Two new heterometallic coordination polymers, [Na4Ln12(stp)8(OH)16(H2O)12]·10H2O [Ln = Dy (1) and Ho (2)], have been prepared from monosodium 2-sulfoterephthalate (NaH2stp), dysprosium acetate, or holmium acetate. They are isostructural, possessing a [Ln12(?3-OH)16](20+) wheel-cluster core based on four vertex-sharing cubane-like [Ln4(?3-OH)4](8+) units. The Ln12 cores are linked by stp ligands into a three-dimensional (3D) architecture. Magnetic studies indicate that complex 1 exhibits slow relaxation of magnetization, and it can be regarded as the first 3D coordination assembly of a Dy12 cluster single-molecule magnet.

Interpenetration is an intrinsic behaviour for the porous coordination networks. To prevent the interpenetration, a common strategy is the imposition of geometric or steric restrictions by incorporating bulky moieties into organic tectons. So far, most of the available incorporations have been achieved through a covalent connection, while few involved in the non-covalent weak interactions. In this paper, we have reported that such interpenetration can be prevented by the less common lone pair-? interactions. By imposing the lone pair-? interactions through the addition of lone-pair-bearing N-methylpyrrolidin-2-one or iodine, combinations of rigid naphthalene diimide tectons bearing two divergently oriented pyridyl units at both imide extremities with ZnSiF6 led to non-interpenetrated cuboid 3-D coordination networks that should have been interpenetrated. In addition, such close-contacting lone pair-? interactions between electron donors and acceptors have also been demonstrated to play a key role in their photochromic properties.

Lanthanide doped ZnO mushroom-like 3D hierarchical structures have been fabricated by polyol-mediated method and characterized by various microstructural and optical techniques. The results indicate that the as-prepared ZnO:Ln(3+) (Ln = Tb, Eu) samples have a hexagonal phase structure and possess a mushroom-like 3D hierarchical morphology. The length of the whole mushroom from stipe bottom to pileus top is about 1.0 ?m, and the diameters of pileus and stipe are about 0.8 ?m and 0.4 ?m, respectively. It is found that the flow of N2 is the key parameter for the formation of the novel ZnO structure and the addition of (NH4)2HPO4 has a prominent effect on the phase structure and the growth of mushroom-like morphology. The potential mechanism of forming this morphology is proposed. The pileus of the formed mushroom is assembled by several radial ZnO:Ln(3+) nanorods, whereas the stipe is composed of over layered ZnO:Ln(3+) nanosheets. Moreover, asymmetrical I-V characteristic curves of ZnO:Ln(3+) mushrooms indicate that the texture composition of the 3D hierarchical morphology might lead to the asymmetrical transport behavior of electrical conductivity. Lanthanide doped ZnO samples can exhibit red or green emission under the excitation of UV light.

Tumor angiogenesis is a critical element of cancer progression, and strategies for its selective blockade are still sought. Here, we examine the angiogenic effects of Globo-H ceramide (GHCer), the most prevalent glycolipid in a majority of epithelial cancers and one that acts as an immune checkpoint. Here, we report that GHCer becomes incorporated into endothelial cells through the absorption of microvesicles shed from tumor cells. In endothelial cells, GHCer addition induces migration, tube formation, and intracellular Ca(2+) mobilization in vitro and angiogenesis in vivo. Breast cancer cells expressing high levels of GHCer displayed relatively greater tumorigenicity and angiogenesis compared with cells expressing low levels of Globo-H. Clincally, GHCer(+) breast cancer specimens contained higher vessel density than GHCer(-) breast cancer specimens. Mechanistic investigations linked the angiogenic effects of GHCer to its endocytosis and binding to TRAX, with consequent release of PLC?1 from TRAX to trigger Ca(2+) mobilization. Ligustrazine, one of the major effective components of the Chinese traditional medicinal herb Ligusticum Chuanxiong Hort, has been reported plenty of biological activities, such as protect cardiovascular and cerebrovascular, neuroprotection and anti-tumor, et al. Because of its remarkable effects, studies on structural modification of ligustrazine have attracted much attention. Ligustrazine synthetic derivatives reported in recent decades are mainly derived from four primary intermediates (TMP-COOH, TMP-OH, TMP-NH2, HO-TMP-OH). To explore the neuroprotection activitiy of ligustrazine intermediates, six ligustrazine intermediates (2, 5, 8, 11, 12, 13) were synthesized and their protective effects against CoCl2-induced neurotoxicity in differentiated PC12 cells were studied. The target compounds were prepared via different chemical methods, including oxidation, substitution, esterification and amidation without changing the structure nucleus of ligustrazine. Compared with TMP (EC50 = 56.03 micromol x L(-1)), four compounds (2, 5, 12 and 13) exhibited higher activity (EC50 < 50 micromol x L(-1)) respectively, of which, compound 2 displayed the highest protective effect against the damaged PC12 cells (EC50 = 32.86 micromol x L(-1)), but target compounds 8 and 11 appeared lower activity (EC50 > 70 micromol x L(-1)). By structure-activity relationships analysis, the introduction of carboxyl, amino to the side chain of ligustrazine and appropriately increase the proportion of ligustrazine may contribute to enhance its neuroprotective activity, which provides a reference for the design, synthesis and activity screening of relevant series of ligustrazine derivatives in the future.

Water pollution worsens the problem of disinfection by-products (DBPs) in drinking water supply. Biodegradation of wastewater organics produces soluble microbial products (SMPs), which can be important DBP precursors. In this laboratory study, a number of enhanced water treatment methods for DBP control, including enhanced coagulation, ozonation, and activated carbon adsorption, were evaluated for their effectiveness in treating SMP-containing water for the DBP reduction purpose. The results show that enhanced coagulation with alum could remove SMPs only marginally and decrease the DBP formation potential (DBPFP) of the water by less than 20%. Although ozone could cause destruction of SMPs in water, the overall DBPFP of the water did not decrease but increased after ozonation. In contrast, adsorption by granular activated carbon could remove the SMP organics from water by more than 60% and reduce the DBPFP by more than 70%. It is apparent that enhanced coagulation and ozonation are not suitable for the removal of SMPs as DBP precursors from polluted water, although enhanced coagulation has been commonly used to reduce the DBP formation caused by natural organic matter. In comparison, activated carbon adsorption is shown as a more effective means to remove the SMP content from water and hence to control the wastewater-derived DBP problem in water supply.

Plant pathogens deploy an array of virulence factors to suppress host defense and promote pathogenicity. Numerous strains of Pseudomonas syringae produce the phytotoxin coronatine (COR). A major aspect of COR function is its ability to mimic a bioactive jasmonic acid (JA) conjugate and thus target the JA-receptor COR-insensitive 1 (COI1). Biological activities of COR include stimulation of JA-signaling and consequent suppression of SA-dependent defense through antagonistic crosstalk, antagonism of stomatal closure to allow bacterial entry into the interior of plant leaves, contribution to chlorotic symptoms in infected plants, and suppression of plant cell wall defense through perturbation of secondary metabolism. Here, we review the virulence function of COR, including updates on these established activities as well as more recent findings revealing COI1-independent activity of COR and shedding light on cooperative or redundant defense suppression between COR and type III effector proteins.

The Baiyangdian Wetland Natural Reserve is an important breeding habitat for oriental great reed warbler (OGRWs), Acrocephalus orientalis, in North China Plain. We investigated the nesting sites of OGRWs by line transect method during June-July, 2011, and 112 nests were found out in total. The ecological-niche factor analysis (ENFA) was applied in nesting habitat suitability assessment for OGRWs in Baiyangdian. The results showed that OGRWs in this reserve preferred nesting in dry land reed landscapes, which located at relatively high altitudes and away from anthropocentric disturbance. In 2011, the suitable and the most suitable nesting habitats of OGRWs in this reserve were 2474.69 hm2 and 1131.19 hm2, accounting 7.6% and 3.5% of the total reserve area, respectively. The most suitable nesting habitats shaped a circle-like structure, and they all clustered together around Damai and Shaoche core area, which located in the east part of the reserve. In order to protect the nesting habitats for OGRWs within Baiyangdian wetland reserve, it was suggested that the functional zone should be reorganized, and that the Damai and Shaoche core area should be integrated into one. This new core area would be bigger and more concentrated, at the same time the buffer zone should also be established appropriately, so as to protect the natural landscapes in this reserve as much as possible.

Autophagy is a tightly regulated lysosomal degradation pathway for maintaining cellular homeostasis and responding to stresses. Beclin 1 and its interacting proteins, including the class III phosphatidylinositol-3 kinase Vps34, play crucial roles in autophagy regulation in mammals. We identified nuclear receptor binding factor 2 (Nrbf2) as a Beclin 1-interacting protein from Becn1(-/-);Becn1-EGFP/+ mouse liver and brain. We also found that Nrbf2-Beclin 1 interaction required the N terminus of Nrbf2. We next used the human retinal pigment epithelial cell line RPE-1 as a model system and showed that transiently knocking down Nrbf2 by siRNA increased autophagic flux under both nutrient-rich and starvation conditions. To investigate the mechanism by which Nrbf2 regulates autophagy, we demonstrated that Nrbf2 interacted and colocalized with Atg14L, suggesting that Nrbf2 is a component of the Atg14L-containing Beclin 1-Vps34 complex. Moreover, ectopically expressed Nrbf2 formed cytosolic puncta that were positive for isolation membrane markers. These results suggest that Nrbf2 is involved in autophagosome biogenesis. Furthermore, we showed that Nrbf2 deficiency led to increased intracellular phosphatidylinositol-3 phosphate levels and diminished Atg14L-Vps34/Vps15 interactions, suggesting that Nrbf2-mediated Atg14L-Vps34/Vps15 interactions likely inhibit Vps34 activity. Therefore, we propose that Nrbf2 may interact with the Atg14L-containing Beclin 1-Vps34 protein complex to modulate protein-protein interactions within the complex, leading to suppression of Vps34 activity, autophagosome biogenesis, and autophagic flux. This work reveals a novel aspect of the intricate mechanism for the Beclin 1-Vps34 protein-protein interaction network to achieve precise control of autophagy.

Magnetic nanoparticles of ferroferric oxide/carbon nanotubes (Fe3O4/CNTs) were synthesized by chemically bonding single-walled carbon nanotubes (SWCNTs) to the Fe3O4 nanoparticles. The pretreated carboxyl SWCNTs were modified on the surface of amino-functioned Fe3O4 nanoparticles through the cross-linking reaction between 1-ethyl-3-( 3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS). The synthesized Fe3O4/CNTs nanoparticles presented high super paramagnetic property and good dispersion ability, and were considered as good candidate adsorbents for dispersive solid phase microextraction. In this work, the synthesized FeO4/CNTs nanoparticles were applied to the extraction of vanillin additives, which was combined with high performance liquid chromatography (HPLC) analysis. The rapid and efficient preconcentration of vanillin and ethyl vanillin from different milk samples were successfully obtained with the detection limits of 10 microg/L and satisfactory recoveries of more than 92%. The results indicated that Fe3O4/CNTs magnetic nanoparticles were good pretreatment alternatives for the concentration of vanillin additives in milk products.

Solar cells incorporated with multi-coloring capability not only offer an aesthetic solution to bridge the gap between solar modules and building decorations but also open up the possibility for self-powered colorful display. In this paper, we proposed a multi-colored semi-transparent organic solar cells (TOSCs) design containing metallic nanostructures with the both high color purity and efficiency based on theoretical considerations. By employing guided mode resonance effect, the multi-colored TOSC behave like an efficient color filter that selectively transmits light with the desired wavelengths and generates electricity with light of other wavelengths. Broad range of coloring and luminosity adjusting for the transmission light can be achieved by simply tuning the period and the duty cycle of the metallic nanostructures. Furthermore, accompanying with the efficient color filtering characteristics, the optical absorption of TOSCs was improved due to the marked suppression of transmission loss at the off-resonance wavelengths and the increased light trapping in TOSCs. The mechanisms of the light guiding in photoactive layer and broadband backward scattering from the metallic nanostructures were identified to make an essential contribution to the improved light-harvesting. By enabling efficient color control and high efficiency simultaneously, this approach holds great promise for future versatile photovoltaic energy utilization.

We utilized a thermal radiation method to synthesize semiconducting hollow ZnO nanoballoons and metal-semiconductor concentric solid Zn/ZnO nanospheres from metallic solid Zn nanospheres. The chemical properties, crystalline structures, and photoluminescence mechanisms for the metallic solid Zn nanospheres, semiconducting hollow ZnO nanoballoons, and metal-semiconductor concentric solid Zn/ZnO nanospheres are presented. The PL emissions of the metallic Zn solid nanospheres are mainly dependent on the electron transitions between the Fermi level (EF) and the 3d band, while those of the semiconducting hollow ZnO nanoballoons are ascribed to the near band edge (NBE) and deep level electron transitions. The PL emissions of the metal-semiconductor concentric solid Zn/ZnO nanospheres are attributed to the electron transitions across the metal-semiconductor junction, from the EF to the valence and 3d bands, and from the interface states to the valence band. All three nanostructures are excellent room-temperature light emitters.

However, it remains unclear whether frontline EGFR TKIs affect subsequent chemo-sensitivity in EGFR-mutant patients. Monosaccharides are the fundamental composition units of saccharides which are a common source of energy for metabolism. An effective and simple method consisting of microwave assisted extraction (MAE), solid phase extraction (SPE) and high performance liquid chromatography-refractive index detector (HPLC-RID) was developed for rapid detection of monosaccharides in plants. The MAE was applied to break down the structure of the plant cells and release the monosaccharides, while the SPE procedure was adopted to purify the extract before analysis. Finally, the HPLC-RID was employed to separate and analyze the monosaccharides with amino column. As a result, the extraction time was reduced to 17 min, which was nearly 85 times faster than soxhlet extraction. The recoveries of arabinose, xylose, fructose and glucose were 85.01%, 87.79%, 103.17%, and 101.24%, with excellent relative standard deviations (RSDs) of 1.94%, 1.13%, 0.60% and 1.67%, respectively. The proposed method was demonstrated to be efficient and time-saving, and had been applied to analyze monosaccharides in tobacco and tea successfully.

A simple and rapid method for multiplex protein assay based on tunable aptamer by microchip electrophoresis has been developed. Different lengths of aptamers can modulate the electrophoretic mobility of proteins, allowing the protein molecules to be effectively separated in hydroxyethyl cellulose buffer with 1.00 mM magnesium ion. A non-specific DNA was exploited as an internal standard to achieve the quantitative assay and to reduce the interference. A fluorescence dye SYBR gold was exploited to improve the sensitivity and to suppress the interference from sample matrix. Under optimum conditions, quantitative assay of PDGF-BB (R(2)=0.9986), VEGF165 (R(2)=0.9909), and thrombin (R(2)=0.9947) were achieved with a dynamic range in the 5.00-150.0 nM and RSDs in the 5.87-16.3% range. The recoveries were varied from 83.6% to 113.1%. Finally, the proposed method was successfully applied to analyze cell secretions, and then the concentration of PDGF-BB and VEGF165 were detected from 5.15 nM to 2.03 nM, and 3.14 to 2.53 nM, respectively, indicating the established method can be used to analyze cell secretions.

A novel polarization rotator (PR) based on mode coupling in plasmonic waveguides is demonstrated by simulation. A silicon waveguide with asymmetric claddings of silicon oxide and metal is applied to induce a hybridization of the polarization modes. Operating at the telecommunication wavelength of 1.55 ?m, polarization conversion efficiency of 99.7% can be achieved in a device at a length of 9.7 ?m with an insertion loss of 2.2 dB. This PR can be easily fabricated by oblique deposition of the claddings after etching the silicon waveguide without precise alignment for two-step lithography as required in a previous design.

Two new Schiff base complexes, [ZnL(Ac)]2 (1) and [CuLCl]·H2O (2) (HL = 2-(2'-hydroxynaphthylmethylene)aminophenylbenzimidazole, Ac = acetate) were synthesized by a one-pot reaction and characterized by elemental analyses, IR and single-crystal X-ray diffraction. Fluorescence of complex 1 showed "on-off-on" switching when Cu(2+) and cyanide were sequentially detected. This phenomenon was investigated by UV-Vis absorption, fluorescence and ESI-MS spectra, and theoretical calculations to explain the response. Complex 1 can selectively respond to Cu(2+) with the fluorescence "turn-off" owing to the formation of complex 2 in the solution. Interestingly, the quenched fluorescence of complex 1 by Cu(2+) ions could be turned on after the addition of cyanide owing to the displacement of L(-) by CN(-) to form [Cu(CN)x](n-), making complex 1 a selective CN(-) probe. In addition, the results of the MTT assay experiment indicate that complex 1 has a low toxicity to cells and can be used to detect intracellular Cu(2+) ion by live cell imaging.

Using distinct ZnO NW arrays to provide positively charged surface, charge effect on cytotoxicity is investigated. 1-D structure of ZnO NWs is the main factor leads to apoptosis accompanied by ROS enrichment and GSH depletion, and electrostatic interaction between positively charged ZnO NWs and negatively charged cells make important contribution to the degree of ZnO NW arrays damaging cell membrane.

A tetrazole derivative 1-[(1H-tetrazol-5-ylimino)methyl]naphthalen-2-ol (H2L) as a fluorescent chemosensor for Al(3+) in DMSO and Zn(2+) in DMF was designed and synthesized. From (1)H NMR data, the Job plot and the ESI-MS spectrum, 1?:?1 stoichiometric complexation between H2L and Al(3+)/Zn(2+) was found in DMSO and DMF, respectively. The theoretical calculations at the level of B3LYP/6-311G** for the ground state and TD-B3LYP/6-311G** for the excited state revealed the sensing mechanism is the inhibition of excited state intramolecular proton transfer (ESIPT). And the possible fluorescent species formed in the DMSO and DMF solutions were deduced to be [Al(HL)(OH)(NO3)(H2O)(DMSO)] and [Zn(HL)(OH)(H2O)(DMF)]. What's more, it is confirmed that H2L could be used to detect Al(3+) and Zn(2+) in cells by bioimaging.

Although breast phyllodes tumors are rare, there is no effective therapy other than surgery. Little is known about their tumor biology. A malignant phyllodes tumor contains heterologous stromal elements, and can transform into rhabdomyosarcoma, liposarcoma and osteosarcoma. These versatile properties prompted us to explore their possible relationship to mesenchymal stem cells (MSCs) and to search for the presence of cancer stem cells (CSCs) in phyllodes tumors.

This paper reports a fluorescence chemosensor, N-(benzimidazol-2-yl)salicylaldimine (H2L), for Zn(II) and Al(III) ions. H2L has high selectivity for Al(III) in dimethyl sulfoxide (DMSO) and for Zn(II) in N,N-dimethylformamide (DMF). In methanol, Zn(II) and Al(III) could also be distinguished by H2L with different excitation wavelengths. The fluorescent species [Zn(HL)(H2O)(CH3OH)](+), [Zn(HL)(H2O)(DMF)](+), [Al(HL)2(OH)(H2O)], and [Al(HL)(OH)2(H2O)(DMSO)] formed in solution were established by a combination of experimental and theoretical methods, including Job's plot, (1)H NMR titration, electrospray inonization mass spectrometry (ESI-MS), and B3LYP-SCRF/6-31(d) and TD-B3LYP-SCRF/6-31G* density functional theory methods. The results show that Zn(II) and Al(III) are all coordinated to the imine nitrogen atom and the hydroxyl oxygen atom from H2L, which is the same as the M(2+) ions in the obtained mononuclear complexes [M(HL)2(CH3OH)2] (where M = Cd, Ni, Co, and Mg). The detection limits of H2L for Zn(II) were 5.98 ?M in methanol and 5.76 ?M in DMF, while the detection limits of H2L for Al(III) were 3.3 ?M in methanol and 5.25 ?M in DMSO. Furthermore, it is also confirmed that H2L has low toxicity for HeLa cells and could be used to detect Zn(II) and Al(III) ions in living cells by bioimaging.

Human METCAM/MUC18, a cell adhesion molecule (CAM) in the immunoglobulin-like gene super family, plays a dual role in the progression of several epithelium cancers; however, its role in the nasopharyngeal carcinoma (NPC) remains unclear. To initiate the study we determined human METCAM/MUC18 expression in tissue samples of normal nasopharynx (NP), NPCs, and metastatic lesions, and in two established NPC cell lines. Immunoblotting analysis was used for the determination in lysates of frozen tissues, and immunohistochemistry (IHC) for expression in formalin-fixed, paraffin-embedded tissue sections of 7 normal nasopharynx specimens, 94 NPC tissue specimens, and 3 metastatic lesions. Human METCAM/MUC18 was expressed in 100% of the normal NP, not expressed in 73% of NPC specimens (or expressed at very low levels in only about 27% of NPC specimens), and expressed again in all of the metastatic lesions. The level of human METCAM/MUC18 expression in NPC tissues was about one fifth of that in the normal NP and metastatic lesions. The low level of human METCAM/ MUC18 expression in NPC specimens was confirmed by a weak signal of RT-PCR amplification of the mRNA. Low expression levels of human METCAM/MUC18 in NPC tissues were also reflected in the seven established NPC cell lines. These findings provided the first evidence that diminished expression of human METCAM/MUC18 is an indicator for the emergence of NPC, but increased expression then occurs with metastatic progression, suggesting that huMETCAM/MUC18, perhaps similar to TGF-?, may be a tumor suppressor, but a metastasis promoter for NPC.

Intertrochanteric fracture is the most common hip fractures in elderly population. Cephalomedullary fixation, including proximal femoral nail antirotation (PFNA) and gamma nail, is commonly used in the treatment of intertrochanteric fractures. One of difficulties encountered in operation is insertion of the guide wire to femoral head. The guide wire would always bend and flow superiorly away from femoral calcar, and the lag screw and helical blade will follow the track and get into superior part of femoral head, not into femoral calcar. Here, we are presenting a surgical technique to direct the guide wire to right track. When guide wire just passed the tack hole of the nail, the rod of nail was slightly tapped down further to allow the superior border of the tack hole to touch the guide wire. Therefore, the superior border of the tack hole can be used as presser to prevent the wire from deviating upward. Through this way, the lag screw and helical blade get a good purchase that docked at femoral calcar. After that, the rod of nail was pulled back a little to make the wire in the middle of the tack hole, which can facilitate the insertion of the PFNA blade or lag screw. Our experience demonstrated that this surgical technique reduced the intraoperative risk for patients.

Intrinsically disordered domains have been reported to play important roles in signal transduction networks by introducing cooperativity into protein-protein interactions. Unlike intrinsically disordered domains that become ordered upon binding, the EF-SAM domain in the stromal interaction molecule (STIM) 1 is distinct in that it is ordered in the monomeric state and partially unfolded in its oligomeric state, with the population of the two states depending on the local Ca(2+) concentration. The oligomerization of STIM1, which triggers extracellular Ca(2+) influx, exhibits cooperativity with respect to the local endoplasmic reticulum Ca(2+) concentration. Although the physiological importance of the oligomerization reaction is well established, the mechanism of the observed cooperativity is not known. Here, we examine the response of the STIM1 EF-SAM domain to changes in Ca(2+) concentration using mathematical modeling based on in vitro experiments. We find that the EF-SAM domain partially unfolds and dimerizes cooperatively with respect to Ca(2+) concentration, with Hill coefficients and half-maximal activation concentrations very close to the values observed in vivo for STIM1 redistribution and extracellular Ca(2+) influx. Our mathematical model of the dimerization reaction agrees quantitatively with our analytical ultracentrifugation-based measurements and previously published free energies of unfolding. A simple interpretation of these results is that Ca(2+) loss effectively acts as a denaturant, enabling cooperative dimerization and robust signal transduction. We present a structural model of the Ca(2+)-unbound EF-SAM domain that is consistent with a wide range of evidence, including resistance to proteolytic cleavage of the putative dimerization portion.

In this work, Mg-Al CO3-layered double hydroxides (LDHs) were used as adsorbent materials for sodium dodecyl benzene sulfonate (SDBS) in aqueous solutions, the enriched SDBS can be directly detected by IO4(-)-H2O2 chemiluminescence (CL) system. The commonly existing cations cannot be enriched by Mg-Al CO3-LDHs due to the structurally positively charged layers of LDHs, while other adsorbed anionic interferents had no effect on the IO4(-)-H2O2 CL reaction. The corresponding linear regression equation was established in the range of 0.1-10 ?M for SDBS. The detection limit at a signal-to-noise (S/N) ratio of 3 for SDBS was 0.08 ?M. The relative standard deviation (RSD) for nine repeated measurements of 0.5 ?M SDBS was 2.6%. This proposed method has been successfully applied to the determination of SDBS in river water samples. To the best of our knowledge, we have first time coupled the high enrichment capacity of LDHs towards anions with CL detection for analytes.

Zhen-wu-tang (ZWT), a well-known formula in China, is widely used to treat chronic kidney diseases. However, very little information on ZWT's mechanism of action is currently available. In this study, we investigated the possible protective role and underlying mechanism of ZWT on nephrotic syndrome (NS) induced by Adriamycin (intravenous injection, 6.0?mg/kg) in rats using biochemical and histopathological approaches. ZWT decreased urine protein excretion and the serum levels of total cholesterol, triglycerides, blood urea nitrogen, and creatinine significantly in diseased rats. A decrease in plasma levels of total protein and albumin was also recorded in nephropathic rats. Pathological results show an improved pathological state and recovering glomerular structure in ZWT treatment groups. ZWT decreased renal IL-8 level but increased renal IL-4 level. In addition, rats subjected to ZWT exhibited less IgG deposition in glomerulus compared with model group. RT-PCR results showed that ZWT decreased the mRNA expression of NF- ? B p65 and increased the mRNA expression of I ? B. Furthermore, ZWT reduced the level of MDA and increased SOD activity. These results demonstrated that ZWT ameliorated Adriamycin-induced NS in rats possibly by inhibiting Adriamycin-induced inflammation damage, enhancing body's antioxidant capacity, thereby protecting glomerulus from injury.

D-Xylose utilization by Clostridium acetobutylicum, an important industrial microorganism used in ABE (Acetone, Butanol and Ethanol) production, has attracted increasing interests. We demonstrated previously that co-overexpression of genes, encoding d-xylose symporter, D-xylose isomerase and xylulokinase, improved D-xylose utilization by C. acetobutylicum (Xiao, H., et al., 2011. Applied and Environmental Microbiology 77, 7886-7895). Here, we further identified genes involved in PPP (Pentose Phosphate Pathway) in C. acetobutylicum and evaluated their contribution to d-xylose utilization. Among all the candidate genes, the CAC1347, CAC1348, CAC1730 and CAC2880 were validated to encode genes tal, tkl, rpe and rpi, four key genes involved in PPP, respectively. The following combined overexpression of these genes conferred a significantly improved xylose-utilizing ability to the recombinant strain, reaching a solvent titer 42% higher than that of the wild-type strain. This finding offers a useful strategy to optimize d-xylose utilization by C. acetobutylicum.

In spite of the advances in the diagnosis and treatment of bladder cancer, the prognosis of bladder cancer remains relatively poor. As a result, it is vital to identify novel diagnostic and prognostic marker of bladder cancer. A growing volume of literature has implicated the vital role of long noncoding RNA in the development of cancer. GHET1, a recently identified lncRNA, was initially characterized in gastric cancer. However, its role in bladder cancer remains largely unknown. In this study, we demonstrated that GHET1 was upregulated in bladder cancer tissues compared to adjacent normal tissues and its over-expression correlates with tumor size, advanced tumor and lymph node status, and poor survival. GHET1 knockdown suppressed the proliferation and invasion of bladder cancer cells in vitro. In the meantime, inhibition of GHET1 reversed the epithelial-mesenchymal-transition in bladder cancer cell line. Taken together, our study suggests that the potential use of GHET1 as a prognostic marker and therapeutic target of bladder cancer.

Soil salinity is an increasingly serious problem worldwide that reduces agricultural output potential. Selected beneficial soil bacteria can promote plant growth and augment tolerance to biotic and abiotic stresses. Bacillus subtilis strain GB03 has been shown to confer growth promotion and abiotic stress tolerance in the model plant Arabidopsis thaliana. Here we examined the effect of this beneficial soil bacterium on salt tolerance in the legume forage crop, white clover. Plants of white clover (Trifolium repens L. cultivar Huia) were grown from seeds with or without soil inoculation of the beneficial soil bacterium Bacillus subtilis GB03 supplemented with 0, 50, 100, or 150 mM NaCl water into soil. Growth parameters, chlorophyll content, malondialdehyde (MDA) content and osmotic potential were monitored during the growth cycle. Endogenous Na(+) and K(+) contents were determined at the time of harvest. White clover plants grown in GB03-inoculated soil were significantly larger than non-inoculated controls with respect to shoot height, root length, plant biomass, leaf area and chlorophyll content; leaf MDA content under saline condition and leaf osmotic potential under severe salinity condition (150 mM NaCl) were significantly decreased. Furthermore, GB03 significantly decreased shoot and root Na(+) accumulation and thereby improved K(+)/Na(+) ratio when GB03-inoculated plants were grown under elevated salt conditions. The results indicate that soil inoculation with GB03 promotes white clover growth under both non-saline and saline conditions by directly or indirectly regulating plant chlorophyll content, leaf osmotic potential, cell membrane integrity and ion accumulation.

Scorpion venom heat-resistant peptide (SVHRP) is a component purified from Buthus martensii Karsch scorpion venom. Although scorpions and their venom have been used in Traditional Chinese Medicine (TCM) to treat chronic neurological disorders, the underlying mechanisms of these treatments remain unknown. We applied SVHRP in vitro and in vivo to understand its effects on the neurogenesis and maturation of adult immature neurons and explore associated molecular mechanisms. SVHRP administration increased the number of 5-bromo-2'-dexoxyuridine (BrdU)-positive cells, BrdU- positive/neuron-specific nuclear protein (NeuN)-positive neurons, and polysialylated-neural cell adhesion molecule (PSA-NCAM)-positive immature neurons in the subventricular zone (SVZ) and subgranular zone (SGZ) of hippocampus. Furthermore immature neurons incubated with SVHRP-pretreated astrocyte-conditioned medium exhibited significantly increased neurite length compared with those incubated with normal astrocyte-conditioned medium. This neurotrophic effect was further confirmed in vivo by detecting an increased average single area and whole area of immature neurons in the SGZ, SVZ and olfactory bulb (OB) in the adult mouse brain. In contrast to normal astrocyte-conditioned medium, higher concentrations of brain-derived neurotrophic factor (BDNF) but not nerve growth factor (NGF) or glial cell line-derived neurotrophic factor (GDNF) was detected in the conditioned medium of SVHRP-pretreated astrocytes, and blocking BDNF using anti-BDNF antibodies eliminated these SVHRP-dependent neurotrophic effects. In SVHRP treated mouse brain, more glial fibrillary acidic protein (GFAP)-positive cells were detected. Furthermore, immunohistochemistry revealed increased numbers of GFAP/BDNF double-positive cells, which agrees with the observed changes in the culture system. This paper describes novel effects of scorpion venom-originated peptide on the stem cells and suggests the potential therapeutic values of SVHRP.

Nasopharyngeal carcinoma (NPC) is one of the common cancers in South China. It can easily invade into cranial nerves, especially in patients with local advanced disease. Despite the fact that the magnetic resonance imaging (MRI) findings are not always consistent with the symptoms of CN palsy, MRI is recommended for the detection of CN involvement (CNI). However, the prognostic impact of MRI-detected CNI in NPC patients is still controversial. To investigate the prognostic value of MRI detected CNI, we performed a retrospective analysis on the clinical data of 375 patients with NPC who were initially diagnosed by MRI. All patients had T3-4 disease and received radical intensity modulated radiation therapy (IMRT) as their primary treatment. The incidence of MRI-detected CNI was 60.8%. A higher incidence of MRI-detected CNI was observed in T4 disease compared with T3 disease (96.8% vs. 42.8%, P<0.001), and a higher incidence was also found in patients with Stage IV disease compared with those with Stage III disease (91.5% vs. 42.3%; P<0.001). The local relapse-free survival (LRFS), distant metastasis-free survival (DMFS), and overall survival (OS) of patients with T3 disease, with or without MRI-detected CNI, was superior to that of patients with T4 disease (P<0.05). No significant differences in LRFS, DMFS or OS were observed between T3 patients with or without MRI-detected CNI. The survival of Stage III patients with or without MRI-detected CNI was significantly superior to that of Stage IV patients (P<0.01), but there was no significant difference between Stage III patients with or without MRI-detected CNI for all endpoints. Therefore, when treated with IMRT, MRI-detected CNI in patients with NPC does not appear to affect the prognosis. In patients with clinical T3 disease, the presence of MRI-detected CNI is not sufficient evidence for defining T4 disease.

Background Lithium has been a first-line choice for maintenance treatment of bipolar disorders to prevent relapse of mania and depression, but many patients do not have a response to lithium treatment. Methods We selected subgroups from a sample of 1761 patients of Han Chinese descent with bipolar I disorder who were recruited by the Taiwan Bipolar Consortium. We assessed their response to lithium treatment using the Alda scale and performed a genomewide association study on samples from one subgroup of 294 patients with bipolar I disorder who were receiving lithium treatment. We then tested the single-nucleotide polymorphisms (SNPs) that showed the strongest association with a response to lithium for association in a replication sample of 100 patients and tested them further in a follow-up sample of 24 patients. We sequenced the exons, exon-intron boundaries, and part of the promoter of the gene encoding glutamate decarboxylase-like protein 1 (GADL1) in 94 patients who had a response to lithium and in 94 patients who did not have a response in the genomewide association sample. Results Two SNPs in high linkage disequilibrium, rs17026688 and rs17026651, that are located in the introns of GADL1 showed the strongest associations in the genomewide association study (P=5.50×10(-37) and P=2.52×10(-37), respectively) and in the replication sample of 100 patients (P=9.19×10(-15) for each SNP). These two SNPs had a sensitivity of 93% for predicting a response to lithium and differentiated between patients with a good response and those with a poor response in the follow-up cohort. Resequencing of GADL1 revealed a novel variant, IVS8+48delG, which lies in intron 8 of the gene, is in complete linkage disequilibrium with rs17026688 and is predicted to affect splicing. Conclusions Genetic variations in GADL1 are associated with the response to lithium maintenance treatment for bipolar I disorder in patients of Han Chinese descent. Cell-to-cell communication is a very important physiological behavior in life entity, and most of human behaviors are related to it. Although cell-to-cell communications are attracting much attention and financial support, rare methods have been successfully developed for in vitro cell-to-cell communication study. In this work, we developed a novel method for cell-to-cell communication study on an integrated microdevice, and signaling molecule and metabolites were online-detected by an electrospray ionization-quadrupole-time-of-flight-mass spectrometer (ESI-Q-TOF-MS) after on-chip solid-phase extraction. Moreover, we presented a "Surface Tension Plug" on a microchip to control cell-to-cell communication. The microdevice consists of three functional sections: cell coculture channel, targets pretreatment, and targets detection sections. To verify the feasibility of cell-to-cell communications on the integrated microdevice, we studied the communication between the 293 and the L-02 cells. Epinephrine and glucose were successfully detected using an ESI-Q-TOF-MS with short analysis time (<10 min). The results demonstrated that the developed microfluidic device is a potentially useful tool for high throughput cell-to-cell communication study.

Plasmonic waveguides essentially support only transverse magnetic modes. A novel plasmonic waveguide consisting of hybrid plasmonic waveguides in both vertical and horizontal directions is proposed to reduce the polarization dependence. In a combined waveguide, surface plasmon polariton (SPP) modes polarized in both vertical and horizontal directions exist in the correspondingly oriented hybrid plasmonic waveguide. In an optimized structure, anticrossing mode coupling is observed between these two SPP modes with a low birefringence by finite-difference time-domain simulation. The energy flux clearly shows the polarization-selective coupling between the polarized guided modes in the feeding silicon waveguide and those in the combined waveguide. Coupling efficiency above 65% is obtained for both polarizations. The proposed plasmonic combined waveguide has a potential application in guiding and processing of light from a fiber with a random polarization state.

A novel graphene photodetector with graphene embedded in a metamaterial perfect absorber (MPA) is proposed and its light absorption property is numerically investigated. Absorption above 40% in graphene is obtained, which is over 17 times that of a bare monolayer graphene. Both horizontally and vertically cascaded MPAs integrated with graphene are also investigated.

We have demonstrated for the first time the suitability of fluorosurfactant-capped spherical gold nanoparticles as HPLC postcolumn colorimetric reagents for the direct assay of cysteine, homocysteine, cystine, and homocystine. The success of this work was based on the use of an on-line tris(2-carboxyethyl)phosphine reduction column for cystine and homocystine. Several parameters affecting the separation efficiency and the postcolumn colorimetric detection were thoroughly investigated. Under the optimized conditions, cysteine, homocysteine, cystine, and homocystine in human urine and plasma samples were determined. Detection limits for cysteine, homocysteine, cystine, and homocystine ranged from 0.16-0.49 ?M. The accuracy in terms of recoveries ranged between 94.0-102.1%. This proposed method was rapid, inexpensive, and simple.

Water pollution by wastewater discharge can cause the problem of disinfection byproducts (DBPs) in drinking water supply. In this study, DBP formation characteristics of soluble microbial products (SMPs) as the main products of wastewater organic biodegradation were investigated. The results show that SMPs can act as DBP precursors in simulated wastewater biodegradation process. Under the experimental conditions, stabilized SMPs had DBPFP (DBP formation potential) yield of around 5.6?molmmol(-1)-DOC (dissolved organic carbon) and DBP speciation profile different from that of the conventional precursor, natural organic matter (NOM). SMPs contained polysaccharides, proteins, and humic-like substances, and the latter two groups can act as reactive DBP precursors. SMP fraction with molecular weight of <1kDa accounted for 85% of the organic carbon and 65% of the DBP formation. As small SMP molecules are more difficult to remove by conventional water treatment processes, more efforts are needed to control wastewater-derived DBP problem in water resource management.

We utilized a metal tantalum (Ta) ball-probe to measure the electrical properties of vertical-aligned one-dimensional (1D) nickel-oxide (NiO) nanorods. The 1D NiO nanorods (on average, ~105 nm wide and ~700 nm long) are synthesized using the hot-filament metal-oxide vapor deposition (HFMOVD) technique, and they are cubic phased and have a wide bandgap of 3.68 eV. When the 1D NiO nanorods are arranged in a large-area array in ohmic-contact with the Ta ball-probe, they acted as many parallel resistors. By means of a rigorous calculation, we can easily acquire the average resistance RNR and resistivity ?NR of a single NiO nanorod, which were approximately 3.1 × 10(13) ? and 4.9 × 10(7) ?.cm, respectively.

A chemosensor for the Al(3+) ion, 1-[(3-hydroxypyridin-2-ylamino)methylene]naphthalen-2(1H)-one (H2L), based on inhibited excited-state intramolecular proton transfer was synthesized. The experimental and theoretical calculations at B3LYP+PCM/6-31G(d) revealed that Al(3+) and H2L form a 1:1 complex, [AlL(OH)(H2O)]2, in dimethyl sulfoxide that exhibits two remarkably enhanced fluorescent emissions at 523 and 553 nm. It is confirmed that H2L could be used to detect Al(3+) ions in cells by bioimaging.

Background: We investigate factors associated with carotid blowout in the cervical portion of head and neck patients in a large cohort. Methods: We reviewed head and neck cancer patients and identified those with carotid blowout. Variables of patients with and without carotid blowout were compared and associated factors were investigated by logistic regression model. Results: A total of 2590 patients were included in the final analysis and 102 patients (3.9%) developed carotid blowout in the cervical region. Body mass index < 22.5 kg/M(2) , primary site of hypopharynx or oropharynx, open wound in the neck requiring wet dressing, radical neck dissection, and total radiation dose to the neck >= 70 Gy were independent factors associated with carotid blowout. Conclusions: After completion of treatment in head and neck cancer patients, carotid blowout was an uncommon complication with catastrophic results. OBJECTIVES: To investigate the protective effects of Sapindus saponins in spontaneously hypertensive rats, and the possible cellular and molecular mechanisms. METHODS: Thirty-two 16-week-old spontaneously hypertensive rats were randomly divided into four groups (8 in each group): model group (placebo), positive control group (27 mg/kg of Captopril Tablets), Sapindus saponins groups (27 mg/kg and 108 mg/kg, respectively). Another 8 healthy Wistar-Kyoto strain (WKY) rats were used as the normal group. The animals were treated for 8 weeks. Blood pressure of rats was determined by non-invasive blood pressure meter (BP-6). Furthermore, the contents of angiotensin II (AngII) in plasma and myocardial tissue were determined by enzyme-linked immunosorbent assay (ELISA), the gene expression of receptor angiotensin type 1 (AT1R) in aorta was determined by quantitative realtime polymerase chain reaction (qRT-PCR). The protein expression of transforming growth factor-?1 (TGF-?1) and AT1R in heart was determined by immunohistochemical staining. The protein expression of p-phosphorylation of p38 mitogen-activated protein kinase (p-p38MAPK) was determined by Western blotting. The contents of interleukin (IL)-1, IL-6 and tumor necrosis factor (TNF) in serum were determined by radioimmunoassay. And the histopathological and morphological changes of aorta and heart tissue samples were assessed semi-quantitatively by hematoxylin-eosin (HE) or Masson staining. RESULTS: Thirty minutes after single or continuous treatment, systolic blood pressure (SBP) was reduced significantly in Sapindus saponins groups. And the contents of AngII, IL-1, IL-6 and TNF-? in serum, the expression of AT1R mRNA, p-p38MAPK and TGF-?1 were significantly suppressed dose-dependently (P<0.05 or P<0.01). With the Sapindus saponins treatment, compared with those of the model group, the cardiac and aortic pathological changes were ameliorated significantly. CONCLUSIONS: Our findings suggest that Sapindus saponins might have protective effects in spontaneously hypertensive rats, the cellular and molecular mechanisms of which might be relevant to the regulation of inflammatory responses mediated by p-p38MAPK signal pathway based on activated Ang II and AT1R.

In developing our video relationships, we compare around 5 million PubMed articles to our library of over 4,500 methods videos. In some cases the language used in the PubMed abstracts makes matching that content to a JoVE video difficult. In other cases, there happens not to be any content in our video library that is relevant to the topic of a given abstract. In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
