1. 表达目的蛋白的稳定细胞系的构建
<ol>
	<li>通过向市售的含 GlutaMAX-I、丙酮酸钠和 4.5&#160;g/L D-葡萄糖的杜尔贝科改良伊格尔培养基（DMEM）中添加热灭活胎牛血清（FCS）和 1% 青霉素/链霉素来制备培养基。</li>
	<li>在 37 °C、5%&#160;CO2 条件下用上述培养基培养 HEK293 细胞。每三天传代一次。吸除旧培养基，将细胞重悬于 15 ml 新鲜培养基中。取 1 ml 细胞悬液加入新的 75&#160;cm2 细胞培养瓶中，再补充 14 ml 培养基。</li>
	<li>使用血细胞计数板分析细胞数量。</li>
	<li>以每孔 2 × 105 个细胞的密度将 HEK293 细胞接种于 6 孔板中，孵育 24 小时。</li>
	<li>转染操作按照转染试剂生产商提供的方案进行。使用转染试剂将 pEGFP-C2 或 pEGFP-C2-CaeB 质粒转染至细胞中。使用前将转染试剂和所需培养基升温至室温（RT）。转染试剂与 DNA 的用量比例为 3:1。
	<ol>
		<li>具体操作：将 1.5 µl 转染试剂直接加入 50 µl 无血清 DMEM 培养基中。为形成复合物，向该混合液中加入 0.5 µg 编码 GFP 或 GFP-CaeB 的 DNA，室温孵育 15&#160;分钟。</li>
		<li>将反应混合物逐滴加入细胞中进行转染，在 37 °C、5%&#160;CO2 条件下继续孵育 24 小时。</li>
	</ol>
	</li>
	<li>在 37 °C、5%&#160;CO2 条件下，加入 1&#160;mg/ml 遗传霉素以筛选 GFP 阳性克隆。</li>
	<li>6&#160;天后，通过流式细胞分选法将单个细胞分选至含有培养基与 HEK293 上清液（体积比 1:1）的 96 孔板中。HEK293 上清液由汇合生长的 HEK293 细胞培养液经 4,966 × g 离心 15&#160;分钟制得。</li>
	<li>次日，更换为含 1.5&#160;mg/ml 遗传霉素的培养基。每周换液一次。若细胞形成汇合单层，则转移至 24 孔板中。每周传代两次，传代比例为 1:5，使用含 1.5&#160;mg/ml 遗传霉素的培养基。最后通过免疫印迹分析和流式细胞术分析 GFP 阳性细胞的比例。</li>
</ol>
2. 通过免疫印迹分析对稳定细胞系进行分析
<ol>
	<li>吸除上清液，用预热的 PBS 洗涤贴壁细胞一次。将细胞重悬于 100 µl 2x Laemmli 样品缓冲液中，95 °C 加热 5&#160;分钟，然后于 -20 °C 保存。</li>
	<li>每份样品上样约 4 × 104 个细胞至 12% SDS-PAGE 凝胶中。同时，上样 6 µl&#160;商品化蛋白 Marker 作为分子量标准。在凝胶电泳系统中，使用 1x&#160;Laemmli 电泳缓冲液，于恒定电压 180&#160;V 条件下电泳 45–60&#160;分钟。</li>
	<li>使用 Trans-Blot SD 半干转印系统，以恒定电压 16&#160;V 将蛋白转移至 PVDF 膜（孔径 0.45 µm），转印时间为 60&#160;分钟。</li>
	<li>用 10 ml 含 5%&#160;脱脂奶粉的 PBS/0.05% Tween 20（MPT）溶液在室温下封闭膜 1 小时。</li>
	<li>将 4 µl 抗 GFP 抗体加入 4 ml MPT 溶液中稀释，4 °C 过夜孵育膜。</li>
	<li>用 10 ml PBS/0.05% Tween 20 溶液洗涤膜三次，每次 10 分钟。</li>
	<li>将膜与 0.8 µl 辣根过氧化物酶标记的二抗在 4 ml MPT 溶液中孵育。</li>
	<li>室温孵育 1 小时后，用 PBS/0.05% Tween 20 溶液洗涤膜三次（同步骤 2.6），然后根据试剂盒说明书使用商品化检测试剂检测辣根过氧化物酶活性。采用常规胶片显影设备显影蛋白条带。</li>
</ol>
3. 使用流式细胞仪分析细胞。
<ol>
	<li>用 PBS 重悬细胞。以 HEK293 细胞作为阴性对照，调节前向散射（FSC）和侧向散射（SSC），使细胞位于量程范围内。通过排除细胞双联体、聚集体和细胞碎片，在活细胞群上圈定门控区域。</li>
	<li>调节光电倍增管（PMT）增益，使未染色细胞位于 FL1 通道（488 nm 氩激光）直方图的最左侧。</li>
	<li>为分析 HEK293-GFP 和 HEK293-GFP-CaeB 细胞的荧光强度，打开 FL1 通道的直方图，并对门控内的细胞群采集 10,000 个事件。</li>
	<li>使用商用 FACS 分析软件分析数据。</li>
</ol>
4. 使用可诱导表达载体系统转染稳定细胞系
<ol>
	<li>以每孔 1 × 105 个细胞的密度，将稳定表达 GFP 或 GFP-CaeB 的 HEK293 细胞接种于 12 孔板中。</li>
	<li>接种后 19 小时，用调控质粒和报告质粒共同转染细胞，报告质粒编码 Bax 或活化型半胱天冬酶&#160;3。
	<ol>
		<li>将 100&#160;ng pWHE125-P 调控质粒（图 2）与 100&#160;ng 报告质粒 pWHE655-hBax 或 pWHE655-revCasp3（图 3）以及 0.4 µl 聚乙烯亚胺共同转染至稳定的 HEK293 细胞中。</li>
		<li>分别将 DNA 和聚乙烯亚胺转染试剂加入 75 µl Opti-MEM 培养基中，于室温准确孵育 5&#160;分钟。为形成复合物，将两种混合液合并后于室温继续孵育 15&#160;分钟。</li>
	</ol>
	</li>
	<li>将聚乙烯亚胺/DNA 溶液逐滴加入细胞中，于 37 °C、5%&#160;CO2 条件下孵育。</li>
</ol>
5. 诱导细胞凋亡
<ol>
	<li>转染后5小时，向培养基中加入1 µg/ml多西环素，诱导促凋亡蛋白的表达。将细胞在37 °C、5%&#160;CO2条件下孵育18小时。</li>
</ol>
6. 通过免疫印迹分析宿主细胞凋亡
<ol>
	<li>在收获细胞前，于光学显微镜下检查细胞，以确认是否出现细胞死亡。凋亡细胞可通过观察到死亡细胞周围存在凋亡小体来识别。</li>
	<li>移除上清液，并用预热的 PBS 洗涤贴壁细胞一次。将细胞重悬于 100 µl 的 2x Laemmli 样品缓冲液中，在 95 °C 下加热 5&#160;分钟，随后于 -20 °C 保存。</li>
	<li>每份样品上样约 4 × 104 个细胞至 12% SDS-PAGE 凝胶中。同时，上样 6 µl 商品化蛋白 Marker 作为分子量标准。在凝胶电泳系统中，使用 1x&#160;Laemmli 电泳缓冲液，在恒定电压 180&#160;V 下电泳 45–60&#160;分钟。</li>

	<li>室温孵育 1 小时后，用 PBS/0.05% Tween 20 溶液洗涤三次（如步骤 6.7 所述），并根据生产商说明书，使用商品化检测试剂盒检测辣根过氧化物酶活性。采用常规胶片显影设备以显示蛋白条带。</li>
	<li>用 7 ml 西部印迹 stripping 缓冲液在室温孵育 30&#160;分钟，以去除已结合的抗体。</li>
	<li>用 PBS/0.05% Tween 20 溶液洗涤三次（如步骤 6.7 所述）后，将膜与 4 µl 抗肌动蛋白（actin）一抗在 4 ml MPT 中 4 °C 过夜孵育。</li>
	<li>用 MPT 溶液洗涤三次（如步骤 6.7 所述）后，将膜与 0.8 µl 辣根过氧化物酶标记的二抗在 4 ml MPT 中稀释后孵育。</li>
	<li>室温孵育 1 小时后，用 PBS/0.05% Tween 20 溶液洗涤三次（如步骤 6.7 所述），并根据生产商说明书，使用商品化检测试剂盒检测辣根过氧化物酶活性。采用常规胶片显影设备以显示蛋白条带。

许多革兰氏阴性细菌病原体的毒力依赖于特化的分泌系统，以劫持真核宿主细胞。细菌利用这些分泌系统将细菌毒力蛋白（效应蛋白）注入宿主细胞内，从而调控多种细胞和生化活动。对效应蛋白的研究不仅极大地增进了人们对宿主/病原体相互作用基本机制的理解，也深化了对真核细胞基本生物学的认识1。研究表明，调控宿主细胞凋亡是许多胞内病原体的重要毒力机制，目前已鉴定出多个调控凋亡的效应蛋白2-9。然而，在许多情况下，这些蛋白精确的分子作用机制仍不清楚。
细胞凋亡是一种程序性细胞死亡形式，在机体对感染的免疫应答中发挥重要作用10。目前已鉴定出两条主要的细胞凋亡通路：一条靶向线粒体（即内在凋亡通路），另一条通过细胞膜上的死亡受体直接传导信号（即外在凋亡通路）。内在或线粒体介导的细胞死亡通路由细胞内信号触发，涉及Bax和Bak的激活，二者是Bcl-2家族中的促凋亡成员。该家族由促凋亡和抗凋亡的调控蛋白组成，共同调控细胞死亡过程11-14。凋亡激活后，Bax和Bak发生寡聚化，进而导致线粒体外膜通透化，促使细胞色素C释放到细胞质中。细胞色素C的释放会激活凋亡小体中的caspase 9，从而启动效应caspase 3和7的活化15。这导致特定底物蛋白的水解，结果之一是细胞表面暴露出磷脂酰丝氨酸16，并释放一种特异性的DNA酶，使染色质发生断裂17,18。
为了确定某个效应蛋白在凋亡级联反应中的作用位点，采用了可诱导表达系统19。用于外源基因条件性表达的调控系统在分析蛋白质在细胞内的功能，或其在组织、器官及生物体发育过程中，以及疾病起始、进展和维持阶段的重要性方面，已成为不可或缺的工具20-23。通常，可诱导控制系统（例如本文所采用的 Tet 系统24）构成一个人工转录单元（见图1）。该系统的一个组分是一种人工构建的转录因子，称为 tTA（四环素依赖性转录激活因子），由细菌转录阻遏蛋白 TetR25 与一个介导转录激活或沉默的哺乳动物蛋白结构域融合而成24,26。第二个组分是一个杂合启动子，称为 TRE（四环素响应元件），由包含至少一个 TATA 盒和一个转录起始位点的真核生物最小启动子，与多个 TetR 特异性 DNA 结合位点 tetO 的重复序列连接而成24,25。第三个组分是 TetR 的天然配体——四环素或其衍生物（如脱水四环素或强力霉素）25。当向培养基中加入配体后，TetR 对 tetO 的亲和力丧失，并从 TRE 上解离，从而阻断目的基因的转录。因此，无论是在细胞培养还是在动物体内，外源基因的表达均可实现时间依赖性和剂量依赖性的精确控制20,23,24。使用 tTA 时，外源基因持续表达，除非存在四环素。这在研究细胞毒性或致癌性蛋白时可能成为劣势，因为必须先将四环素从系统中去除，才能启动外源基因表达，进而监测目标蛋白对细胞的影响。这一过程耗时较长，且去除往往不完全，尤其是在转基因动物中27。为克服这一局限，研究人员利用对强力霉素具有相反响应特性的 TetR 突变体，构建了一种新的转录因子 rtTA（反向 tTA）28。rtTA 仅在强力霉素存在时结合 TRE 并同时激活转录。针对系统中残余的“渗漏”表达（即在无 TRE 结合转录因子时仍发生的外源基因表达），其来源可能包括：(i) 基因组整合位点的位置效应，(ii) TRE 本身29，或 (iii) tTA/rtTA 的非特异性结合28，研究人员在系统中引入了一个额外的转录沉默因子，称为 tTS（四环素依赖性转录沉默因子）30。tTS 与 rtTA 共同构成一个双重调控网络（见图1）。在无强力霉素时，tTS 结合 TRE 并主动抑制任何残余的转录；而在强力霉素存在时，tTS 从 TRE 上解离，同时 rtTA 结合并诱导目的基因表达。这种额外的严格调控机制对于高效表达高活性的细胞毒性蛋白往往是必需的31-34。

以下实验的总体目标是研究细菌毒力因子对细胞内信号转导的干扰作用。首先建立稳定表达目标蛋白的细胞系，以在整体细胞水平上研究其活性；作为第二步，构建一个可诱导表达载体系统，该系统可在特定步骤激活宿主细胞的信号级联反应。随后分析目标蛋白是否能够干扰宿主细胞信号级联的激活过程，从而确定目标蛋白发挥作用的具体位点。结果表明，柯兹黄杆菌（*Coxiella burnetii*）毒力因子KAB可干扰凋亡级联反应中Bax下游、Caspase-3上游的信号传导，该结论基于Western blot分析得出。该方法有助于回答感染生物学领域的关键问题，例如确定特定细菌效应蛋白干扰信号转导级联反应的关键步骤。这不仅有助于我们更好地理解病原微生物如何操控其宿主细胞，还能揭示这些基本细胞过程的功能机制。尽管该方法可用于深入了解细菌如何阻止细胞凋亡的发生，但它也可应用于细胞死亡领域的其他信号转导系统，如坏死性凋亡（necroptosis）或细胞焦亡（pyroptosis），以及参与细胞增殖、基因调控或免疫系统反应的信号网络。本实验操作将由 Mann 实验室的研究生 Stephanie Besler 演示。首先，在 12 孔板中接种稳定表达 GFP 或 GFP-SayB 的 HEK 293 细胞，每孔接种密度为 100,000 个细胞。接下来，分别在75 µL的Opti-MEM培养基中制备DNA和聚乙烯亚胺转染试剂，并在室温下孵育5分钟以形成复合物。将两种混合物一起在室温下共同孵育15分钟。随后，将聚乙烯亚胺-DNA溶液逐滴加入细胞中，并在37°C、5% CO₂条件下孵育。转染后5小时，向培养基中加入终浓度为1微克/毫升的多西环素，以诱导促凋亡蛋白的表达。随后将细胞在37摄氏度、5%二氧化碳条件下孵育18小时，并在光学显微镜下观察细胞，于收获前检查细胞死亡的诱导情况。观察诱导的凋亡细胞，这些细胞可通过凋亡小体的存在进行检测。接着，移除上清液并洗涤贴壁细胞。用预热的PBS洗涤一次后，将细胞重悬于100微升的2×拉姆利样品缓冲液中。然后在95摄氏度加热五分钟，之后置于负20摄氏度保存以备后续使用。随后，加载六微升商用蛋白ladder作为分子量标记。然后在12% SDS-PAGE凝胶上每样品加载约40,000个细胞。使用180伏恒定电压，在凝胶电泳系统中运行凝胶45至60分钟，采用一倍拉利运行缓冲液。随后，使用转印 blot SD 半干转移槽，在16伏恒定电压下将蛋白质转印至PVDF膜上，持续60分钟。然后在室温下用10毫升MPT封闭膜，孵育1小时。将7微升抗裂解PARP抗体加入7毫升MPT中稀释。将PVDF膜与抗体溶液在4摄氏度下孵育过夜。移除抗体溶液后，用含0.05%吐温20的PBS缓冲液洗涤三次，每次10分钟；随后在室温下用7毫升MPT稀释1.4微升辣根过氧化物酶标记的二抗溶液，与膜孵育1小时。用含0.05%吐温20的磷酸盐缓冲液（PBS）洗涤膜三次，根据生产商说明书，使用商用试剂盒检测辣根过氧化物酶活性，并使用标准设备进行胶片显影以观察蛋白条带。随后，在用含0.05%吐温20的PBS洗涤三次后，将膜与7毫升Western blot stripping缓冲液在室温下孵育30分钟，以去除结合的抗体，再将膜与4毫升MPT缓冲液中的4微升抗肌动蛋白（anti-actin）抗体在4摄氏度下孵育过夜。第二天，用约10毫升含0.05%吐温20的磷酸盐缓冲液对膜进行三次洗涤，每次10分钟；然后将膜与1.4 µL辣根过氧化物酶标记的二抗在7毫升MPT中稀释后共同孵育，室温孵育1小时。用含0.05%吐温20的PBS缓冲液洗涤膜三次，使用商用试剂盒检测辣根过氧化物酶活性，并采用标准设备进行胶片显影以显示蛋白条带。本实验中，对HEK 293 GFP和HEK 293 GFP sabe细胞的全细胞裂解物进行GFP免疫印迹，以验证其稳定表达。HEK 293 GFP 和 HEK 293 GFP SA B 细胞的代表性流式细胞术分析显示了GFP表达的强度。HEK 293 GFP 和 HEK 293 GFP SA B 细胞经4 µM孢霉素处理6小时以诱导内在性细胞凋亡。通过裂解PARP免疫印迹分析检测细胞凋亡，结果显示，与HEK 293 GFP细胞相比，HEK 293 GFP SA B细胞中PARP的裂解减少。CB 可保护细胞免受 Bax 诱导的细胞凋亡，但不能保护细胞免受 Caspase-3 诱导的细胞凋亡。在实施该实验流程时，应尽可能检测信号通路中的多个组分，以帮助确定相互作用发生的步骤。此外，建议对在静息状态和激活状态下均被激活的蛋白质进行检测，以判断所选的效应蛋白是干扰了通路本身，还是干扰了该流程之后的激活步骤。还可以采用其他方法，如基因敲低、基因敲除、酵母双杂交、下拉实验、翻译后修饰或蛋白质水解稳定性分析，以回答更多问题，例如：这些微生物效应蛋白操纵宿主正常细胞生理活动的确切分子机制是什么？这种操纵如何有利于病原体的存活与传播？
