Mechanisms of cellular and intra-cellular scaling remain elusive. The use of Xenopus embryo extracts has become increasingly common to elucidate mechanisms of organelle size regulation. This method describes embryo extract preparation and a novel nuclear scaling assay through which mechanisms of nuclear size regulation can be identified.
Een fundamentele vraag in de celbiologie is hoe cel en organel maten worden gereguleerd. Het is al lang bekend dat de grootte van de kern algemeen schalen met de grootte van de cel, met name tijdens embryogenese als dramatische daling van zowel cellen en nucleaire grootte voorkomen. Mechanismen van grootte regelgeving nucleaire zijn grotendeels onbekend en kunnen kanker relevant zijn waar veranderde nucleaire grootte is een belangrijke diagnostische en prognostische parameter. In vivo zal het identificeren van nucleaire grootte toezichthouders worden bemoeilijkt door de essentiële en complexe aard van de nucleaire functie. De in vitro benadering beschreven afmetingen control nucleaire studie maakt gebruik van de normale verlaging kerngrootte die tijdens Xenopus laevis ontwikkeling. Eerst worden kernen geassembleerd in X. laevis ei extract. Vervolgens worden deze kernen geïsoleerd en opnieuw gesuspendeerd in cytoplasma van embryo laat stadium. Na 30-90 min incubatieperiode nucleaire oppervlaktegebieddaalt met 20-60%, waardoor een bruikbare test om cytoplasmatische componenten aanwezig zijn in een vergevorderd stadium embryo's die bijdragen aan de ontwikkeling van nucleaire grootte schalen te identificeren. Een belangrijk voordeel van deze aanpak is het relatieve gemak waarmee de eieren en embryo-extracten biochemisch kan worden gemanipuleerd, waardoor de identificatie van nieuwe eiwitten en activiteiten die kerngrootte regelen. Zoals bij elke in vitro benadering van validatie resulteert in een in vivo systeem belangrijk is en micro-injectie van X. laevis embryo's is bijzonder geschikt voor deze studies.
De afmetingen van cellulaire organellen typisch schalen met de grootte van de cel, en dit is wellicht het best beschreven voor de schaal van nucleaire grootte met celgrootte 1-10. Dit geldt met name tijdens embryogenese en celdifferentiatie bij dramatische vermindering van zowel de cel kerngrootte vaak waargenomen 11,12. Bovendien veranderde kerngrootte is een belangrijke parameter voor kanker diagnose en prognose 13-17. Mechanismen die bijdragen tot de grootte regelgeving nucleaire zijn grotendeels onbekend, voor een deel te wijten aan de complexiteit en de essentiële aard van de nucleaire structuur en functie. De hier beschreven methode werd ontwikkeld als een in vitro assay voor kerngrootte schalen die vatbaar is voor biochemische manipulatie en opheldering van mechanismen van regulatie nucleaire grootte.
Xenopus laevis ei extract is een goed opgezet systeem te herhalen en te studeren complexe cellulaire processen in een in vitro </em> Context. Deze extracten zijn nieuwe fundamentele informaties verscheidene cellulaire processen waaronder de aanmaak en functie van de mitotische spoel, endoplasmatisch reticulum en kern 18-22 geopenbaard. Een belangrijk voordeel van het extractiesysteem dat X. laevis ei extracten geven een nagenoeg onverdund cytoplasma waarvan de samenstelling is makkelijk te veranderen, bijvoorbeeld door toevoeging van recombinante eiwitten of immunodepletie. Verder is men in staat om essentiële processen te manipuleren door gebruik behandelingen die anderszins letale in een in vivo context. Wijzigingen van het ei extract procedure mogelijk maken voor de isolatie van uittreksels uit X. laevis embryo's in plaats van eieren, en deze embryo-extracten zijn even vatbaar voor biochemische manipulatie 23. Tijdens X. laevis ontwikkeling, de eencellige bevruchte embryo (~ 1 mm diameter) ondergaat een reeks van twaalf snelle celdelingen (stappen 1-8) genereren duizenden 50 μm diameter en kleinere cellen, het bereiken van een ontwikkelingsstadium aangeduid als de midblastula overgang (MBT) of stadium 24-26 augustus. De MBT wordt gekenmerkt door het begin van zygote transcriptie, celmigratie, asynchrone celdelingen, verwerving van gap fasen en vaststelling van nucleaire steady-state en zijn geen continue nucleaire expansie zoals in de pre-MBT embryo. Van stap 4 tot gastrulatie (stadia 10,5-12), het volume van individuele kernen daalt met meer dan 10-voudig 11.
Hier is het doel om mechanismen die aan deze verlaging van nucleaire grootte tijdens ontwikkeling progressie identificeren. De aanpak is om eerst te assembleren kernen in X. laevis ei extract en die kernen uit het ei cytoplasma / extract te isoleren. Deze kernen worden vervolgens opnieuw gesuspendeerd in cytoplasma van eind gastrulastadium embryo's. Na een incubatieperiode, de kernen van ei extract kleiner worden in een vergevorderd stadium embryo extract. We redeneerden dat deze would een nuttige test voor het identificeren van cytoplasmatische componenten die aanwezig zijn in een vergevorderd stadium embryo's die bijdragen aan de ontwikkeling van nucleaire grootte schalen zijn. Gebruik van deze assay, in combinatie met in vivo validatie we aangetoond dat proteïne kinase C (PKC) draagt ontwikkelingsstoornissen verlaging kerngrootte in X. laevis 23.
Here is presented a novel method to study mechanisms of nuclear size regulation during X. laevis development. Developmental progression is associated with dramatic changes in cell physiology, metabolism, division rates, and migration, as well as alterations in the sizes of cells and intracellular structures.
