现在，在真空干燥器中对混合物进行脱气，直到所有气泡都被除去，混合物看起来又透明了。为了获得400微米厚的微流控芯片模板地板，将三克混合物倒在硅主板上，并将硅母模放在旋转涂布机上，以21倍G旋转涂覆5秒，54倍G旋转涂覆10秒。如前所述，再次脱气以去除被困的气泡。

现在，切割PDMS并将其从硅母板上剥离，并将PDMS层切割成更小的碎片，其微流体通道的尺寸相同，这些微流体通道将粘合在模板顶部。使用1%洗涤剂溶液轻轻擦拭模板和微孔五分钟，然后用去离子水冲洗。接下来，用异丙醇冲洗模板和微通道，然后用去离子水冲洗它们。
现在，在室温下用压缩空气在一个酒吧中干燥模板和微通道一分钟。将模板和微通道置于等离子清洗机中，粘合表面朝上。打开等离子清洗机后，等离子体处理模板和微通道40秒，然后从等离子清洗器中取出，并立即将微通道粘合在模板顶部。
将微流控芯片储存在70摄氏度的烘箱中五天，以确保PDMS疏水回收。在实验当天，在实验前几个小时将盒式培养箱设置为37摄氏度，然后在显微镜台上设置注射器泵和加热的玻璃板，设置与盒式培养箱相同的温度。实验前90分钟，将微流控芯片放入装有100%乙醇的容器中，用100%乙醇冲洗通道至少10分钟，然后将微流控芯片置于真空干燥器中并脱气至少30分钟。
现在将乙醇与蒸馏水交换，并对芯片进行至少30分钟的真空处理。将微流控芯片放入70摄氏度的烤箱中10分钟，以除去通道中残留的任何液体痕迹。将一毫升MOPS培养基移入离心机小瓶中，加入10微升0.132摩尔磷酸钾。
将100微升过夜培养物等分到离心机小瓶中，并以2，300倍G离心培养物两分钟。轻轻丢弃上清液，将沉淀重悬于一毫升新鲜MOPS培养基中，其中含有0.015%的吐温20和0.01%的磷酸钾，并将细菌悬浮液装入一毫升注射器中。要固定注射器和管道连接，请直接将针头插入管道。

将900微升的0.015%吐温20水溶液移液到离心机小瓶中，然后将100微升的原始胶体悬浮液移入其中。以13，500倍G离心悬浮液一分钟，并用吐温20水溶液轻轻地替换上清液。将胶体悬浮液装入一毫升注射器中，并通过微流体管将注射器连接到芯片上。
通过位于通道上游部分中央部分的入口将悬浮液注入微流控芯片中，并逐渐推动悬浮液直到模板被覆盖。以每分钟0.07至0.2微升的流速取出胶体悬浮液，并通过显微镜软件对图案化过程进行成像。一旦半月板快速到达模板的末端，就增加流速。
利用直通道几何形状来图案化荧光大肠杆菌菌株的固定相位细胞，沉积5， 000个分析陷阱中的83%。图案细菌在通道充满新鲜LB后的1.5小时内的不同时间恢复生长。一旦恢复生长，单个细菌细胞开始形成单个菌落，这些菌落膨胀直到形成表层并且单细胞分辨率丧失。超过55， 000个捕集器的分析表明，在分析的捕集器中，分别有93%和89%的捕集器形成了由两微米和一微米直径颗粒制成的绿红色二聚体。
