Effects of Supplemental Boron on Growth Performance and Meat Quality in African Ostrich Chicks.

To investigate the effects of boron on growth performance and meat quality, 10-day-old Africa ostrich chicks were randomly divided into 6 groups with 6 replicates in each group. For 80 days, birds in the treatments were fed the same basal diet but given different concentrations of boron-supplemented water. The highest final BW (33.4 ± 0.30 kg), ADFI (376 ± 1.83 g), and ADG (224 ± 1.01 g) appeared in the group receiving 160 mg/L boron (group 4). 160 mg/L boron also decreased drip loss (2.20 ± 0.59), cooking loss (35.3 ± 1.14), and elevated pH value (6.13 ± 0.28) of meat (P < 0.05). Ostrich chicks in the 640 mg/L treatment group (group 6) had the lowest final BW (30.8 ± 1.05 kg) and ADG (208 ± 0.74 g) (P < 0.05). The highest ash (1.35 ± 0.01%) and pH (6.18 ± 0.03) and the lowest protein (20.4 ± 1.74%), drip loss (2.10 ± 0.76%), cooking loss (35.0 ± 0.41%), C18:1 (28.2 ± 0.65%), and C18:3?3 (2.60 ± 0.51%) appeared in group 6 (P < 0.05) as well. Overall, the optimum concentration of 160 mg/L supplemental boron improved ostrich growth performance and meat quality; however, high concentrations of boron decreased both performance and meat quality.

5,10-Methylenetetrahydrofolate Reductase Polymorphisms and Colon Cancer Risk: a Meta-analysis.

Previous studies investigating the association between 5,10-methylenetetrahydrofolate reductase (MTHFR) gene polymorphisms and colon cancer risk have generated conflicting results. The aim of our meta-analysis was to clarify the precise association. A systematic literature search was conducted to identify all relevant studies. Pooled odds ratio (ORs) with 95% confidence interval (CI) were used to estimate the strength of the association. In this meta-analysis, a total of 13 articles, involving 5,386 cases and 8,017 controls met the inclusion criteria. Overall, a significant association was found between colon cancer risk and the MTHFR C667 polymorphism (TT vs CC+CT: OR=0.79; 95%CI=0.65-0.96; p=0.017). Stratification by ethnicity revealed that MTHFRC667 was associated with colon cancer risk in the non-Asian group (TT vs CC+CT:OR=0.77, 95%CI=0.68-0.89, p=0.000; TT vs CC: OR=0.84, 95%CI=0.73-0.97, p=0.016). Stratification by source of control indicated that MTHFR C667 also correlated with colon cancer risk in the population-based subgroup (TT vs CC: OR=0.85, 95%CI=0.74-0.97, p=0.017; TT vs CC+CT: OR=0.78, 95%CI=0.68-0.89, p=0.000) and hospital-based subgroup (TT vs CC+CT: OR=0.65, 95%CI=0.49-0.86, p=0.003). However, risk was significantly increased for MTHFR A1298C polymorphisms and colon cancer risk in hospital-based studies (C vs A: OR=1.52, 95%CI=1.26-1.83, p=0.000; CC+AC vs AA: OR=1.93, 95%CI=1.47-2.49, p=0.000) but reduced in population-based studies (CC vs AA: OR=0.83, 95%CI=0.70-0.99, p=0.042). In conclusion, the results of our meta-analysis suggest that the MTHFR C667 polymorphism is associated with reduced colon cancer risk, especially for non-Asian populations.

This study was aimed to investigate the abnormal expression of PDGFRA gene in eosinophilia by FISH. Translocations of PDGFRA gene in 13 patients with eosinophilia were detected by using 4q12 three-color probe and FISH technology. Fifteen people were used as control to establish the normal cut-off value of fluorescence signal of PDGFRA. The results indicated that 1 out of 13 patients with eosinophilia was corrected and was diagnosed as CML. The fusion gene of FIP1L1-PDGFRA (F/P) was found in 2 patients and the positive rate of F/P fusion gene detected by probe 4q12 was 17% in the 12 patients with eosinophilia. Other translocation forms involving PDGFRA gene were not found. It is concluded that a variety of translocation forms of PDGFRA gene can be detected in patients with eosinophilia by using 4q12 three-color probe and FISH technology, which can provide important information for assessing diagnosis and treatment.

The condition of water cycles in Tibet Plateau is a complex process, and the hydrogen and oxygen isotopes contain important information of this process. Based on the analysis of isotopic composition of freshwater lake, saltwater lake and geothermal water in the southern Tibetan Plateau, this study investigated water cycling, composition and variation of hydrogen and oxygen isotopes and the influencing factors in the study area. The study found that the mean values of delta18O and deltaD in Daggyaima lake water (-17.0 per thousand for delta18O and -138. 6 per thousand for deltaD), Langcuo lake water (-6.4 per thousand for delta18O and -87.4 per thousand for deltaD) and Dagejia geothermal water (-19.2 per thousand for delta18 and -158.2 per thousand for deltaD) all showed negative delta18O and deltaD values in Tibetan Plateau by the influence of altitude effects. Lake water and geothermal water were influenced by evaporation effects in inland arid area, and the slope of evaporation line was less than 8. Deuterium excess parameters of lake water and geothermal water were all negative. The temperature of geothermal reservoirs in Dagejia geothermal field was high,and oxygen shift existed in the relationship of hydrogen and oxygen isotopes.

The aim of this study was to evaluate the outcomes of multiple advanced surgical treatments (i.e. microsurgery, laparoscopic surgery and endoscopic surgery) for acquired obstructive azoospermia. We analyzed the surgical outcomes of 51 patients with suspected acquired obstructive azoospermia consecutively who enrolled at our center between January 2009 and May 2013. Modified vasoepididymostomy, laparoscopically assisted vasovasostomy and transurethral incision of the ejaculatory duct with holmium laser were chosen and performed based on the different obstruction sites. The mean postoperative follow-up time was 22 months (range: 9 months to 52 months). Semen analyses were initiated at four postoperative weeks, followed by trimonthly (months 3, 6, 9 and 12) semen analyses, until no sperm was found at 12 months or until pregnancy was achieved. Patency was defined as >10,000 sperm ml-1 of semen. The obstruction sites, postoperative patency and natural pregnancy rate were recorded. Of 51 patients, 47 underwent bilateral or unilateral surgical reconstruction; the other four patients were unable to be treated with surgical reconstruction because of pelvic vas or intratesticular tubules obstruction. The reconstruction rate was 92.2% (47/51), and the patency rate and natural pregnancy rate were 89.4% (42/47) and 38.1% (16/42), respectively. No severe complications were observed. Using multiple advanced surgical techniques, more extensive range of seminal duct obstruction was accessible and correctable; thus, a favorable patency and pregnancy rate can be achieved.

Scientific research output of clinical trials in Chinese medicine (CM) is insufficient, but international papers hold an important scientific position in China. Based on the current situation, we analyzed the present publication situation of international medical papers in our country, and the feasibility and urgency of publishing international papers on clinical trials of CM. Finally, we proposed to use the PDCA (plan-do-check-action) cycle method to improve the quality control and management of CM clinical trials. Moreover, by combining our experience in relevant scientific research launched at our department, we expounded strategies for improving the quantity and quality of international papers in CM.

Occurrence of new toxic chemicals in sludge from wastewater treatment plants (WWTPs) is of concern for the environment and human health. Alternative halogenated flame retardants (HFRs) are a group of potentially harmful organic contaminants in the environment. In this study, a nationwide survey was carried out to identify the occurrence of HFRs and their potential dehalogenated products in sewage sludge from 62 WWTPs in China. Of all 20 target chemicals analyzed, decabromodiphenyl ethane (DBDPE), hexabromocyclododecane (HBCD) and 1, 2-bis (2,4,6-tribromophenoxy)-ethane (BTBPE) were detected in all sludge samples, and the concentrations were in the range of 0.82-215, 0.09-65.8, and 0.10-2.26 ng g(-1) d.w., respectively. Dechlorane Plus (DP) was found in 60 of 62 samples, and the concentration ranged from nd-298 ng g(-1) with a mean of 18.9 ng g(-1) d.w. The anti-DP fractional abundance fanti (0.79) in the samples was much higher than the commercial DP composition (fanti = 0.59), indicating a stereoselective degradation. Comparison with global sludge concentrations of HFRs indicate that China is at the medium pollution level in the world. Principal components analysis revealed that strong correlations existed between ln-transformed concentrations (natural logarithm) of the dominant BFRs and total organic carbon (TOC) as well as industrial wastewater proportion, influent volume and serving population. Significant linear relationships (R = 0.360-0.893, p < 0.01) were found among emerging brominated flame retardants (BFRs), suggesting their common commercial applications and release sources to the environment. Two kinds of dehalogenated products, pentabromocyclododecane (PBCD) and undecachloropentacyclooctadecadiene (Cl11-DP), derived from HBCD and DP, were also identified in sewage sludge for the first time.

Two new ionone glycosides, named frehmaglutoside G (1) and frehmaglutoside H (2), together with six known compounds, rehmapicroside (3), sec-hydroxyaeginetic acid (4), dihydroxy-?-ionone (5), trihydroxy-?-ionone (6), rehmaionoside A (7) and rehmaionoside C (8), were isolated from the 95% EtOH extract of the dried roots of Rehmannia glutinosa Libosch. Their structures were determined on the basis of extensive spectroscopic analyses, including HR-ESI-MS, UV, IR, 1D and 2D NMR ((1)H-(1)H COSY, HSQC, HMBC and NOESY) methods. The absolute configurations were confirmed via the circular dichroism spectra.

Hepatitis B surface antigen (HBsAg) quantification has garnered attention because of its high predictive value in determining treatment responses. The HBsAg quantification assays, such as Architect and Elecsys, are commercially available, and more assays are in development. We aimed to compare the results of the Architect and Elecsys assays with those of a new assay, WTultra. The WTultra HBsAg assay is a sandwich chemiluminescent microplate enzyme immunoassay and provides an alternative choice which is more cost-effective and potentially applicable in developing or resource-constrained countries and areas. A total of 411 serum samples were collected from patients during various phases of chronic hepatitis B (CHB) infection. The samples were assessed using the three assays, and the results were compared and analyzed. The results for the Architect, Elecsys, and WTultra assays were well correlated according to the overall results for the samples (correlation coefficients, rArchitect versus WTultra = 0.936, rArchitect versus Elecsys = 0.952, and rWTultra versus Elecsys = 0.981) and the various infection phases (rArchitect versus WTultra ranging from 0.67 to 0.975, rArchitect versus Elecsys ranging from 0.695 to 0.982, and rWTultra versus Elecsys ranging from 0.877 to 0.99). Additionally, consistent results were observed according to genotype (genotype B: rArchitect versus WTultra = 0.976, rArchitect versus Elecsys = 0.978, and rWTultra versus Elecsys = 0.979; genotype C: rArchitect versus WTultra = 0.950, rArchitect versus Elecsys = 0.963, and rWTultra versus Elecsys = 0.981) and hepatitis B virus (HBV) DNA levels (rArchitect = 0.540, rWTultra = 0.553, and rElecsys = 0.580). In conclusion, the Elecsys and WTultra assays were well correlated with the Architect assay, irrespective of the CHB infection phase or genotype. All of these assays are reliable for HBsAg quantification.

Ephrin type-A receptor 1 (EPHA1) (11771145) was documented to be one of the most strongly associated locus with Alzheimer's disease (AD) in a recent meta-analysis of five genome wide association studies. However, its contribution to the pathogenesis of AD remains unclear to date. Here, we addressed the role of EPHA1 in AD by investigating the influence of EPHA1 on cerebrospinal fluid and neuroimaging biomarkers in three clinical stages from the Alzheimer's Disease Neuroimaging Initiative (ADNI) database. We did not detect significant association of EPHA1 with amyloid-? deposition or tau protein. However, the A-allele in the mild cognitive impairment group remarkably prevented hippocampal atrophy (partial correlation coefficient 2.812, 95% CI 0.651 to 4.973) at two-year follow-up. Additionally, AD subjects with the A-allele displayed less atrophy and greater cerebral metabolic rate for glucose (CMRgl) in the right lateral occipitotemporal gyrus (volume: partial correlation coefficient 540.10, 95% CI 247.26 to 832.95; CMRgl: partial correlation coefficient 0.056, 95% CI 0.024 to 0.087) and inferior temporal gyrus (volume: partial correlation coefficient 327.98, 95% CI 11.65 to 644.31; CMRgl: partial correlation coefficient 0.055, 95% CI 0.019 to 0.091) at baseline. This study suggests EPHA1 (rs11771145) interferes with the pathological alteration of the hippocampus and the lateral occipitotemporal and inferior temporal gyri throughout the AD process, leading to a lower risk of AD. However, the limited sample size and follow-up as well as the diversity across ethnicities precluded explanation of these findings.

The objective of this study was to compare the efficacy and safety of levofloxacin 750 mg for 5 days versus 500 mg for 7-14 days intravenous (IV) in the treatment of community-acquired pneumonia (CAP). This clinical trial was the first of its kind conducted in Chinese people and also in Asian population. A total of 241 were enrolled and randomized to 750 mg group (n = 121) or 500 mg (n = 120) group from 10 study centers. The median treatment duration was 5.0 days in 750 mg and 9.0 days in 500 mg group. The median total dose was 3750 mg in 750 mg and 4500 mg in 500 mg group. The bacterial eradication rate was 100% in both groups. The overall efficacy rate in 750 mg group was 86.2% (94/109), and 84.7% (94/111), in 500 mg group of full analysis set visit 4, 95% confidence interval of 1.6% (-7.8-10.9%); the statistical results showed that 750 mg group was non-inferior to 500 mg group. The most common clinical adverse drug reactions were injection site adverse reactions in both 750 mg group and 500 mg group; the other common adverse drug reactions were insomnia, nausea, skin rash, etc. The most common drug-related laboratory abnormalities were neutrophil percentage decreased, decreased white blood cell count, alanine aminotransferase, and aspartate aminotransferase elevation in both 750 mg group and 500 mg group. Most of adverse drug reactions were mild in severity and well-tolerated. In summary, the regimen of levofloxacin 750 mg IV for 5 days was at least as effective and well tolerated as 500 mg IV for 7-14 days for the treatment of CAP.

Avian ?-defensins (AvBDs) are a family of small antimicrobial peptides that play important roles in the innate immunity of birds. Herein, we report on two new ostrich AvBD genes, AvBD2 and AvBD7, which were isolated from the bone marrow of ostriches (Struthio camelus). The coding regions of ostrich AvBD2 and AvBD7 comprised 195 bp and 201bp, which encoded 64 and 66 amino acids, respectively. Homology analysis showed that ostrich AvBD2 had the highest similarity (up to 86%) with the swan goose (Anser cygnoides) AvBD2, while ostrich AvBD7 shared the highest similarity (up to 81%) with chicken AvBD7. Analysis of the codon-usage bias showed that the two ostrich AvBDs had different codon-usage patterns from other AvBDs. The two synthetic AvBD peptides exhibited antibacterial activities against both Gram-positive and Gram-negative bacteria, and these activities decreased significantly in the presence of 100mM NaCl (P<0.01). Real-time reverse transcription-polymerase chain reaction analysis showed that AvBD2 and AvBD7 were widely expressed at different levels in 17 different tissues. This is the first report of the nucleotide sequences of ostrich AvBDs. Further investigations of these two AvBDs may help us to gain new insights into the immune defense system of the ostrich and to make subsequent therapeutic use of ostrich defensins.

Abstract Objectives: Previous published data indicated that interleukin-21 (IL-21) gene polymorphisms were shown to associate with multiple autoimmune diseases (ADs), but the results remain inconclusive. The aim of this study was to perform a meta-analysis to assess the overall association between IL-21 gene polymorphisms (rs907715, rs2221903) and multiple ADs. Methods: All eligible case-control studies were searched in the PubMed and Embase database. A meta-analysis was conducted on the association between the IL-21 gene variants and ADs using: (1) allelic contrast, (2) homozygote contrast, (3) the recessive model, and (4) the dominant model. Results: A total of 12 relevant studies including 10?535 cases and 19?356 controls were enrolled in this meta-analysis. A significant association between IL-21 rs907715 gene polymorphism and AD was found under the allelic (OR: 1.102, 95% CI: 1.057-1.149, p?=?0.000), homozygous (OR: 1.220, 95% CI: 1.089-1.368, p?=?0.001), dominant (OR: 1.160, 95% CI: 1.027-1.309, p?=?0.017), and recessive genetic model (OR: 1.119, 95% CI: 1.055-1.187, p?=?0.000) among Caucasian populations. However, there was no significant association between IL-21 rs2221903 polymorphism and AD in different genetic models. Conclusions: Data from the present study suggest that the IL-21 rs907715 polymorphism might be associated with multiple ADs susceptibility in Caucasians. Especially, the allele G of intronic rs907715 in IL-21 confers increased risk of ADs.

No randomised trials have been done to establish the best treatment for the disease. Ghrelin alleviates early brain injury after subarachnoid hemorrhage via the PI3K/Akt signaling pathway.

Early brain injury (EBI) plays a key role in the pathogenesis of subarachnoid hemorrhage (SAH). Although the neuroprotective effects of ghrelin have been demonstrated in several studies, whether ghrelin reduces EBI after SAH remains unknown. In this study, we hypothesized that treatment with ghrelin would attenuate EBI after SAH, and that this protection would be mediated, at least in part, by activation of the PI3K/Akt signaling pathway. Adult male Sprague-Dawley rats (n=100) were randomly divided into the following groups: control group (n=20), SAH group (n=20), SAH+vehicle group (n=20), SAH+ghrelin group (n=20) and SAH+ghrelin+LY294002 group (n=20). The rats were injected with autologous blood (0.3mL) into the prechiasmatic cistern to induce SAH. Ghrelin (80?g/kg, IP), or an equal volume of vehicle, was administered immediately after surgery. The PI3K inhibitor, LY294002, was applied to manipulate the proposed pathway. Mortality, neurological scores, brain edema, cell apoptosis, and the expression of p-Akt, and cleaved caspase-3 proteins were assayed after 24h SAH. Ghrelin significantly improved neurological function and reduced neuronal apoptosis and brain edema at 24h after SAH. The level of p-Akt, expressed mainly in neurons, was markedly up-regulated. Additionally, the level of cleaved caspase-3 was decreased by ghrelin treatment. The beneficial effects of ghrelin in SAH rats were partially suppressed by LY294002. These results demonstrate that ghrelin may reduce EBI after SAH, via a mechanism involving the PI3K/Akt signaling pathway.

Measurement of the sound transmission characteristics of normal neck tissue using a reflectionless uniform tube.

Understanding the sound transmission of the neck tissue is necessary and important in areas such as vocal function evaluation and electrolarynx improvement. In this paper, a simple method using a reflectionless tube was proposed to measure the neck frequency response function (NFRF) of ten normal subjects (five males and five females) during Mandarin vowel production. The NFRFs across different subjects producing different vowels were measured at different neck positions and compared to confirm the effectiveness of the method, and determine the NFRF variations in normal subjects. The results showed that the proposed method offered an easy and effective way to obtain an accurate NFRF. For normal subjects, the neck tissue can be treated as a low-pass filter, with a maximum gain at 310?Hz and a roll-off at a slope of -8.4?dB/octave, flattening out above 2000?Hz. The measurement position on the neck did not influence the shape of the NFRF, but did change the overall gains of the NFRF. In addition, there was a significant gender difference in NFRFs at the low frequencies. Finally, some potential applications of this method and the results are suggested.

Sugar beet (Beta vulgaris) is a biennial root crop that grows vegetatively in the first year and starts shoot elongation (bolting) and flowering after exposure to cold temperatures over winter. Early bolting before winter is controlled by the dominant allele of the B locus. Recently, the BOLTING time control 1 (BTC1) gene has been cloned from this locus. BTC1 promotes early bolting through repression of the downstream bolting repressor B. vulgaris flowering locus T1 (BvFT1) and activation of the downstream floral activator BvFT2. We have identified a new bolting locus B2 acting epistatically to B. B2 houses a transcription factor which is diurnally regulated and acts like BTC1 upstream of BvFT1 and BvFT2. It was termed BvBBX19 according to its closest homolog from Arabidopsis thaliana. The encoded protein has two conserved domains with homology to zinc finger B-boxes. Ethyl methanesulfonate-induced mutations within the second B-box caused up-regulation of BvFT1 and complete down-regulation of BvFT2. In Arabidopsis, the expression of FT is promoted by the B-box containing protein CONSTANS (CO). We performed a phylogenetic analysis with B-box genes from beet and A. thaliana but only BvCOL1 clustered with CO. However, BvCOL1 had been excluded as a CO ortholog by previous studies. Therefore, a new model for flowering induction in beet is proposed in which BTC1 and BvBBX19 complement each other and thus acquire a CO function to regulate their downstream targets BvFT1 and BvFT2.

Systemic lupus erythematosus (SLE) is a systemic autoimmune disease, with heterogeneous presentation. The aim of this study was to examine the association of a single-nucleotide polymorphism, rs7396562, of the interferon induced with single immunoglobulin IL-1-related receptor (SIGIRR) gene with SLE in a Chinese population. A total of 741 SLE patients and 731 healthy control subjects were enrolled in the present study. The genotyping of polymorphism (rs7396562) was determined by TaqMan allele discrimination assay on the 7,300 real-time polymerase chain reaction system. The frequency of T allele for rs7396562 in patients was significantly higher than in controls (T versus G, OR=1.318, 95 % confidence interval (CI)=1.139-1.525, P<0.001). Fortunately, some significant difference in genotype distribution was found between cases and controls (P<0.001). We also found some significant evidence for the association of the SIGIRR rs7396562 polymorphism with SLE between dominant and recessive model (TG+TT versus GG, P=0.002; TT versus TG+GG, P=0.002). We also analyzed the association of the SIGIRR rs7396562 T allele with clinical features; luckily, photosensitivity and malar rash had some significant signal with the SNP. In conclusion, our study represents the first report demonstrating an association of the SIGIRR rs7396562 polymorphism with SLE susceptibility in a Chinese population.

There have been few studies of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) in environmental water because of the large volume of water required for PCDD/Fs analysis. Water quality directly affects aquatic organisms, and little is known about how PCDD/Fs are transported in aquatic environments. PCDD/Fs were analyzed in eight water samples from Dongting Lake, China, which was contaminated with PCDD/Fs because of sodium pentachlorophenate use between the 1960s and the 1980s. The total PCDD/F concentrations in the samples were 36-345 pg L(-1), and the mean was 191 pg L(-1). Octachlorodibenzo-p-dioxin was the most abundant PCDD/F congener in every sample, contributing 67-95% of the total 2,3,7,8-chlorinated PCDD/F concentrations. The toxic equivalent (WHO-TEQ) concentrations in the samples were 0.17-0.37 pg L(-1), and the mean was 0.28 pg L(-1), which is higher than the Canadian environmental quality guideline (0.038 pg L(-1) WHO-TEQ for freshwater) and the United States Environmental Protection Agency water quality criterion (0.014 pg L(-1) WHO-TEQ). PCDD/Fs were also determined in six fish species collected from Dongting Lake, to assess the concentrations, accumulation patterns, and potential for toxic effects. The total 2,3,7,8-chlorinated PCDD/F concentrations in the fish samples were 2.2-17.9 pg g(-1) (wet weight), and the dominant congeners were octachlorodibenzo-p-dioxin, 1,2,3,4,6,7,8-heptachlorodibenzo-p-dioxin, 1,2,3,4,7,8-hexachlorodibenzo-p-dioxin, and 2,3,4,7,8-pentachlorodibenzofuran. The PCDD/F WHO-TEQs were 0.10-0.92 ww (3.3-65.3l w) pg g(-1) in different species of fish. PCDD/F congener patterns in fish may be affected by food chain biomagnification and the lipid content of the species.

With the ion-exchange resin method, the atmospheric nitrogen wet deposition in Beijing urban area within the Fifth Ring Road was investigated from June to October, 2012. The relationship between atmospheric nitrogen wet deposition and rainfall precipitation was investigated, the differences of nitrogen wet deposition in different months, different ring roads (the Fifth Ring Road, the Fourth Ring Road, the Third Ring Road and the Second Ring Road) and different functional areas (institutes and colleges district, ring-road, residential areas, railway station and public garden) were also investigated. The results showed that the average value and standard deviation of ammonia-nitrogen, nitrate-nitrogen and nitrite-nitrogen were significantly different during different months in 2012. The atmospheric nitrite nitrogen deposition first decreased and then increased, the maximum value appeared in September. The positive relationships between ammonia nitrogen (nitrate nitrogen) and mean monthly precipitation and negative relationships between nitrite nitrogen and mean monthly precipitation were both significant (P < 0.05). The three nitrogen depositions of ring-road and railway station were higher than other functional areas, but only the nitrite nitrogen deposition had obvious regional difference. The differences of the three nitrogen depositions among different ring roads were all not significant and it meant that the nitrogen wet deposition was equally distributed in Beijing urban area.

Triptolide is a diterpene triepoxide, extracted from the Chinese herb Tripterygium wilfordii Hook F, which has been shown to have antitumor activity in a number of cancers. Neuroblastoma is an aggressive extracranial pediatric solid tumor, with significant chemotherapeutic resistance. In this study, triptolide was hypothesized to be a potential therapeutic agent for neuroblastoma. The effects of triptolide on neuroblastoma cell growth and tumor development were investigated. Cell growth and proliferation were evaluated using a cell counting kit?8 assay and a 5-bromo-2-deoxyuridine staining assay. Cell cycle and apoptosis were detected by flow cytometry. Reverse transcription?quantitative polymerase chain reaction was conducted to detect the expression levels of the apoptosis?associated proteins, caspase?3 and caspase?9. The tumorigenicity of neuroblastoma cells was assessed by a soft agar clonogenic assay and an in vivo tumorigenic assay. The results demonstrated that exposure of BE(2)?C human neuroblastoma cells to triptolide resulted in a reduction in cell growth and proliferation, and the induction of cell death and apoptosis, together with cell cycle arrest in the S phase. A soft agar assay indicated that triptolide inhibited the colony?forming ability of BE(2)?C neuroblastoma cells. The xenograft experiment showed that triptolide significantly reduced tumor growth and development in vivo. The data suggested that this Chinese herb may be a potential novel chemotherapeutic agent for neuroblastoma.

Polychlorinated naphthalenes (PCNs) were nominated as persistent organic pollutants candidate in the Stockholm Convention in 2011. In this study, the profiles, concentrations and spatial distributions of PCNs were analyzed in 30 sewage sludge samples from wastewater treatment plants (WWTPs) in China. Concentrations of ?75PCNs in sludge samples were in the range of 1.05-10.9 ng/g dry weight (dw) with a mean value of 3.98 ng/g dw. The predominant homologues in the sludge were mono- to tetra-CNs, accounting for approximately 85% of total PCNs. The total toxic equivalent quantities (TEQs) of dioxin-like PCN congeners ranged from 0.04 to 2.28 pg/g dw with a mean value of 0.36 pg/g dw, which were lower than the maximum permissible TEQ concentrations in sludge for land application in China. Levels of PCNs and TEQs in sludge were relatively higher in samples from highly industrialized and developed cities in eastern China, implying a possible link between PCN contamination and the local economic development, but more studies are warranted to corroborate this. Industrial sources might be important contributors of PCNs to sewage sludge in China.

Graphene is widely used as promising electronic material and devices, owing to its exceptional electronic and optoelectronic properties. Up to now, defect-free graphene has been limited to the method for controllable, reproducible and scalable mass production. A simple, green, and nontoxic approach for large-scale preparation of high quality graphene is produced by exfoliation of graphite sheets collaborated with intercalant (FeCl2) under hydrothermal conditions, the absence of defects or oxides in graphene with a yield up to 10 wt% can be a practical application and industrial process such as optical limiters, transparent conductors, and sensors. This new process could potentially be improved to give a yield of up to 35 wt% of the starting graphite mass with sediment recycling. We show with experiments and theories that exfoliation graphene is the result of a combined action by diminishing the van der Waals interactions between graphite layers and the shear force drove by the Brownian motion of H2O and FeCl2 molecules. Hydrothermal exfoliation has potential applications in the exfoliation of other layered materials (e.g. BN, MoS2) and carbon nantubes, and in the synthesis of intercalation compounds, nanoribbons, and nanoparticles, thus opening new ways of exfoliation engineering.

This study was purposed to establish a new inducing system for differentiation of human embryonic stem cells into hematopoietic progenitor cells and to explore the effect of different extracellular matrices (DEM) on production of hematopoietic cells. The 3 kinds of extracellular matrices-matrigel, fibronectin and IV type collagen (collagen IV) were chosen to package cultured plates, the direct adherent culture on extracellular matrix was used, and the hematopoietic growth factors were added into cultured plates to induce the differentiation of human embryonic stem cells into hematopoietic progenitor cells. The hematopoietic colony forming unit assay was used to determine the yielded colony forming cells, the flow cytometry and real-time quantitative PCR were used to detect the expression of markers specific to hematopoiesis and the effect of 3 extracellular matrices on production of hematopoietic progenitor cells was compared. The results showed that after being induced for 14 days, the total yield of colony forming cells, the ratio of CD34(+) cells and the expression level of SCL and CD34 on collagen IV were significantly higher than those on matrigel and fibronectin groups (P < 0.05). It is concluded that human embryonic stem cells can efficiently differentiate into hematopoietic progenitor cells by direct adherent culture on extracellular matrices, and the collagen IV can improve the hematopoietic differentiation of human embryonic stem cells.

The purpose of the present study is to determine if visfatin is involved in inflammation or apoptosis induced by LPS in rat. Forty Wistar rats were divided into four groups: saline group, LPS group, visfatin group and Visfatin?+?LPS co-stimulated group. Spleen samples from each group of rats were collected for study. The spleen structure was examined by histological imaging. Apoptosis was evaluated with TUNEL reaction. Caspase-3 was detected with immunohistochemistry and western blot. The apoptosis-related genes were detected by qPCR and inflammatory cytokines were tested by ELISA. Our main findings were as follows. (1) Macrophages were markedly increased in the visfatin group compared with the saline group. This finding was confirmed when spleen samples were examined with western blot using CD68 antibody. (2) Visfatin promoted the expression of CD68 and caspase-3 in rat spleen, whereas visfatin could inhibit the expression of CD68 and activated caspase-3 in spleen of LPS-induced acute inflammation. (3) Visfatin had a pro-apoptotic effect on normal rat spleen, whereas it exerted an anti-apoptotic effect during LPS-induced lymphocytes apoptosis in rat spleen. Moreover, the effect of visfatin on cell apoptosis was mediated by the mitochondrial pathway. (4) Visfatin could modulate both the anti-inflammatory cytokines and pro-inflammatory cytokines in rat spleen, such as IL-10, IL-4, IL-6, TNF-? and IL-1?. Taken together, we demonstrate that visfatin could participate in the inflammatory process in rat spleen by modulating the macrophages and inflammatory cytokines. Also, visfatin plays a dual role in the apoptosis in rat spleen, which is mediated by the mitochondrial pathway.

Unintentionally produced persistent organic pollutants (UP-POPs) were determined in ambient air from around five secondary non-ferrous metal processing plants in China, to investigate the potential impacts of the emissions of these plants on their surrounding environments. The target compounds were polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), dioxin-like polychlorinated biphenyls (dl-PCBs), and polychlorinated naphthalenes (PCNs). The PCDD/F, dl-PCB, and PCN concentrations in the ambient air downwind of the plants were 4.70-178, 8.23-7520 and 152-4190 pg/m(3), respectively, and the concentrations upwind of the plants were lower. Clear correlations were found between ambient air and stack gas concentrations of the PCDD/Fs, dl-PCBs, and PCNs among the five plants, respectively. Furthermore, the UP-POPs homolog and congener patterns in the ambient air were similar to the patterns in the stack gas samples. These results indicate that UP-POPs emissions from the plants investigated have obvious impacts on the environments surrounding the plants.

Neuroblastoma is a common solid malignant tumor of the sympathetic nervous system, which contributes to 15% of cancer?related mortality in children. The differentiation status of neuroblastoma is correlated with clinical outcome, and the induction of differentiation thus constitutes a therapeutic approach in this disease. However, the molecular mechanisms that control the differentiation of neuroblastoma remain poorly understood. The present study aimed to define whether GATA3 is involved in the differentiation of neuroblastoma cells. The results demonstrated that GATA3 is a prognostic marker for survival in patients with neuroblastoma, and that high?level GATA3 expression is associated with increased self?renewal and proliferation of neuroblastoma cells. Retinoic acid treatment led to GATA3 downregulation together with neuronal differentiation, suppression of cell proliferation and inhibition of tumorigenecity in neuroblastoma cells. These findings suggest that GATA3 is a key regulator of neuroblastoma differentiation, and provide a novel potential therapeutic strategy for the induction of neuroblastoma differentiation.

Unintentionally produced persistent organic pollutants (UP-POPs) including polychlorinated dibenzo-p-dioxins, and dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), and polychlorinated naphthalenes (PCNs) were characterized and quantified in stack gas and fly ash from the second ventilation systems in five typical converters in five different steelmaking plants. The 2378-substituted PCDD/Fs (2378-PCDD/Fs) and dioxin-like PCB (dl-PCBs) toxic equivalents (TEQs) were 1.84-10.3 pg WHO-TEQ Nm(-3) in the stack gas and 5.59-87.6 pg WHO-TEQ g(-1) in the fly ash, and the PCN TEQs were 0.06-0.56 pg TEQ Nm(-3) in the stack gas and 0.03-0.08 pg TEQ g(-1) in the fly ash. The concentrations of UP-POPs in the present study were generally lower than those in other metallurgical processes, such as electric arc furnaces, iron ore sintering, and secondary metallurgical processes. Adding scrap metal might increase UP-POP emissions, indicating that raw material composition was a key influence on emissions. HxCDF, HpCDF, OCDF, HpCDD, and OCDD were the dominant PCDD/Fs in the stack gas and fly ash. TeCB and PeCB were dominant in the stack gas, but HxCB provided more to the total PCB concentrations in the fly ash. The lower chlorinated PCNs were dominant in all of the samples. The 2378-PCDD/F, dl-PCB, and PCN emission factors in stack gases from the steelmaking converter processes (per ton of steel produced) were 1.88-2.89, 0.14-0.76, and 229-759 ?g t(-1), respectively.

Accumulating research has shown that chronic D-galactose (D-gal) exposure induces symptoms similar to natural aging in animals. Therefore, rodents chronically exposed to D-gal are increasingly used as a model for aging and delay-of-aging pharmacological research. Mitochondrial dysfunction is thought to play a vital role in aging and age-related diseases; however, whether mitochondrial dysfunction plays a significant role in mice exposed to D-gal remains unknown. In the present study, we investigated cognitive dysfunction, locomotor activity, and mitochondrial dysfunction involved in D-gal exposure in mice. We found that D-gal exposure (125?mg/kg/day, 8 weeks) resulted in a serious impairment in grip strength in mice, whereas spatial memory and locomotor coordination remained intact. Interestingly, muscular mitochondrial complex I deficiency occurred in the skeletal muscle of mice exposed to D-gal. Mitochondrial ultrastructure abnormality was implicated as a contributing factor in D-gal-induced muscular impairment. Moreover, three combinations (A, B, and C) of nutrients applied in this study effectively reversed D-gal-induced muscular impairment. Nutrient formulas B and C were especially effective in reversing complex I dysfunction in both skeletal muscle and heart muscle. These findings suggest the following: (1) chronic exposure to D-gal first results in specific muscular impairment in mice, rather than causing general, premature aging; (2) poor skeletal muscle strength induced by D-gal might be due to the mitochondrial dysfunction caused by complex I deficiency; and (3) the nutrient complexes applied in the study attenuated the skeletal muscle impairment, most likely by improving mitochondrial function.

Systemic lupus erythematosus (SLE) is complex autoimmune disease which involves various facets of the immune system. Signaling transducers and activators of transcription 1 (STAT1) belongs to the family of STAT transcription factors that mediate various biological responses. Recently, studies in both experimental animal models of lupus and patients with SLE have revealed expression and activation of STAT1 is closely associated with the pathogenesis of SLE. Moreover, increased production of interferons (IFNs) and aberrant activation of IFNs signaling, which is mechanistically linked to increased level of STAT1, are crucial for the development of SLE. Therefore, we will focus on the association of STAT1 and SLE based on recent understandings to render more information about the mechanisms of STAT1 might perform in. Hopefully, the information obtained will lead to a better understanding of the development and pathogenesis of systemic autoimmune diseases, as well as its clinical implications and therapeutic potential.

Minimal-invasive procedure and one-step surgery offer autologous mesenchymal stem cells derived from peripheral blood (PB-MSCs) a promising prospective in the field of cartilage regeneration. We report a case of a 19-year-old male athlete of kickboxing with ICRS grade IV chondral lesions at the 60° region of lateral femoral trochlea, which was repaired by activating endogenous PB-MSCs plus autologous periosteum flap transplantation combined with correcting the patellofemoral malalignment. After a 7.5 year follow-up, the result showed that the patient returned to competitive kickboxing. Second-look under arthroscopy showed a smooth surface at 8 months postoperation. The IKDC 2000 subjective score, Lysholm score and Tegner score were 95, 98 and 9 respectively at the final follow up. CT and MRI evaluations showed a significant improvement compared with those of pre-operation.

Development and Evaluation of a Novel and Rapid Detection Assay for Botrytis cinerea Based on Loop-Mediated Isothermal Amplification.

Botrytis cinerea is a devastating plant pathogen that causes grey mould disease. In this study, we developed a visual detection method of B. cinerea based on the Bcos5 sequence using loop-mediated isothermal amplification (LAMP) with hydroxynaphthol blue dye (HNB). The LAMP reaction was optimal at 63°C for 45 min. When HNB was added prior to amplification, samples with B. cinerea DNA developed a characteristic sky blue color after the reaction but those without DNA or with DNA of other plant pathogenic fungi did not. Results of HNB staining method were reconfirmed when LAMP products were subjected to gel electrophoresis. The detection limit of this LAMP assay for B. cinerea was 10-3 ng µL-1 of genomic DNA per reaction, which was 10-fold more sensitive than conventional PCR (10-2 ng µL-1). Detection of the LAMP assay for inoculum of B. cinerea was possible in the inoculated tomato and strawberry petals. In the 191 diseased samples, 180 (94.2%) were confirmed as positive by LAMP, 172 (90.1%) positive by the tissue separation, while 147 (77.0%) positive by PCR. Because the LAMP assay performed well in aspects of sensitivity, specificity, repeatability, reliability, and visibility, it is suitable for rapid detection of B. cinerea in infected plant materials prior to storage and during transportation, such as cut flowers, fruits and vegetables.

An efficient enzymatic process was developed to produce optically pure D-phenylalanine through asymmetric resolution of the racemic DL-phenylalanine using immobilized phenylalanine ammonia-lyase (RgPAL) from Rhodotorula glutinis JN-1. RgPAL was immobilized on a modified mesoporous silica support (MCM-41-NH-GA). The resulting MCM-41-NH-GA-RgPAL showed high activity and stability. The resolution efficiency using MCM-41-NH-GA-RgPAL in a recirculating packed-bed reactor (RPBR) was higher than that in a stirred-tank reactor. Under optimal operational conditions, the volumetric conversion rate of L-phenylalanine and the productivity of D-phenylalanine reached 96.7 mM h?¹ and 0.32 g L?¹ h?¹, respectively. The optical purity (eeD) of D-phenylalanine exceeded 99%. The RPBR ran continuously for 16 batches, the conversion ratio did not decrease. The reactor was scaled up 25-fold, and the productivity of D-phenylalanine (eeD>99%) in the scaled-up reactor reached 7.2 g L?¹ h?¹. These results suggest that the resolution process is an alternative method to produce highly pure D-phenylalanine.

Histone methylation plays an important role in gene transcription and chromatin organization and is linked to the silencing of a number of critical tumor suppressor genes in tumorigenesis. G9a is a histone methyltransferase (HMTase) for histone H3 lysine 9. In this study, we investigated the role of G9a in neuroblastoma tumor growth together with the G9a inhibitor BIX01294. The exposure of neuroblastoma cells to BIX01294 resulted in the inhibition of cell growth and proliferation, and BIX01294 treatment resulted in the inhibition of the tumorigenicity of neuroblastoma cells in NOD/SCID mice. Therefore, G9a may be a potential therapeutic target in neuroblastoma. Moreover, we found several specific characteristics of autophagy after BIX01294 treatment, including the appearance of membranous vacuoles and microtubule-associated protein light chain 3 (LC3B). Similar results were observed in G9a-knockdown cells. In conclusion, our results demonstrated that G9a is a prognostic marker in neuroblastoma, and revealed a potential role of G9a in regulating the autophagy signaling pathway in neuroblastoma.

This study was purposed to evaluate the efficacy of autologous peripheral blood hematopoietic stem cell transplantation ( auto-PBHSCT) for patients with malignant lymphoma. From January 1992 to June 2012, 32 malignant lymphoma (ML) patients were treated with auto-PBHSCT in our hospital. There were 56 cases of non-Hodgkins lymphoma(NHL) and 16 cases of Hodgkins lymphoma(HD). The preconditionging regimens of total body irradiation(TBI) plus cyclophosphamide combined with etoposide/pharmorubicin were used in 50 patients, and the BEAM (BCNU+etoposide+Ara-C+melphalan) were used in the other 22 patients. The average number of CD34(+) cells was (6.6 ± 4.9) ×10(6)/kg. The results showed that transplanted cells were engrafted and hematopoiesis was reconstituted in all patients. The median time of neutropilic granulocyte recovery up to ? 0.5×10(9)/L was 12 ± 4.4 days, and the platelet recovery up to ? 20×10(9)/L was 14 ± 4.9 days . The transplantation related mortality was 4.2%. The patients were followed up for 6-217 months. The median follow-up time was 63.5 months. 1-year OS rate was 92.3 ± 3.2%. 3-years OS rate was 81.4 ± 4.9%. 5-years OS rate was 77.6 ± 5.3%. 10-years OS rate was 68.9 ± 6.8%. It is concluded that auto-PBHSCT is effective for malignant lymphoma, and long time follow-up shows that the OS rate is still high. Furthermore, the safety of auto-PBHSCT can also be received. Before transplantation the patients with CR can get higher OS rate.

This study was aimed to explore the clinical features and related prognostic factors of follicular lymphoma (FL). The total of 59 newly diagnosed FL patients were analyzed retrospectively in term of age, sex, clinical manifestation, laboratory data, pathological examination, clinical stage and so on, so as to find out the related prognostic factors. The results showed that the median age of the 59 patients was 55 years, among them the male-female ratio was 1: 1.36. Among the 64.4% cases with the involvement outside lymphonode , the bone marrow involvement accounted for 28.8%, while the involvement of gastrointestinal tract and skeleton was in the second place. The rate of BCL-2/JH gene rearrangement was 30.3%, III-IV of clinical stage accounted for 71.2% in these FL patients. During the follow up of the 56 patients, OR and CR rates were 96.4% and 80.3% respectively, 5 year-and 10 year overall survival rates were 88.2% and 74.1% respectively, the relapse rate was 33.9%. Univariate analysis showed that the age, the longest diameter of involved node, gastrointestinal involvement, serum lactate dehydrogenase, ECOG, IPI, aaIPI, FLIPI, FLIPI2 and whether initial therapy could achieve CR or not were influencing factors of prognosis for FL patients. Multivariant analysis showed that the gastrointestinal involvement, IPI, aaIPI and whether initial therapy could achieve CR or not were independent prognostic factors for OS. The relative mortality risk of patients without CR was 70 times of patient with CR. It is concluded that the newly diagnozed FL is more observed in middle to old aged females, displaying frequent bone marrow involvement, less frequent BCL-2/JH gene rearragement, III-IV of clinical staging, and high relapse rate, but longer survival time. The gastrointestinal involvement, IPI, aaIPI and whether CR can be achieved or not after initial therapy are independeut prognostic factors for FL patients.

This study is to observe the protection effect of amentoflavone (AMT) in Selaginella tamariscina against TNF-alpha-induced vascular inflammation injury of endothelial cells. On the basis of TNF-alpha induced human umbilical vein endothelial cell, observe the influence of AMT on endothelial active factor, the contents of SOD and MDA, the protein expression of vascular endothelial adhesion molecules and inflammatory factor; study the effect of its common related signal pathways such as NF-kappaB; research the effect of AMT against TNF-a induced human umbilical vein endothelial cell injury by means of MTT, ELISA, Western blotting and the cell immunofluorescence. The results showed that AMT could increase the content of NO and decrease the levels of VCAM-1, E-selectin, IL-6, IL-8 and ET-1; enhance the activity of SOD, reduce the content of MDA; downregulate the protein expressions of VCAM-1, E-selectin, NF-kappaBp65 and up-regulate IkappaBalpha, attenuate the NF-kappaBp65 transfer to cell nucleus. AMT has the effect of protect vascular endothelial and maybe via the signal pathway of NF-kappaB to down-regulate the inflammation factor and oxidative damage factor of downstream.

Two city ecosystem research stations were selected for continuous monitoring of atmospheric fine particulate matter (PM2.5) concentration from June to September 2008, when the Beijing Olympic Games was held. The relationship between meteorological factors, artificial control management measures and PM2.5 dynamic characteristics were analyzed. The results showed that the daily average concentration of PM2.5 at RCEES Station, which is near the 5th North Ring, was 0.067 mg x m(-3) during the study period, and the particle concentration during the Olympic Games was 0.060 mg x m(-3), which was decreased by about 26% compared with the content before the Olympic Games (0.081 mg x m(-3)). As for JX Station located in the 2th South Ring, its daily average concentration of PM2.5 was 0.078 mg x m(-3). Similar to RCEES Station, PM2.5 average content during the Olympic Games (0.069 mg x m(-3)) was 27% lower compared with the content before the Olympic Games (0.095 mg x m(-3)). Two peaks could be found when daily distribution of PM2.5 was displayed, one appeared between 08:00 and 10:00 (the average content for RCEES and JX were 0.068 and 0.089 mg x m(-3), respectively), and the other appeared at 20:00-22:00, which could be attributed to traffic emission and road dust pollution, especially the emission from the growing number of cars in rush hours. Meteorological condition during the whole period was typical in summer in Beijing with both high temperature and humidity. Correlation analysis revealed that the concentrations of PM2.5 had a positive correlation with the air temperature (P < 0.01), while it had no significant relationship with wind speed, relative humidity or precipitation (P > 0.05). Our monitoring results suggested that the artificial air pollution control strategies were effective measures for air pollution control during 2008 Beijing Olympic Game.

Background/Aims: The aim of this study was to identify useful prognostic factors for patients with distal cholangiocarcinoma. Methodology: The records of 84 patients with distal cholangiocarcinoma undergoing pancreatoduodenectomy were retrospectively reviewed. Potential clinicopathological prognostic factors that may affect survival were examined by univariate and multivariate analyses. Results: There were two patients died within 30 days of surgery. Overall survival rates were 69.51%, 42.68%, and 36.59% for 1, 3 and 5 years, respectively (median survival time, 32.74 months). Univariate analysis found that alanine aminotransferase/aspartate aminotransferase (AST/ALT) ratio less than and equal to 2, serum bilirubin less than and equal to 171 ?mol/L, CA19-9 level less than 150 U/L, tumor size less than 2 cm, absent neural invasion, and absent lymph node involvement were associated with higher survival rate (p <0.05). Furthermore, multivariate analysis found that AST/ALT ratio more than 2, present lymph node involvement and present neural invasion were the independent risk factors of poor survival (p <0.01). Conclusions: These results suggest that not only the well-known lymph node involvement, but also neural invasion and AST/ALT ratio more than 2 might be useful prognostic factors for long-term survival in distal cholangiocarcinoma.

Systemic lupus erythematosus (SLE) is a complex autoimmune disease arising from the action of multiple genetic and environmental risk factors. The aim of this study was to examine the association of a single-nucleotide polymorphism, rs1990760, of the interferon induced with helicase C domain 1 (IFIH1) gene with SLE in a Chinese population. A total of 877 SLE patients and 978 healthy control subjects were enrolled in the present study. The genotype of the IFIH1 rs1990760 polymorphism was determined by Sequenom MassARRAY technology. The IFIH1 rs1990760 T allele was significantly increased in patient group compared with control subjects (T versus C, Odds ratio?=?1.20, 95 % confidence interval?=?1.02-1.40). However, no significant difference in genotype distribution was found between cases and controls (P?=?0.07). No significant evidence was detected for the association of the IFIH1 rs1990760 polymorphism with SLE under neither dominant nor recessive model (TT + TC versus CC, P?=?0.06; TT versus TC + CC, P?=?0.08). We also analyzed the association of the IFIH1 rs1990760 T allele with clinical features, whereas no significant signal was found. In conclusion, our study represents the first report demonstrating an association of the IFIH1 rs1990760 polymorphism with SLE susceptibility in a Chinese population.

Mono- to Octa-chlorinated PCDD/Fs in stack gas from typical waste incinerators and their implications on emission.

Mono- to octa-chlorinated dibenzo-p-dioxins and dibenzofurans (mono- to octa-CDD/Fs) were determined in 14 stack gas samples from two municipal solid waste incinerators and two medical waste incinerators. The total PCDD/F concentrations were 5.1-390 ng/Nm(3), and the mono- to trichlorinated homologues contributed 53.2-94.5% of the total concentrations. The homologue profiles were dominated by the MoCDF, ranged from 1.51 to 113.1 ng/Nm(3), and the proportion that each PCDF homologue group contributed to the total concentration decreased with increasing chlorination level. The toxic equivalent concentrations (I-TEQs) were 0.01-2.81 ng I-TEQ/Nm(3), with 2,3,4,7,8-PeCDF being the biggest contributor, at 30.6-60.0%. Correlations were found among the PCDD/PCDF ratios (D/F ratios), the degree of chlorination, and the TEQ. Stack gases with low I-TEQs had higher proportions of the less chlorinated homologues and lower D/F ratios, which could be attributed to the removal of the more chlorinated isomers by the air pollution control systems used by the incinerators. 2,8-DiCDF, 2,4,8-TrCDF, DiCDF, and TrCDF can be used as TEQ indicators for monitoring PCDD/Fs. 2,8-DiCDF and 2,4,8-TrCDF correlated well with the TEQ because they strongly correlated with 2,3,4,7,8-PeCDF, implying possible correlations in their formation mechanisms.

Tremendous strides have been made in the diagnosis and treatment of human immunodeficiency virus (HIV); perhaps now the largest barrier to controlling HIV is retaining those diagnosed in care. Data on out-of-care populations are needed to develop effective retention methods, yet obtaining these remains methodologically challenging due to inherent difficulties in sampling. The purpose of this study was to evaluate whether individuals identified by two sampling methods commonly used to approximate out-of-care populations were significantly different from a sample of newly identified out-of-care persons. We compared medical records of 345 out-of-care persons identified by a novel population-based health information exchange who had not received CD4 or viral load monitoring in >1 year with: medical records from a randomly selected, time-matched sample of 488 HIV-infected persons with at least one HIV care visit in the past 5 years, and interviews with 382 participants from a time-matched clinic-based convenience sample. Newly identified out-of-care persons were significantly different from both proxies with respect to demographic, clinical, and utilization characteristics, suggesting that samples of in-care proxy persons are inadequate to describe those not engaged in care. Novel approaches to sampling out-of care populations are urgently needed in order to better understand these populations and ways to improve retention and slow the HIV/AIDS epidemic.

A new apiose-containing kaempferol trioside, kaempferol-3-O-?-l-rhamnosyl-(1? ? 6?)-O-?-d-galactopyranosyl-7-O-?-d-apiofuranoside, along with 16 known compounds, were isolated from 50% acetone extract of Silphium perfoliatum L. Their structures were elucidated by acid hydrolysis and spectroscopic techniques including UV, IR, MS, (1)H, (13)C, and 2D-NMR. In addition, the pharmacological activity of compound 1 was tested with HepG2 and Balb/c mice (splenic lymphocytes and thymic lymphocytes) in vitro, and it exhibited inhibitory effect on the proliferation of HepG2 cells and showed the immunosuppressive activity.

The concentrations of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), dioxin-like polychlorinated biphenyls (dl-PCBs), and polychlorinated naphthalenes (PCNs) were determined in workplace air from eight secondary nonferrous metal processing plants to investigate occupational exposure to these toxic compounds. The total estimated daily intakes of PCDD/Fs and dl-PCBs for workers by inhalation in the workplace were in the range of 0.15-9.91 and 0.13-8.59 pg of WHO-TEQ/kg of body weight (bw) for moderate and light activities, respectively. The daily inhalation doses for workers in the workplaces of three investigated plants exceeded the tolerable daily intake recommended by the World Health Organization. These results indicate that the risk of occupational exposure to dioxins by inhalation in the workplace of plants investigated was considerably high. For PCNs, the daily inhalation doses for workers in the workplace were in the range of 0.005-4.46 and 0.004-3.87 pg of TEQ/kg of bw for moderate and light activities, respectively, which were lower than those of dioxins. To identify the source of PCDD/Fs, PCBs, and PCNs in workplace air, their homologue profiles were compared with those in stack gas from the plants investigated. It was found that significant dioxin contamination in workplace air was mainly attributed to the emission of fugitive gas from smelting furnaces during reclamation processes.

Male factor is a common cause of infertility and the male partner must be systematically evaluated in the workup of every infertile couple. Various Eastern medical strategies have been tried with variable success. This article describes the clinical effects of Eastern medicine approaches including acupuncture, Chinese herbal medicine, massage, yoga, tai chi, and qi gong, which could improve the sperm parameters and motility, genital inflammatory conditions, as well as immune system disorders, sexual dysfunction, and varicocele. Acupuncture reduces inflammation, increases sperm motility, improves semen parameters, modulates the immune system, and improves sexual and ejaculatory dysfunction in male infertility. The clinical effects may be mediated via activation of somatic afferent nerves innervating the skin and muscle. Chinese herbal medicines may also exert helpful effects in male infertility, and it is worth noting that some herbal drugs may result in male infertility. Massage also exerts positive effects in male infertility. Nevertheless, the mechanisms of clinical effects are unclear. Tai chi, qi gong, and yoga have not been investigated in male infertility, but it has been reported to regulate endocrine and central or autonomic nervous systems. In conclusion, Eastern medical approaches have beneficial on reproductive effects in male infertility. However, future well-designed, randomized, clinical control trials are needed to evaluate the safety, efficacy, and mechanisms of Eastern medical approaches for male infertility.

The effects of cooling conditions on graphene growth by the atmospheric pressure chemical vapor deposition (APCVD) method on platinum are investigated. It is found that the cooling conditions are the key to better control of the coverage, thickness and uniformity of the graphene films. A series of experiments were carried out with different cooling rates and gas atmospheres. Various characterization techniques (SEM, Raman, AFM and TEM) were applied to examine the graphene morphology and quality. A "three competitive reactions" mechanism is proposed to explain the cooling condition effects. Based on optimized growth conditions, a high-coverage, single-layer graphene film could be grown on Pt using the APCVD method. Back-gated field effect transistors were fabricated and the measured carrier mobility is about 1600 cm(2) V(-1) s(-1).

In order to investigate the effect of PD-L1 on lymphoma cell proliferation and chemotherapy resistance, and explore a new and promising approach for therapy of lymphomas. The level of PD-L1 expression in tissues of patients with lymphoma and cell lines were detected by immunohistochemistry and flow cytometry. Both the stable high-PDL1-A20 cell strain and low-PDL1-A20 cell strain were sorted by flow cytometry, then inoculated into BALB/c mice as to establish a lymphoma bearing mouse model for observing tumor genesis. Finally, the effect of PD-L1 on routinely chemotherapy drug-cisplatin were observed. The results showed that the PD-L1 highly and widely expressed in lymphoma tissue and cell strains, the stable high-PD-L1 cell strain sorted by flow cytometry have higher lymphoma genesis than that of low-PD-L1 cell strain. It is concluded that PD-L1 can promote lymphoma genesis, meanwhile the PD-L1 may resist effect of cisplatin on lymphoma cells through anti-apoptosis.

Dechlorane Plus (DP) is a high-production volume, chlorinated flame retardant comprising two major isomers, syn- and anti-DP. In this study, levels of DP were measured in soil and earthworm samples in farmland from a typical E-waste dismantling area in China. The concentrations of total DP ranged from 0.17 to 1,990 ng g(-1) dw in soil samples and 3.43 to 89.2 ng g(-1) lw in earthworm samples. Higher DP levels were found in some main E-waste dismantling sites than those in other sites. The ratios of anti-DP to total DP (f anti) ranged from 0.57 to 0.80 in soil samples and 0.47 to 0.81 in earthworm samples, respectively. The f anti in most samples in this study was in the range of the reported technical DP values. These results showed that improper E-waste dismantling activities could result in the emission of DP. Most earthworm samples showed very low BSAFs (biota-to-soil accumulation factors) for total DP. The values of BSAF were in the range of 0.0007-1.85, with an average value of 0.23. This study presents the first report of the DP in earthworms, which would be useful for ecological risk assessment of DP in terrestrial ecosystem.

This study was undertaken to replicate some of these associations and further test for genetic interactions among these genes in SLE in a Chinese population. Ten single-nucleotide polymorphisms (SNPs) in NFKB1, REL, inhibitor of ?B-like (I?BL), I?B kinase ? (IKBKB), tumor necrosis factor receptor associated factor 6 (TRAF6), tumor necrosis factor a-induced protein 3 (TNFAIP3), TNFAIP3 interacting protein 1 (TNIP1) were genotyped in 898 Chinese patients with SLE and 988 healthy controls by Sequenom MassArray technology. Single-marker genetic association analysis was performed, and additive and multiplicative interactions were analyzed. Associations of TNFAIP3 rs2230926 (p=1.43 × 10(-3)) and TNIP1 rs10036748 (p=4.33 × 10(-3)) with SLE were replicated in our study. Two other SNPs, NFKB1 rs28362491 and I?BL rs2071592, showed nominal evidence for association (p=4.70 × 10(-2) and p=5.90 × 10(-3), respectively) but these were not significant after applying Bonferroni correction. Additive interaction analysis revealed significant interaction between NFKB1 rs28362491 and TNFAIP3 rs2230926 (RERI=0.98, 95%CI=0.02-1.93; AP=43.2%, 95%CI=0.12-0.74). Significant multiplicative interaction was observed between NFKB1 rs28362491 and TNIP1 rs3792783 (p=0.03). Our results provide evidence for gene-gene interactions, which further support the important role of NF-?B signaling pathway in the genetic basis of SLE and the notion of genetic interactions accounting for missing heritability.

To motivate more people to participate in vaccination campaigns, various subsidy policies are often supplied by government and the health sectors. However, these external incentives may also alter the vaccination decisions of the broader public, and hence the choice of incentive needs to be carefully considered. Since human behavior and the networking-constrained interactions among individuals significantly impact the evolution of an epidemic, here we consider the voluntary vaccination on human contact networks. To this end, two categories of typical subsidy policies are considered: (1) under the free subsidy policy, the total amount of subsidy is distributed to a certain fraction of individual and who are vaccinated without personal cost, and (2) under the partial-offset subsidy policy, each vaccinated person is offset by a certain amount of subsidy. A vaccination decision model based on evolutionary game theory is established to study the effects of these different subsidy policies on disease control. Simulations suggest that, because the partial-offset subsidy policy encourages more people to take vaccination, its performance is significantly better than that of the free subsidy policy. However, an interesting phenomenon emerges in the partial-offset scenario: with limited amount of total subsidy, a moderate subsidy rate for each vaccinated individual can guarantee the group-optimal vaccination, leading to the maximal social benefits, while such an optimal phenomenon is not evident for the free subsidy scenario.

The purpose of this study was to compare the efficacy and safety of a single subcutaneous injection of pegylated filgrastim with daily filgrastim as a prophylaxis for neutropenia induced by commonly used chemotherapy regimens. Fifteen centers enrolled 337 chemotherapy-naive cancer patients with normal bone marrow function. All patients randomized into AOB and BOA arms received two cycles of chemotherapy. Patients received a single dose of pegylated filgrastim 100 µg/kg in cycle 1 (AOB) or cycle 2 (BOA) and daily doses of filgrastim 5 µg/kg/day in cycle 1 (BOA) or cycle 2 (AOB). Efficacy and safety parameters were recorded. The primary end point was the rate of protection against grade 4 neutropenia after chemotherapy [defined as the rate at which the absolute neutrophil count (ANC) remained >0.5×10(9)/l throughout the entire cycle]. Ninety-four percent of patients receiving pegylated filgrastim or filgrastim did not develop grade 4 neutropenia. The incidence of ANC<1.0×10(9)/l was 16.0% (50/313) after support with either pegylated filgrastim or filgrastim. The incidences of febrile neutropenia and antibiotic administration were similar in both groups. Notably, faster ANC recovery was observed with pegylated filgrastim support. The ANC nadir was also earlier with pegylated filgrastim (day 7) support than with filgrastim support (day 9), although the depth of nadir was not significantly different. A single subcutaneous injection of pegylated filgrastim 100 ?g/kg provided adequate and safe neutrophil support comparable with daily subcutaneous injections of unmodified filgrastim 5 ?g/kg/day in patients receiving commonly used standard-dose mild-to-moderate myelosuppressive chemotherapy regimens.

Three new ursane-type triterpenes, glutinosalactone A-C (1-3), were isolated from the 50% aqueous acetone extract of the leaves of Rehmannia glutinosa. Their structures were elucidated on the basis of spectral analysis (IR, NMR and MS spectroscopy). The cytotoxic effects of compounds 1-3 against three human cancer cell lines (MCF-7, MG63 and HepG2) were also evaluated. Compound 3 showed cytotoxic activities with IC50 values of 8.35-39.25 ?M.

Rapid increase of the urban impervious underlying surfaces causes a great increase of urban runoff and the accumulation of pollutants on the roof and road surfaces brings many pollutants into the drainage system with the runoff, and it thus becomes a great threat to the urban water environment. To know the runoff pollution process and to build scientific basis for pollutant control, runoff processes from the roof and road surfaces were monitored and analyzed from 2004 to 2006, and the runoff EMC (Event Mean Concentration) was calculated. It was found that two types of runoff were seriously polluted by COD and TN. The COD and TN of roof runoff exceeded the fifth level of the surface water environmental quality standard (GB 3838-2002) by 3.64 and 4.80 times, respectively, and the COD and TN of road runoff exceeded by 3.73 and 1.07 times, respectively. M (V) curve was used to determine the relation between runoff volume and runoff pollution load. Various degrees of the first flush phenomenon were found for TSS, COD, TN and TP in roof runoff. But this phenomenon occurred only for TSS and TP of the road runoff, and on the whole it was not obvious. Properties of the underlying surfaces, rainfall intensity, and pollutant accumulation are all important factors affecting the roof and road runoff pollutant emission characteristics.

Rheumatoid arthritis (RA) is an autoimmune disease, characterized by chronic inflammation in synovial joints. Effective treatment for RA is lacking because the clear etiology and pathogenesis of RA have not been fully elucidated. Cytokine-mediated immunity has been found to play an important role in the pathogenesis of various autoimmune diseases such as RA. Recently, IL-32 is identified with high expression in RA patients and mice models of experimental inflammatory arthritis. IL-32 is recognized to play a crucial role in RA with pro-inflammatory effects. Furthermore, interventions for blocking IL-32 in RA seem possible and applicable. Therefore, targeting IL-32 may give therapeutic potential. In this article, we discuss the biological features of IL-32 and summarize recent advances in understanding the role of IL-32 in disease onset of and treatment for RA. Hopefully the information obtained will benefit for developing novel therapeutic strategies.

The aim of the present study was to clarify the distribution and relative frequencies of somatostatin (SST)-producing cells in the stomach and the small intestine of the ostrich by using immunohistochemistry. The results indicated that somatostatin-immunoreactive (SST-IR) cells were distributed in mucosal layers of the proventriculus, duodenum, jejunum and ileum. However, no immunoreactivity was observed in the gizzard. SST-IR cells were found at the lower part of glandular lobule in the proventriculus, which were oval and round generally. SST-IR cells were present in the mucous membrane of entire small intestine of the ostrich. SST-IR cells had round and spherical shapes (closed-type cells), or spindle and pyriform shapes (open-type cells) in the small intestine. SST-positive cells were localized preferentially in the proventriculus of the 60-day-old ostrich. These results indicated that SST might be involved in functional and developmental regulation of gastrointestinal tract of the ostrich.

Sewage treatment plants (STP) are an important source of short chain chlorinated paraffins (SCCPs) to the ambient environment through discharge of effluent and application of sludge. In this work, a field study was conducted to determine the behavior and possible removal of SCCPs during the sewage treatment process in an advanced municipal STP in Beijing, China. SCCPs were detected in all sewage water and sludge samples, and 97% of the initial mass loading in raw sewage was found to be associated with suspended matter. The total concentrations in raw influent, tertiary effluent, and dewatered sludge were 184 ± 19 ng/L, 27 ± 6 ng/L, and 15.6 ± 1.4 ?g/g dry weight (d.w.), respectively. The dissolved concentrations of total SCCPs (?SCCPs) significantly decreased during mechanical, biological, and chemical treatments. SCCP homologue profiles in aqueous phase were distinctly different from those in solid phase. Along the treatment process, the relative abundance of shorter chain and lower chlorinated congeners gradually increased in sewage water, but no obvious variations of homologue profiles were found in sludge. Mass flow analysis indicated, the removal efficiency in aqueous phase for ?SCCPs was 82.2%, and the congener-specific removal efficiencies were positively related to their solid-water partition coefficients (K(d)). Mass balance results indicated that 0.8% and 72.6% of the initial SCCP mass loading were ultimately found in the effluents and dewatered sludge, respectively, while the remaining 26.6% was lost mainly due to biodegradation/biotransformation. It was suggested that the activated sludge system including basic anaerobic-anoxic-aerobic processes played an effective role in removing SCCPs from the wastewater, while the sorption to sludge by hydrophobic interactions was an important fate of SCCPs during the sewage treatment.
