Cotton rats are extremely excitable and have a strong flight-or-fight response. A handling method optimized to reduce the stress of the animals is described which will make cotton rats more accessible as a preclinical model.
Oncolytic viruses are a novel anticancer therapy with the ability to target tumor cells, while leaving healthy cells intact. For this strategy to be successful, recent studies have shown that involvement of the host immune system is essential. Therefore, oncolytic virotherapy should be evaluated within the context of an immunocompetent model. Furthermore, the study of antitumor therapies in tolerized animal models may better recapitulate results seen in clinical trials. Cotton rats, commonly used to study respiratory viruses, are an attractive model to study oncolytic virotherapy as syngeneic models of mammary carcinoma and osteosarcoma are well established. However, there is a lack of published information on the proper handling procedure for these highly excitable rodents. The handling and capture approach outlined minimizes animal stress to facilitate experimentation. This technique hinges upon the ability of the researcher to keep calm during handling and perform procedures in a timely fashion. Finally, we describe how to prepare cotton rat mammary tumor cells for consistent subcutaneous tumor formation, and how to perform intratumoral and intraperitoneal injections. These methods can be applied to a wide range of studies furthering the development of the cotton rat as a relevant pre-clinical model to study antitumor therapy.
Onkolytiska virus (OV) selektivt replikera i tumörceller genom att utnyttja biokemiska skillnader mellan normala och tumörceller 1,2. Det finns två typer av OVS: de som inte kräver en mutation för att uppnå selektiv onkolys, kallat naturligt förekommande vildtyp virus och sådana som måste konstrueras för att uppnå selektiv onkolys. Insamlingen av mutationer inom en given tumörtyp bestämmer vilken typ av selektiv tillväxtfördel över normala celler för en OV 2. Säkerheten och nyttan av att använda OVS har påvisats i kliniska prövningar 3-7. Bovint herpesvirus typ 1 (BHV-1) är en medlem av Herpesviridae familjen, och Alphaherpesviridae underfamilj. BHV-1 initiates bovina luftvägsinfektioner komplex i boskap, manifesterar sig i en mängd olika symptom som liknar en dålig kall 8,9. BHV-1 binder fäst och inträdes receptorer används av HSV-1, såsom heparansulfat och nectin-1 10. Men binder CD155 i stället för nectin-2 10. BHV-1 har en mycket smal värdområde så att den inte kan effektivt komma in och initiera replikering i normala och transformerade murina celler 3,4,10. Detta gör användningen av konventionella musmodeller problematisk. Den onkolytiska förmåga hos BHV-1 har visats in vitro 11,12. BHV-1 har visat att initiera replikering i och döda humana tumörceller från en mängd olika histologiska ursprung, inklusive bröstcancerceller och bröstcancer initierande celler 11,12. Emellertid måste det antitumörkapacitet hos BHV-1 utvärderas in vivo inom ramen för en immunkompetent värd.
Humant adenovirus (Ad), för vilkenDet finns 57 identifierade serotyper, oftast orsakar respiratorisk sjukdom hos människor. Onkolytisk Ad vektorer har utvärderats för sin antitumör effekt med flera avancera in i kliniska prövningar 13-15. Trots lovande prekliniska data, har kliniska resultat nått upp till förväntningarna. Mänsklig tumör xenograft modeller används vanligtvis för att studera antitumör effekten av Ad-vektorer, även om de uppvisar försvagat immunsvar mot viruset 16,17. Dessutom syngena musmodeller är icke tillåtna att Ad-infektion, vilket gör utvärdering av värdimmunsvar som använder dessa modeller opraktiska 17,18.
Värdens immunsystem har identifierats som den mest inflytelserika mekanism genom vilken OVS framkallar tumörcelldöd 19. Antitumörsvar mellan toleransstimulerade och icke-toleransstimulerade tumörassocierat antigen (TAA) modeller skiljer och kan kraftigt påverka framgången för OV terapi. HSV-1 OV KM100 (ICP0 n212VP16 1814 20) 20,21 framkallade tumörregression i 80% av tumörbärande möss i en mus Polyoma Mellanöstern T-antigen bröstcancer modell 22. Men i HER-2 / neu-modeller, antitumör effekten av KM100 varierade mellan 20% färdigt regression i syngena möss och tumör stasis i transgena, HER2-toleranta möss. Tillsammans har dessa uppgifter betona vikten av fullt utvärdera OVS använder djurmodeller som bäst sammanfatta människans immun landskapet till fullo förstå vilka funktioner bestämmer terapeutisk framgång.
Bomulls råtta (Sigmodon hispidus), infödda på Nord- och Sydamerika, används oftast som en modell av respiratory syncytial virus infektion (som ses i 5). Bomulls råttor används också i anti-BHV-1 vaccination forskning som de rekapitulera patologin associerad med bovin respiratorisk sjukdom komplex 6,23. Vidare, BHV-1 infektion i bomullsråttorär immunogent, förmå ihållande mucosal och systemiska immunsvar 6,23-25. Cellinjer har härletts från spontana fibrosarkom och osteosarkom av bröstkörteln (LCRT) och ben (CCRT och VCRT), respektive 26. Bomullsråttor har använts för att utvärdera effektiviteten av onkolytiska Ad-vektorer in vivo eftersom de är mottagliga för Ad-infektion och uppvisar en liknande patologi till människor 27-29. Användningen av nedsatt immunförsvar modeller för preklinisk utvärdering av OVS är inte bara mindre tecken på kliniska svar på terapi men de misslyckas med att ta hänsyn till den roll i immunsystemet i onkolytiskt virotherapy 30,31. Därför syngena och tumör toleranta bomullsråttmodeller av bröstkarcinom och osteosarkom är relevanta modeller i vilka för att utvärdera för-kliniska effekten av OVS, såsom BHV-1 och Ad som inte kan studeras med användning av konventionella musmodeller.
