Denne artikkelen viser en optimalisert prosedyre for avbildning av de nevrale substrater av auditiv stimulering i songbird hjernen ved hjelp av funksjonell Magnetic Resonance Imaging (fMRI). Den beskriver utarbeidelsen av lyd stimuli, plassering av faget og oppkjøp og påfølgende analyse av fMRI data.
Nevrobiologi av fuglesang, som en modell for menneskelig tale, er en uttalt område av forskning i atferdsmessige nevrovitenskap. Mens elektrofysiologiske og molekylære metoder tillate etterforskningen av enten ulike stimuli på noen nevroner, eller en stimulans i store deler av hjernen, oksygentilførsel i blodet nivå avhengig (BOLD) funksjonell Magnetic Resonance Imaging (fMRI) kan kombinere både fordeler, dvs. sammenligne nevrale aktivering indusert av ulike stimuli i hele hjernen samtidig. fMRI i sangfugler er utfordrende på grunn av den lille størrelsen på sine hoder og fordi deres ben og spesielt deres skallen omfatter en rekke luft hulrom, indusere viktige mottakelighet gjenstander. Gradient-ekko (GE) BOLD fMRI har blitt brukt til sangfugler 1-5 (for en gjennomgang, se 6). Disse studiene fokuserte på de primære og sekundære auditive hjerneområder, som er områder uten mottakelighet gjenstander. Men fordi processes av interesse kan forekomme utover disse regionene, er hele hjernen BOLD fMRI nødvendig å bruke en MR-sekvens mindre utsatt for disse gjenstandene. Dette kan oppnås ved hjelp av spinn-ekko (SE) BOLD fmri 7,8. I denne artikkelen beskriver vi hvordan du bruker denne teknikken i sebra finker (Taeniopygia guttata), som er små sangfugler med en kroppsvekt på 15-25 g grundig studert i atferdsmessige nevrovitenskap av fuglesang. Hovedtema for fMRI studier på sangfugler er sang oppfatning og sang læring. Den auditive natur stimuli kombinert med svak BOLD følsomhet SE (i forhold til GE) baserte fMRI sekvenser gjør gjennomføringen av denne teknikken svært utfordrende.
I denne rapporten beskriver vi en optimalisert protokoll for detaljert in vivo karakterisering av nevrale substrater av auditiv stimulering i bedøvede sebra finker.
I tråd med presenterte protokollen, de fleste av funksjonelle hjerneaktivitet hos dyr ved hjelp BOLD fMRI, anaesthetize dyrene under oppkjøpet. Opplæring dyr for å venne dem til magneten miljø og skanneren støy under studieopphold er også mulig, men ganske tidkrevende og utfordrende, og derfor sjelden brukt.
