Cotton rats are extremely excitable and have a strong flight-or-fight response. A handling method optimized to reduce the stress of the animals is described which will make cotton rats more accessible as a preclinical model.
Oncolytic viruses are a novel anticancer therapy with the ability to target tumor cells, while leaving healthy cells intact. For this strategy to be successful, recent studies have shown that involvement of the host immune system is essential. Therefore, oncolytic virotherapy should be evaluated within the context of an immunocompetent model. Furthermore, the study of antitumor therapies in tolerized animal models may better recapitulate results seen in clinical trials. Cotton rats, commonly used to study respiratory viruses, are an attractive model to study oncolytic virotherapy as syngeneic models of mammary carcinoma and osteosarcoma are well established. However, there is a lack of published information on the proper handling procedure for these highly excitable rodents. The handling and capture approach outlined minimizes animal stress to facilitate experimentation. This technique hinges upon the ability of the researcher to keep calm during handling and perform procedures in a timely fashion. Finally, we describe how to prepare cotton rat mammary tumor cells for consistent subcutaneous tumor formation, and how to perform intratumoral and intraperitoneal injections. These methods can be applied to a wide range of studies furthering the development of the cotton rat as a relevant pre-clinical model to study antitumor therapy.
Onkolytiske virus (OV) selektivt å replikere i tumorceller ved å utnytte biokjemiske forskjeller mellom normale og tumorceller 1,2. Det er to typer av OVS: de som ikke krever en mutasjon for å oppnå selektiv oncolysis, referert til som naturlig forekommende vill-type virus og de som må være konstruert for å oppnå selektiv oncolysis. Samlingen av mutasjoner innenfor et gitt tumortype bestemmer arten av den selektive vekst fordel i forhold til normale celler til et OV-2. Den sikkerhet og nytte av hjelp OVS har blitt vist i kliniske studier 3-7. Til tross for fremskritt innen onkolytisk virotherapy det finnes gap mellom pre-kliniske og kliniske resultater, noe som tyder på at bedre modeller for å evaluere antitumor effekt av OVS.
Bovine herpesvirus type 1 (BHV-1) er et medlem av Herpesvirus familien, og Alphaherpesviridae underfamilie. BHV-1 tt ig ngater luftveiene hos storfe kompleks i storfe, manifestere i et bredt spekter av symptomer som ligner en dårlig kald 8,9. BHV-1 binder feste- og inngangs reseptorer som brukes av HSV-1, slik som heparansulfat og nectin-1 10. Men binder det CD155 i stedet for nectin-2 10. BHV-1 har et meget snevert vertsområde, slik at det er i stand til effektivt å legge inn og initiere replikasjon i normale og transformerte museceller 3,4,10. Menneskelig Adenovirus (Ad), somDet er 57 identifisert serotyper, oftest forårsaker respiratorisk sykdom hos mennesker. Onkolytiske Ad vektorer har blitt evaluert for sin antitumor effekt med flere fremmarsj i kliniske studier 13-15. Til tross for lovende pre-kliniske data, har kliniske resultater falt kort av forventningene. Menneskelig svulst xenograft modeller er vanligvis brukes til å studere antitumor effekt av Ad vektorer, selv om de viser svekket immunresponser mot viruset 16,17. Videre syngene murine modeller er umottagelige til Ad infeksjon, noe som gjør evaluering av vertsimmunresponser ved hjelp av disse modellene upraktisk 17,18.
Vertens immunsystemet har blitt identifisert som den mest innflytelsesrike mekanismen som OVS lokke fram tumorcelledød 19. Antitumor respons mellom toleriserte og ikke-toleriserte tumorassosiert antigen (TAA) modellene skiller og kan i stor grad påvirke suksessen av OV terapi. HSV-1 OV KM100 (ICP0 n212VP16 i 1814 20) 20,21 fremkalte tumor regresjon i 80% av tumorbærende mus i en muse Polyoma Middle T antigen jursvulster modell 22. Men i HER-2 / neu-modeller, den antitumor effekt av KM100 variert mellom 20% komplett regresjon i syngene mus og svulst stasis i transgene, HER2-toleriserte mus. Sammen disse dataene markere betydningen av fullt evaluere OVS bruker dyremodeller som best rekapitulere menneskets immun landskapet å fullt ut forstå hvilke funksjoner bestemme terapeutisk suksess.
Bomullen rotte (Sigmodon hispidus), innfødt til Nord-og Sør-Amerika, er mest brukt som en modell av respiratorisk syncytialt virus infeksjon (som anmeldt i 5). Bomullsrotter blir også brukt i anti-BHV-1 vaksinasjon forskning som de rekapitulere patologien forbundet med bovin respiratorisk sykdom kompleks 6,23. Videre BHV-1-infeksjon av bomullsrotterer immunogent, indusere vedvarende slimhinne og systemiske immunresponser 6,23-25. Cellelinjer er avledet fra spontan fibrosarcoma og osteosarkomer av melkekjertlene (LCRT) og bein (CCRT og VCRT), henholdsvis 26. Bomullsrotter har blitt brukt for å evaluere in vivo effekt av onkolytiske Ad vektorer som de er utsatt for Ad-infeksjon og oppviser lignende patologi for mennesker 27-29. Bruken av immunkompromitterte modeller for pre-klinisk evaluering av OVS er ikke bare mindre indikerer kliniske respons på behandling, men som ikke tar hensyn til den rolle i immunsystemet i onkolytisk virotherapy 30,31. Derfor syngenisk og tumor-toleriserte bomullsrottemodeller av brystkreft og osteosarkom er relevante modeller som å evaluere pre-kliniske effekten av OVS, slik som BHV-1 og Ad, som ikke kan studeres ved hjelp av konvensjonelle murine modeller.
Cotton rats are highly excitable and have a strong flight response. Therefore, special care should be taken to minimize any undue stress on the animal. The cage setup described will allow for safe and easy capture of the animals, with the placement of the enrichment tube being of the utmost importance.
