Investigation of how diatoms cope with the rapid fluctuations in iron bioavailability in marine environments may facilitate a better understanding of the mechanisms underlying their ecological success, in particular their ability to proliferate rapidly during favorable conditions. In this study, using in vivo biochemical markers and whole-cell iTRAQ-based proteomics analysis, we explored the cellular responses associated with reactive oxygen species (ROS) production and cell fate decision during the early response to Fe limitation in the centric diatom Thalassiosira pseudonana. Fe limitation caused a significant decrease in Photosystem (PS) II photosynthetic efficiency, damage to the photosynthetic electron transport chain in PS I, and blockage of the respiratory chain in complexes III and IV, which could all result in excess ROS accumulation. The increase in ROS likely triggered programmed cell death (PCD) in some of the Fe-limited cells through synthesis of a series of proteins involved in the delicate balance between pro-survival and pro-PCD factors. The results provide molecular-level insights into the major strategies that may be employed by T. pseudonana in response to Fe-limitation: the reduction of cell population density through PCD to reduce competition for available Fe, the reallocation of intracellular nitrogen and Fe to ensure survival, and an increase in expression of antioxidant and anti-PCD proteins.

With more and more attention and investment on acupuncture scientific researches, considerable outcomes and achievements has been acquired, but the shortcoming of low transformation rate of acupuncture research achievements is gradually exposed. Nowadays there is no related report on this problem, so based on achievement translational research in other areas and practical situation of acupuncture, the existing problems and solutions are analyzed. As a result, the existing problems include (1) the research content is mainly basic research and clinical research but less acupuncture device research, leading to limited transformation efficiency; (2) the evaluation system and transformation pattern are still needed to be perfect. The solutions are (1) to properly evaluate the research achievements of acupuncture, (2) to advocate the concept and method of translational medicine, (3) to reform the policy and system, and (4) to establish valid platforms covering research, outcomes and transformation.

Functional dyspepsia (FD) has been a worldwide complaint. More effective therapies are needed with fewer adverse effects than are seen with conventional medications. Acupuncture, as a traditional therapeutic method, has been widely used for functional gastrointestinal disorders in the East. Manual acupuncture and electroacupuncture have been recognized treatments for FD, but to date, no robust evidence has been found for the effectiveness and safety of these interventions in the treatment of this condition.

Until now, there has been little data on the isotope fractionation of nickel (Ni) in higher plants and how this can be affected by plant Ni and zinc (Zn) homeostasis. A hydroponic cultivation was conducted to investigate the isotope fractionation of Ni and Zn during plant uptake and translocation processes. The nonaccumulator Thlaspi arvense, the Ni hyperaccumulator Alyssum murale and the Ni and Zn hyperaccumulator Noccaea caerulescens were grown in low (2 ?M) and high (50 ?M) Ni and Zn solutions. Results showed that plants were inclined to absorb light Ni isotopes, presumably due to the functioning of low-affinity transport systems across root cell membrane. The Ni isotope fractionation between plant and solution was greater in the hyperaccumulators grown in low Zn treatments (?(60)Niplant-solution = -0.90 to -0.63‰) than that in the nonaccumulator T. arvense (?(60)Niplant-solution = -0.21‰), thus indicating a greater permeability of the low-affinity transport system in hyperaccumulators. Light isotope enrichment of Zn was observed in most of the plants (?(66)Znplant-solution = -0.23 to -0.10‰), but to a lesser extent than for Ni. The rapid uptake of Zn on the root surfaces caused concentration gradients, which induced ion diffusion in the rhizosphere and could result in light Zn isotope enrichment in the hyperaccumulator N. caerulescens. In high Zn treatment, Zn could compete with Ni during the uptake process, which reduced Ni concentration in plants and decreased the extent of Ni isotope fractionation (?(60)Niplant-solution = -0.11 to -0.07‰), indicating that plants might take up Ni through a low-affinity transport system of Zn. We propose that isotope composition analysis for transition elements could become an empirical tool to study plant physiological processes.

The clinical efficacy and safety of auricular acupuncture (AA) for treatment of primary insomnia was evaluated. After a comprehensive retrieval in domestic and foreign databases, literatures were strictly screened and Revman 5.2 software was applied to perform a Meta-analysis on eligible randomized controlled trials (RCTs). The evidence quality was assessed with GRADE profiler 3.6 software. As a result, 8 articles were included involving 894 patients. Compared among AA and sham AA, placebo AA, blank control, there was significant difference in Pittsburgh sleep quality index (PSQI) [WMD = -3.48, 95% CI (-3.96, -3.00)], sleep latency LWMD = -10.14, 95% CI (-17.16, -3.12)] and sleep awakening times [WMD = -9.98, 95% CI (-1.10,-0.48)]. Compared between AA and western medication, there was significant difference in PSQI [WMD = -3.62, 95% CI (-4.59, -2.65)]. The evidence quality was moderate in AA vs. sham AA, placebo AA or blank control, while that of the rest was extremely low. No reports of adverse events were described in all studies. In conclusion, for the treatment of primary insomnia, AA could effectively improve sleep quality, but due to the low evidence quality, cautious attitude should be taken on this conclusion, and clinical trials with large sample and high quality were needed in the further.

Heart-brain correlation is an important component of Chinese medicine about the theory of zang-fu organs, which is still valuable for acupuncture clinical practice. Nowadays, increasing evidence supports the close association between the heart-brain axis, central autonomic nerve network and cardiovascular diseases, as well as the extensive regulative effects of acupuncture intervention on the heart-brain axis, functional connectivity of the brain, automatic nerve activities and cardiac functions. Therefore, the authors of the present paper hold that from the viewpoint of the heart-brain relationship, and by combining non-invasive functional brain imaging techniques with the patients' subjective and objective clinical indexes, our researchers will possibly and systematically reveal the underlying central mechanisms of acupuncture therapy in the treatment of chronic stable angina pectoris. However, the concrete biochemical mechanism should be proved via other advanced biological techniques.

Essential hypertension (EH) is a very common clinical disorder affecting the patient's health. Accumulating evidence indicates that immunological factors play an important role in the pathogenesis of hypertension. In the present paper, the authors introduce 1) progress of researches on the pathogenesis of hypertension from cellular immune and body fluid immune (multiple immuno-humoral factors); 2) effects of acupuncture intervention on natural killer cell activity, exercise-induced immunosuppression, circulating inflammatory factor levels and balance of cytokines; 3) blood-pressure reduction effect of acupuncture intervention by lowering circulating TNF-alpha, IL-6, matrix metalloproteinases-9, angiotensin convertase and endothelin levels, and up-regulating serum opioid peptide content, etc. to decrease inflammatory injury of the cardiovascular system. Many researches have demonstrated that acupuncture may have a positive role in improving EH in clinical practice, which may be associated with its regulative effect on immune system, but its mechanism has not been fully elucidated.

Neurological Recovery and Antioxidant Effects of Curcumin for Spinal Cord Injury in the Rat: A Network Meta-Analysis and Systematic Review.

Spinal cord injury (SCI) is a devastating condition affecting young, healthy individuals worldwide. Existing agents have inadequate therapeutic efficacy, and some are associated with side effects. Our objective is to summarize and critically assess the neurological recovery and antioxidant effects of curcumin for treatment of SCI in rat models. PUBMED, EMBASE and Chinese databases were searched from their inception date to February 2014. Two reviewers independently selected animal studies that evaluated neurological recovery and antioxidant effects of curcumin compared with placebo in rats with SCI, extracted data and assessed the methodological quality. A pairwise analysis and a network meta-analysis were performed. Eight studies with adequate randomization were selected and included in the systematic review. Two studies had a higher methodological quality. Overall, curcumin appears to significantly improve neurological function, as assessed using Basso, Beattie, Bresnahan (BBB) locomotor rating scale (4 studies, n=132, pooled MD=3.09, 95% CI=3.40 to 4.45, P=0.04), in a random effects model and decrease the malondialdehyde (MDA) using a fixed effects model (4 studies, n=56, pooled MD=-1.00, 95% CI=-1.59 to -0.42, P=0.00008). The effect size, assessed using the BBB scale, increased gradually with increasing curcumin dosage. The difference between low-dose and high-dose curcumin using the BBB scale was statistically significant. Neurological recovery and antioxidant effects of curcumin were observed in rats with SCI despite poor study methodological quality. An assessment of the methodological quality of animal model SCI studies is required, and good methodological quality should be valued in systematic reviews of animal studies. The modern literatures on the diseases treated with acupuncture at Shenmen (HT 7) were collected. Using literature statistical analysis in bibliometrics, the common compatibility of Shenmen (HT 7) and the disease spectrum were analyzed so as to provide the references of the diseases that can be treated with acupuncture at Shenmen (HT 7). It was found that 39 kinds of diseases and symptoms, mainly nerve system disedses, could be treated with acupuncture at Shenmen (HT 7), including insomnia, depression, anxiety and dementia. Shenmen (HT 7) was seldom used alone. Instead, it was commonly used along with Baihui (GV 20), Sanyinjiao (SP 6), Neiguan (PC 6), Sishencong (EX-HN 1) and Taicong (LR 3).

The research on indications of acu-moxibustion is very important for effectively guiding clinical practice. In the present paper, the authors retrieved domestic literature about researches on the disease-spectrum or indications of acu-moxibustion therapy in recent 10 years, and made a bibliometrics analysis according to the described research methods, publishing year, quantity and their proportionate relationship. A total of 18 qualified original papers involving 414 illnesses or clinical conditions were included in the present paper. Among the 414 kinds of clinical conditions or illnesses, 72 were considered to be suitable candidates for acu-moxibustion therapy, including functional dyspepsia, herpes zoster, irritable bowel syndrome, gastrointestinal dysfunction, scapulohumeral periarthritis, cervical syndrome, dyssomnia, acne (cyst type), chronic urticaria, regional neurodermatitis, pain, itching, etc. which belong to illness-spectrum grade-I. Generally, the acu-moxibustion therapy is, in clinical effects, relatively poorer for structural diseases which are attributed to grade-II or grade-III type. Nevertheless, the diversity of clinical indications of acu-moxibustion has been confirmed nowadays. Current studies on illness-spectrum are of certain uniformity and comprehensiveness, but need further extending and perfecting, and need more standardized methods corresponding to principles of the evidence-based medicine.

All-trans retinoic acid (ATRA) exerts various effects on physiological processes such as cell growth, differentiation, apoptosis and inflammation. Prohibitins (PHB), including prohibitin 1 (PHB1) and prohibitin 2 (PHB2), are evolutionary conserved and pleiotropic proteins implicated in various cellular functions, including proliferation, tumor suppression, apoptosis, transcription, and mitochondrial protein folding. The renin-angiotensin-aldosterone system plays a pivotal role in the regulation of blood pressure and volume homeostasis. All these factors and systems have been implicated in renal interstitial fibrosis. Therefore, the objective of this study was to investigate the effect of ATRA treatment on the renin-angiotensin-aldosterone system and expression of prohibitins to further understand its role in the processes leading to renal interstitial fibrosis.

[Characteristics of acupoint selection of acupuncture-moxibustion for vertigo in history: a data mining research].

The data mining technique is adopted to analyze characteristics and rules of acupoint and meridian selection of acupuncture-moxibustion for treatment of vertigo at different time periods in the ancient. The data is collected from literature regarding acupuncture-moxibustion from the pre-Qin period to the end of Qing Dynasty, so as to establish a clinical literature database of ancient acupuncture-moxibustion for treatment of vertigo. Data mining method is applied to analyze the commonly used meridians, acupoints and special acupoints in different dynasties, also possible rules are explored. Totally 82 pieces of prescription of acupuncture-moxibustion for treatment of vertigo are included. In the history the leading selection of acupoitns are Fengchi (GB 20), Hegu (LI 4), Shangxing (GV 23) and Jiexi (ST 41) while that of meridians are mainly three yang meridians of foot and the Governor Vessel, especially the acupoints on the Bladder Meridian of foot yangming had the highest utilization rate, accounting for 23.04%. The acupoint selection is characterized by special acupoint, accounting for 80.6%, among which the crossing points are the most common choice. Distal-proximal acupoints combination is the most frequent method. The results indicate that the ancient acupuncture-moxibustion for treatment of vertigo focused on acupoints in the yang meridians, and the specific acupoints play an essential role in prescription; also the principle of syndrome differentiation and selecting acupoints along the meridians could be seen.

Phthalic acid esters (PAEs), a class of organic pollutants with potent endocrine-disrupting properties, are widely present in municipal sludge. Study of PAEs biodegradation under different anaerobic biological treatment processes of sludge is, therefore, essential for a safe use of sludge in agricultural practice. In this study, we selected two major sludge PAEs, i.e. di-n-butyl phthalate (DBP) and di-(2-enthylhexyl) phthalate (DEHP), to investigate their biodegradation behaviors in an anaerobic sludge digestion system and a fermentative hydrogen production system. The possible factors influencing PAEs biodegradation in relation to changes of sludge properties were also discussed. The results showed that the biodegradation of DBP reached 99.6% within 6 days, while that of DEHP was 46.1% during a 14-day incubation period in the anaerobic digestion system. By comparison, only 19.5% of DBP was degraded within 14 days in the fermentative hydrogen production system, while no degradation was detected for DEHP. The strong inhibition of the degradation of both PAEs in the fermentative hydrogen production system was ascribed to the decreases in microbial biomass and ratios of gram-positive bacteria/gram-negative bacteria and fungi/ bacteria, and the increase of concentrations of volatile fatty acids (e. g. acetic acid, propionic acid and butyric acid) during the fermentative hydrogen-producing process.

This study investigated genome-wide gene expressions and the cardioprotective effects of electro-acupuncture pretreatment at the PC6 Neiguan acupoint on myocardial ischemia reperfusion (I/R) injury. Male SD rats were randomly divided into four groups: sham operation (SO), I/R, electro-acupuncture at the PC6 Neiguan acupoint pretreatment (EA) and electro-acupuncture at non-acupoint pretreatment (NA). Compared with the I/R group, the survival rate of the EA group was significantly increased, the arrhythmia score, infarction area, serum concentrations of CK, LDH and CK-Mb and plasma level of cTnT were significantly decreased. RNA-seq results showed that 725 genes were up-regulated and 861 genes were down-regulated under I/R conditions compared to the SO group; both EA and NA reversed some of these gene expression levels (592 in EA and 238 in NA group). KEGG pathway analysis indicated that these genes were involved in multiple pathways, including ECM, MAPK signaling, apoptosis, cytokine and leukocyte pathways. In addition, some pathways were uniquely regulated by EA, but not NA pretreatment, such as oxidative stress, cardiac muscle contraction, gap junction, vascular smooth muscle contraction, hypertrophic, NOD-like receptor, and P53 and B-cell receptor pathways. This study was first to reveal the gene expression signatures of acute myocardial I/R injury and electro-acupuncture pretreatment in rats.

Photo-induced chemical reaction of trans-resveratrol has been studied. UV B, liquid state and sufficient exposure time are essential conditions to the photochemical change of trans-resveratrol. Three principal compounds, cis-resveratrol, 2,4,6-phenanthrenetriol and 2-(4-hydroxyphenyl)-5,6-benzofurandione, were successively generated in the reaction solution of trans-resveratrol (0.25mM, 100% ethanol) under 100?Wcm(-2) UV B radiation for 4h. cis-Resveratrol, originated from isomerization of trans-resveratrol, resulted in 2,4,6-phenanthrenetriol through photocyclisation reaction meanwhile loss of 2 H. 2,4,6-Phenanthrenetriol played a role of photosensitizer producing singlet oxygen in the reaction pathway. The singlet oxygen triggered [4+2] cycloaddition reaction of trans-resveratrol, and then resulted in the generation of 2-(4-hydroxyphenyl)-5,6-benzofurandione through photorearrangement and oxidation reaction. The singlet oxygen reaction was closely related to the substrate concentration of trans-resveratrol in solution.

Acupoint compatibility is not only a foundation for formulating acupoint recipes in clinical practice, but also a core of meridian and acupoint theory. Many studies have revealed that the application of multiple-acupoints combination in the treatment of different clinical conditions or illnesses may lead to three outcomes, i.e., synergistic effect, antagonistic effect and no effect. Therefore, the application of multiple acupoints combination is definitely not a simple result of 1 + 1 = 2, or 2 - 1 = 1 in the management of many clinical disorders, and rather, the reasonable combination of multiple acupoints is a non-liner, dynamic and complicated issue. It has been well documented that the complex biological network consists of modules/communities, a set of nodes that are more densely inter-connected among themselves than with the rest of the network, which can be detected by community detection according to their modularity. After analyzing the current status and development trends of acupoint comparability researches in both China and other countries of the world, the authors of the present paper put forward some ideas and methods for the research of correlation, composing characteristics and dynamically evolutionary regularities of acupoint combination from the macroscopic and local aspects, dynamic and static angles on the basis of the theory of community structure theory of complex networks.

Comprehensive assessment of the association of WNK4 polymorphisms with hypertension: evidence from a meta-analysis.

The relationship between with-no-lysine [K] kinase 4 (WNK4) gene polymorphisms and hypertension has been widely investigated, However, the studies yielded contradictory results. To evaluate these inconclusive findings comprehensively, we therefore performed a meta-analysis. Ten articles encompassing 16 independent case-control studies with 6089 hypertensive cases and 4881 normotensive controls were selected for this meta-analysis. Four WNK4 gene polymorphisms were identified (G1155942T, G1156666A, T1155547C, and C6749T). The results showed statistically significant associations of G1155942T polymorphism (allelic genetic model: odds ration or OR = 1.62, 95% confidence interval or CI: 1.11-2.38, P = 0.01; dominant model: OR = 1.85, 95% CI: 1.07-3.19, P = 0.03) and C6749T polymorphism (allele contrast: OR = 2.04, 95% CI: 1.60-2.59, P<0.01; dominant model: OR = 2.04, 95%CI: 1.59-2.62, P<0.01; and homozygous model: OR = 5.01, 95% CI: 1.29-19.54, P = 0.02) with hypertension risk. However, neither C1155547T nor G1156666A was associated significantly with hypertension susceptibility. In conclusion, this meta-analysis suggested that WNK4 G1155942T and C6749T gene polymorphisms may contribute to the susceptibility and development of hypertension. Further well-designed studies with larger sample size are required to elucidate the association of WNK4 gene multiple polymorphisms with hypertension risk.

Granulosa cells (GCs) are essential for proper oocyte, follicular development, and steroidogenesis in the ovary. Transforming growth factor ? (TGF-?) superfamily members are critical in regulating GCs growth and differentiation. Smad3 is known to serve as a signaling intermediate for the TGF-?; however, the functions of Smad3 in the human GCs remain unidentified. In this study, the luteinized GCs collected from follicular aspirates from patients undergoing in vitro fertilization were cultured and engineered to overexpress and knockdown Smad3, which were validated by RT-PCR and Western blotting. Immunocytochemistry showed that Smad3 protein was strongly expressed in human ovarian luteinized GCs. EdU incorporation demonstrated that Smad3 promoted the proliferation of GCs, and the expression of PCNA was also enhanced by Smad3. ELISA analysis indicated that the secretion of both estradiol and progesterone was stimulated by Smad3. In addition, Smad3 upregulated the level of follicle-stimulating hormone receptor (FSHR), luteinizing hormone receptor (LHR), and protein kinase A (PKA) proteins. We subsequently added special PKA inhibitor H89 into the GCs and found that the stimulating effect on the growth of GCs by Smad3 was blocked partly. The morphology of cultured GCs was changed by Smad3, and the expression level of integrin ?1 was enhanced by Smad3. Kindlin-2, an important cellular mediating molecule of integrin ? signaling, was expressed in human ovarian luteinized GCs and was upregulated by Smad3. Our results indicated that Smad3 promoted the proliferation and steroidogenesis of human ovarian luteinized GCs, and these effects may be mediated by the FSHR/LHR-PKA signaling pathway.

The PAD regime, composed of bortezomib, adriamycin and dexamethasone, improves the outcomes of patients with advanced multiple myeloma (MM), but at the same time produces high frequency of serious toxic side effects. For the first time, we evaluated the efficacy and safety of a bortezomib-dose-reduced PAD regime in the treatment of relapsed/refractory MM in this clinical study. Forty-five patients were treated with two to six 21-day cycles of PAD, comprising bortezomib at 1.3 mg/m(2) (P1AD, n?=?21) or 1.0 mg/m(2) (P2AD, n?=?24) (days 1, 4, 8, 11), adriamycin at 9 mg/m(2) (days 1-4) and dexamethasone at 40 mg/day (days 1-4). Overall, 36 patients (80 %) showed at least partial remission (PR), in which 9 cases (20 %) showed complete remission (CR) and 10 cases (22 %) showed very good partial remission (VGPR). The efficacy of PAD regimen in advanced MM patients was not related to the traditional prognostic factors. There was no significant difference between P1AD and P2AD in the rates of PR, CR or VGPR, 1.5-year progression-free survival (PFS), and overall survival (OS) (81 % vs. 79 %, 48 % vs. 38 %, 64 % vs. 59 %, and 85 % vs. 73 %, respectively). However, the grade 3-4 toxic effects, including thrombocytopenia (13 % vs. 38 %), peripheral neuropathy (8 % vs. 33 %) and 3-4 grade gastrointestinal reaction (13 % vs. 43 %), were markedly inhibited after P2AD compared to P1AD (P?

Ho(3+)/Yb(3+) codoped TeO?-WO?-ZnO-ZnX?(X=F, Cl) glasses were prepared by melt-quenching method. The absorption spectra, transmittance spectra, X-ray diffraction (XRD) curves, Raman spectra and mid-infrared fluorescence spectra were measured, along with the Judd-Ofelt intensity parameters, stimulated emission and absorption cross-sections were calculated to evaluate the effects of halide amount of the spectroscopic properties. It is shown that the introduction of an appropriate amount of halide can further improve the mid-infrared fluorescence intensity through an enhanced phonon-assisted energy transfer between Ho(3+)/Yb(3+) ions and the energy transfer mechanisms are investigated quantitatively in detail by calculating energy transfer microparameters and phonon contribution ratios. The results indicate that this kind of glasses is a promising material for mid-infrared optical fiber.

Aplastic anemia (AA) is an autoimmune disease in which T cell activation is suspected to play an important role. T cell immunoglobulin and ITIM (immunoreceptor tyrosine-based inhibition motif) domain (TIGIT) is an inhibitory receptor, which exhibits inhibitory functions on the immune response. However, its role in AA has not been clearly determined. In the current study, we showed that the frequency of TIGIT-positive CD4(+) T cells was reduced in the vast majority of AA patients (85%, 17/20). In TIGIT-silenced human CD4(+) T cells, stimulation of agonistic anti-TIGIT monoclonal antibody significantly facilitated cell proliferation, increased production of IL-2 and IFN-?, and inhibited production of IL-10. However, in TIGIT-overexpressed human CD4(+) T cells, cell proliferation and the production of IL-2, IFN-?, and TNF-? were significantly hindered; in contrast, the secretion of IL-10 was improved. RT-PCR and Western blotting showed that T-bet expression in human CD4(+) T cells was significantly decreased by TIGIT overexpression, but only slightly altered by TIGIT knockdown. In mouse models, lentivirus-mediated TIGIT-overexpressed CD4(+) T cell transfer significantly rescued the decreased red blood cell count, attenuated the increase in serum INF-? and TNF-? levels, and lengthened the median survival time. The mRNA levels of CD34, stem cell factor (SCF), and granulocyte/macrophage-colony-stimulating factor (GM-CSF) in bone marrow mononuclear cells were also up-regulated. In conclusion, increased expression of TIGIT could inhibit the function of CD4(+) T cells in vitro and ameliorate immune-mediated bone marrow failure of AA in vivo providing a new potential strategy for the treatment of AA.

Uroporphyrinogen III methyltransferase (UMT) is a novel reporter owing to the catalytic products in the cells that emit strong red fluorescence under UV light. Here, we engineered the gene encoding the functional barley UMT (bUMT) by error-prone PCR and broadened the application UMT as a red fluorescent reporter in Escherichia coli. A variant, termed mbUMT, was selected and emitted stronger cell fluorescence than the wild type bUMT expressed in different E. coli strains, under different promoters and induction conditions respectively. The constructed mbUMT with a C-terminal ssrA tag was degraded in cells by the protease ClpXP encoded by E. coli chromosome, whereas the bUMT was expressed as active aggregates. Before they are exported to the periplasm, both proteins catalyze the substrate in the cytoplasm and emit cell fluorescence. The results suggested that the evolved bUMT is a better candidate to monitor in vivo degradation by E. coli ClpXP.

The marine diatom Chaetoceros muelleri grown under air (0.03% CO2), 10%, 20%, and 30% CO2 conditions was evaluated to determine its potential for CO2 reduction coupled with biodiesel production. The results indicated that C. muelleri grows well with high CO2 aeration levels (10-20%) and is induced to accumulate lipids under 10-30% CO2. In particular, the highest values of the maximum biomass concentration (1.059gL(-1)), maximum specific growth rate (0.868d(-1)), maximum biomass productivity (0.272gL(-1)d(-1)), maximum CO2 biofixation (0.428gL(-1)d(-1)), and total lipid (43.40% dry weight) and neutral lipid contents were all obtained with 10% CO2 aeration. Moreover, the analysis of the fatty acid composition of C. muelleri revealed the predominance of C14-C18 fatty acids (>90%) and saturated and monounsaturated fatty acids (>80%) under all CO2 levels. The results suggest that C. muelleri has great potential to biodiesel production using flue gases.

3-Deoxy-d-arabino-heptulonate-7-phosphate (DAHP) synthase is one of the key enzymes, which catalyzes the first step in the aromatic amino acid biosynthetic pathway and yields the three amino acids tyrosine (Tyr), tryptophan (Typ), and phenylalanine (Phe). In Escherichia coli (E. coli), three differently regulated DAHP synthases carry out the first regulated step in the aromatic amino acid biosynthetic pathway. The three DAHP synthases encoded by the genes aroG, aroF, and aroH are inhibited by phenylalanine, tyrosine and tryptophan, respectively. In this work, the aroG gene was cloned and mutated by site-directed mutagenesis using overlap extension PCR (SOE-PCR) technique. The feedback-resistant DAHP synthase encoded by aroG was achieved by replacing the residue Pro150 of aroG with Leu as to increase net carbon flow down the common pathway. SDS-PAGE and Western blots were used to assess the protein expression level of aroGM which showed the strain harboring the mutated aroGM150 gene achieving over-expression compared to the strain containing an empty plasmid pET-28b((+)).

The development of more selective chelators for the washing of heavy metal contaminated soil is desirable in order to avoid excessive dissolution of soil minerals. Speciation and mobility of Cu, Zn, Pb, and Ni in a contaminated soil washed with phenyldiaminetetraacetic acid (PDTA), a derivative of EDTA, were investigated by batch leaching test using a range of soil washing conditions followed by sequential extraction. With appropriate washing conditions, PDTA significantly enhanced extraction of Cu from the contaminated soil. The primary mechanisms of Cu extraction by PDTA were complexation-promoted dissolution of soil Cu and increased dissolution of soil organic matter (SOM). PDTA showed high selectivity for Cu(II) over soil component cations (Ca(II), Mg(II), Fe(III), Mn(II), Al(III)), especially at lower liquid-to-soil ratios under PDTA deficiency, thus avoiding unwanted dissolution of soil minerals during the soil washing process which can degrade soil structure and interfere with future land use. PDTA-enhanced soil washing increased the exchangeable fractions of Cu, Zn, and Pb and decreased their residual fractions, compared to their levels in unwashed soil.

Paired box gene 2 (PAX2) can regulate tissue development and cellular differentiation, and it is associated with renal diseases. CD2-associated protein (CD2AP) is an adaptor protein involving in a variety of physiological and disease processes. Renal interstitial fibrosis (RIF) is a hallmark of common progressive chronic diseases which lead to renal failure. This study was performed to investigate whether there was a potential signal pathway between PAX2 and CD2AP in RIF rats induced by unilateral ureteral obstruction (UUO). Eighty Wistar male rats were divided into two groups randomly: sham operation group (SHO) and model group subjected to UUO (GU), n?=?40. The model was established by left ureteral ligation. Renal tissues were collected at 14 d and 28 d after surgery. The objective of this study was to analyze the influence of thermal-acid treatment on the formation of resistant starch (RS). The maximum RS content in citric acid-heat treated starches (CAHT) reached 36.55%, which was 7 times higher of that in native starch. According to HPSEC-MALLS-RI analysis, amylopectin was more susceptible to hydrolysis than amylose during citric acid-heat treatment (CAH). X-ray measurement revealed that even though the starch crystalline pattern was changed from A-type to a more resistant B-type after CAH, the fraction of crystalline region decreased from 21.16% to 8.37%. The hydroxyls on the starch chains were substituted by the citric acid anhydrides during CAH according to FT-IR analysis, which led to the formation of ester bond cross-linking structures in starch granules, and it could be the main contribution to the increase of RS content in CAHT samples.

In this study a comparative proteomics approach involving a mass spectrometric analysis of synchronized cells was employed to investigate the cellular-level metabolic mechanisms associated with siliceous cell wall formation in the pennate diatom Pseudo-nitzschia multiseries. Cultures of P. multiseries were synchronized using the silicate limitation method. Approximately 75% of cells were arrested at the G2+M phase of the cell cycle after 48 h of silicate starvation. The majority of cells progressed to new valve synthesis within 5h of silicon replenishment. We compared the proteome of P. multiseries at 0, 4, 5, and 6h of synchronization progress upon silicon replenishment using two-dimensional gel electrophoresis. Forty-eight differentially expressed protein spots were identified in abundance (greater than two-fold change; P<0.005), some of which are predicted to be involved in intracellular trafficking, cytoskeleton, photosynthesis, lipid metabolism, and protein biosynthesis. Cytoskeleton proteins and clathrin coat components were also hypothesized to play potential roles in cell wall formation. The proteomic profile analysis suggests that P. multiseries most likely employs multiple synergistic biochemical mechanisms for cell wall formation. These results improve our understanding of the molecular mechanisms underlying silicon cell wall formation and enhance our understanding of the important role played by diatoms in silicon biogeochemical cycling.

Interleukin-6 (IL-6) is a key pro-inflammatory cytokine involved in different physiologic and pathophysiologic processes, and circulating levels of IL-6 differ greatly between individuals. The Chinese Hui is one of the largest ethnic minorities, little is known about the distribution of IL-6 genetic variations and their effects on serum levels in Hui population. The aim of the present study is to determine the prevalence of -174G/C (rs1800795), -597G/A (rs1800797), and -634C/G (rs1800796) polymorphisms in the IL-6 gene promoter region and their association with IL-6 serum levels in the Ningxia Hui population. A total of 96 Hui subjects, (57 men and 39 women; mean age 49.65 ± 19.73 years) unrelated nationality residents in Ningxia Hui Autonomous Region were enrolled. Genotyping of the three polymorphisms were performed by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) combined with gel electrophoresis and then confirmed by direct sequencing. The -174G/C (97.92% GG, 2.08% GC, and 0% CC) and -597G/A (98.96% GG, 1.04% GA, and 0% AA) polymorphisms were rare. The frequencies of -634C/G genotypes CC, CG, and GG were found to be 54.17%, 40.62%, and 5.21%, respectively in total studied subjects, the derived allele frequencies for the C and G alleles were 74.48% and 25.52%. Increased IL-6 levels were correlated with the IL-6 -634G allele carriers (CG+GG genotypes). The results suggest that IL-6 -174G/C and -597G/A are rare but -634C/G is common in the Ningxia Hui population, and the -634G allele is associated with circulating levels of IL-6.

A Large scale of full-thickness skin defects is lack of auto-grafts and which requires the engineered skin substitutes for repair and regeneration. One major obstacle in skin tissue engineering is to expand epidermal stem cells (ESCs) and develop functional substitutes. The other one is the scaffold of the ESCs. Here, we applied type I collagen-modified chitin membrane to form collagen-chitin biomimetic membrane (C-CBM), which has been proved to have a great biocompatibility and degraded totally when it was subcutaneously transplanted into rat skin. ESCs were cultured, and the resulting biofilm was used to cover full-thickness skin defects in nude mice. The transplantation of ESCs- collagen- chitn biomimetic membrane (ESCs-C-CBM) has achieved in situ skin regeneration. In nude mice, compared to controls with collagen-chitin biomimetic membrane (C-CBM) only, the ESCs-C-CBM group had significantly more dermatoglyphs on the skin wound 10 w after surgery, and the new skin was relatively thick, red and elastic. In vivo experiments showed obvious hair follicle cell proliferation in the full-thickness skin defect. Stem cell markers examination showed active ESCs in repair and regeneration of skin. The results indicate that the collagen-modified chitin membrane carry with ESCs has successfully regenerated the whole skin with all the skin appendages and function.

This paper aims to achieve Bootstraping in hierarchical data and to provide a method for the estimation on confidence interval(CI) of intraclass correlation coefficient(ICC).First, we utilize the mixed-effects model to estimate data from ICC of repeated measurement and from the two-stage sampling. Then, we use Bootstrap method to estimate CI from related ICCs. Finally, the influences of different Bootstraping strategies to ICCs CIs are compared. The repeated measurement instance show that the CI of cluster Bootsraping containing the true ICC value. However, when ignoring the hierarchy characteristics of data, the random Bootsraping method shows that it has the invalid CI. Result from the two-stage instance shows that bias observed between cluster Bootstrapings ICC means while the ICC of the original sample is the smallest, but with wide CI. It is necessary to consider the structure of data as important, when hierarchical data is being resampled. Bootstrapping seems to be better on the higher than that on lower levels.

Acupuncture, as a modality treatment, has gained increasing popularity and acceptance between public and health-care professionals worldwide. Recently, there has been intensive debate about the efficacy of acupuncture therapy due to the conflicting outcome of clinical trials. Acupoint specificity was regarded as one of the core scientific issues with respect to acupuncture practice at the Society for Acupuncture Research international symposium held in 2007. In this chapter, we reviewed the recent development in basic science and clinical studies on the role of acupoint specificity. The evidence cumulated from brain imaging and many biological studies showed that the point specificity in acupuncture does exist, although acupoint specificity-related issues such as sham acupoint and placebo phenomenon need to be seriously considered. How to optimize the efficacy of acupoint and minimize the impact of sham acupuncture is an urgent issue faced by acupuncture community, and more studies are warranted on the subjects.

The acupuncture has been practiced in China for more than 3000 years and was spread to Europe and American from the sixteenth to the nineteenth century. The history of acupuncture research was initiated in the eighteenth century and developed rapidly since then. In the past, physicians tried hard to apply acupuncture into clinical practice, while scientists were focused on the possible characteristics of acupoints and meridians. In the modern time, scientists have strived hard to evaluate the real effectiveness of acupuncture and the underlying physiological and biological mechanisms of acupuncture. Reviewing research history from past to present, we are delighted to witness this wonderful development. Accumulated evidences that acupuncture is beneficial in various conditions significantly enhanced our understanding the mechanisms of acupuncture treatment. However, there is still no conclusive evidence in acupuncture clinical studies. The clinical research still needs great improving, while the basic research results need to be appropriately transformed into clinical outcomes. Based on current achievements, we believe that although the challenges and difficulties exist, a more collaborative, innovative, and integrated approach will help us to achieve further progress in future acupuncture research.

Based on comprehensive arrangement and analysis of edition origin and development, system, characteristic, archive holding situation of Zhenjiu Fengyuan (Source of Acupuncture and Moxibustion writlen) by LI Xue-chuan in Qing dynasty, the error and mistakes in Summary of TCM Ancient books in China, Union Catalog of TCM Books and Grand Chinese Dictionary of Medical Books are pointed out. On the other hand, under full evaluation and comparison among three types of block-printed editions, Dihua Caotang edition in 2 years of Daoguang emperor, Qing Dynasty (about 1822) that is collected in the library of Beijing University of CM is considered as good edition.

Siderophore-producing rhizobacteria (SPR) are a group of plant growth-promoting rhizobacteria, being able to play an important role in assisting the phytoremediation of heavy metals-contaminated soils. Based on the comprehensive analysis of related researches at home and abroad, this paper elaborated the functions of SPR in alleviating the heavy metals stress and toxicity to plants and the mechanisms of SPR in improving the heavy metals bioavailability in soil, and indicated that SPR had good application potential in promoting the plant growth in heavy metals-contaminated soils and reinforcing the heavy metals accumulation in plants. The contradictory phenomena of SPR in increasing or decreasing heavy metals accumulation in plants, which existed in current researches, were also analyzed. Aiming at the deficiencies in current researches, it was suggested that in the future researches, the mechanisms of the interactions between SPR and plants, especially hyperaccumulators, should be further studied, the key factors affecting the heavy metals complexation and mobilization in soil by siderophores should also be further clarified, the effects of siderophores on the heavy metals bioavailability and its subsequent influence on the heavy metals uptake by plants should be comprehensively considered, and the measures for improving the colonization of SPR in heavy metals-contaminated soil should be explored.

This study was aimed to investigate the expression and role of 14-3-3? in the AML cell lines: sensitive HL-60 and drug-resistant HL-60/VCR cells. Semi-quantitative RT-PCR and Western blot were respectively used to examine the expression of mdr1 mRNA and Pgp in AML cell lines to validate the results of microarray. Western blot was performed to investigate the expression of Pgp, 14-3-3?, and anti-apoptosic protein BCL-2, MCL-1 proteins. Immunofluorescence assay was used to detect the subcellular location of 14-3-3? protein in HL-60 and HL-60/VCR cells by laser scanning confocal microscopy. Transduction with siRNA was used to silence 14-3-3? in AML cell lines. Cell count method and flow cytometry of cell cycle were used to analyze the changes of growth of AML cells. The results found that mdr1 mRNA and Pgp did not expressed in HL-60 cells, but significantly overexpressed in HL-60/VCR cells. Except 14-3-3?, the expression of other subtypes of 14-3-3 was higher in HL-60/VCR cells than that in HL-60 cells, especially 14-3-3?. The higher expression of 14-3-3?, BCL-2, MCL-1 protein was observed in HL-60/VCR cells than that in HL-60 cells. These results were same results from gene chip. It was also noticed that 14-3-3? was located in the cytoplasma and nuclei of AML cell lines, especially over-expressed in HL-60/VCR cells. Furthermore, suppression of 14-3-3? by RNA interference resulted in inhibition of the proliferation of AML cells with decreased protein expression of BCL-2 and MCL-1, especially in HL-60/VCR cells. It is concluded that 14-3-3? plays an important role in proliferation of AML cells and associates with BCL-2 and MCL-1 expression. These results suggested that development of therapy targeting 14-3-3? may provide novel, effective strategies for refractory and relapsed AML.

Mantle cell lymphoma (MCL) remains incurable due to its inevitable pattern of relapse after treatment with current existing therapies. However, the promise of a cure for MCL lies in the burgeoning area of novel agents. In this study, we elucidated the therapeutic effect and mechanism of carfilzomib, a novel long-acting second-generation proteasome inhibitor, in MCL cells. We found that carfilzomib induced growth inhibition and apoptosis in both established MCL cell lines and freshly isolated primary MCL cells in a dose-dependent manner. In contrast, carfilzomib was less toxic to normal peripheral blood mononuclear cells from healthy individuals. The carfilzomib-induced apoptosis of MCL cells was mediated by the activation of JNK, Bcl-2, and mitochondria-related pathways. In addition, carfilzomib inhibited the growth and survival signaling pathways NF-?B and STAT3. Interestingly, we discovered that expression of immunoproteasome (i-proteasome) subunits is required for the anti-MCL activity of carfilzomib in MCL cells. In MCL-bearing SCID mice/primary MCL-bearing SCID-hu mice, intravenous administration of 5 mg/kg carfilzomib on days 1 and 2 for 5 weeks slowed/abrogated tumor growth and significantly prolonged survival. Our preclinical data show that carfilzomib is a promising, potentially less toxic treatment for MCL. Furthermore, an intact i-proteasome, especially LMP2, appears to be necessary for its anti-MCL activity, suggesting that i-proteasome could serve as a biomarker for identifying patients who will benefit from carfilzomib.

Diffuse large B cell lymphoma (DLBCL) is the most common non-Hodgkin lymphoma in the world. Clinically, biologically, and pathologically, DLBCL is a heterogeneous entity with a range of potential outcomes. Immunochemotherapy regimens, consisting of the chimeric monoclonal anti-CD20 antibody rituximab in combination with chemotherapy, have improved the outcomes. Relapsed DLBCL is generally treated with salvage immunochemotherapy followed by high-dose therapy and autologous stem cell transplantation; however, DLBCL is not yet curable in up to a third of patients. The real promise for cure lies in novel agents and their rational combinations. The improved understanding of DLBCL subtypes and gene expression profiling has led to the identification of targeted drugs that may allow for subtype specific therapy. We have summarized the existing data on the prognostic factors and the treatment of DLBCL, including the use of novel agents such as lenalidomide, carfilzomib, and ibrutinib. We also share our thoughts on the direction of future clinical trials.

As a famous TCM physician and acupuncturist in our country, professor GUAN Ji-duo, who was determined to study medicine and save life from a child, has been engaged in teaching and clinical work of acupuncture and moxibustion for more than sixty years. With a lifetime writing and tireless teaching, he has made every effect, from small clue to see what is coming, to be well-versed in the learning of both ancient and modern times and learn widely from others points to devote all his life for the development and innovation of acupuncture and moxibustion. With deep study on syndrome differentiation of meridians, he has focused on acupoint combination, advocated on painless insertion of needle, and made "smokeless moxa-stick" to form a unique academic thought. At the age of more than 90, professor GUAN still insists on outpatient clinic, which is highly respected.

Polymicrobial infections are responsible for significant mortality and morbidity in adults and children. Staphylococcus epidermidis and Candida albicans are the most frequent combination of organisms isolated from polymicrobial infections. Vascular indwelling catheters are sites for mixed species biofilm formation and pose a significant risk for polymicrobial infections. We hypothesized that enhancement of biofilms in a mixed species environment increases patient mortality and morbidity.

The interaction between buttermilk and resveratrol (trans-3,5,4-trihydroxystilbene) was examined using fluorescence spectroscopy. Ultracentrifugation of buttermilk (10% total solids, TS)-resveratrol (100-1600 ?M) mixtures yielded three fractions comprising cream (14.79% w/w, 1.12% TS), milk serum (75.94% w/w, 5.56% TS), and a casein-rich precipitate (9.27% w/w, 2.94% TS). The majority of the added resveratrol was partitioned into the casein-rich precipitate (50.5-56.8%), with lesser amounts in the milk serum (35.3-41.2%) and cream layer (7.9-8.7%), demonstrating that most of the resveratrol interacted with the proteins. The interaction of the milk proteins with resveratrol was investigated by measuring the quenching of protein intrinsic fluorescence. Complex formation was spontaneous and exothermic. The apparent binding constants between milk proteins in buttermilk and resveratrol were ?2.47 × 10(4), 1.65 × 10(4), 1.11 × 10(4), and 0.72 × 10(4) M(-1), respectively, at 278, 288, 298, and 308 K. The increased aqueous solubility of resveratrol by complexation to whole buttermilk makes it useful as a food vehicle for carrying resveratrol.

Acupuncture as an essential component of complementary and alternative medicine is gradually recognized and accepted by the mainstream of contemporary medicine. For obtaining preferable clinical effectiveness, Deqi is commonly regarded as efficacy predictor and parameter which is necessary to be achieved. Influential factors for acupuncture efficacy, like Deqi sensation as well as propagated sensation along channels (PSCs), enjoyed a long history in acupuncture basic research. Concerning this study, taking into account different positions on acupuncture Deqi sensation and PSCs, we would like to attest whether different body positions for subjects during needling procedure yield differed acupuncture Deqi sensation, particularly in terms of intensity, and PSCs. Methods. We used self-controlled method and selected 30 healthy subjects to perform needle insertion at Futu point (ST32) bilaterally. Then they were instructed to record the value of intensity of acupuncture sensation and the length and width of PSCs after removing the needle. Results. In regard to intensity of Deqi, kneeling seat position is stronger than supine position, accounting for 90% of the total number of subjects. In length of PSCs, kneeling seat position is greater than supine position, accounting for 56.7%. In width of PSCs, kneeling seat position is greater than supine position, accounting for 66.7%. Conclusion. Our findings show that needle inserting at Futu point (ST32) in kneeling seat position achieve better needle sensation and provide reference for clinical.

A five-year field trial was conducted at the surrounding area of Dabao Mountain Mine to explore the feasibility and availability of using kenaf (Hibiscus cannabinus) , a fiber crop with strong heavy metals tolerance and potential economic value, to reclaim the multi-metal contaminated acidic farmland soil. Different amendments were applied prior to the kenaf planting to evaluate their effects on the soil properties and kenaf growth. After the amendments application, the kenaf could grow well on the heavy metals contaminated soil with the Pb, Zn, Cu, Cd, and As concentrations being 1600, 440, 640, 7. 6, and 850 mg . kg-1, respectively. Among the amendments, dolomite and fly ash had better effects than limestone and organic fertilizer. With the application of dolomite and fly ash, the aboveground dry mass production of kenaf reached 14-15 t . hm-2, which was similar to that on normal soils, and the heavy metal concentrations in the bast fiber and stem of kenaf decreased significantly, as compared with the control. The mass of the bast fiber accounted for 32% -38% of the shoot production, and the extractable heavy metal concentrations in the bast fiber could meet the standard of technical specifications of ecological textiles in China, suggesting that the bast fiber had potential economic value. It was suggested that planting kenaf combining with dolomite/fly ash application could be an effective measure to reclaim the multi-metal contaminated acidic farmland soil.

Mechanisms of cadmium (Cd)-induced inhibition of photosynthesis in the Zn/Cd hyperaccumulator Picris divaricata were investigated using photosynthesis limitation analysis. P. divaricata seedlings were grown in nutrient solution containing 0, 5, 10, 25, 50, or 75 ?M Cd for 2 weeks. Total limitations to photosynthesis (TL) increased from 0% at 5 ?M Cd to 68.8% at 75 ?M Cd. CO2 diffusional limitation (DL) made the largest contribution to TL, accounting for 93-98% of TL in the three highest Cd treatments, compared to just 2-7% of TL attributable to biochemical limitation (BL). Microscopic imaging revealed significantly decreased stomatal density and mesophyll thickness in the three highest Cd treatments. Chlorophyll fluorescence parameters related to photosynthetic biochemistry (Fv/Fm, NPQ, ?PSII, and qP) were not significantly decreased by increased Cd supply. Our results suggest that increased DL in leaves is the main cause of Cd-induced inhibition of photosynthesis in P. divaricata, possibly due to suppressed function of mesophyll and stomata. Analysis of chlorophyll fluorescence showed that Cd supply had little effect on photochemistry parameters, suggesting that the PSII reaction centers are not a main target of Cd inhibition of photosynthesis in P. divaricata.

The feasibility of the case-based learning (CBL) of Acupuncture-Moxibustion Techniques is discussed in the aspects of the connotation application, implementation process and effect evaluation of CBL in teaching this course. The result shows that compared with the traditional teaching model, CBL achieves the same effects on the mastery of theoretic knowledge on the techniques of acupuncture, moxibustion and cupping for the undergraduates. Moreover, CBL presentes the obvious advantages on the improvements of the ability of the manipulation selection based on the clinical symptoms and the practical manipulative skills of the students. Hence, CBL is deserved to be promoted as an experimental unit in the professional technique courses of acupuncture-moxibustion.

In these cases, our algorithms are trying their best to display videos with relevant content, which can sometimes result in matched videos with only a slight relation.
