Transporte iónico celular a menudo puede evaluarse mediante el control de pH intracelular (pH). Transporte iónico epitelial es fundamental para la homeostasis de iones sistémica así como el mantenimiento de gradientes electroquímicos celulares esenciales. PH intracelular (pH) está influenciado por muchos transportadores de iones y así control de pH es una herramienta útil para evaluar la actividad de transportista. Moderno codificado genéticamente pH-indicadores (GEpHIs) ofrecen cuantificación óptica de pH en las células intactas a escala celular y subcelular. Este protocolo describe la cuantificación en tiempo real del pH celular Reglamento en túbulos de Malpighi (MTs) de Drosophila melanogaster por ex vivo vivo-proyección de imagen de pHerry, un pseudo-radiométrica GEpHI con un pKun adecuadas para el seguimiento de los cambios de pH en el citosol. El objetivo de este protocolo es describir cuantificación de pH intracelular (pHi) utilizando un genéticamente codificado-indicador de pH (GEpHI) y demostrar cómo este método puede utilizarse para evaluar transporte basolateral H+ un insecto modelo (D. melanogaster) estructura renal, los túbulos de Malpighi (TM). MTs sirven como los órganos excretores de la mosca de la fruta y son funcionalmente similares a la nefrona mamífera en varios aspectos clave1. MTs se arreglan como 2 pares de túbulos (anteriores y posteriores) en el tórax y el abdomen de la mosca. En la escala celular transporte trans epitelial ion en el lumen se logra por una membrana plasmática apical V-ATPasa3 y un intercambiador+ de álcali-metal/H así como un basolateral Na+-K+–4de la ATPasa, hacia adentro-rectificador K+ canales5, Na+-conducido Cl−/HCO3− intercambiador (NDAE1)6y Na+-K+-2 Cl− cotransporter (NKCC; Ncc69)7, mientras que células radiadas median Cl– y agua transporte8,9. Este sistema fisiológico complejo pero accesible ofrece excelentes oportunidades para la investigación de mecanismos de transporte iónico endógeno cuando se combinan con los diversos conjuntos de herramientas genéticas y de comportamiento de Drosophila.
La justificación de este protocolo fue describir un sistema genéticamente maleable para el estudio de transporte iónico epitelial con potencial para la integración de la célula de a conducta y exportación de herramientas para otros sistemas modelo. Expresión de pHerry10, un GEpHI derivada de una fusión de verde pH-sensible Super eclíptica pHluorin11,12 (SEpH) y rojo mCherry insensible pH13, MTs permite la cuantificación del transporte de H+ en MT las células a través de la alta K+/nigericin calibración técnica14. Medida que muchos transportadores de iones equivalentes de H+ , cuantificación de pH intracelular sirve como una representación funcional de movimiento de iones a través de una variedad de transportadores.
