Experimental Screening Protocols, Immunocytochemistry and Microscopy-based Imaging Techniques for Penium margaritaceum

P. margaritaceumの細胞壁を抗ペクチンモノクローナル抗体(JIM5など)で標識すると、カルシウム錯体繊維のネットワークと、規則的なパターンまたは格子を形成する突起が明らかになります(図1)。ペクチンは細胞中心または峡部に沈着し、そこで古いペクチンを極に向かって移動させます(図2)。異なるペクチン抗体様JIM7で標識すると、峡部の狭いバンドにおける高メチルエステル化ペクチンの初期分泌が強調されます(図3A、B)。細胞壁に位置する他のポリマーには、mAbで標識されたアラビノガラクタンタンパク質、JIM13が含まれます(図4)。大量のEPSが細胞壁の外部から分泌され(図5)、滑走と細胞凝集体の形成が可能になります。これらの標識技術のいくつかは、細胞壁の定量的検査や細胞増殖などの発生研...
