Hier presenteren we een eenvoudige, goedkope en selectieve chemische spot test-protocol voor de detectie van synthetische cathinones, een klasse van nieuwe psychoactieve stoffen. Het protocol is geschikt voor gebruik in verschillende gebieden van de rechtshandhaving die optreden van illegaal materiaal.
Synthetische cathinones zijn een grote klasse van nieuwe psychoactieve stoffen (NPS) die steeds in drugsvangsten gemaakt door wetshandhaving en andere grens bescherming agentschappen wereldwijd heersen. Kleur testen is een voorlopige identificatie-techniek met vermelding van de aanwezigheid of afwezigheid van een bepaald geneesmiddel klasse met behulp van de snelle en ongecompliceerde chemische methoden. Vanwege hun relatief recente opkomst is een test van de kleur voor de specifieke identificatie van synthetische cathinones momenteel niet beschikbaar. In deze studie, introduceren we een protocol voor de vermoedelijke identificatie van synthetische cathinones, dienst van drie waterige reagens oplossingen: Natriumacetaat, copper(II) nitraat en 2,9-dimethyl-1,10-phenanthroline (neocuproine). Kleine pin-hoofd formaat bedragen (ongeveer 0,1-0,2 mg) van de vermoedelijke drugs worden toegevoegd aan de putten van een plek porseleinen plaat en elk reagens wordt dan toegevoegd ontkleuring sequentieel voordat de verwarming op een kookplaat. Een kleurverandering van zeer licht blauw tot geel-oranje na 10 min de waarschijnlijk aanwezigheid van synthetische cathinones aangeeft. Het zeer stabiel en specifieke test reagens heeft het potentieel voor gebruik in de vermoedelijke screening van onbekende monsters voor synthetische cathinones in een forensisch laboratorium. Echter de overlast van een stap extra verwarming voor het resultaat van de wijziging van kleur de test laboratorium toepassing beperkt en vermindert de kans op een eenvoudige vertaling naar veldproeven.
De illegale drugsmarkt werkt ook aan een traditionele business door te blijven evolueren en zich aanpassen aan een veranderende markt. Vooruitgang in de moderne technologie, speciaal, de wereldwijde verspreiding van krachtige communicatie heeft meer online aankopen via de donkere netto1 en uitgebreide kennis gedeeld worden tussen gebruikers via online forums2gezien. Gecombineerd met de vooruitgang in de chemie, de snelle opkomst van nieuwe psychoactieve stoffen (NPS) gemaakt een ernstige uitdaging voor nationale en internationale drugsbestrijding.
NPS zijn potentieel gevaarlijke stoffen van misbruik die soortgelijke gevolgen aan drugs onder internationale controle hebben. In eerste instantie verkocht als “legale” alternatieven, werden 739 NPS gemeld aan het Bureau van de Verenigde Naties over Drugs en misdaad (UNODC) tussen 2009 en 20163. Volgens het meest recente jaarverslag, een record aantal NPS werden in beslag genomen aan de Australische grens, met de meerderheid van de geanalyseerde, verder aangeduid als synthetische cathinones4. Op een wereldwijde schaal, inbeslagnames van synthetische cathinones zijn gestaag toegenomen sinds het eerst gemeld in 2010, en zijn een van de meest in beslag genomen NPS-5.
De uitdagingen van NPS zijn een grotendeels gepubliceerde onderwerp van discussie6,7. Forensische laboratoria en rechtshandhavingspersoneel bleven benadeeld zonder passende methoden om te detecteren en identificeren van NPS tijdens hun snelle opkomst. Uitgebreid onderzoek naar de opsporing van de NPS, met inbegrip van synthetische cathinones, in de in beslag genomen materiaal, is nog werkzaam zijn gaschromatografie-massaspectrometrie (GC-MS)8 en vloeistof-chromatografie-hoge resolutie massa spectrometrie (LC-HRMS)9 voor bevestigende analyse. Toenemende vraag naar minimale monstervoorbereiding heeft gezien, infrarood en Raman spectroscopie10 studies alsook ambient ionisatie massaspectrometrie analyses, zoals directe analyse in real-time massaspectrometrie (DART-MS)11, 12. de behoefte aan snelle en gevoelige analyse in het veld heeft ook gezien de opneming van papier spray ionisatie-massaspectrometrie (PSI-MS) in draagbare apparaten voor gebruik door wetshandhaving13. Vele instrumentale technieken bieden bevestigende analyse met gevoelige opsporing en kwantitatieve resultaten. Voor high-throughput analyse, kunnen zij echter tijdrovend zijn als gevolg van de bereiding van de monsters, lopen tijden, en instrument-training en onderhoud.
Vermoedelijke kleur tests zijn bedoeld om te suggereren de aanwezigheid of afwezigheid van bepaalde klassen van de drug in een test monster14. De wetenschappelijke werkgroep voor de analyse van in beslag genomen Drugs (SWGDRUG) classificeert kleur testen als de laagste discriminerende techniek van de macht, naast ultraviolet spectroscopie en immunoassay15. Echter, zijn zij nog steeds veel in dienst van wetshandhavings- en andere veiligheidspersoneel als een middel om te zorgen voor snelle resultaten op een aanzienlijk lagere kosten in vergelijking met andere technieken. Het belangrijkste voordeel aangeboden door kleur ter plaatse testmethoden is de mogelijkheid om ze uit te voeren in het veld met behulp van draagbare testkits.
De selectiviteit van kleur tests is afhankelijk van individuele chemische reacties tussen het test-reagens en de klasse van de drug van belang zijn voor het maken van een kleur te veranderen. Huidige vermoedelijke testprotocollen gebrek aan een specifieke test voor het opsporen van synthetische cathinones gebruikte reagentia die gebrek aan specificiteit en bevatten gevaarlijke stoffen zijn vaak tewerkgesteld. Andere aanbevolen reagentia hebben niet onderzocht op een groot aantal mogelijke synthetische cathinon stoffen16.
Het doel van dit werk is om een eenvoudige kleur testprotocol die gemakkelijk kan worden ingezet door de belanghebbende partijen voor de oriënterende van synthetische cathinones in illegale stoffen van onbekende samenstelling. Belanghebbende partijen zou omvatten rechtshandhaving, grens bescherming agentschappen, forensische laboratoria en andere relevante veiligheidspersoneel. De voorgestelde methoden hanteren een reductie-oxidatie reactie tussen het elektron-accepteren koperen complexe reagens en het elektron rijke synthetische cathinon drug moleculen voorkomende. Gebruik van deze chemische methoden ontwikkeld, kunt een toepassen in de vorm van een vermoedelijke kleur test te suggereren de aanwezigheid van synthetische cathinones.
Deze kleur test-protocol werd aangepast van experimenteel werk gepubliceerd door Al-Obaid et al. 18 waarin de auteurs aangetoond een kleurverandering vindt plaats in aanwezigheid van cathinon geëxtraheerd uit de khat plant. Aangebrachte wijzigingen in het gepubliceerde protocol waren nodig om te voorzien in de toepassing ervan in de detectie van de vermoedelijke illegale drugs.
