in decreased growth, migration, and invasion. Informatics analysis of a large patient cohort showed that decreased miR-152 expression correlated with increased metastasis and a decrease in biochemical recurrence free survival. Analysis of 39 prostate cancer tissues with matched controls (20 AA and 19 CA), showed that 50% of AA patients had statistically significant lower miR-152 expression compared to only 35% of CA patients. Ectopic expression of miR-152 in LNCaP, PC-3, and MDA-PCa-2b cells down-regulated DNA (cytosine-5)-methyltransferase 1 (DNMT1) through direct binding in the DNMT1 3'UTR. There appeared to be a reciprocal regulatory relationship of miR-152/DNMT1 expression, as cells treated with siRNA DNMT1 caused miR-152 to be re-expressed in all cell lines. In summary, these results demonstrate that epigenetic regulation of miR-152/DNMT1 may play an important role in multiple events that contribute to the aggressiveness of PCa tumors, with an emphasis on AA PCa patients .

Sirtuin 1 (SIRT1) regulates numerous neuronal processes, including metabolism, antioxidation and aging, through activation of peroxisome proliferator-activated receptor coactivator 1-? (PGC-1?), an upstream regulator of mitochondrial biogenesis and function. However, the role of SIRT1 in the oxidative stress induced by seizures has yet to be elucidated. The present study aimed to investigate whether SIRT1 was involved in the activation of the PGC-1?/mitochondrial antioxidant system following status epilepticus (SE) in rats. The data demonstrated that SIRT1 expression and activity were enhanced in the rat hippocampus following SE. SIRT1 inhibition effectively blocked the SE-associated increase in PGC-1? and mitochondrial antioxidant enzymes, including superoxide dismutase 2 (SOD2) and uncoupling protein 2 (UCP2). Additionally, it was also demonstrated that the activation of SIRT1 enhanced mitochondrial electron transport chain complex I activity and increased ATP content. In conclusion, the present results suggest that SIRT1 activation may alleviate mitochondrial oxidative stress induced by seizures partially via PGC-1? signaling.

Some published evidence has revealed that the dendritic cells can interact with pathogens that exist in the inner foreskin. This information provides a new vision that pathogens could play a role through the redundant prepuce; numerous studies have failed to find pathogens in prostates of patients who had chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS). However, no studies have reported an association between foreskin length and CP/CPPS. Hence, we conducted a retrospective case-control study of clinical data from 322 CP/CPPS patients (case group) and 341 nonCP/CPPS patients (control group). Demographic characteristics, lifestyle factors, and foreskin lengths were collected and analyzed. Multivariate logistic regression was adopted to calculate the odds of foreskin length for CP/CPPS. According to the multivariate logistic regression results, when the foreskin length covered up more than half of the glans penis, the odds for CP/CPPS were higher with an increased foreskin (odds ratio (OR): 1.66, 95% confidence interval (CI): 1.04-2.66). In comparison, when the glans penis was completely covered by the foreskin, the OR value increased to 1.86 (95% CI, 1.2-2.88). The study results showed an association between foreskin length and the odds of CP/CPPS. When the foreskin length covered up more than half of the glans penis, there were greater odds for CP/CPPS. This possible mechanism might result from interaction between pathogens and DCs in the inner foreskin, consequently activating T-cells to mediate allergic inflammation in the prostate and producing the autoimmunizations causing CP/CPPS.

This meta-analysis aims to evaluate the relationships between serum vascular endothelial growth factor (VEGF) level and radiosensitivity in patients with nonsmall cell lung cancer (NSCLC) among Asians. We searched CISCOM, CINAHL, Web of Science, PubMed, Google Scholar, EBSCO, Cochrane Library, and CBM databases from their inception through October 1, 2013. Meta-analysis was performed using the STATA 12.0 software. Fourteen clinical studies were included in this meta-analysis, including five case-control studies and nine cohort studies. Our meta-analysis results revealed that levels of serum VEGF in NSCLC patients were higher than that of healthy controls. There was a significant difference in serum VEGF levels between before and after radiotherapy in NSCLC patients. Further, we found significant differences in serum VEGF levels between effective and noneffective clinical response groups pre- and postradiotherapy. Serum VEGF levels showed no significant associations with tumor-node-metastasis (TNM) stage and histologic grade in NSCLC patients. NSCLC patients with positive VEGF expression had shorter overall survival than those with negative VEGF expression. Our meta-analysis suggests that serum VEGF level may be a useful biomarker in predicting radiosensitivity and prognosis of NSCLC patients among Asians.

Influenza A virus has a wide range of hosts. It has not only infected human, but also been reported interspecies transmission from humans to other animals, such as pigs, poultry, dogs and cats. However, prevalence of A (H1N1) pdm09 influenza virus infections in cats in northeastern China is unknown. Therefore, the prevalence of A (H1N1) pdm09 influenza virus infections was performed among cats in northeastern China in this study.

From January to July 2012, we looked for evidence of subclinical A (H1N1) pdm09 and seasonal human influenza viruses infections in healthy dogs in China. Sera from a total of 1920 dogs were collected from Guangdong, Guangxi, Fujian and Jiangxi provinces. We also examined archived sera from 66 dogs and cats that were collected during 2008 from these provinces. Using hemagglutination inhibition (HI) and microneutralization (MN) assays, we found that only the dogs sampled in 2012 had elevated antibodies (?1:32) against A(H1N1)pdm09 virus and seasonal human influenza viruses: Of the 1920 dog sera, 20.5 % (n = 393) had elevated antibodies against influenza A(H1N1) pdm09 by the HI assay, 1.1 % (n = 22), and 4.7 % (n = 91) of the 1920 dogs sera had elevated antibodies against human seasonal H1N1 influenza virus and human seasonal H3N2 influenza virus by the HI assay. Compared with dogs that were raised on farms, dogs that were raised as pets were more likely to have elevated antibodies against A(H1N1)pdm09 and seasonal human influenza viruses. Seropositivity was highest among pet dogs, which likely had more diverse and frequent exposures to humans than farm dogs. These findings will help us better understand which influenza A viruses are present in dogs and will contribute to the prevention and control of influenza A virus. Moreover, further in-depth study is necessary for us to understand what roles dogs play in the ecology of influenza A.

Temporal lobe epilepsy is characterized by spontaneous recurrent seizures (SRS) and associated with behavioral problems. However, the molecular mechanisms underlying these problems are not yet clear. In this study, kainic acid (KA) was systemically administered to immature male Wistar rats to induce SRS. The behavior of the immature rats was evaluated with a water maze, elevated-plus mazes, and open field tests. The expression patterns of synaptophysin, SNAP-25, and synaptotagmin 1 (Syt 1) were examined by reverse-transcriptase polymerase chain reaction (RT-PCR) and Western blot analysis. KA-treated rats with SRS demonstrated learning and memory deficits, reduced anxiety, and increased locomotor activity, compared with placebo-treated rats and KA-treated rats without SRS. No neuronal cell loss was observed in the hippocampus 6 weeks after exposure to KA. However, RT-PCR and Western blot analyses revealed decreased synaptophysin, SNAP-25, and Syt 1 expression in KA-treated rats with SRS. Synaptophysin, SNAP-25, and Syt1 expression levels were found to be positively correlated with learning and memory but negatively correlated with anxiety and locomotor activity. These data suggested that SRS may induce changes in synaptophysin, SNAP-25, and Syt1 expression and may be functionally related to SRS-induced behavioral deficits.

Normal mammalian terminal erythroid differentiation is a precisely regulated process during which the progenitor cells execute particular programs to form a mature erythrocytic phenotype. In the present study, it was found that RbAp48, a histone-binding protein associated with retinoblastoma protein, was upregulated during terminal erythroid maturation in vivo and in vitro. This indicated that RbAp48, at least in part, participated in the regulation of murine erythropoiesis. Following sodium butyrate (SB) induction, murine erythroleukemia (MEL) cells began to re-enter erythroid differentiation and the ratio of differentiated cells reached ~80% at 72 h. The erythroid maturation-related mRNA expression of ?-globin, ?-globin and glycophorin A (GPA) was increased markedly, which indicated that SB induced MEL differentiation. During MEL differentiation, the RbAp48 level showed a 1.5-fold increase at 72 h, and the globin transcription factor (GATA)-1 level was also upregulated in the early stage of differentiation. By contrast, the c-Myc level was gradually downregulated in MEL differentiation. Using an immunofluorescence assay, the results of the study directly showed that the average fluorescence intensity of RbAp48 in each cell reached an almost 1.7-fold increase at 72 and 96 h. This was consistent with the western blot results of RbAp48 during MEL differentiation. In addition, reduced expression of RbAp48 by RNA inference decreased SB-induced MEL differentiation by ~20%, indicating that a high level of RbAp48 was essential for MEL differentiation. Taken together, these results established a functional link between RbAp48 and erythroid differentiation.

Men with azoospermia or severe oligospermia (< 5 x 10(6)/ml) should have genetic testing to identify the reason for male infertility before treatment. Identification of obstructive azoospermia (OA) or non-obstructive azoospermia (NOA) is essential because genetic testing differs for OA (which has normal testicular function, testicular volume, and FSH) versus NOA (which has small, soft testes and increased FSH). Among patients with NOA, history and physical examination along with laboratory testing is required to choose genetic testing specifically for primary testicular failure or congenital hypogonadotropic hypogonadism (HH). Genetic testing options include cystic fibrosis transmembrane conductance regulator (CFTR) testing for men with OA due to congenital absence of the vas, while karyotype, Y chromosome microdeletions (YCMD), and other specific genetic tests may be indicated if patient has severe oligospermia or NOA. These genetic tests help to identify which patients may benefit from medical and/or surgical intervention. The most recent techniques for genetic analysis will improve diagnosis and management of male infertility.

Environmental problems as well as their related ecosystem stress and human health risk in China have raised wide concerns along with the rapid economic development in recent years. Numerous studies with a sharp increase in publication number have addressed the ubiquitous of anthropogenic chemicals in various environmental compartments and human tissues. However, very few data were available to clarify the temporal trend and to give the retrospective analysis of chemical pollution in China. Environmental Specimen Bank (ESB) is a system for the systematic collection and long-term storage of specimens, which has been established since the 1970s in developed counties and recognized as a fundamental complement for environmental monitoring and scientific research. Currently, the value of ESB is becoming more broadly recognized globally, and China is still at the early stage. This article described the history and status and put forwarded the future key points of Chinese ESB development for illustrating the intensive environmental changes in China and the world.

The insular cortex is located deep within the Sylvian fissure between multi-functional and structurally-compressed cerebral structures, and has been suggested to play an important role in both basic sensorimotor and complex social-emotional functions. Such structural and functional complexity presents a challenge for neurosurgeons to remove tumors within the insula safely. It has therefore not yet been documented how neurosurgical resection of insular gliomas would impact social-emotional functions. In this study, we examined empathy, a high-level social-emotional function, in four patients with localized insular gliomas pre- and post-operatively. The patients completed an empathy-for others pain task in which they viewed another person's hand or foot in painful or non-painful situations and made judgments about either pain (explicit empathy) or laterality of the hand or foot (implicit empathy). They also completed questionnaires assessing general emotional processing and personality. Deficits in both explicit and implicit empathetic pain processing were found in patients before the operations. However, the operations significantly improved their empathetic ability after surgery, accompanied by unchanged personality traits. These results confirmed previous findings that the insula plays a critical role for empathetic pain perception. Importantly, the current results suggest that surgical resection is not only a suitable treatment for insular gliomas for clinical consideration, but also effective in improving high-level functions such as empathetic pain perception.

The present study aimed to investigate the biological functions of miR-96 in the processes of proliferation and clonogenicity in the prostate cancer cells. miR-96 was identified to be markedly up-regulated in prostate cancer cell and cancer tissues compared with normal prostate cell and normal prostate tissues by microarray method and RT-PCR analysis. Down-regulation of miR-96 expression reduced the proliferation and colony formation ability of PC3 prostate cancer cells, while over-expression of miR-96 induced proliferation and colony formation ability of LNCaP prostate cancer cells. Forkhead box protein O1 (FOXO1) is key tumor suppressors and has been shown to play key roles in the regulation of diverse cellular processes, including cell proliferation, differentiation, cell cycle progression and apoptosis. The expression level of FOXO1 was strikingly up-regulated in PC3 cells after transfected with miR-96 inhibitor, and FOXO1 expression was down-regulated in LNCaP cells after transfected with miR-96 mimics. miR-96 may play a vital role in promoting cell proliferation in human prostate cancer cells. Inhibition of miR-96 caused expression increase of tumor suppressor gene FOXO1, thus manipulating miR-96 expression may be a promising approach in treatment of prostate cancer.

This study aimed to evaluate the relationship of serum levels of vascular endothelial growth factor (VEGF) and transforming growth factor-?1 (TGF-?1) with radiosensitivity of elderly patients with unresectable non-small cell lung cancer (NSCLC) receiving three-dimensional conformal radiation therapy (3D-CRT). Fifty-eight elderly patients with unresectable NSCLC and 40 healthy controls were enrolled in this study. The deprivation of sensory input after hearing damage results in functional reorganization of the brain including cross-modal plasticity in the sensory cortex and changes in cognitive processing. However, it remains unclear whether partial deprivation from unilateral auditory loss (UHL) would similarly affect the neural circuitry of cognitive processes in addition to the functional organization of sensory cortex. Here, we used resting-state functional magnetic resonance imaging to investigate intrinsic activity in 34 participants with UHL from acoustic neuroma in comparison with 22 matched normal controls. In sensory regions, we found decreased regional homogeneity (ReHo) in the bilateral calcarine cortices in UHL. However, there was an increase of ReHo in the right anterior insular cortex (rAI), the key node of cognitive control network (CCN) and multimodal sensory integration, as well as in the left parahippocampal cortex (lPHC), a key node in the default mode network (DMN). Moreover, seed-based resting-state functional connectivity analysis showed an enhanced relationship between rAI and several key regions of the DMN. Meanwhile, lPHC showed more negative relationship with components in the CCN and greater positive relationship in the DMN. Such reorganizations of functional connectivity within the DMN and between the DMN and CCN were confirmed by a graph theory analysis. These results suggest that unilateral sensory input damage not only alters the activity of the sensory areas but also reshapes the regional and circuit functional organization of the cognitive control network.

Since 2006, more and more cases of the infectious H3N2 canine influenza virus (CIV) in pet dogs have been reported in Southern China. However, little is known about the prevalence situation of H3N2 CIV infections in farmed dogs in China. This is the first systematic epidemiological surveillance of CIV in different dog populations in Southern China. Two virus strains A/Canine/Guangdong/1/2011(H3N2) and A/canine/Guangdong/5/2011(H3N2) were isolated from canine nasal swabs collected at one dog farm in Guangzhou and the other farm in Shenzhen. Sequence and phylogenetic analysis of eight gene segments of these viruses revealed that they were most similar to the newly isolated canine H3N2 viruses in dogs and cats from Korea and China, which originated from avian strain. This indicates that H3N2 CIV may be a common pathogen for pet and farmed dog populations in Southern China at present. Serological surveillance has shown that the infection rate of this avian-origin canine influenza in farmed dogs and in pet dogs were 12.22% and 5.3%, respectively; as determined by the ELISA. The data also suggested that transmission occurred, most probably by close contact, between H3N2 CIV infected dogs in different dog populations in recently years. As H3N2 outbreaks among dogs continue in the Guangdong Province (located very close to Hong Kong), the areas where is densely populated and with frequent animal trade, there is a continued risk for pet H3N2 CIV infections and for mutations or genetic reassortment leading to new virus strains with increased transmissibility among dogs. Further in-depth study is required as the H3N2 CIV has been established in different dog populations and posed potential threat to public health.

In order to study the effects of biochar on the release of pollutants from sediment, Arundo donax, Phragmites australis, Arachis hypogaea and Zea mays were pyrolyzed into biochar. Biochar was used to cover the contaminated sediment in the simulated reactors. Concentrations of NH4(+) -N, NO3(-) -N, NO2(-) -N, COD and P4(3-) -P were continuously monitored, and the cumulative release amount and release rate were calculated. Besides that, the release amount of water-soluble NH4(+) -N and PO4(3-) -P from biochars were determined. In the blank control group, which was not covered with any kinds of biochar, the ammonia-nitrogen concentration of the overlying water reached a maximum value of 4.27 mg x L(-1) on the twenty-fifth day, and then stabilized at about 4.02 mg x L(-1). Ammonia-nitrogen concentrations of the treatment groups with the four kinds of biochar all maintained below 0.3 mg x L(-1) after 25 days. Particularly, the Phragmites australis treatment group showed the best ability to inhibit ammonia-nitrogen release, and the cumulative release amount of ammonia-nitrogen was reduced by 85.61%. The cumulative release amount of COD was reduced by 28.83% to 30%. Phosphate concentration of the Zea mays group was higher than that of the blank group. On the contrary, Arundo donax and Phragmites australis groups showed a great potential in the inhibition of phosphate release. Biochar released the majority of NH4(+) -N and PO4(3-) -P in the first 3 days. The release amount of NH4(+) -N from Arachis hypogaea group was larger than those of the other three groups, reaching 36.79 mg x kg(-1). Similarly, Zea mays group had the largest release of PO4(3-) -P, which was 70.64 mg x kg(-1). All suggest that biochar is a potential in-situ capping material to reduce the release of NH4(+) -N, COD and PO4(3-) -P in polluted sediments, and has the ability of applying to the remediation of sediment in the polluted water.

Findings of coesite and microdiamond in metamorphic rocks of supracrustal protolith led to the recognition of continental subduction to mantle depths. The crust-mantle interaction is expected to take place during subduction of the continental crust beneath the subcontinental lithospheric mantle wedge. This is recorded by postcollisional mafic igneous rocks in the Dabie-Sulu orogenic belt and its adjacent continental margin in the North China Block. These rocks exhibit the geochemical inheritance of whole-rock trace elements and Sr-Nd-Pb isotopes as well as zircon U-Pb ages and Hf-O isotopes from felsic melts derived from the subducted continental crust. Reaction of such melts with the overlying wedge peridotite would transfer the crustal signatures to the mantle sources for postcollisional mafic magmatism. Therefore, postcollisonal mafic igneous rocks above continental subduction zones are an analog to arc volcanics above oceanic subduction zones, providing an additional laboratory for the study of crust-mantle interaction at convergent plate margins.

Highly ordered channel structure SBA-15 was widely concerned as new adsorbents in environmental protection, in order to increase its heavy metal ions adsorption ability from aqueous solution, the diamine -modified porous silicate SBA-15 was synthesized by a hydrothermal grafting method and characterized by TEM, X-ray diffraction, FTIR and N2 adsorption-desorption. The SBA-15 and modified SBA-15 samples were used as sorbents to adsorb Cd(II) ions from aqueous solution. The effect of experimental parameters, such as pH, contact time, sorbent dosage and temperature were examined, and the maximum adsorption amount was also calculated. The results showed that under same conditions, the Cd(II) removal rate was higher for 2N-SBA-15 than that of the unmodified SBA-15. The adsorption process was controlled by system pH. The highest removal rate could reached about 95% after pH was higher than 4. Adsorption equilibrium was reached within 30 minutes,and more than 95% Cd2+ was adsorbed when 7.5-20 mg sorbent was added into 100 mL solution contained 25 mg x L(-1) Cd2+. The adsorption capacity increased from 94.73% to 98.22% with temperature increased from 25 degrees C to 35 degrees C. The Langmuir model can be used to describe adsorption isotherms. The adsorption capacity of Cd2+ was 0.9 mmol x g(-1) which is comparable to the adsorption capacity of various adsorbents reported in the literature, and 0.1 mol x L(-1) HCl could remove nearly 93% Cd2+ from 2N-SBA-15 particles. Based on the thermodynamic, pH, XPS and Zeta potential analysis results in this study, it could be concluded that the adsorption process was an endothermic and spontaneous reaction which contained physical adsorption, ion exchange and chelating reaction etc. The study indicated that the diamine -modified ordered mesoporous material SBA-15 is a potential sorbent which could be used for the aqueous Cd2+ removal.

MicroRNAs (miRNAs) are a class of endogenous non-coding small RNAs of 18-22-nucleotides in length that regulate gene expression at the post-transcriptional level. The objective of this study was to examine the differences in the miRNA expression profiles of the lungs and trachea of beagle dogs infected with canine influenza virus (CIV). Total RNA was isolated from lung and trachea tissues of beagle dogs infected and non-infected with H3N2 CIV at 4dpi. A total of 41,512,315 and 39,107,475 reads were obtained from the lung and trachea, respectively. Out of a total 288 dog miRNAs available in miRBase, 227 and 236 miRNAs were detected in the infected (Fg) and the non-infected lungs (Fc), respectively, whereas 242 miRNAs were detected in both the infected (Qg) and the non-infected trachea (Qc). From these, 34 and 45 miRNAs were differentially expressed in the lungs and trachea between the infected and non-infected dogs, respectively. More miRNAs were highly expressed in the non-infected tissues than in the infected tissues. miR-143 was the most abundantly expressed miRNA in the four samples, followed by let-7. In total, 252, 234, 196 and 235 novel miRNAs were identified in the Fc, Fg, Qc, and Qg groups, respectively. To our knowledge, this is the first study examining the miRNA gene expression in CIV infected dogs using the Solexa sequencing approach. We have revealed the existence of a large number miRNAs that are affected by CIV infection as well as identified some potentially new miRNAs. These findings will help us better understand the host-CIV interaction and its relationship to pathogenesis, as well as contribute to the prevention and control of CIV.

Two-micrometer thulium laser resection of the prostate-tangerine technique (TmLRP-TT) has been introduced as a minimally invasive treatment for benign prostatic hyperplasia (BPH). Acute urinary retention (AUR) is a common and serious complication of BPH. The study was undertaken to assess the clinical efficacy and safety of TmLRP-TT in the treatment of patients with AUR secondary to BPH. A prospective evaluation of 52 patients undergoing TmLRP-TT from December 2011 to November 2012 was carried out. Preoperative status, surgical details, and perioperative complications were recorded. The follow-up outcome was evaluated with subjective and objective tests at 1 and 6 months. Mean age was 70.3?±?7.8 years old. Mean prostate volume was 69.6?±?31.6 ml, and mean residual volume with retention was 274.5?±?150.7 ml. Mean operative time was 64.1?±?30.4 min. Mean catheterization duration was 5.4?±?1.1 days. The mean International Prostate Symptom Score, quality of life score, and postvoid residual urine volume decreased significantly at 6-month follow-up (21.6?±?6.8 vs. 6.8?±?3.2, 4.4?±?1.2 vs. 0.9?±?0.8, 274.5?±?150.7 vs. 40.6?±?22.5 ml). The mean maximum urinary flow rate was 18.7?±?6.9 ml/s postoperative. Two (3.8 %) of the patients required blood transfusion in operation. Five (9.6 %) of the patients had transient hematuria postoperative, and two (3.8 %) of them received 3 days recatheterization due to clot retention. The early clinical results suggest that the TmLRP-TT is a promising safe, effective, and minimally invasive treatment for patients with AUR secondary to BPH. The incidence of complications was low.

[Parameter sensitivity analysis of runoff simulation and model adaptability research based on HSPF].

Based on the sensitivity analysis of key parameters, we calibrated and validated HSPF model in a typical small watershed of hilly region in Tai Lake, and then evaluated the adaptability of the model in the hilly region of Tai Lake. In this study, 5 sensitivity parameters AGWRC, UZSN, INFILT, LZSN, and DEEPFR were determined by using the relative sensitivity method which is based on perturbation analysis, and then the annual runoff from 2005 to 2010 was simulated. The results showed good agreement with percent differences of - 6.39%, - 8.76% and 9.66%, respectively, which were all below 10% during the calibration period, and the simulation results were in a good range with percent differences of -13.31% and 5.22% during the validation period. The Nash-Sutcliffe (Ens) coefficients reached 0.87 and 0.69, respectively, and relative errors were 1.63% and 4.14%, which met the simulation requirements and could reflect the variation of annual runoff in Zhongtian River Basin. The results show that the HSPF model in the study area has better adaptability to watershed runoff simulation which has scientific significance to the simulated hydrological processes in the region and also provides reference for application and promotion of HSPF model in different regions in our country.

The occurrence and removal of six pharmaceuticals and personal care products (PPCPs) including caffeine (CF), N, N-diethyl-meta-toluamide (DEET), carbamazepine, metoprolol, trimethoprim (TMP), and sulpiride in a municipal wastewater treatment plant (WWTP) in Shanghai, China were studied in January 2013; besides, grab samples of the influent were also taken every 6 h, to investigate the daily fluctuation of the wastewater influent. The results showed the concentrations of the investigated PPCPs ranged from 17 to 11,400 ng/L in the WWTP. A low variability of the PPCP concentrations in the wastewater influent throughout the day was observed, with the relative standard deviations less than 25 % for most samples. However, for TMP and CF, the slight daily fluctuation still reflected their consumption patterns. All the target compounds except CF and DEET, exhibited poor removal efficiencies (<40 %) by biological treatment process, probably due to the low temperature in the bioreactor, which was unfavorable for activated sludge. While for the two biodegradable PPCPs, CF, and DEET, the anaerobic and oxic tank made contributions to their removal while the anoxic tank had a negative effect to their elimination. The tertiary UV treatment removed the investigated PPCPs by 5-38 %, representing a crucial polishing step to compensate for the poor removal by the biologic treatment process in winter.

Thulium laser resection of the prostate-tangerine technique (TmLRP-TT) dissects whole prostatic lobes off the surgical capsule, similar to peeling a tangerine. The present study aimed to evaluate the safety and efficacy of TmLRP-TT for older symptomatic benign prostatic hyperplasia patients with large prostates during 18 months of follow-up. A prospective analysis of 95 consecutive patients with large prostates (>80 ml) who underwent surgical treatment using TmLRP-TT was carried out. All patients were evaluated preoperatively and at 1, 6, 12, and 18 months postoperatively by the International Prostate Symptom Score (IPSS), quality of life (QoL), maximum urinary flow rate (Q max), postvoid residual urine volume (PVR), International Index of Erectile Function 5 (IIEF-5), urine analysis, and urine culture. Perioperative complications were recorded and graded by the modified Clavien classification system (CCS). Mean preoperative prostate volume was 106.81?±?24.79 ml. TmLRP-TT was successfully completed in all patients. The mean operative duration, catheterization time, and hospital stay were 95.36?±?27.06 min, 2.25?±?0.9 days, and 5.39?±?1.18 days, respectively. The decrease in mean hemoglobin level was 1.23?±?0.72 g/dl, and that in mean serum sodium level was 0.71?±?2.56 mmol/l. Within the observation period of 18 months, the patients showed an improvement in IPSS (20.01?±?7.08 vs. 4.96?±?3.68), QoL (4.10?±?1.16 vs. 1.23?±?1.30), Q max (8.14?±?3.81 ml/s vs. 18.33?±?2.56 ml/s) and PVR (102.70?±?70.64 ml vs. 20.28?±?30.02 ml), compared with baseline values (P?

A "basophilic method" for phenolic hydroxyl bridged lanthanide coordination polymers (CPs) was developed. With this method, eleven CPs with the general formula of [Ln(HL1)L1·H(2)O](n) (Ln = Tb (1), Nd (2), Eu (3), Gd (4), La (5), Er (6), Y (7), H(2)L1 = 4-methyl salicylic acid) and [Ln(HL2)L2·2MeOH](n) (Ln = Eu (8), Tb (9), Gd (10), La (11), H(2)L2 = 3-hydroxy-2-naphthoic acid) were synthesized based on two ligands, and five of them (1-4 and 8) were characterised by X-ray single crystal diffraction. The powder X-ray diffraction patterns (PXRD) of complexes showed that 1-7 are isostructural, 8-11 are isostructural. Furthermore, 1 was characterised by Fourier transform infrared (FT-IR), thermogravimetric analysis (TGA), differential thermal analysis (DTA), elemental analysis (EA), one and two photon luminescence were investigated in detail.

Paclitaxel, one of the broadest-spectrum anticancer agents, is currently being used in the treatment of patients with solid tumors. In the present study, we compared the effect of paclitaxel on two types of leukemia cells. Our results showed that paclitaxel could inhibit the proliferation of MEL and K562 cells in a dose- and time-dependent manner. The mechanism of proliferative inhibition in K562 cells treated by paclitaxel was related to the cell cycle arrest in the G2/M phase, as well as the induction of apoptosis. By contrast, MEL cells treated by paclitaxel showed significant characteristics of necrosis, which indicated that the mode of cell death induced by paclitaxel in these two types of leukemia cells differed. Advances in research of the cell cycle, apoptosis and necrosis will extend our understanding of the mechanisms of paclitaxel-induced cell death, particularly in leukemia cells. Further elucidation of the mechanisms of necrosis in MEL cells may expedite the development of improved paclitaxel-based regimens for cancer therapy.

Abdominal wall desmoid tumors: A case report.

Desmoid tumors (DTs) are rare lesions that do not possess any metastatic potential. However, they have a strong tendency to invade locally and recur. They constitute 3% of all soft tissue tumors and 0.03% of all neoplasms. Abdominal DTs occur sporadically or are associated with certain familial syndromes, such as familial adenomatous polyposis (FAP). The single form of this neoplasm most frequently occurs in females of reproductive age and during pregnancy. A female patient with a DT of the abdominal wall who had no relevant family history was admitted to hospital. The patient, who presented with a painless mass in the left anterolateral abdomen, had no history of trauma, surgery or childbearing. According to the medical history, physical examination and CT report, the patient was diagnosed with DT. Radical resection of the affected abdominal wall musculature was performed, and the defect was replaced with a polypropylene mesh. The histological diagnosis was of DT. The patient remains in good health and complete remission without any other treatment following surgery. DTs exhibit aggressive growth and have a high rate of recurrence. Surgery is the optimal treatment, and subsequent radiotherapy may decrease the local recurrence rate. Further research into their aetiology is required combined with multicentre clinical trials of new treatments in order to improve management of this disease. This case report provides general knowledge of DT, and may be used as a guidance for diagnosis and treatment.

Pollution levels of perfluorochemicals in eggs purchased from the markets in Beijing had been investigated. The egg samples of chicken and duck were collected from the 59 stalls of 14 main eggs wholesale markets in Beijing, respectively. Systematic analyses were made for seventeen kinds of perfluorochemicals (11 perfluorinated carboxylates (PFCAs), 3 perfluorinated sulfonates (PFSAs), perfluorooctane sulfonamide (FOSA), 2-perfluorooctylethanoic acid (FOEA) and 2H-perfluoro-2-decenoic acid (FOUEA) by a high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The results showed that there was a certain perfluorochemical contamination in all egg samples. Nine kinds of perfluorochemicals were detected in chicken eggs, perfluorononanoic acid (PFNA), perfluoroheptanoic acid (PFHpA) and perfluorooctanoate acid (PFOA) are dominant, and their average concentrations were 0.105, 0.073 and 0.069 ng x g(-1), respectively. Ten kinds of perfluorochemicals were detected in the duck eggs, perfluorooctane sulfonate (PFOS) and PFOA are dominant, and their average concentrations were 0.378 and 0.296 ng x g(-1), respectively. Perfluoropentanoic acid ( PFPeA), perfluorotetradecanoic acid (PFTA), perfluorobutane sulfonate (PFBS) and 2-perfluorooctylethanoic acid (FOEA) were not detected in all samples. The total concentration of PFCs in the duck eggs was 3.4 times of that in the chicken eggs. A strong positive correlation (r = 0.954) was not only observed between of PFNA and PFHpA in chicken eggs, but also found between perfluoroundecanoic acid (PFUnDA) and perfluorotridecanoic acid (PFTrDA) in duck eggs (r = 0.915). The results of health-based risk assessment showed that there was little immediate risk of exposure to PFOS and PFOA via the consumption of chicken eggs and duck eggs purchased from the markets in Beijing.

Inflammatory pseudotumor of the spleen (IPTS) is an extremely rare condition. To the best of our knowledge, only ?113 cases have been reported in the literature since the first 2 cases were reported in 1984. The present study reports the case of an IPTS in a 72-year-old male patient. The splenic tumor was identified incidentally 1 year prior to the patient being admitted to the Second Affiliated Hospital of Dalian Medical University (Dailan, China). There were no specific clinical symptoms. The initial diagnosis was of splenic lymphoma based on the pre-operative radiological findings. However, the patient underwent a splenectomy and the final pathological diagnosis of IPTS was declared. The present study also highlighted the difficulty of forming accurate pre-operative diagnoses, even when using modern imaging techniques. A partial resection of the spleen or splenectomy was considered to be the required treatment to form a definitive diagnosis and exclude malignancy. The prognosis of IPTS is generally considered to be favorable following splenectomy. The clinical and pathological features of previously reported cases are also briefly reviewed in the present study to aid in improving the accuracy of the diagnosis of this rare disease.

This study aims to explore the probable mechanism of better result of dural reconstruction by CO2 laser soldering and the effect of exogenous basic fibroblast growth factor (bFGF) or transforming growth factor-beta1(TGF?1) on wound healing. In part I of the study, ten minipigs were randomized into two equal groups, and the dural defects were reconstructed by conventional fibrin glue (FG) bonding (group I a) or by CO2 laser soldering (group Ib). In part II, 36 minipigs were randomized into three equal groups, and the dural defect was reconstructed by CO2 laser soldering; then exogenous bFGF or TGF?1 was administered in group IIb and group IIc, respectively, while group IIa served as control group. The dural specimens were harvested at 1st week postoperatively in part I; and at 1st, 2nd, 3rd, and 4th week postoperatively in part II, they were examined for healing condition and subjected to hematoxylin-eosin (HE) staining and immunohistochemical (IHC) staining with antibodies against bFGF and TGF?1. Anoplin is a recently discovered antimicrobial peptide (AMP) isolated from the venom sac of the spider wasp Anoplius samariensis, and it is one of the shortest ?-helical AMP found naturally to date consisting of only ten amino acids. Previous results showed that anoplin exhibits potent antimicrobial activity but little hemolytic activity. In this study, we synthesized anoplin, studied its cytotoxicity in Friend virus-induced leukemia cells [murine erythroleukemia (MEL) cells], and proposed its possible mechanism. Our results showed that anoplin could inhibit the proliferation of MEL cells in a dose-dependent and time-dependent manner via disrupting the integrity of cell membrane, which indicated that anoplin exerts its cytotoxicity efficacy. In addition, the cell cycle distribution of MEL cells was arrested in the G?/G? phase significantly. However, anoplin could not induce obvious apoptosis in MEL cells, as well as anoplin could not induce visible changes on morphology and quantity in the bone marrow cells isolated from normal mice. All of these results indicate that anoplin, as generally believed, is a selective AMP, a value characteristic in the design of safe therapeutic agents. The cytotoxicity of anoplin on MEL cells was mainly attributable to the plasma membrane perturbation and also to the intracellular events such as the arrest of cell cycle. Although this is an initial study that explored the activity of anoplin in vitro rather than in vivo, with the increasing resistance of conventional chemotherapy, there is no doubt that anoplin has desirable feature to be developed as a novel and selective anticancer agent.

Chlamydia spp. are obligate intracellular gram-negative bacteria that cause a wide range of significant diseases in humans and animals worldwide, resulting in significant economic losses. Chlamydial infection in cattle has been reported in many countries including China. However, there has been no survey of chlamydial infection of dairy cattle in Guangzhou, southern China. The objective of the present investigation was to examine the chlamydial seroprevalence in dairy cattle in Guangzhou, subtropical southern China by using an indirect hemagglutination assay (IHA). The overall seroprevalence of chlamydial infection in dairy cattle was 7.25% (29/400). Greater than or equal to eight-yr-old dairy cattle had the highest seroprevalence (10.34%), followed by those that were???6 years old or??0.05). Dairy cattle with 5 pregnancies had the highest seroprevalence (10.81%). These results indicate that chlamydial infection was present in dairy cattle in Guangzhou, subtropical southern China, and integrated strategies and measures should be executed to control and prevent chlamydial infection and disease outbreak in the study region.

Hemokinin-1 is a peptide encoded by Pptc, which belongs to the family of mammalian tachykinins. Our previous results showed that rat/mouse hemokinin-1 (r/m HK-1) produced striking analgesia after intracerebroventricular (i.c.v.) injection in mice, and the analgesia could be blocked by the NK1 receptor antagonist and the opioid receptor antagonist, respectively. However, the precise distribution sites and the molecular mechanism involved in the analgesic effect after i.c.v. administration of r/m HK-1 are needed to be further investigated deeply. Using the fluorescence labeling method, our present results directly showed that r/m HK-1 peptides were mainly distributed at the ventricular walls and several juxta-ventricular structures for the first time. Our results showed that the mRNA expressions of NK1 receptor, PPT-A, PPT-C, KOR, PDYN, DOR and PENK were not changed markedly, as well as the protein expression of NK1 receptor was hardly changed. However, both the transcripts and proteins of MOR and POMC were up-regulated significantly, indicating that the analgesic effect induced by i.c.v. administration of r/m HK-1 is related to the activation of NK1 receptor first, then it is related to the release of endogenous proopiomelanocortin, as well as the increased expression level of ? opioid receptor. These results should facilitate further the analysis of the analgesia of r/m HK-1 in the central nerval system in acute pain and may open novel pharmacological interventions.

Toxoplasma gondii is a protozoan parasite infecting almost all warm-blooded animals and humans. There are three infective stages of T. gondii: the tachyzoites, the bradyzoites and the oocysts. The tachyzoite is a rapidly multiplying stage and the main pathogenic factor. In North America and Europe, T. gondii is consisted of four major clonal lineages (namely Types I, II, III and Type 12). In this study, we explored the proteomic profiles of different genotypes (Type I-RH strain, Type II-PRU strain, Type II-TgQHO strain and ToxoDB 9-TgC7 strain) of T. gondii tachyzoites by using two-dimensional fluorescence difference gel electrophoresis (2D DIGE) combined with MALDI-TOF MS. Totally, 110 differentially abundant protein spots were selected. Of these, 98 spots corresponding to 56 proteins from T. gondii were successfully identified. These included surface antigen (SAG1), heat shock protein 70 (Hsp70), disulfide isomerase, coronin, heat shock protein 60 (Hsp60), pyruvate kinase, receptor for activated C kinase 1 and peroxiredoxin. Gene ontology (GO) enrichment analysis revealed that most of the differentially abundant proteins were involved in biological regulation, metabolic process, response to stress, binding, antioxidant activity and transporter activity. According to the Kyoto Encyclopedia of Genes and Genomes (KEGG) metabolic pathway maps of T. gondii, some identified proteins were involved in the glycolytic/gluconeogenesis pathway. The present study identified differentially abundant proteins among different genotypes of T. gondii and these findings have implications for the better understanding of the phenotypic differences among the examined T. gondii genotypes which in turn may contribute to the better control of toxoplasmosis. ?-Poly-L-lysine (?-PL), one of the only two homo-poly amino acids known in nature, is used as a preservative. In this study, strategies of feeding precursor L-lysine into 5 L laboratory scale fermenters, including optimization of L-lysine concentration and time, was investigated to optimize the production of ?-PL by Streptomyces sp. M-Z18. The optimized strategy was then used in ?-PL fed-batch fermentation in which glucose and glycerol served as mixed carbon sources. In this way, a novel ?-PL production strategy involving precursor L-lysine coupled with glucose-glycerol co-fermentation was developed. Under optimal conditions, ?-PL production reached 37.6 g/l, which was 6.2 % greater than in a previous study in which glucose and glycerol co-fermentation was performed without added L-lysine (35.14 g/l). To the best of our knowledge, this is the first report of the enhancement of ?-PL production through L-lysine feeding to evaluate the use of fermenters. Meanwhile, the role of L-lysine in the promotion of ?-PL production, participating ?-PL synthesis as a whole, was first determined using the L-[U-(13)C] lysine labeling method. It has been suggested that the bottleneck of ?-PL synthesis in Streptomyces sp. M-Z18 is in the biosynthesis of precursor L-lysine. The information obtained in the present work may facilitate strain improvement and efficient large-scale ?-PL production.

Aging is a major risk factor for prostate cancer (PCa), and prostatic stromal cells may also promote PCa progression. Accordingly, stromal cells do not equally promote PCa in older males and younger males. Therefore, it is also possible that the expression of androgen receptors (ARs) by prostatic stromal cells in older versus younger males plays different roles in PCa progression. Using a gene knockdown technique and coculture system, we found that the knockdown of the AR in prostatic stromal cells obtained from younger males could promote the invasiveness and metastasis of cocultured PC3/LNCaP cells in vitro. By contrast, the invasiveness and metastasis of LNCaP cells was inhibited when cocultured with prostatic stromal cells from older males that when AR expression was knocked down. Moreover, after targeting AR expression with small hairpin RNA (shRNA), matrix metalloproteinase (MMP) expression in stromal cells was observed to increase in the younger group, but decreased or remained unchanged in the older group. One exception, however, was observed with MMP9. In vivo, after knocking down AR expression in prostatic stromal cells, the incidence of metastatic lymph nodes was observed to increase in the younger age group, but decreased in the older age group. Together, these data suggest that the AR in prostatic stromal cells played opposite roles in PCa metastasis for older versus younger males. Therefore, collectively, the function of the AR in prostatic stromal cells appears to change with age, and this may account for the increased incidence of PCa in older males.

Eight-stranded ?-barrel outer membrane proteins can confer bacterial virulence via resistance to host innate defenses. This resistance function of OmpW, which was recently identified as an eight-stranded ?-barrel protein, was investigated in this study. Our results demonstrated that upregulation of OmpW correlated with increased bacterial survival during phagocytosis. Bacterial mutants harboring a deletion of ompW exhibited a significantly increased phagocytosis rate. Both observations suggest that the OmpW protein protects bacteria against host phagocytosis. In addition, expression of ompW is regulated by iron, which implies that the resistance provided by OmpW may be an important factor in iron-related infectious diseases. Concentrations of representative organochlorines (OCs), including organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) in a batch of farmed mandarin fish from a cultivation pond in Wujiang City, Jiangsu Province, were determined with GC-ECD. Meanwhile, a preliminary human health risk assessment was conducted. Results showed that, the concentrations of dichlorodiphenyltrichloroethanes (DDTs), hexachlorocyclohexanes (HCHs), hexachlorobenzene (HCB) and PCBs in the farmed mandarin fish were in the range of 1.3-4.57 ng/g, 0.13-1.24 ng/g, 0.07-0.44 ng/g and 0-5.22 ng/g, respectively, with an average value of 2.96, 0.40, 1.27 and 0.7 ng/g, respectively. These values were far below the corresponding residue limits set by Chinas Ministry of Health and the U.S. Food and Drug Administration, and at the low end of the global pollution bar. Referring to the reference values given by USEPA Integrated Risk Information System, calculated non-carcinogenic hazard ratios (HRs) of DDTs, HCB and PCBs were all <1 and carcinogenic HRs of DDTs, HCHs, HCB and PCBs were all >1. The results indicated that DDTs, HCHs and HCB in farmed mandarin fish did not have negative human health influence, but existed potential carcinogenic risk to humans. Compared to rural residents, urban residents were more vulnerable to DDTs, HCHs, HCB and PCBs threats.

The objective of this paper is to reveal to what degree biobased jet fuels (biojet) can reduce greenhouse gas (GHG) emissions from the U.S. aviation sector. A model of the supply and demand chain of biojet involving farmers, biorefineries, airlines, and policymakers is developed by considering factors that drive the decisions of actors (i.e., decision-makers and stakeholders) in the life cycle stages. Two kinds of feedstock are considered: oil-producing feedstock (i.e., camelina and algae) and lignocellulosic biomass (i.e., corn stover, switchgrass, and short rotation woody crops). By factoring in farmer/feedstock producer and biorefinery profitability requirements and risk attitudes, land availability and suitability, as well as a time delay and technological learning factor, a more realistic estimate of the level of biojet supply and emissions reduction can be developed under different oil price assumptions. Factors that drive biojet GHG emissions and unit production costs from each feedstock are identified and quantified. Overall, this study finds that at likely adoption rates biojet alone would not be sufficient to achieve the aviation emissions reduction target. In 2050, under high oil price scenario assumption, GHG emissions can be reduced to a level ranging from 55 to 92%, with a median value of 74%, compared to the 2005 baseline level.

Toll-like receptor-4 (TLR-4) is integrally involved in lipopolysaccharide (LPS) signaling and has a requisite role in the activation of nuclear factor-?B (NF-?B). The exact mechanisms that lend perfluorocarbon (PFC) liquids a cytoprotective effect have yet to be elucidated. Therefore we examined in an in vitro model the cytoprotective effect of PFC on LPS-stimulated alveolar epithelial cellls (AECs).
