A two-step procedure for the synthesis of pharmaceutically active indole-derivatives by C-H functionalization with anilines is described, using photo- and Brønsted acid catalysis.
The direct functionalization of C-H bonds is an important and long standing goal in organic chemistry. Such transformations can be very powerful in order to streamline synthesis by saving steps, time and material compared to conventional methods that require the introduction and removal of activating or directing groups. Therefore, the functionalization of C-H bonds is also attractive for green chemistry. Under oxidative conditions, two C-H bonds or one C-H and one heteroatom-H bond can be transformed to C-C and C-heteroatom bonds, respectively. Often these oxidative coupling reactions require synthetic oxidants, expensive catalysts or high temperatures. Here, we describe a two-step procedure to functionalize indole derivatives, more specifically tetrahydrocarbazoles, by C-H amination using only elemental oxygen as oxidant. The reaction uses the principle of C-H functionalization via Intermediate PeroxideS (CHIPS). In the first step, a hydroperoxide is generated oxidatively using visible light, a photosensitizer and elemental oxygen. In the second step, the N-nucleophile, an aniline, is introduced by Brønsted-acid catalyzed activation of the hydroperoxide leaving group. The products of the first and second step often precipitate and can be conveniently filtered off. The synthesis of a biologically active compound is shown.
Den direkte funktionalisering af CH-obligationer er et vigtigt og langvarigt mål i organisk kemi 1. Sådanne transformationer kan være meget stærke for at strømline syntese ved at spare steps, tid og materialer sammenlignet med konventionelle metoder, der kræver indføring og fjernelse af aktiverende eller lede grupper. Derfor funktionalisering af CH-obligationer er også attraktiv for grøn kemi 2. Under oxidative betingelser, to CH obligationer eller en CH, og et heteroatom-H-binding kan omdannes til CC og C-heteroatom obligationer, (figur 1) 3-9. Ofte er disse oxidative koblingsreaktioner kræver syntetiske oxidanter, dyre katalysatorer eller høje temperaturer. Derfor er mange forsøg på at udvikle metoder, der bruger billige katalysatorer, godartede tilstande og ilt eller luft som terminal oxidant 10.

Mange organiske forbindelser reagerer langsomt med ilt fra luften i autoxidation reaktioner, som kan functionalize CH bindinger ved effektivt at indsætte O 2, der danner et hydroperoxid del 11,12. Autoxidation bruges på industrielt plan genererede oxiderede forbindelser fra kulbrinter, men autoxidation er også en uønsket proces, hvis det fører til nedbrydning af værdifulde forbindelser eller materialer. I nogle tilfælde, for eksempel diethylether, hydroperoxider dannet i luften kan også være eksplosive. For nylig opdagede vi en reaktion, der anvender en autooxidation at danne en ny CC binding fra CH bindinger uden behov for en redox-aktiv katalysator 13,14 </sup>. Simply omrøring substraterne under oxygen i nærvær af en syrekatalysator fører til dannelsen af de nye produkter. Nøglen til reaktionen er den lette dannelse af mellemliggende hydroperoxider, som er substitueret med det andet substrat ved syrekatalyse 15. Reaktionen er imidlertid begrænset til xanthen og nogle beslægtede forbindelser, som er let oxideret under en atmosfære af oxygen, og produkterne har hidtil ikke fundet applikationer. Ikke desto mindre er inspireret af denne opdagelse, har vi udviklet en beslægtet oxidativ kobling metode der udnytter princippet om CH funktionalisering via mellemliggende peroxider (chips) til at syntetisere farmaceutisk aktive indolderivater 16.
Indoler, især tetrahydrocarbazoles 1 kan let oxideres til hydroperoxiderne 2 i nærvær af singlet oxygen 17-19, som kan dannes ved hjælp af en sensibilisator og synligt lys 20. En hydroperoxide del kan i princippet fungere som en leaving-gruppe, hvis aktiveret af syre katalyse og give mulighed for at indføre en nukleofil 21,22. Hydroperoxider er også kendt for at undergå syrekatalyseret omlejring reaktioner som anvendt i den industrielle syntese af phenolen fra cumen Hock processen 23. Ved omhyggelig optimering undersøgelser, kunne vi finde betingelser for at favorisere den ønskede substitution reaktion med N-nucleofiler som aniliner 3 over de uønskede nedbrydningsprodukter veje ved omlejring 16. Her beskriver vi denne to-trins CHIPS procedure i detaljer, der kun bruger synligt lys sensibiliserende, oxygen og syre. Blandt de udvalgte produkter er indolderivater 4, som viser høj antiviral aktivitet eller inhiberer vaskulær endotel vækstfaktor (VGF), hvilket kan være vigtigt for tumorterapi 24-26.
Sammenfattende kunne vi påvise, at en CH-binding i tetrahydrocarbazoles kan hensigtsmæssigt funktionaliseret til at generere CN-koblingsprodukter i en to-trins procedure.
