细胞转移后 10 天，使用一毫升注射器在每只实验动物的尾巴基部向小鼠注射 100 微克在完全弗氏佐剂中乳化的甲基化 BSA。细胞转移后 17 天，通过关节内注射 20 μg 甲基化 BSA 和 100 μg 甲基化 BSA 和 100 μg 全卵清蛋白到左膝关节中诱导关节炎在 10 微升 PBS 中注入每个麻醉过继转移动物的右膝关节。诱发关节炎后 7 天，再次测量膝盖以计算膝关节直径增加的百分比，并手术切除髌骨。
用福尔马林固定关节。然后，在 EDTA 中脱钙后，将膝盖包埋在石蜡中，并获得四个微米切片用于组织化学染色和分析。第 28 天，抗原特异性 T 调节细胞表达高水平的 CD3 和抗原特异性 T 细胞受体。
这些细胞中的大多数还表达 CD25、CD127 和 CTLA-4，所有这些细胞在天然存在的 T 调节细胞中通常以较高的水平表达。事实上，FoxP3 在 IPSC 衍生的 T 调节细胞中仍然存在，即使在用 Notch 配体进行长期体外刺激后，如细胞内染色所检测到的那样。此外，这些抗原特异性 IPSC T 调节细胞在体外受到抗原脉冲脾细胞刺激时会产生抑制性细胞因子，表明这些细胞具有潜在的抑制性表型。
在 OVA 处理的膝关节中观察到 FoxP3 阳性细胞，在接受对照载体转导的 IPSC 的膝关节中没有明显的 FoxP3 阳性细胞。此外，与单独使用 FoxP3 载体相比，在接受 TCR-FoxP3 载体转导的 IPSC 的小鼠的膝盖中可以看到更多的 CD4 阳性、FoxP3 阳性、TCR3 Beta 5 阳性细胞，这表明抗原特异性 IPSC-Treg 细胞在过继转移到受体动物后迁移到抗原诱导的关节炎膝关节。当 OVA 存在时，IPSC 衍生的细胞过继转移也影响炎性膝关节肿胀的显着减少，但对对照甲基化 BSA 注射膝关节没有影响。
高分辨率 Micro-CT 成像进一步证实了肿胀减轻的积极效果，与仅注射 MBSA 的对照动物相比，在细胞转移治疗的膝关节中观察到的骨质流失减少。一旦集合起来，如果执行得当，这些深入的程序可以在六周内完成。在尝试这种技术时，重要的是要记住诱导 TCR-FoxP3 抗原转导的 IPSC 的灌注分化。
