Her presenterer vi en enkel, billig og selektiv kjemiske stikkprøve protokoll for påvisning av syntetiske katinoner, en klasse med nye psykoaktive stoffer. Protokollen er egnet for bruk i ulike områder av rettshåndhevelse støte ulovlig materiale.
Syntetisk katinoner er en stor klasse av nye psykoaktive stoffer (NPS) som er stadig mer utbredt i narkotikabeslag gjort av politi og andre grensen beskyttelse globalt. Farge testing er en presumptive identifikasjon teknikk som indikerer tilstedeværelse eller fravær av en bestemt legemiddel klasse med raske og ukompliserte kjemiske metoder. På grunn av deres relativt nylige framveksten er en farge test for bestemt identifikasjon av syntetisk katinoner ikke tilgjengelig. I denne studien vi innføre en protokoll for presumptive identifikasjon av syntetiske katinoner, sysselsetter tre vandig reagens løsninger: copper(II) nitrat, 2,9-dimethyl-1,10-phenanthroline (neocuproine) og natrium acetate. Liten pin-hode størrelse beløp (ca 0,1-0.2 mg) av mistanke om narkotika er lagt til brønnene av en porselen spot plate, og hver reagens legges deretter dropwise sekvensielt før oppvarming på en kokeplate. En farge endre fra lys blå til gul-oransje etter 10 min angir at sannsynlig syntetiske katinoner. Svært stabil og bestemt test reagensen har potensial for bruk i presumptive screening av ukjent prøver for syntetisk katinoner i et rettsmedisinske laboratorium. Men ordensforstyrrelser av et ekstra Varmeapparat skritt for farge endre resultatet begrenser testen laboratorium program og reduserer sannsynligheten for en lett oversettelse til feltet testing.
Det illegale narkotikamarkedet fungerer på lignende måte en tradisjonell virksomhet ved å fortsette å utvikle seg og tilpasse seg et skiftende marked. Fremskritt innen moderne teknologi, spesielt den globale spredningen av kraftige kommunikasjons har sett økt onlinekjøp via mørke netto1 og omfattende kunnskap deling blant brukerne via nettfora2. Kombinert med fremskritt innen kjemi, rask fremveksten av nye psykoaktive stoffer (NPS) opprettet en alvorlig utfordring for internasjonale og nasjonale narkotika-kontroll.
NPS er potensielt farlige stoffer misbruk som har liknende effekter til narkotika internasjonal kontroll. Opprinnelig ble markedsført som “lovlig” alternativer, ble 739 NPS rapportert til FNs kontor for narkotika og kriminalitet (UNODC) mellom 2009 og 20163. Ifølge den nyeste årsrapporten beslaglagt et rekordhøyt antall NPS på australske grensen, med fleste av disse analysert, ytterligere identifisert som syntetisk katinoner4. På en global skala, beslag av syntetisk katinoner har økt jevnt siden først rapportert i 2010, og er en av de oftest beslaglagt NPS5.
Utfordringene med NPS har vært et stort sett publiserte diskusjon6,7. Rettsmedisinske laboratorier og rettshåndhevelse personell var igjen en ulempe uten egnede metoder å oppdage og identifisere NPS under rask fremveksten. Omfattende forskning på deteksjon av NPS, inkludert syntetisk katinoner, beslaglagt materiale, har ansatt gass kromatografi-massespektrometri (GC-MS)8 og flytende kromatografi høy oppløsning massespektrometri (LC-HRMS)9 for bekreftende analyse. Økende etterspørsel etter minimal eksempel forberedelse har sett infrarød og Raman spektroskopi10 studier som ambient ionisation masse spectrometric analyser, som direkte analyse i sanntid massespektrometri (DART-MS)11, 12. behovet for rask, følsom analyse i feltet har også sett inkorporering av papir spray ionisering-massespektrometri (PSI-MS) bærbare enheter for bruk av loven håndhevelse13. Mange medvirkende teknikker tilbyr bekreftende analysen med følsom deteksjon og kvantitative resultater. Men for høy gjennomstrømming analyse, kan de være tidkrevende eksempel forberedelse, operasjonstid, og apparatet opplæring og vedlikehold.
Presumptive farge testene foreslå tilstedeværelse eller fravær av visse narkotika klasser i en test prøve14. Vitenskapelige arbeider gruppen for analyse av beslaglagt narkotika (SWGDRUG) klassifiserer farge testing som det laveste kresne makt teknikken, ultrafiolett spektroskopi og immunanalyser15. Men er de fortsatt mye ansatt av politi og andre sikkerhetspersonell å gi raske resultater til en vesentlig lavere pris sammenlignet med andre teknikker. Den største fordelen tilbys av fargen flekk testmetoder er muligheten til å utføre dem i feltet bruker bærbare test kits.
Selektivitet av farge tester er avhengig av individuelle kjemiske reaksjoner som oppstår mellom test reagensen og narkotika klassen rundt opprette fargeendring. Gjeldende presumptive testing protokoller mangler en bestemt test for å oppdage syntetiske katinoner. brukte reagenser som mangler spesifisitet og inneholder farlige stoffer er ofte ansatt. Andre anbefalte reagenser har ikke blitt vist på en rekke mulige syntetisk cathinone stoffer16.
Formålet med dette arbeidet er å presentere en enkel farge testprotokollen som lett kan brukes av interesserte for foreløpige screening av syntetisk katinoner i illegale stoffer moirene. Interesserte parter omfatter politi, grensen beskyttelse byråer, rettsmedisinske laboratorier og andre relevante sikkerhetspersonell. Metodene foreslåtte ansette en reduksjon-oksidasjon reaksjonen forekommende mellom elektron-akseptere kobber komplekse reagensen og elektron rik syntetisk cathinone stoffet molekyler. Benytter disse kjemiske metoder utviklet, kan man bruke dem i form av en presumptive farge test foreslå tilstedeværelsen av syntetiske katinoner.
Denne fargen testprotokollen ble tilpasset fra eksperimentelt arbeid publisert av Al Obaid et al. 18 der forfatterne viste en fargeendring oppstår i nærvær av cathinone utdraget fra khat anlegget. Endringer i publiserte protokollen var nødvendig å forutse anvendelsen i presumptive illegale gjenkjenning. Det viktigste hensynet var å redusere omfanget av reaksjonen. Protokollen beskrevet i denne utredningen er utformet for å brukes på gaten prøver og narkotikabeslag.
