Updated Protocol for the Assembly and Use of the Minibioreactor Array (MBRA)

대변 슬러리는 인간의 대변을 혐기성 방식으로 해동한 다음 인산염 완충 식염수(PBS)와 최종 농도 25% w/v로 혼합하여 제조했습니다. 무균성을 확인한 후, 9개의 바이오리액터 챔버에 각각 3mL의 동일한 대변 슬러리를 접종했습니다. 미생물 군집은 배지 투입이나 폐기물 제거 없이 밤새 성장하여 16시간 동안 별도의 배치 배양을 수행했습니다. 그런 다음 공급 펌프를 켜고 시간당 1.92mL의 유량으로 설정했습니다. 4일간의 연속 흐름 후, 바이오리액터에서 샘플을 수집하고 16S rRNA 유전자 시퀀싱을 사용하여 미생물 군집 구성을 분석한 다음 Deblur로 노이즈 제거를 수행하고 QIIME 213의 SILVA 138 SSU 데이터베이스를 사용하여 분류학적 분류를 수행했습니다. 총 65개의 속이 9개의 반복실험 모두에서 검출되었지만 생물반응기는 18개의 속만이 지배적이었고 각 속은 9개의 반복실험 중 하나에서 최소 2%의 풍부도를 구성했습니다(그림 5). 생물반응기는 65개 속 중 22개가 9개의 반복실험 모두에서 검출되었고 추가로 17개의 속이 복제의 절반 이상에서 검출될 정도로 높은 재현성을 보였다. 적어도 하나의 반응기에 없는 대부분의 속(43속 중 37속)은 희귀종이었으며 각각 생물반응기에서 상대적 풍부도가 2% 미만이었습니다. 요약하면, 연속 흐름 MBRA 배양은 각 바이오리액터 챔버 내에서 16시간 동안 별도의 배치 배양을 한 후에도 동일한 대변 샘플에서 파생된 복잡하고 재현 가능한 미생물 군집을 지원했습니다.
