当所有切片都就位后，加入 2 到 3 微升 L 15，将胶原蛋白适中到最早的位置，然后盖上培养皿。让胶原蛋白凝固 20 分钟。胶原蛋白凝固后，小心地加入 2 毫升切片培养基，该培养基已在 5% 二氧化碳和 37 摄氏度中平衡至少一小时。
必须注意不要在 37 摄氏度 5% 二氧化碳培养箱中将胶原蛋白从盖玻片处脱落，并在成像前让切片恢复至少三个小时。以高时间和空间分辨率对组织内的细胞行为进行长时间成像具有挑战性。最佳培养条件和白场显微镜的使用相结合对于获得良好结果至关重要。
使用装有气象站环境室的 delta vision core 宽视场显微镜对切片进行成像。腔室始终保持在 37 摄氏度，二氧化碳灌注仪将显微镜载物台保持在 5% 二氧化碳和 95% 空气成像通常使用 40 倍 1.30 NA 油浸镜头进行，并使用核心快照 HQ 两个称为 CCD 相机 Z 切片从 1.5 微米到 45 微米的组织暴露捕获一次。时间应尽可能短，例如 5 到 50 毫秒。
