Spatial inlärning och minne av smågnagare bedöms ofta via navigationsförmåga i labyrinter, mest populära av som är vatten och torka-land (Barnes) labyrinter. Förbättrad prestanda över sessioner eller försök är tänkt att reflektera inlärning och minne av utrymnings bur / plattform plats. Betraktas mindre stressande än vatten labyrinter är Barnes labyrint en relativt enkel design av en cirkulär plattform topp med flera hål jämnt fördelade runt omkretsen kant. Alla utom ett av hålen är falsk bottnade eller blinda sinande, medan en leder till en flykt bur. Svagt aversiva stimuli (t.ex. ljus taklampa) ger motivation att lokalisera fly buren. Latens för att lokalisera flykt buren kan mätas under sessionen, men ytterligare effektmått kräver normalt videoinspelning. Flera typer av labyrinter har utvecklats som icke-invasiv bedömning av denna hippocampus beroende spatial inlärning och minne i små och stora gnagare (t.ex. vatten labyrint, flera T-labyrinten, radial arm labyrint och torr-mark labyrinter) 3-6. Här fokuserar vi på den runda plattformen eller Barnes labyrint, som först beskrevs 1979 av Dr Carol Barnes 7. Denna labyrint har använts för att testa rumslig navigering inlärning och minne i en rad olika gnagarmodeller, inklusive råttor (RaTTU norvegicus), möss (Mus musculus), rådjur möss (Peromyscus maniculatus bairdii), Kalifornien möss (Peromyscus californicus), och hystricomorph gnagare (t.ex. degus [Octodon degus]) 8-13. Andra arter som bedöms med hjälp av Barnes labyrinten inkluderar amerikanska kackerlackor (Periplaneta americana) 14, majs ormar (Elaphe guttata guttata) 15, squamate reptiler (t.ex. sido-fläckig ödlor [Uta stansburiana]) 16 och icke-mänskliga primater (t.ex. mus lemurer [Microcebus murinus]) 17. En primär fördel med Barnes labyrint användning är att den framkallar mindre stress i ämnena i förhållande till vatten labyrinter, såsom Morris water maze 43, även om båda kan framkalla akuta ökningar i plasma kortikosteron koncentrationer i möss 44. Som torra land labyrint kan Barnes labyrinten vara mer etologiskt relevant för markbundna gnagare 45. Även om vatten labyrint prestanda har visat sig vara känsligare för genetiska förändringar i möss 3,46,47, är mer känsliga för vissa andra ändringar 48,49 Barnes labyrint prestanda. I gnagarmodeller där vatten labyrint användning inte är möjlig, får den Barnes labyrinten ger en finjusteras bedömning av rumsliga minne lagring 31. De milt aversiva stimuli som normalt används i Barnes labyrint (dvs. starkt ljus),dock inte kan ge tillräcklig motivation för gnagare att lokalisera flykt buren 45. Dessutom kan gnagare lära sig att inget straff uppstår om de inte kommer in i flykt buren. I stället för att aktivt söka efter flykt buren, vissa gnagare aktivt utforska en labyrint för långa löptider för varje försök. Som granskas av Kennard och Woodruff-Pak 24, kommer denna ökade prospekterings förlänga latens för att lokalisera utrymnings bur, våglängd, och öka antalet fel. Således, mätning av flera parametrar, bland latens, felfrekvens, tiden i de korrekta och felaktiga kvadranter, hastighet, tid att flytta, tid att vila, och sökstrategi, tillsammans kan ge en bättre indikator för varje persons rumslig navigering inlärning och minnesförmåga 8 -10. Vissa har argued att primära mått på funktion är en mer korrekt bild av spatial inlärning än de totala åtgärderna 50. De flesta studier, inklusive de exempel som beskrivs här, använder latens för att komma in på flykt buren för att fastställa felfrekvens och sökstrategi. Vidare, några spårningsmjukvarusystem har en trepunkts kroppsdetektionssystem som kan mäta frekvenser sniffa rätt vs felaktiga hål. Barnes maze mönster varierar men i allmänhet var och en har 12 eller 20 potentiella utloppshål, av vilka endast en leder till hemmet eller en flykt bur. Flykt bur kan vara belägen antingen direkt under utloppshål på labyrinten topp (till labyrinter utan väggar) eller inbyggd i den omgivande väggen hos labyrinten. Figurerna 1-5 visar exempel på Barnes maze designer för Peromyscus arter (Figur 1) och råttor (figur 2-5). Pluggar eller bottnar måste täcka nonescape hål för att förhindra djuret från att falla ut ur labyrinten.
