我们感谢京都大学的Matsushita博士和Oka博士提供了由35S启动子驱动表达PHYB-GFP的phyB突变体。本研究部分得到了创新领域科学研究所专项基金、提议研究领域项目（19H05713至K.Y.）的资助。

在本研究中，建立了两种不同的方法用于制备活体胚乳细胞层样品：一种适用于发育中的种子，另一种适用于成熟种子。根据种皮的坚硬程度不同，所需的操作方法略有差异。所用试剂和设备的详细信息列于下文 材料表.
1. 从发育中的种子制备完整的胚乳样品
<ol>
	<li>角果采集
	<ol>
		<li>培养 拟南芥 在土壤或岩棉上种植植物直至开花。</li>
		<li>用彩色线标记完全开放的花朵（开花后0天，0 DAF）。 
		注意：避免使用绿色、黄色或棕色的线标记花朵，因为这些颜色与正在生长或成熟的植株及角果难以区分。</li>
		<li>在果柄处切下标记的正在发育的角果（如图所示） 图1(1) 并将其收集至 1.5 mL 离心管中。 
		注意：发育中的角果在授粉后12-16天适用于本方案的制备。</li>
	</ol>
	</li>
	<li>发育中种子的解剖 
	注意：以下步骤必须在湿润的滤纸上进行，以防止样品干燥。操作应在体视显微镜下进行。
	<ol>
		<li>分裂一个阀门（如图所示） 图1(2)）从隔膜（图中所示） 图1（1）使用两把镊子：一把尖端较粗，用于夹持；另一把尖端尖锐，用于撕开。</li>
		<li>用镊子夹住尖端闭合的镊尖，轻轻从角果中取出正在发育的种子，以避免损伤种子（图1(2)).</li>
		<li>用镊子夹住种子，避免压碎种子，然后进行操作&#160;使用注射针（27 G，0.40 mm × 19 mm）在包围胚的种皮和胚乳上制造一个约0.2 mm大小的切口图1(3)). 
		注意：制作切口的最佳位置位于子叶与胚根的交界处。</li>
		<li>用镊子夹住种子以挤出胚胎图1(4) 不要压碎由种皮和胚乳组成的空种荚，尽量保持其圆形形态。</li>
		<li>将注射针插入空的种子囊膜的种脐处，从内向外穿刺图1(5)).</li>
		<li>保持针头位置不变，用闭合的镊子尖端刮擦种皮表面，并剪开空种荚的一侧，使其打开（图1(6)).</li>
		<li>使用尖头镊子将空的种子信封展开成一张纸图1（7））。此时样品应被分离为由胚乳和种皮层组成的双层片状结构（图1(8)). 
		注意：如果在下方步骤3中使用水作为封片介质时样品容易卷曲，可将片状样品分割成两部分。从约授粉后18天（18 DAF）的角果中收获的种子（此阶段种皮呈褐色但尚未完全干燥）也可采用本方案处理，但在制样前需将种子浸水数分钟。</li>
	</ol>

2. 成熟种子完整胚乳样品的制备
<ol>
	<li>成熟种子的吸胀作用
	<ol>
		<li>向含有干燥物的1.5 mL离心管中加入1 mL双蒸水 拟南芥 种子。</li>
		<li>将种子在室温下浸润至少40分钟（图2(1)). 
		注意：干燥种子及吸水后40分钟内的种子在步骤2.2.1和2.2.2中难以对种皮和胚乳进行划伤，且难以在不损坏空种壳的情况下取出胚；而较长的吸水时间有利于操作。</li>
		<li>使用1000 µL移液器将吸胀的种子转移到湿润的滤纸上。</li>
	</ol>
	</li>
	<li>成熟种子的解剖 
	注意：以下步骤必须在湿润的滤纸上进行，以防止样品干燥。操作应在体视显微镜下进行。
	<ol>
		<li>用镊子夹住种子，避免压碎，使用注射针在包围胚的种皮和胚乳上制造一个约0.2 mm大小的切口图2(2)).&#160;</li>
		<li>用镊子夹住种子，将胚胎挤出图2（3）不要压碎空的种皮囊，其包含种皮和胚乳。尽量保持其圆形形状。</li>
		<li>用注射针将空的种子囊的上下两侧切开，使其形成圆筒状（图2(4)).</li>
		<li>沿圆柱形空种皮的中心轴切开，将其分为两部分图2(5)。样品应分离为由胚乳层和种皮层组成的双层片状结构（图2(6)).</li>
	</ol>

3. 显微镜观察
<ol>
	<li>将胚乳样品以片状置于载玻片上，并用水或全氟十氢化萘（PFD）封片。 
	注意：如果样品与盖玻片之间残留气泡，使用全氟十氢化萘（PFD）会特别有效，已有文献报道其在成像含有气室的样品（如叶片）时效果良好18,19。PFD具有最低的表面张力，能够轻易填充样品表面的空气间隙。然而，对于延时成像，建议使用水作为封片介质，因为成熟种子中的水分含量应保持充足，以维持细胞的流动性。</li>
	<li>轻柔地将盖玻片覆盖在样品上，确保胚乳层朝向盖玻片。 
	注意：可使用指甲油或封片膏密封盖玻片边缘，以防止样品和封片介质脱水。</li>

最近的多项研究表明，胚乳在调控种子最佳萌发过程中起着至关重要的作用14,15,16,17。例如，胚乳细胞中的光感受器光敏色素B（PHYB）能够感知&#160;红光（R）或&#160;远红光（FR），从而调控萌发反应15。胚乳还具有温度感知组织的功能，在高温条件下抑制萌发反应16。胚乳的质量控制对于种子的最佳萌发至关重要，尤其是在长期贮藏的种子中17。

Preparation of Intact Tissue for Microscopic Analysis of the Endosperm Cell Layer in Developing and Mature Arabidopsis Seeds

根据图1所示的方案，从开花后14天（DAF）采集的角果中分离发育中的种子以制备胚乳样品（在此阶段，种皮仍为绿色）。在较大区域内观察到大量胚乳细胞及其细胞内结构（图3A）。本实验使用C末端融合GFP的PHYB蛋白（PHYB-GFP）表达的种子。众所周知，PHYB在红光激活后会转位至细胞核，并形成称为光体（photobodies, PBs）的PHYB阳性斑点，这些结构参与调控种子萌发4,20,21,22。样品取自在22 °C连续白光条件下生长的植株。如图3B所示，在胚乳细胞的细胞核内检测到了PBs。该结果与先前研究一致，表明胚乳中的PHYB可调控种子的最适萌发15...

我们的研究领域之一是种子科学。我们旨在解答胚乳如何影响种子萌发这一科学问题。一种名为种子生长抑制试验的体外检测方法，揭示了胚乳在种子萌发过程中的生理重要性。拟南芥种子非常微小，其胚乳细胞层位于种皮（testa）下方，即细胞内部。在不进行化学固定的条件下制备胚乳样品具有较大挑战性。以往对胚乳的研究分析均采用化学固定的样品，因此无法观察活体蛋白的动态变化。本实验方案可实现对胚乳细胞动态过程的五维细胞成像。该方案仅需使用常规实验室设备，即可深入探究胚乳细胞间的细胞事件、分子机制及其正常功能。[旁白] 首先将拟南芥植株种植于土壤或岩棉中，直至开花阶段。用彩色线标记完全开放的花朵，以指示开花后零天。选择开花后12至16天的角果用于下一步操作。使用精细剪刀在果柄处剪下标记的发育中角果，并将其收集到1.5毫升离心管中。准备湿润的滤纸用于解剖，以防止样品干燥，并将样本置于体视显微镜下。使用两把镊子，一把尖端较粗用于固定，另一把尖端尖锐用于撕开。从果瓣与果柄连接处（replum）分离出一个果瓣。然后使用闭合尖端的镊子，轻柔地从角果中夹取发育中的种子，避免损伤。使用27号注射针头，在包围胚的种皮（testa）和胚乳（endosperm）上制造一个约0.1至0.2毫米大小的切口。用镊子夹住种子，注意不要压碎，并在子叶与胚根交界处进行划痕。接着，用镊子轻轻挤压种子，将胚推出，注意不要压碎空的种皮囊，并保持其圆形形态。将注射针头从切口插入空的种皮囊内，并由内向外穿刺。随后保持针头位置不动，用闭合尖端的镊子刮擦种皮表面。然后剪开种皮囊的一侧，使其能够展开。现在使用尖头镊子将种皮囊展开成片状。分离后的样品应为由胚乳和种皮组成的双层片状结构。将干燥的拟南芥种子放入1.5毫升离心管中，加入1毫升双蒸水。在室温下浸泡种子至少40分钟。在体视显微镜载物台上准备一张湿润的滤纸用于解剖，使用1000微升移液器将吸胀后的种子转移至湿润滤纸上。将样品置于体视显微镜下观察。使用注射针头在包围胚的种皮和胚乳上制造一个约0.2毫米大小的切口。用镊子夹住种子，避免压碎，并在子叶与胚根交界处进行划痕。随后用镊子轻压种子，将胚推出。避免压碎空的种皮囊，保持其圆形形态。使用注射针头切割空种皮囊的上下两侧，将其塑造成圆柱形。接着沿圆柱形种皮囊的中心轴剪开，将其分为两部分。最终应得到由胚乳和种皮组成的双层片状结构。将分离出的胚乳样品以片状置于载玻片上，并用水封片。最后，轻轻盖上盖玻片，确保胚乳层朝上贴近盖玻片。使用指甲油或封片胶密封盖玻片边缘，防止干燥，然后在显微镜下观察样品。利用既定的制备方案，成功观察到开花后14天收获种子中的大量胚乳细胞及其细胞内结构。在白光条件下，表达PHYB-GFP的胚乳细胞核内检测到光体（photo bodies），证实其存在及核内定位。在交替红光与远红光照射下，胚乳细胞表现出可逆的光体形成，证实PHYB具有光可逆性及生物学活性，至少可持续至解剖后4.5小时。对种子抑制3小时后完整胚乳细胞中的线粒体进行荧光延时成像，检测到线粒体运动。在种子抑制1天后，胚乳细胞中观察到更活跃的线粒体运动，表明其活性随时间逐渐增强。
