Prion-like propagation of protein aggregates has recently emerged as being implicated in many neurodegenerative diseases. The goal of this protocol is to describe, how to use the nematode C. elegans as a model system to monitor protein spreading and to investigate prion-like phenomena.
Prions are unconventional self-propagating proteinaceous particles, devoid of any coding nucleic acid. These proteinaceous seeds serve as templates for the conversion and replication of their benign cellular isoform. Accumulating evidence suggests that many protein aggregates can act as self-propagating templates and corrupt the folding of cognate proteins. Although aggregates can be functional under certain circumstances, this process often leads to the disruption of the cellular protein homeostasis (proteostasis), eventually leading to devastating diseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD), Amyotrophic lateral sclerosis (ALS), or transmissible spongiform encephalopathies (TSEs). The exact mechanisms of prion propagation and cell-to-cell spreading of protein aggregates are still subjects of intense investigation. To further this knowledge, recently a new metazoan model in Caenorhabditis elegans, for expression of the prion domain of the cytosolic yeast prion protein Sup35 has been established. This prion model offers several advantages, as it allows direct monitoring of the fluorescently tagged prion domain in living animals and ease of genetic approaches. Described here are methods to study prion-like behavior of protein aggregates and to identify modifiers of prion-induced toxicity using C. elegans.
Många neurodegenerativa sjukdomar, däribland Alzheimers sjukdom (AD), Parkinsons sjukdom (PD), amyotrofisk lateralskleros (ALS), och transmissibla spongiforma encefalopatier (TSE), är förknippade med aggregering benägna proteiner och är därmed kollektivt kallas protein misfolding störningar (PMDs ). TSE eller prionsjukdomar utgör en unik klass av PMDs genom att de kan vara smitt hos både människor och djur en. På molekylär nivå, prioner replikera genom att rekrytera och konvertera mono α-helix-rika värd-kodad cellulär PrP (PrP C) i den patologiska β-sheet-rika PrP konforma 2,3. Självförökningsproteinaggregat har också identifierats i svampar, som delar viktiga egenskaper med däggdjurs prioner 4,5. Dessutom däggdjurs prioner kan röra sig från cell till cell och infektera naiva celler 6,7.
Medan PMDs other än TSE inte smittsam, de delar en gemensam patogena principen med prionsjukdomar 8,9. Även proteinerna knutna till var och en av PMDs inte är relaterade i struktur eller funktion, de alla formulär aggregat via en kristalliseliknande process som kallas kärn eller seedade polymerisation; Dessutom proteinhaltiga frön växa genom att rekrytera sina lösliga isoformer 2,10,11. Verkningsgraden att själv propagerar varierar in vivo, beroende på de inneboende egenskaperna hos proteinet, som tillsammans med ytterligare cellulära faktorer såsom molekylära chaperoner slutligen avgör andelen aggregerad kärnbildning, sådd, fragmentering och spridning 12-15. Därför måste det finnas en fin balans mellan dessa faktorer som gör att en effektiv spridning av proteinaggregering. Detta kan också förklara varför bara vissa amyloidogena aggregat hysa kännetecken på en prion, och därmed inte alla PMDs är smittsam. Prioner tycks representera "topp-artister" ofa brett spektrum av självreplikerande proteinaggregat, vilket gör dem till ett kraftfullt verktyg för att studera PMDs 8,13.
Fängslande, toxiciteten i samband med sjukdomsrelaterade aggregat har ofta en icke cell självständig komponent 16,17. Detta innebär att de påverkar angränsande celler som inte uttrycker motsvarande gen, i motsats till en strikt cell autonom verkan, vilket innebär att endast de celler som uttrycker genen uppvisar den specifika fenotypen. Detta compellingly framgår av vävnadsspecifika uttryck eller slå ner på de respektive proteiner i många modeller av neurodegenerativa sjukdomar 18-26. Olika mekanismer har föreslagits som grund för denna icke-cell självständig toxicitet i PMDs, inklusive minskad näringstillförsel, obalans i neuronal signalering, glutamatexcitotoxicitet och neuroinflammation 16,27,28. Dessutom, en prion-liknande rörelse av sjukdomsbundna aggregat mellan celler might bidra till denna aspekt 29,30. Ökande bevis tyder på att andra proteininneslutningar än prioner kan sända från cell-till-cell, vilket kan förklara den karakteristiska spridning av patologi observerats i många PMDs 30-36. Men det har ännu inte fastställts om det finns ett klart orsakssamband mellan intercellulära rörelse sjukdoms proteiner och toxisk effekt på angränsande celler. Därför är det nödvändigt och viktigt för utvecklingen av nya behandlingar en bättre förståelse av de cellulära vägar som ligger bakom cell till cell överföring och icke cell autonoma toxicitet. Men många aspekter av prion-liknande spridning och cellulära faktorer som påverkar cell-till-cell överföring av felveckade proteiner i metazoans inte förstått, i synnerhet på organismnivå.
Den nematod Caenorhabditis elegans har flera fördelar som ger potentialen att upptäcka nya aspekter av prion-liknande spreading i metazoans 17. Den är transparent, vilket möjliggör in vivo-spårning av fluorescerande märkta proteiner i den levande organismen. Dessutom är många cellulära och fysiologiska processer som påverkas av sjukdomen bevarad från maskar för människors, och C. elegans är också mottaglig för en mängd olika genetiska manipulationer och molekylära och biokemiska analyser 37-39. Exakt 959 somatiska celler utgör den vuxna hermafrodit med en enkel plan kropp som fortfarande har flera olika vävnadstyper, såsom muskel, neuroner och tarmen.
Att etablera en ny prionmodell i C. elegans, valde vi att exogent uttrycka väl karaktäriseras glutamin / asparagin (Q / N) -rika priondomän NM i cytosoliska jästprionprotein Sup35, eftersom det inte finns några kända endogena prionproteiner i maskar 4,40. Jäst prioner har varit ovärderliga för att belysa grundläggande mekanismer prionreplika 41-44. Dessutom är NM på granst cytosoliskt prion-liknande protein som har visat sig rekapitulera hela livscykel av en prion i däggdjurscellkultur 45,46. Likaledes, när den uttrycks i C. elegans, det Sup35 prion domänen antagits anmärkningsvärt väl till de olika kraven för förökning i metazoiska celler jämfört med jästceller och uppvisade viktigaste funktionerna i prionbiologi 40. NM aggregering associerades med en djup giftig fenotyp, inklusive störningar av mitokondrie integritet och utseende olika autophagy relaterade blåsor på cellnivå, samt embryonala och larvstillestånd, utvecklingsförsening och en utbredd störning av proteinveckmiljön på organismnivå. Påfallande, uppvisar priondomäncellen självständiga och icke cell autonoma toxicitet, påverkar grann vävnader där transgenen inte uttrycks. Vidare är den vesikulär transport av prion domän inom och mellan cellerna övervakas realtid <em> In vivo 40.
Här beskriver vi hur du undersöka prionliknande spridning i C. elegans. Vi kommer att förklara hur man övervaka intra- och intercellulära transport av vesiklar som innehåller prion domän med time-lapse fluorescensmikroskopi. Vi kommer att betona användningen av vävnadsspecifika fällbara sensorer och ubiquitously uttryckt spännings reportrar att utvärdera cell autonoma och icke cell autonoma effekter på cellulär kondition. Slutligen kommer vi att beskriva förfarandet för ett nyligen genomfört genomet bred RNA-interferens (RNAi) skärmen för att identifiera nya modifierare av prion-inducerad toxicitet. I kombination kan dessa metoder bidra till att retas isär genetiska vägar som är involverade i den intercellulära rörelsen av proteiner och deras icke cell autonoma toxicitet.
