Elektrofysiologische opnamen van<em> Drosophila</em> Embryo's laten analyses van de ontwikkeling van spier-en neuron elektrische eigenschappen, evenals karakteriseren van functionele synaptogenese op de glutamaat neuromusculaire verbinding en centrale cholinerge en GABA-erge synapsen.
Drosophila is een vooraanstaande genetisch model voor de studie van zowel de embryonale ontwikkeling en functionele neurowetenschappen. Traditioneel worden deze velden geïsoleerd nogal van elkaar, met grotendeels onafhankelijke geschiedenis en wetenschappelijke gemeenschappen. Echter, de interface tussen deze vaak uiteenlopende terreinen is de ontwikkelingsprogramma's ten grondslag liggen aan de verwerving van functionele elektrische signaal-eigenschappen en de differentiatie van de functionele chemische synapsen tijdens de laatste fasen van neurale circuit formatie. Deze interface is een uitermate belangrijk gebied voor onderzoek. In Drosophila, deze fasen van de functionele ontwikkeling optreden tijdens een periode van <8 uur (bij 25 ° C) tijdens het laatste derde deel van de embryogenese. Deze late ontwikkelingsfase werd lang beschouwd als hardnekkige het onderzoek als gevolg van de afzetting van een zware, ondoordringbare epidermale cuticula. Een doorbraak van tevoren was de toepassing van de water-polymeriseren chirurgische lijm die lokaal kan worden toegepast op de cuticula om gecontroleerde dissectie van de laat-stadium embryo's mogelijk te maken. Met een dorsale longitudinale incisie, kan het embryo te plat, waardoor de ventrale zenuw snoer en de lichaamswand spieren aan experimenteel onderzoek. Whole-cell patch-clamp technieken kunnen dan worden gebruikt om op te nemen van individueel herkenbare neuronen en somatische spieren. Deze opname configuraties zijn gebruikt om het uiterlijk en de rijping van de ionische stromen en actiepotentiaal voortplanting in zowel neuronen en spieren te volgen. Genetische mutanten die deze elektrische eigenschappen werden gekenmerkt om de moleculaire samenstelling van ionenkanalen en de bijbehorende signalering complexen onthullen, en om de verkenning van de moleculaire mechanismen van functionele differentiatie te beginnen. Een bijzondere focus is de montage van synaptische verbindingen, zowel in het centrale zenuwstelsel en periferie. De glutamaat neuromusculaire junctie (NMJ) is het meest toegankelijk is voor een combinatie van optische beeldvorming en elektrofysiologische opname. Een glas zuig elektrode wordt gebruikt om de perifere zenuw te stimuleren, met prikkelende knooppunt huidige (EJC) opnames gemaakt in de voltage-opgespannen spieren. Deze opname configuratie is gebruikt om de functionele differentiatie van de synaps grafiek en het uiterlijk en de rijping van presynaptische glutamaat afgifte eigenschappen track. Daarnaast kunnen postsynaptische eigenschappen onafhankelijk van elkaar worden getest via iontoforetische of druk de toepassing van glutamaat rechtstreeks naar de spier oppervlak, het uiterlijk en de rijping van de glutamaat receptor velden te meten. Zo kunnen zowel pre-en postsynaptische elementen afzonderlijk of in combinatie worden gevolgd tijdens de embryonale synaptogenese. Dit systeem is zwaar gebruikt voor het isoleren en karakteriseren van genetische mutanten dat embryonale synapsvorming aantasten, en dus onthullen de moleculaire mechanismen voor de specificatie en differentiatie van synaps verbindingen en functionele synaptische signalering eigenschappen.
Elektrofysiologische opname van Drosophila embryo's moet handmatig manipulatie en dissectie. De gezondheid van de voorbereiding, en de daaruit voortvloeiende de kwaliteit van de opnames, hangt af van een in staat om snel en netjes voor te bereiden de kwetsbare embryonale weefsels voor het opnemen, en voer vervolgens het experiment. Onderzoekers moet bedreven zijn in zowel de embryonale ontledingen en patch clamp electrofysiologie voordat je probeert om zowel aan te pakken tegelijk.
