[Effects of drought stress on growth and water use efficiency of two medicinal plants].

Growth characteristics, stable carbon isotope discrimination (Delta13C), water use efficiency (WUE), and their correlation of Cassia obtusifolia and Isatis indigotica were measured at three soil water levels, i. e., 30%, 50% and 75% of field water holding capacity (FWHC), and at three growth stages. The growth indices of the two medicinal plants at 75% of FWHC were higher than those at 30% and 50% of FWHC, suggesting that the two medicinal plants could obtain high production under sufficient moisture condition. The Delta13C(A) (aboveground biomass-based Delta13C) and Delta13C(T) (total biomass-based Delta13C) decreased, and the WUE(A) (aboveground biomass-based WUE) and WUE(T) (total biomass-based WUE) of C. obtusifolia and I. indigotica increased with the increasing degree of drought stress. The growth indices of the two medicinal plants had little difference in the different water treatments, which indicated that the two medicinal plants were insensitive to drought stress. Water use efficiency of I. indigotica had significant negative relationships with aboveground biomass and total biomass, while that of C. obtusifolia had a significant positive correlation with the root/shoot ratio.

The interleukin (IL)-6/gp130 family of cytokines (e.g., IL-6, IL-11, leukemia inhibitory factor (LIF) and oncostatin M (OSM)) play important roles in the central nervous system (CNS) during neuroinflammation and neurodevelopment. However, little is known regarding the responses by astroglia and microglia to this family of cytokines. Here the expression of the IL-6/gp130 cytokine receptors and subsequent signal pathway activation was examined in murine astrocytes and microglia in vitro. Astrocytes had high levels of OSMR mRNA while lower levels of IL-6R, LIFR and IL-11R mRNAs were also present. In comparison, in microglia there was no detectable OSMR mRNA, higher levels of IL-6R mRNA and lower levels of the LIFR and IL-11R mRNAs. The OSMR protein was present in astrocytes but was undetectable in microglia. Conversely, the IL-6R protein was present in microglia but not detectable in astrocytes. In astrocytes but not microglia, phosphorylation of STAT1 and STAT3 occurred in response to OSM, whereas both microglia and astrocytes responded to hyper-IL-6 (IL-6 linked to the soluble IL-6 receptor). Finally, in both microglia and astrocytes, OSM failed to activate NF?B or induce iNOS and nitrite production. We conclude: (1) notable differences exist in the expression of receptors utilized by the IL-6/gp130 family of cytokines in astrocytes and microglia, and (2) the findings provide a molecular basis for the differential response to OSM by astrocytes versus microglia and demonstrate a fundamental means for achieving cellular specificity in the response of these glial cells to this cytokine. The Chinese surf clam Mactra chinensis Philippi, 1846 is a commercially important marine bivalve belonging to the family Mactridae (Mollusca: Bivalvia). In this study, the M. chinensis mitochondrial genomic features are analyzed. The genome has 34 genes on the same strand, lacking atp8 and both trnS (trnS1 and trnS2) as compared with the typical gene content of metazoan mitochondrial genomes. The A+T content of M. chinensis mitochondrial genome is 63.72%, which is slightly lower than that of M. veneriformis (67.59%) and Coelomactra antiquata (64.33% and 64.14% for the samples from Ri Zhao, Shandong Province, and Zhang Zhou, Fujian Province, China, respectively) in the same family. There are 22 NCRs in the M. chinensis mitochondrial genome, accounting for 12.91% of the genome length. The longest NCR (1,075bp in length) is located between trnT and trnQ. A TRS (127bp×8.15) accounts for 96.3% (1,035/1,075) of this NCR. The occurrence of TRS in NCR is shared by the two Mactra mitochondrial genomes, but is not found in the two Coelomactra mitochondrial genomes. A phylogenetic tree constructed based on 12 PCGs of 25 bivalve mitochondrial genomes shows that all seven genera (Mactra, Coelomactra, Paphia, Meretrix, Solen, Mytilus, and Crassostrea) constitute monophyletic groups with very high support values. Pairwise genetic distance analyses indicate that the genetic distance of C. antiquata from the two localities is 0.084, which is greater than values between congeneric species, such as those in Mactra, Mytilus, Meretrix, and Crassostrea. The results show that the C. antiquata from the two localities represent cryptic species.

Abstract: Four land use types of cropland, abandoned cropland, 10-year-old and 43-year-old Robinia psedudoacia plantations in the Grain-for-Green Program in hilly area of North China were studied to investigate the effects of returning cropland to forestland on soil quality by using integrated soil quality index. The results showed that the nutrients of topsoil increased significantly with increasing tree age, and soil properties in 0-5 cm soil layer improved. Compared with the cropland, soil physical and chemical properties of the two R. psedudoacia plantations improved, and the soil microbial biomass C and N increased significantly. The integrated soil quality index decreased in order of 43-year-old R. psedudoacia plantation (0.542) > 10-year-old R. psedudoacia plantation (0. 536)> the abandoned cropland (0.499) > the cropland (0.498), suggesting the soil quality was improved during the conversion from cropland to forestland.

Based on quinoline-2,3-dicarboxylic acid (H2L), two metal(II) complexes formulated as MnL(phen)(H2O)·H2O (phen = 1,10-phenanthroline) (1) and Co(HL)2(PPA)·4H2O (PPA = N(1),N(4)-di(pyridin-4-yl)terephthalamide) (2) were synthesized and structurally characterized by single-crystal X-ray diffraction. Both complexes 1 and 2 exhibit one-dimensional (1D) chain-like structures, in which stable five-membered rings are observed. Different chains are linked by strong ?-? stacking interactions into a three-dimensional (3D) supramolecular architecture. Both complexes can increase the degradation rate of methyl orange (MO), which is expected to be associated with their electrocatalytic activities for the H2 evolution reaction from water.

A novel rhodamine based sensor 1 has been synthesized and characterized by NMR, FT-IR and mass. The binding properties of sensor 1 were investigated in the presence of various metal ions by UV-Vis spectroscopy and fluorescence spectroscopy. Sensor 1 showed colorimetric and high emission intensity enhancement in the presence of Pb(2+) in chloroform-THF (7:3).

A novel rhodamine based fluorescent chemosensor RQP was prepared and characterized by (1)HNMR, (13)CNMR and HR-MS. The properties of RQP were studied through UV-Vis spectroscopy and fluorescence spectroscopy. RQP showed highly selectivity toward Hg(2+) over other metal ions, including Ag(+), Cd(2+), Cu(2+), Na(+), Mg(2+), Ni(2+), Pb(2+), Fe(3+), and Zn(2+) in aqueous solutions. The recognition process is reversible and confirmed by EDTA experiment.

Estrogen has been reported to affect pain perception, although the underlying mechanisms remain unclear. In this investigation, pain behavior testing, patch clamp recording, and immunohistochemistry were used on rats and transgenic mice to determine which estrogen receptors (ERs) and the related signaling pathway are involved in the rapid modulation of estrogen on P2X3 receptor-mediated events. The results showed that 17?-estradiol (E2) rapidly inhibited pain induced by ?,?-methylene ATP (?,?-me-ATP), a P2X1 and P2X3 receptor agonist in ovariectomized rats and normal rats in diestrus. The ER? agonist 4,49,499-(4-propyl-[1H]-pyrazole-1,3,5-triyl) trisphenol (PPT) and G protein-coupled receptor 30 (GPR30) agonist G-1 mimicked the estrogen effect, whereas the ER? agonist diarylpropionitrile (DPN) had no effect. In cultured rat dorsal root ganglion (DRG) neurons, PPT and G-1 but not DPN significantly attenuated ?,?-me-ATP-mediated currents, with the dose-response curve of these currents shifted to the right. The inhibitory effect of E2 on P2X3 currents was blocked by G-15, a selective antagonist to the GPR30 estrogen receptor. E2 lacked this effect in DRG neurons from ER?-knockout mice but partly remained in those from ER?-knockout mice. The P2X3 and GPR30 receptors were coexpressed in the rat DRG neurons. Furthermore, the ERK1/2 inhibitor U0126 reversed the inhibitory effect of E2 on ?,?-me-ATP-induced pain and of PPT or G-1 on P2X3 receptor-mediated currents. The cAMP-protein kinase A (PKA) agonist forskolin, but not the PKC agonist phorbol-12-myristate-13-acetate (PMA), mimicked the estrogen-inhibitory effect on P2X3 receptor currents, which was blocked by another ERK1/2 inhibitor, PD98059. These results suggest that estrogen regulates P2X3-mediated peripheral pain by acting on ER? and GPR30 receptors expressed in primary afferent neurons, which probably involves the intracellular cAMP-PKA-ERK1/2 pathway.

The early-diverging eudicot order Trochodendrales contains only two monospecific genera, Tetracentron and Trochodendron. Although an extensive fossil record indicates that the clade is perhaps 100 million years old and was widespread throughout the Northern Hemisphere during the Paleogene and Neogene, the two extant genera are both narrowly distributed in eastern Asia. Recent phylogenetic analyses strongly support a clade of Trochodendrales, Buxales, and Gunneridae (core eudicots), but complete plastome analyses do not resolve the relationships among these groups with strong support. However, plastid phylogenomic analyses have not included data for Tetracentron. To better resolve basal eudicot relationships and to clarify when the two extant genera of Trochodendrales diverged, we sequenced the complete plastid genome of Tetracentron sinense using Illumina technology. The Tetracentron and Trochodendron plastomes possess the typical gene content and arrangement that characterize most angiosperm plastid genomes, but both genomes have the same unusual ?4 kb expansion of the inverted repeat region to include five genes (rpl22, rps3, rpl16, rpl14, and rps8) that are normally found in the large single-copy region. Maximum likelihood analyses of an 83-gene, 88 taxon angiosperm data set yield an identical tree topology as previous plastid-based trees, and moderately support the sister relationship between Buxaceae and Gunneridae. Molecular dating analyses suggest that Tetracentron and Trochodendron diverged between 44-30 million years ago, which is congruent with the fossil record of Trochodendrales and with previous estimates of the divergence time of these two taxa. We also characterize 154 simple sequence repeat loci from the Tetracentron sinense and Trochodendron aralioides plastomes that will be useful in future studies of population genetic structure for these relict species, both of which are of conservation concern.

The relationships between self-efficacy, self-care behavior, anxiety, and depression for Taiwanese individuals with type 2 diabetes were determined in this study. Depression and anxiety are common symptoms that can contribute toward adverse medical outcomes. A descriptive, cross-sectional, correlational design was used. The sample comprised 201 patients with type 2 diabetes from diabetes outpatient clinics at three teaching hospitals in Taiwan. Four new lignans, strebluslignanol F (1), (7R,8S,7?R,8?S)-erythro-strebluslignanol G (2), isomagnaldehyde (3) and isostrebluslignanaldehyde (4), along with 12 known lignans (5-16) were isolated from the ethyl acetate-soluble part of MeOH extract of the root of Streblus asper. Their structures were elucidated through various spectroscopic methods, including 1D NMR ((1)H NMR, (13)C NMR), 2D NMR (HMQC, HMBC and NOESY) and HRMS. The stereochemistry at the chiral centers was determined using CD spectra, as well as analyses of coupling constants and optical rotation data. The isolated lignans were evaluated for their anti-HBV activities in vitro using the HBV transfected HepG2.2.15 cell line. The most active lignans, (7R,8S,7?R,8?S)-erythro-strebluslignanol G, magnolol, isomagnolol and isolariciresinol, exhibited significant anti-HBV activities with IC50 values of 1.58, 2.03, 10.34 and 3.67 ?M, respectively, for HBsAg with no cytotoxicity, and of 3.24, 3.76, 8.83 and 14.67 ?M, respectively, for HBeAg with no cytotoxicity. (7R,8S,7?R,8?S)-erythro-Strebluslignanol G and magnolol showed significant anti-HBV activities to inhibit the replication of HBV DNA with the IC50 values of 9.02 and 8.67 ?M, respectively.

Antimicrobial peptides have been widely identified from amphibian skins except salamanders. A novel antimicrobial peptide (CFBD) was isolated and characterized from skin secretions of the salamander, Cynops fudingensis. The cDNA encoding CFBD precursor was cloned from the skin cDNA library of C. fudingensis. The precursor was composed of three domains: signal peptide of 17 residues, mature peptide of 41 residues and intervening propeptide of 3 residues. There are six cysteines in the sequence of mature CFBD peptide, which possibly form three disulfide-bridges. CFBD showed antimicrobial activities against Staphylococcus aureus, Bacillus subtilis, Candida albicans and Escherichia coli. This peptide could be classified into family of ?-defensin based on its seqeuence similarity with ?-defensins from other vertebrates. Evolution analysis indicated that CFBD was close to fish ?-defensin. As far as we know, CFBD is the first ?-defensin antimicrobial peptide from salamanders.

Temporal and spatial variations in the water status of walnut trees (Juglans regia L.) and the soil in which they were growing were traced by analyzing the differences in hydrogen isotopes during spring and summer in a 7-year-old walnut stand. Walnut root dynamics were measured in both dry and wet seasons. Walnut roots were mainly distributed in the upper soil (0-30 cm depth), with around 60% of the total root mass in upper soil layers and 40% in deep soil layers (30-80 cm depth). The upper soil layers contributed 68% of the total tree water requirement in the wet season, but only 47% in the dry season. In the wet season, total roots, living roots and new roots were all significantly more abundant than in the dry season. There were significant differences in pre-dawn branch percentage loss of hydraulic conductance (PLC), pre-dawn leaf water potential and transpiration between the dry and wet seasons. Water content in the upper soil layers remarkably influenced xylem water stable-hydrogen isotope (?D) values. Furthermore, there were linear relationships between the xylem water ?D value and pre-dawn branch PLC, pre-dawn leaf water potential, transpiration rate and photosynthetic rate. In summary, J. regia was compelled to take a larger amount of water from the deep soil layers in the dry season, but this shift could not prevent water stress in the plant. The xylem water ?D values could be used as an indicator to investigate the water stress of plants, besides probing profiles of soil water use.

Transpiration is a major component of vegetation evapotranspiration, and a core in the study of plant water physiological ecology. Its measurement methods attracted extensive attention, among which, thermal dissipation is considered as an optimal method for measuring tree transpiration. Numerous studies showed that thermal dissipation method was relatively accurate in measuring individual tree transpiration and stand-scale water consumption. However, there exist potential errors between the true value and the measurements during measurement process. In this paper, the potential errors of thermal dissipation method in measuring sap flux density and of the temperature difference determination from single tree to stand-scale were reviewed, and the research prospects on the potential errors of thermal dissipation method in China were discussed. The corresponding solutions were also proposed.

The negative effects of above problems are discussed in this paper. In order to relieve the conflict between the increase of genetics contents and the decrease of teaching hours in genetics teaching of undergraduates and provide reference for future textbook compilation, some approaches on solving repetition of teaching content and suggestions on joining problems are put forward.

The correlation of glutathione S-transferase (GST) M1/T1 genetic polymorphisms with oxidative stress-related chronic diseases was proved recently. The aim of the present study was to investigate the association of GSTM1/T1 genetic polymorphisms with antioxidant biomarkers and consumption of fruits and vegetables (F&V) in healthy subjects. In this study, for conducting a 3 d dietary survey, 190 healthy adults were recruited. After DNA extraction, a multiple PCR method was used for GSTM1/T1 genotyping. A spectrophotometer method was applied for the determination of plasma total antioxidant capacity (T-AOC), vitamin C level and erythrocyte GST enzyme activity. A general linear model was used to compare the mean values of antioxidant parameters for different GSTM1/T1 genotypes and consumption of F&V. Polymorphisms of GSTM1/T1 had no effects on plasma T-AOC and vitamin C levels. Deletion of the GSTM1 gene decreased the erythrocyte GST activity. There was correlation between plasma T-AOC and consumption of F&V in the GSTM1? or GSTT1? subjects. A similar pattern was evident for erythrocyte GST activity in the GSTM1? subjects. No association was found among consumption of F&V and GSTM1/T1 genotypes and plasma vitamin C level. Different consumption of F&V had no impact on plasma T-AOC and vitamin C levels in the GSTM1?/GSTT1? or GSTM1?/GSTT1? subjects. The erythrocyte GST activity was more sensitive to consumption of F&V in the individuals with the GSTM1?/GSTT1? genotype. Association was found among GSTM1/T1 genotypes, antioxidant parameters and consumption of F&V. Large-scale and multiple ethnic studies are needed to further evaluate the relationship.

To investigate the association between the single nucleotide polymorphisms (SNPs) in adiponectin gene promoter and essential hypertension (EH) in Chinese Korean and Han of Yanbian area, 220 EH patients and 268 normotensive control individuals were enrolled. PCR and direct DNA sequencing were used to determine the -11426A>G (rs16861194), -11391G>A (rs17300539), -11377C>G (rs62620185), -11156insCA (rs60806105), and -11043C>T (rs76786086) SNPs in the promoter region of adiponectin gene. Total cholesterol (TC), the triglyceride (TG), fasting plasma glucose (FPG), low density lipoprotein-cholesterol (LDL-C), and high density lipoprotein-cholesterol (HDL-C) levels were examined by oxi-dase method. The plasma adiponectin and insulin were measured by enzyme linked immunosorbent assay (ELISA). The results showed that: (1) SNPs of -11426A>G, -11377C>G, and -11156insCA were found and in Hardy-Weinberg equilib-rium (P>0.05), but not the case in -11391G>A and -11043C>T. (2) -11426A>G and -11156insCA were perfectly in link-age disequilibrium (D=1; r2=1). (3) The allele G frequency of -11426A>G polymorphism in Chinese Korean (21.10%) was significantly higher than that in Chinese Han (12.50%), and also higher in EH group than in the control group of Chinese Han. The genotype and allele frequencies of -11377C>G showed no significant difference between the two groups ob-served. (4) The haplotype -11426G -11377C frequency in EH of Chinese Han was higher than in the control group (P<0.05). (5) The EH showed lower plasma adiponectin level compared with the control group (P<0.001) in both Chinese Korean and Han. Our results indicate that: (1) the perfect linkage disequilibrium of -11426A>G and -11156insCA is first reported, and the SNP of -11426A>G is associated with Chinese Han and Korean. (2) -11426 G and -11426G -11377C are risk factor and risk haplotype in Yanbian Chinese Han, but not in Chinese-Korean. (3) The lower hypoadiponectinemia is the important risk factors for EH in Chinese Korean and Han of Yanbian area. (4) There is no relationship between -11426A>G polymorphism and the plasma adiponectin level.

Gallbladder cancer is the most common malignant tumor of the biliary tract. Early diagnosis of gallbladder cancer is difficult because of the latent onset and lack of good biomarkers. To identify new biomarkers that improve the early diagnosis and/or serve as possible therapeutic targets in gallbladder cancer is essential. In the present study, serum proteins were separated by two-dimensional gel electrophoresis (2-DE) in 3 patients with gallbladder cancer and 3 healthy volunteers. The differentially expressed spots were identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Western blotting and immunohistochemistry were performed to verify the expression of certain candidate proteins. Protein expression and clinical correlation was evaluated. We found that 64 protein spots were significantly changed in gallbladder cancer. Twenty-four proteins including S100A10, haptoglobin, cystatin-B, profilin-1 and superoxide dismutase were successfully identified. Among these proteins, S100A10 and haptoglobin were validated using Western blotting. Immunohistochemically, the expression of S100A10 and haptoglobin proteins was found to be higher in gallbladder cancer tissues compared to that in gallbladder adenoma, liver cholangiocarcinoma and cholecystitis tissue. Patients with high expression of S100A10 and haptoglobin were linked to late stage disease and poor clinical prognosis. Our data suggest that combined comparative proteomic analysis by 2-DE and MALDI-TOF-MS is an effective method for identifying differentially expressed proteins in serum samples. These proteomic approaches could be used for identifying new serum biomarkers in gallbladder cancer. S100A10, haptoglobin and other identified proteins may be potential molecular targets for early gallbladder cancer diagnostics and therapeutic applications.

Motion information scalability is an important requirement for a fully scalable video codec, especially in low bit rate or small resolution decoding scenarios, for which the fully scalable motion model (SMM) has been proposed. SMM can collaborate flawlessly with other scalabilities, such as spatial, temporal and quality, in a scalable video codec. It performs better than the nonscalable motion model. To further improve the SMM, this paper extends the algorithm to support the hierarchical B frame structure and bidirectional or multidirectional motion estimation. Furthermore, the corresponding rate distortion optimized estimation for improved efficiency in several scenarios is discussed. Several simulation results based on the updated framework are presented to verify the advantage of this extension.

Three molecularly imprinted polymers were synthesized with different functional monomers respectively, using methyl parathion as the template. These monomers are methacrylic acid, acrylamide and 4-vinylpyridine. According to the UV spectrum study the interaction between methyl parathion and 4-vinylpyridine was stronger than that of the others. Comparably, the infrared spectrum study showed the same results which indicated that 4-vinylpyridine could associate with the template at two different kinds of binding sites, the P-O-C and the -NO2 site, and is most likely to form steady covalent bonds with methyl parathion, while the other two monomers could only associate with the template at the P-O-C site. Furthermore, the infrared spectra of the synthesized polymers confirmed the existence of the functional groups in each kind of polymer, which might interact with the template.

Motion scalability is designed to improve the coding efficiency of a scalable video coding framework, especially in the medium to low range of decoding bit rates and spatial resolutions. In order to fully benefit from the superiority of motion scalability, a rate-distortion optimized bitstream extractor, which determines the optimal motion quality layer for any specific decoding scenario, is required. In this paper, the determination process first starts off with a brute force searching algorithm. Although guaranteed by the optimal performance within the search domain, it suffers from high computational complexities. Two properties, i.e., the monotonically nondecreasing property and the unimodal property, are then derived to accurately describe the rate-distortion behavior of motion scalability. Based on these two properties, modified searching algorithms are proposed to reduce the complexity (up to five times faster) and to achieve the global optimality, even for those decoding scenarios outside the search domain.

The study purpose was to evaluate the effectiveness of a structural intervention in reducing unprotected sexual behaviours, increasing condom accessibility and their relationship to the prevalence of HIV infection and sexually transmitted infections (STIs) in gay bathhouse attendees. A quasi-experimental study with a non-equivalent control group for comparison was conducted at nine gay bathhouses in Taiwan from 2004 to 2006. A structural intervention designed to increase condom distribution and accessibility inside the bathhouses was implemented at one bathhouse in each city. Bathhouse attendees were invited to complete a questionnaire and to be screened for HIV infection and STIs. Of 632 study participants, 270 were surveyed six months after the intervention was conducted. At the time of follow-up after controlling for baseline data, patrons attending bathhouses for which intervention was performed were more likely to report accessing condoms inside bathhouses than those attending control bathhouses (92.6% versus 81.3%, P = 0.016), and condoms were more likely to be available at the reception desk of the bathhouse entrance (87.5% in intervention bathhouses versus 69.4% in control, P = 0.047). In a multivariate analysis adjusted for age, access to condoms inside bathhouses (adjusted odds ratio (AOR): 4.35; 95% confidence interval (CI): 1.48-12.78) and attendance at bathhouses with intervention (AOR: 2.07; 95% CI: 1.07-3.97) were independently associated with consistent condom use during anal intercourse. There were no significant differences in the prevalences of HIV infection and STIs at the six-month follow-up. Bathhouses with structural intervention were associated with consistent condom use during anal intercourse among bathhouse patrons. Our findings highlight the importance of extension of condom distribution inside the bathhouses to increase condom use among bathhouse patrons.

A rapid, sensitive and selective ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method with hydrophilic interaction chromatography has been developed and validated for the simultaneous determination of didanosine and valdidanosine (L-valine amino acid ester prodrug of didanosine) in rat plasma. Solid-phase extraction (SPE) column was employed to extract the analytes from rat plasma, with high extraction recovery (>85%) for both didanosine and valdidanosine. The analytes were then separated by hydrophilic interaction chromatography (HILIC column) and detected by a triple-quadrupole mass spectrometry equipped with an electrospray ionization (ESI) source. The method was linear over the concentration ranges of 2-20,000 ng/mL for didanosine and 4-300 ng/mL for valdidanosine. The lower limit of quantitation (LLOQ) of didanosine and valdidanosine was 2 and 4 ng/mL, respectively. The intra-day and inter-day relative standard deviation (RSD) were less than 15% and the relative errors (RE) were all within 15%. Finally, the validated UPLC-MS/MS method was successfully applied to the pharmacokinetic study after either didanosine or valdidanosine orally administrated to the Sprague-Dawley rats.

Chinese jujube (Ziziphus jujube Mill.) is important economically for its fruit and also bears attractive medical value. Its flower development concurs with the growth of current fruit bearing shoots (FBSs). However, events involved in current FBSs, including FBS differentiation, flower development, and reliable housekeeping gene are still unknown. In this study, the morphology of FBS development and floral formation were examined and expression profiles of several potential internal control genes was determined. These included genes encoding proteins involved in protein translation (translation elongation factor 1alpha, ZjEF1), protein folding (cyclophilin, ZjCyP) or degradation (ubiquitin extension protein, ZjUBQ), and proteins involved in the structure of the cytoskeleton (beta-actin, ZjACT) or nucleosome (histone3, ZjH3). Our results showed that the floral development in early growing FBSs (less than 20 mm in length) or shoot apices was not complete. Among ZjACT, ZjEF1, ZjCyP, ZjUBQ, and ZjH3, ZjH3 was the most suitable housekeeping gene to evaluate FBS development, based on their expression in early growing FBSs, shoot apices, and different organs. These results will be useful for further molecular mechanism study about FBS development in Chinese jujube.

The objective of the present investigation was to develop and evaluate a drug-in-adhesive transdermal patch for S-amlodipine (S-AM) free base. Initial in vitro experiments were conducted to optimize the formulation parameters before transdermal delivery in rats. The effects of the type of adhesive and the content of permeation enhancers on S-AM free base transport across excised rat skin were evaluated. For in vivo studies, patches were administered transdermally to rats while orally administered S-AM in suspension and intravenously administered S-AM solution were used as controls. The plasma level of S-AM following transdermal application could be maintained for 72 h. After transdermal administration to rats, the absolute bioavailability was 88.8% for S-AM free base. After dose normalization, the areas under the plasma concentration-time curve (AUC) and mean residence times (MRT) were evidently increased and extended, respectively. This suggests that the transdermal application of S-AM in a drug-in-adhesive transdermal patch may be used for the treatment for hypertension.

Objective of this study is to develop and evaluate the new solid self-emulsifying (SE) pellets of poorly soluble nitrendipine (NTD). These pellets were prepared via extrusion/spheronization technique, using liquid SEDDS (NTD, Miglyol 812, Cremophor RH 40, Tween 80, and Transcutol P), adsorbents (silicon dioxide and crospovidone), microcrystalline cellulose and lactose. The resulting SE pellets with 30% liquid SEDDS exhibited uniform size (800-1000 microm) and round shape, droplet size distribution following self-emulsification was nearly same to the liquid SEDDS (72+/-16 nm and 64+/-12 nm). The in vitro release was similar for the two SE formulations (over 80% within 30 min), both significantly higher than the conventional tablets (only 35% within 30 min). The oral bioavailability was evaluated for the SE pellets, liquid SEDDS and conventional tablets in fasted beagle dogs. AUC of NTD from the SE pellets showed 1.6-fold greater than the conventional tablets and no significant difference compared with the liquid SEDDS. In conclusion, our studies illustrated that extrusion/spheronization technique could be a useful large-scale producing method to prepare the solid SE pellets from liquid SEDDS, which can improve oral absorption of NTD, nearly equivalent to the liquid SEDDS, but better in the formulation stability, drugs leakage and precipitation, etc.

A full-length cDNA coding lipoprotein lipase (LPL) was cloned from liver of adult common carp (Cyprinus carpio Var. Jian) by RT-PCR and rapid amplification of cDNA ends (RACE) approaches. The cDNA obtained was 2,411 bp long with a 1,524 bp open reading frame (ORF) encoding 507 amino acids. This amino acid sequence contains two structural regions: N-terminus (24-354 residues) and C-terminus (355-507 residues). Before N-terminus, 1-23 residues is signal peptide, 6-23 residues is transmembrance helix. At N-terminus, some conversed functional sites were found, including two N-linked glycosylation sites Asn(41) and Asn(88); one catalytic triad Ser(174), Asp(198) and His(283); one conserved heparin-binding site Arg(321) to Arg(324) (RKNR); eight cysteines residues Cys(69) and Cys(82), Cys(258) and Cys(281), Cys(306) and Cys(325), Cys(317) and Cys(320) which are involved in four disulfide bridges; one polypeptide "lid" that participates in substrate specificity. At C-terminus, Asn(401) is another N-linked glycosylation site, and Trp(434) and Trp(435) (WW) is lipid-binding site. The amino acid sequence has a high similarity, and shows similar structural features to LPL of other species. Tissue distribution of LPL mRNA in liver, head kidney, mesenteric adipose tissue, heart and white muscle of common carp was analyzed by semi-quantitative RT-PCR method using beta-actin gene as internal control. The result showed that the expressions of LPL mRNA were detected in all examined tissues of common carp. The expression levels of LPL in the mesenteric adipose tissue was highest among these tissues, following in liver and head kidney, and the lowest expression was found in heart and white muscle.

We report the favorable outcome of intravitreal injection of bevacizumab (Avastin) to treat choroidal neovascularisation (CNV) secondary to angioid streaks in an 82-year-old Chinese man. Within 2 weeks of bevacizumab injection, visual acuity improved from 20/40 to 20/25 and optical coherence tomography revealed complete resolution of CNV and retinal edema. The juxtafoveal hemorrhage nearly disappeared, and the size of CNV markedly regressed from 1,200 microm to unremarkable on fluorescein angiography. These improvements were maintained after 2 additional doses of intravitreal bevacizumab administered at 4-week intervals. No ocular or systemic side effects were observed. Neither recurrent CNV lesion nor new hemorrhage was noted during 18 months of follow-up. The short-term results suggest that intravitreal injection of bevacizumab for the treatment of CNV in patients with angioid streak is well tolerated and highly effective. It may be a promising pharmacologic treatment option since limited therapies are available for this disease with a rather aggressive course.

Progesterone (P) has been suggested as a bone-trophic hormone. Previous studies have shown that P promoted bone formation by stimulating the proliferation and differentiation of osteoblasts. But, the effect of P on apoptosis of osteoblast in vitro has not been reported. We propose that P may promote bone formation by suppressing the apoptosis of osteoblast. The present study was performed to investigate the effect of P on apoptosis of murine MC3T3-E1 osteoblastic cells. Cell apoptosis was measured by acidine orange/ethidium bromide (AO/EB) staining and sandwich-enzyme-immunoassay. Progesterone receptor (PR), cytochrome c, caspase-9 and caspase-3 protein levels were determined by Western blot analysis. The enzyme substrate was also used to assess the activation of caspase-3 and caspase-9. Progesterone suppressed MC3T3-E1 cells apoptosis induced by serum deprivation, and this effect was blocked by a PR antagonist RU486. Furthermore, the suppressive effects of P on cytochrome c release and caspase-9 and caspase-3 activation in serum-deprived MC3T3-E1 cells were also reversed by RU486. Our study demonstrated that P protects osteoblast against apoptosis through PR and the downstream mitochondrial pathway. Thus, the data suggest that the effects of P on osteoblast apoptosis may contribute to the mechanisms by which P exerts its action on bone formation.

Although it is a crucial cellular process required for both normal development and to face stress conditions, the control of programmed cell death in plants is not fully understood. We previously reported the isolation of ATXR5 and ATXR6, two PCNA-binding proteins that could be involved in the regulation of cell cycle or cell death. A yeast two-hybrid screen using ATXR5 as bait captured AtIPS1, an enzyme which catalyses the committed step of myo-inositol (MI) biosynthesis. atips1 mutants form spontaneous lesions on leaves, raising the possibility that MI metabolism may play a role in the control of PCD in plants. In this work, we have characterised atips1 mutants to gain insight regarding the role of MI in PCD regulation.

This paper presents the design, fabrication, and characterization of an electrostatically actuated user-reconfigurable elastomer microfluidic system intended for very large scale integration (VLSI) microfluidics. By adding thin film metal flexures into the PDMS polymer, individual elastomer channels were made to self-close without the use of pneumatics via the application of 15-20 V, 5 MHz signals synthesized digitally by a microcontroller and a radio-frequency amplifier IC. These valves were arranged into hexagonal or quadricular arrays with 75% fill factor. During use, valves were selected to be permanently closed, permanently open or addressable; this allowed for the on-the-fly determination of channels, valves and pumps. We present characterization of flow and pressure data for valves and pumps and demonstrate various multi-component configurations of the system capable of pumping, mixing, splitting and circulating flow. The presented technology is compatible with standard PDMS microfluidics, has actuation voltages low enough to be driven by commercial CMOS ICs and can be used to displace aqueous, gaseous and lipid phases. The primary contribution of this technology is to provide a scalable non-pneumatic platform for the very large scale integration of microfluidic total analysis systems.

A full-length cDNA coding lipoprotein lipase (LPL) was cloned from liver of adult Pengze crucian carp (Carassius auratus var. Pengze) by RT-PCR and rapid amplification of cDNA ends (RACE) approaches. The cDNA obtained was 1877 bp long with a 1524 bp open reading frame (ORF) encoding 507 amino acids, including a putative signal peptide of 23 amino acids long. The deduced amino acid sequence has a high similarity and shows similar structural features to LPL of other species. The LPL protein has a calculated molecular mass of 57.7 kDa and isolectric point of 7.85. Tissue distribution of LPL mRNA in mesenteric adipose tissue, liver, heart, head kidney and white muscle of adult Pengze crucian carp was analyzed by semi-quantitative RT-PCR method using beta-actin gene as internal control, the result showed that this gene was ubiquitously expressed in all tissues tested with the highest abundance in mesenteric adipose tissue, following in head kidney and liver, and the lowest expression was found in heart and white muscle.

Aiming at the deficiencies in the researches about the probability distribution model for mixed forests tree measurement factors, a joint maximum entropy probability density function was put forward, based on the maximum entropy principle. This function had the characteristics of 1) each element of the function was linked to the maximum entropy function, and hence, could integrate the information about the probability distribution of measurement factors of main tree species in mixed forests, 2) the function had a probability expression of double-weight, being possible to reflect the characteristics of the complex structure of mixed forests, and accurately and completely reflect the probability distribution of tree measurement factors of mixed forests based on the fully use of the information about the probability distribution of measurement factors of main tree species in mixed forests, and 3) the joint maximum entropy probability density function was succinct in structure and excellent in performance. The model was applied and tested in two sampling plots in Tianmu Mountain Nature Reserve. The fitting precision (R2 = 0.9655) and testing accuracy (R2 = 0.9772) were both high, suggesting that this model could be used as a probability distribution model for mixed forests tree measurement factors, and provided a feasible method to fully understand the comprehensive structure of mixed forests.

Integrated care for severely disabled long-term care facility residents: is it better?

The demands of long-term care facilities (LTCFs) residents are complex which usually require a range of professionals and caregivers to provide treatment and care. To reduce this fragmentation of care, integrated care models are developed in modern health care system, and a gradual change from traditional LTCF care to integrated care has occurred in many countries. Although integrated care is assumed to improve the quality of care, evidences supporting these effects are insufficient. We recruited 7 private LTCF (74 residents) in northern Taipei and randomized them into integrated care model (N=42, mean age=82.8+/-8.0 years, 54.8% males) and traditional model (N=32, 81.7+/-8.8 years, 43.8% males). Integrated care model group was provided an actively working interdisciplinary team in addition to traditional nursing and personal care in traditional model group. Physical function, nutritional status and several quality indicators (unplanned feeding tube replacement, unplanned urinary catheter replacement, pneumonia, urinary tract infection and so on) were compared with both groups. Overall, LTCF residents in the integrated care model group showed significant improvement in serum levels of albumin (3.78+/-0.32 vs. 3.60+/-0.45, p=0.004) and hemoglobin (12.62+/-1.58 vs. 12.03+/-1.24, p=0.004) during the study period. Among selected quality indicators, subjects in integrated care model group were similar to traditional model group except that integrated care model group had a significantly reduced unplanned feeding tube replacement rate. In conclusion, the clinical effectiveness of integrated care model among severly disabled LTCF residents is minimal and a further cost-effectiveness study is needed to promote optimal quality of care in this setting.

The purpose of this study was to investigate the relationships among the self-efficacy, professional commitment, and job satisfaction of diabetic health-care personnel. The research design was cross-sectional. Three teaching hospitals, one from each of northern, middle and southern parts of Taiwan, were selected for data collection and used questionnaires to collect data; 202 participants were recruited. The demographic data for job title and job satisfaction were not significantly different (F = 2.13, P = 0.090). Self-efficacy was significantly positively correlated with professional commitment (r = 0.29, P = 0.000) and with job satisfaction (r = 0.14, P = 0.041). A total of 34.1% of the variance in job satisfaction was explained by age, years of service in the medical or nursing field, the actual number of years caring for patients with diabetes, self-efficacy, and professional commitment. Understanding the self-efficacy and professional commitment of medical and nursing personnel can help increase job satisfaction and improve the quality of medical and nursing care.

The grasshoppers are ideal materials to study various meiotic stages of spermatogenesis due to their easy availability, fairly large chromosomes, and fewer numbers of chromosomes. It is easy to make temporary squash preparation of grasshopper testes; however, it is usually difficult for the beginners to differentiate between stages of meiosis. In view of this, we demonstrated the method of identification of meiotic stages by chromosome number and chromosome conformation, taking spermatogonial meiosis of Locusta migratoria manilensis as an example. We described briefly the mitosis of spermatogonia and the spermatogenesis of this species as well.

We describe the phylogenetic analysis and expression pattern of the Xenopus radial spoke protein 3 (RSP3) gene during early development. The Xenopus RSP3 protein presents characteristic features of the RSP3 family. It contains a radial spoke domain, which is 75 and 72 % identical to the corresponding region of human and Chlamydomonas RSP3 proteins, respectively. Examination of the phylogenetic relationship between the Xenopus RSP3 protein and its known homologues from different deuterostomes indicates that the RSP3 proteins are highly conserved among deuterostomes. Whole-mount in situ hybridization analyses show that Xenopus RSP3 is a maternal mRNA enriched in the animal hemisphere during cleavage stages. The expression is detected in the dorsal region of the embryo during gastrulation, then in the presumptive neuroectoderm at the end of gastrulation. During neurulation and at the subsequent stages, the expression of RSP3 mRNA is detected in the entire multiciliated cells of epidermis. At tail-bud stages, it is progressively expressed in the otic vesicles and sequentially expressed in the nephrostomes. Expression could be also detected in the floor plate of the neural tube. This expression pattern persists until at least late tail-bud stages.

This paper assessed the coastal environmental quality along the Beibu Gulf using a statistical approach, multi-biomarker pollution index (MPI). Samples of clam (Paphia undulata) and sediment were collected at nine sites from the intertidal zone of Beibu Gulf in April 2011. Nine biomarkers of response were measured both in gill and digestive gland in Paphia undulata, and twelve kinds of contaminants were measured in different sediment samples. According to the Pearson s correlation coefficients between biomarker responses in Paphia undulata and contaminant levels in sediments, five biomarkers either in gill for oxidized glutathione (GSSG), reduced glutathione (GSH), glutathione S-transferase (GST) or in digestive gland for thiobarbituric acid reactive substances (TBARS) and glutathione peroxidase (GPx) were selected for MPI calculation. For each biomarker at each site, a response index was allocated, and the MPI value of this site was calculated as the sum of the response index of the five biomarkers. The results of the calculation (MPI values from 18 to 39) showed significant differences among sampling sites. The environmental quality of all sites ranged from class 1 (clean) to class 3 (lightly contaminated), and no site fell into class 4 (contaminated) or class 5 (heavily contaminated), indicating a good environmental quality in the intertidal zone of Beibu Gulf. However, the environmental quality at some sites (S1, S3, 54 and S7) fell into class 3 (lightly contaminated), indicating mild interference from human activities has occurred.

Wild Secale species, Secale africanum Stapf., serve as a valuable source for increasing the diversity of cultivated rye (Secale cereale L.) and provide novel genes for wheat improvement. New wheat - S. africanum chromosome 1R(afr) addition, 1R(afr)(1D) substitution, 1BL.1R(afr)S and 1DS.1R(afr)L translocation, and 1R(afr)L monotelocentric addition lines were identified by chromosome banding and in situ hybridization. Disease resistance screening revealed that chromosome 1R(afr)S carries resistance gene(s) to new stripe rust races. Twenty-nine molecular markers were localized on S. africanum chromosome 1R(afr) by the wheat - S. africanum introgression lines. Twenty markers can also identically amplify other reported wheat - S. cereale chromosome 1R derivative lines, indicating that there is high conservation between the wild and cultivated Secale chromosome 1R. Nine markers displayed polymorphic amplification between S. africanum and S. cereale chromosome 1R(afr) derivatives. The comparison of the nucleotide sequences of these polymorphic markers suggested that gene duplication and sequence divergence may have occurred among Secale species during its evolution and domestication.

By using Circumference Dendrometer 2 (DC2), this paper studied the diurnal variation of trunk diameter in Quercus variabilis plantation at the south aspect of Taihang Mountains. During seasonal drought, the diurnal variation of Q. variabilis trunk diameter was quite evident. The time of the diameter shrinkage followed the start-up time of sap flow, but the appearance of the minimum trunk diameter lagged behind the maximum sap flow flux about 3-4 h. The maximum daily shrinkage (MDS) value of the trunk diameter presented a trend low-high-low, being significantly correlated with the diurnal differences of cumulative sap flow flux and leaf water potential and having a significant quadratic relationship with soil moisture content. The MDS value was affected by the variations of meteorological factors, being significantly correlated with the diurnal variations of air temperature, vapor pressure deficit, and relative humidity, but less correlated with the diurnal variation of solar radiation. After successive precipitation, soil moisture content was no longer the limiting factor of the diurnal variation of Q. variabilis trunk diameter. The MDS value had less correlations with the diurnal differences of cumulative sap flow flux, leaf water potential, soil moisture content, and other meteorological factors. Stepwise regression analysis indicated that the soil moisture content and air temperature in seasonal drought and rain seasons were the key factors affecting the diurnal variation of Q. variabilis trunk diameter.

Based on the canopy temperature and micrometeorological data of Quercus variabilis in its main growth season (from May to August) in hilly areas of North China in 2011, this paper analyzed the variations of canopy temperature (T(c)) in Q. variabilis plantation and their relations with micrometeorological factors in typically clear days and cloudy days. From 9:00 to 17:00 in clear days, the boundary layer of canopy was unstable, and the mean T(c) was 3.55 degrees C higher than the mean air temperature (T(a)). In cloudy days, the variations of T(c) were gentler than those in clear days. The T(c) was significantly correlated with T(a), net solar radiation (R(n)), relative humidity, and wind speed, with a multiple correlation coefficient being 0. 825. The T(a) and R(n) were the dominant meteorological factors controlling T(c), and their affecting degree on T(c) was associated with weather condition.

Large-scale forward genetic screening analysis of development of hematopoiesis in zebrafish.

Zebrafish is a powerful model for the investigation of hematopoiesis. In order to isolate novel mutants with hematopoietic defects, large-scale mutagenesis screening of zebrafish was performed. By scoring specific hematopoietic markers, 52 mutants were identified and then classified into four types based on specific phenotypic traits. Each mutant represented a putative mutation of a gene regulating the relevant aspect of hematopoiesis, including early macrophage development, early granulopoiesis, embryonic myelopoiesis, and definitive erythropoiesis/lymphopoiesis. Our method should be applicable for other types of genetic screening in zebrafish. In addition, further study of the mutants we identified may help to unveil the molecular basis of hematopoiesis.

The choice of internal control genes is important since it may affect the study outcome in RT-qPCR. Indeed, it is well-known that expression levels of traditional internal control genes can vary across tissue types and across experimental settings within one specific tissue type. The aim of this study is an evaluation of a set of housekeeping genes (HKGs) to be used in the normalization of gene expression in vitro different cultured cells, THP-1 and K562. The transcriptional stability of eleven potential internal control genes (RPL37A, ACTB, GAPDH, B(2)M, PPIB, PGK1, PPIA, SDHA, TBP, HPRT1 and RPL13A) were evaluated using RT-qPCR and were compared in different treatment, that was un-stimulated or LPS-stimulated cells. The raw Ct values were determined for each candidate gene at different time points following LPS-stimulated or unstimulated cells. Furthermore, all data were analyzed by the geNorm, BestKeeper, and NormFinder validation programs. Results indicated that PPIB and PGK1 were the most stable internal control genes in this study. RPL13A was found to be the least stable. This study provides the comprehensive reported assessment of internal control genes for use in expression studies in vitro cultured cells. These findings further emphasize the need to accurately validate candidate internal control genes in the study before use in gene expression studies using RT-qPCR.

In order to understand the accuracy of Granier s thermal dissipation method in measuring tree water consumption, a comparative study was made from May to June, 2010. The sap flow density of potted Platycladus orientalis was measured with thermal dissipation probe, which was compared with the whole-plant gravimetric measurement. There were significant linear relationships (R2 > 0.825) between the sap flow velocity in both north and south directions of P. orientalis measured by thermal dissipation probe and the transpiration rate measured by gravimetric method. The average daily sap flux in the north and south directions of P. orientalis were 10.6% and 15.1% lower than the daily average transpiration of P. orientalis, respectively, but the differences were not significant. Therefore, Graniers method had high reliability in the measurement of P. orientalis transpiration at daily scale, though the large temperature fluctuation between day and night could result in a lower daily sap flux than daily transpiration.

Biomarkers are applied worldwide in marine environmental assessment due to their "early warning" function to chemical pollutants. Several integrative index approaches such as multi-marker pollution index (MPI), integrated biomarker response (IBR), bioeffect assessment index (BAI), biomarker response index (BRI), and health assessment index (HIS), have been developed based on biomarkers. By transforming the complex alterations of biomarkers into a single class or value, these approaches have been so far the useful tools for assessing the environmental quality. This review summarized the establishment of evaluation indicator system, the calculation of integrative index, the grading of pollution status, and the practical applications of each approach, and discussed the existing problems in marine environmental assessment based on biomarker index. Some improving suggestions were also proposed.

A novel bioactive peptide (polypedarelaxin 1) was identified from the skin secretions of the tree frog, Polypedates pingbianensis. Polypedarelaxin 1 is composed of 21 amino acid residues with a sequence of QGGLLGKVSNLANDALGILPI. Its primary structure was further confirmed by cDNA cloning and mass spectrometry analysis. Polypedarelaxin 1 was found to elicit concentration-dependent relaxation effects on isolated rat ileum. It has no antimicrobial and serine protease inhibitory activities. BLAST search revealed that polypedarelaxin 1 did not show similarity to known proteins or peptides. Especially, polypedarelaxin 1 do not contain conserved structural motifs of other amphibian myotropic peptides, such as bradykinins, bombesins, cholecystokinin (CCK), and tachykinins, indicating that polypedarelaxin 1 belongs to a novel family of amphibian myotropic peptide.

Strain-insensitive temperature sensing with a dual polarization fiber grating laser.

In this paper, a strain-insensitive temperature sensor based on a dual polarization fiber grating laser is demonstrated. The laser is fabricated by inscribing two wavelength-matched Bragg gratings in an Er-doped fiber. It emits single-longitudinal-mode output in wavelength domain and generates a RF-domain signal as a beat note between the two polarization modes. A temperature sensor has been exploited by monitoring the beat frequency. The measured temperature sensitivity is -78.46 kHz/°C. Theoretical analysis suggests that the temperature response is a result of both the differences in thermo-optic coefficient and thermal expansion between the core and cladding. In contrast, the sensor is almost insensitive to applied axial strain. We found that the strain insensitivity is due to the compensation between the strain-induced birefringence change and the effect of the elongation/material index change. The proposed sensor can be applied for reliable and precise measurement of temperature independently, towards the applications in structural integrity, oil-well monitoring, aerospace engineering, and process control.

Effectiveness of a community-based health promotion program targeting people with hypertension and high cholesterol.

The purpose of this study was to evaluate the effectiveness of a community-based health promotion program targeting people with hypertension and high cholesterol. A pre-experimental study was conducted. A total of 60 residents were recruited to participate. Participants were assessed at baseline and at a 6 month follow up at a regional hospital in northern Taiwan. The questionnaires used for data collection consisted of an assessment of self-efficacy, self-care activities, health outcomes, and physical fitness. Several teaching resources were used, including a DVD, a self-care booklet, group support (exercise and counseling sessions), and telephone follow up. A significant decrease in waist circumference (t = 2.20, P = 0.03) and high density lipoprotein-cholesterol level (t = 4.71, P < 0.00) was found at follow up. Moreover, the level of physical fitness activity, and sit-ups specifically (t = 3.10, P < 0.00), was increased. Participants also showed significant increases between baseline and 6 month follow up in their efficacy expectation score (t = -5.81, P < 0.00), outcome expectation scores (t = -4.76, P < 0.00) and self-care behavior scores (t = -2.78, P = 0.007). The community-based health promotion program is an effective means of helping people with hypertension and high cholesterol and should be instituted regularly and evaluated in clinical practice.

In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
